1A practical guide to amplicon and metagenomic analysis of microbiome data显示文摘Advances in high-throughput sequencing(HTS)have fostered rapid developments in the field of microbiome research,and massive microbiome datasets are now being generated.However,the diversity of software tools and the complexity of analysis pipelines make it difficult to access this field.Here,we systematically summarize the advantages and limitations of microbiome methods.Then,we recommend specific pipelines for amplicon and metagenomic analyses,and describe commonly-used software and databases,to help researchers select the appropriate tools.Furthermore,we introduce statistical and visualization methods suitable for microbiome analysis,including alpha-and betadiversity,taxonomic composition,difference comparisons,correlation,networks,machine learning,evolution,source tracing,and common visualization styles to help researchers make informed choices.Finally,a stepby-step reproducible analysis guide is introduced.We hope this review will allow researchers to carry out data analysis more effectively and to quickly select the appropriate tools in order to efficiently mine the biological significance behind the data.Yong-Xin Liu Yuan Qin Tong Chen Meiping Lu Xubo Qian Xiaoxuan Guo Yang Bai 2021Protein & Cell2021,12,5:20

3Paclitaxel formulation with stable sustained-release behavior and its biological safety evaluation显示文摘Biological safety and stable sustained-release of the drug are two crucial issues involved in the formulation of paclitaxel.Focusing on these issues, by using the FDA approved polylactide as carrier material, soybean lecithin as surfactant and maltodextrin as thickener, paclitaxel loaded PLA microspheres were simply prepared by solvent evaporation, thus guaranteeing the biological safety. The introduction of maltodextrin as a thickener aided to a stable sustained-release of paclitaxel. Surface morphology, particle size, drug loading rate, encapsulation efficiency and in vitro drug release behavior were investigated.Biological safety evaluations such as acute toxicity, allergies, hemolysis, skin stimulation and genotoxicity test were also carried out. 4Boosting of the enhanced permeability and retention effect with nanocapsules improves the therapeutic effects of cetuximab显示文摘Objective:The introduction of therapeutic antibodies(tAbs)into clinical practice has revolutionized tumor treatment strategies,but their tumor therapy efficiency is still far below expectations because of the rapid degradation and limited tumor accumulation of tAbs.Methods:We developed a nanocapsule-based delivery system to induce the self-augmentation of the enhanced permeability and retention(EPR)effect.This system constantly penetrated across the blood-tumor barrier into the tumor while avoiding the attack of tAbs by the immune system.The biodistribution and therapeutic effect were tested with single dose administration of nanocapsule-tAbs in vivo.Results:The accumulation of Nano(cetuximab)within subcutaneous PC9 tumors was gradually enhanced over 6 days after single dose administration,which was contrary to the biodistribution of native cetuximab.Nano(cetuximab)accumulated in tumor tissues via the EPR effect and released cetuximab.The released cetuximab acted on vascular endothelial cells to destroy the blood-tumor barrier and induce self-augmentation of the EPR effect,which in turn contributed to further tumor accumulation of long-circulating Nano(cetuximab).Compared with single dose administration of native cetuximab,Nano(cetuximab)showed an effective tumor suppressive effect for 3 weeks.Conclusions:The nanocapsule-based delivery system efficiently delivered tAbs to tum or tissues and released them to boost the EPR effect,which facilitated further tumor accumulation of the tAbs.This novel self-augmentation of the EPR effect facilitated by the biological characteristics of tAbs and nanotechnology contributed to the improvement of the therapeutic effect of tAbs,and stimulated new ideas for antibody-based tumor therapy.Chao Yang Yanli Tan Hongzhao Qi Junhu Zhou Lixia Long Qi Zhan Yunfei Wang Xubo Yuan Chunsheng Kang 2020Cancer Biology & Medicine2020,17,2:1

10Surface photodynamic ion sterilization of ITO-Cu_(2)O/ZnO preventing touch infection显示文摘Pathogenic microbial infections are threatening the people’s health and even life.The most common channel of infections can be caused by skin contact,especially hand touching facilities such as touching screen.In this work,Cu_(2)O covered with ZnO nanofilm was prepared on the surface of indium tin oxide conductive glass by electrodeposition and the followed atomic layer deposition process.This composite coating had a light transmittance of 71.5%,which met the light transmission needs of touch screen device.Electron spin resonance spectra showed that composite materials can generate more reactive oxygen species(ROS)than a single component under solar light irradiation.This was because a p-n junction with a built-in electric field was formed at the interface after Cu_(2)O contacting with ZnO.In the process of photocatalysis,photogenerated electrons and holes migrated at the interface driven by the built-in electric field,which promoted the separation of carriers.The antibacterial rate against Staphylococcus aureus reached 92.5%after 3 min of light irradiation with simulated sunlight due to the synergy of ROS and Cu ions,Zn ions.Therefore,this work may provide a potential method for antibacterial application of preventing hand touch infections.Zexin Liu Xiangmei Liu Zhenduo Cui Yufeng Zheng Zhaoyang Li Yanqin Liang Xubo Yuan Shengli Zhu Shuilin Wu 2022Journal of Materials Science & Technology2022,,27:1

13Growth of large scale PtTe,PtTe_(2) and PtSe_(2) films on a wide range of substrates显示文摘1T phase of transition metal dichalcogenides(TMDCs)formed by group 10 transition metals(e.g.Pt,Pd)have attracted increasing interests due to their novel properties and potential device applications.Synthesis of large scale thin films with controlled phase is critical especially considering that these materials have relatively strong interlayer interaction and are difficult to exfoliate.Here we report the growth of centimeter-scale PtTe,1T-PtTe2 and 1T-PtSe2 films via direct deposition of Pt metals followed by tellurization or selenization.We find that by controlling the Te flux,a hitherto-unexplored PtTe phase can also be obtained,which can be further tuned into PtTe2 by high temperature annealing under Te flux.These films with different thickness can be grown on a wide range of substrates,including NaCl which can be further dissolved to obtain free-standing PtTe2 or PtSe2 films.Moreover,a systematic thickness dependent resistivity and Hall conductivity measurements show that distinguished from the semiconducting PtSe2 with hole carriers,PtTe2 and PtTe films are metallic.Our work opens new opportunities for investigating the physical properties and potential applications of group 10 TMDC films and the new monochalcogenide PtTe film.Kenan Zhang Meng Wang Xue Zhou Yuan Wang Shengchun Shen Ke Deng Huining Peng Jiaheng Li Xubo Lai Liuwan Zhang Yang Wu Wenhui Duan Pu Yu Shuyun Zhou 2021Nano Research2021,14,6:1

16Functionalization of PEG-PM PC-based polymers for potential theranostic applications显示文摘The synergistic effect of polyethylene glycol(PEG)and poly(2-methacry-loyloxyethyl phosphorytcholine)(PMPC)can effectively reduce the protein absorption,which is beneficial to theranostics.However,PEG-PMPC-based polymers have rarely been used as nanocarriers in the theranostic field due to their limited modifiability and weak interaction with other materials.Herein,a plain method was proposed to endow them with the probable ability of loading small active agents,and the relationship between the structure and the ability of loading hydrophobic agents was explored,thus expanding their applications.Firstly,mPEG-PMPC or 4-arm-PEG-PMPC polymer was synthesized by atom transfer radical polymerization(ATRP)using mPEG-Br or 4-arm-PEG-Br as the macroinitiator.Then a strong hydrophobic segment,poly(butyl methacrylate)(PBMA),was introduced and the ability to load small hydrophobic agents was further explored.The results showed that linear mPEG-PMPC-PBMA could form micelles 50-80 nm in size and load the hydrophobic agent such as Nile red efficiently.In contrast,star-like 4-arm-PEG-PMPC-PBiyiA,a monomolecular micelle(10-20 nm),could hardly load any hydrophobic agent.This work highlights effective strategies for engineering PEG-PMPC-based polymers and may facilitate the further application in numerous fields.Ning CHEN Sidi LI Xueping LI Lixia LONG Xubo YUAN Xin HOU Jin ZHAO 2021Frontiers of Materials Science2021,15,2:0
17The effect of umbrella-type branching on the blood circulation and tumor targeting of star-branched PLA-PMPC copolymer micelles显示文摘The effect of branching on the blood circulation and tumor targeting of polymer nanovehicles in vivo was investigated in this study. For the purpose, star-branched poly(lactic acid) and poly(2-methacryloyloxyethyl phosphorylcholine)(PLA-PMPC)copolymers with umbrella-type AB3,(AB3)_(2), and(AB3)_(3) architecture were synthesized by branching at the PLA core. Micelles self-assembled from these copolymers were used to evaluate the effect of core branching on blood circulation and tumor targeting. The results showed that branching changed the behavior of polymeric self-assembly in solution, thereby changing the size and surface anti-fouling performance of the polymeric micelles. Moreover, star-branched copolymer micelles with a higher branching degree allowed their payload to persist better in blood(half-time prolonged from 7.1, 8.6 to 13.8 h) and for a 1.72-fold higher content at the tumor site. 18Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND 19Controlled release of dexamethasone from porous PLGA scaffolds under cyclic loading显示文摘Poly(L-lactide)-b-poly(ethylene glycol)(PLLA-PEG) microspheres containing dexamethasone(Dex) have been fabricated using a spray-drying technique.Porous poly(lactic-co-glycolic acid)(PLGA) scaffolds were prepared using a method combining thermally induced phase separation and porogen leaching.A post-seeding technique was used to immobilize Dex-containing PLLA-PEG microspheres on porous PLGA scaffolds,and drug-containing microspheres-scaffolds(MS-S) were obtained.Simple Dex-containing scaffolds(D-S) were also made as the control by directly dissolving Dex in the PLGA solution during scaffold fabrication.The morphologies of microspheres and scaffolds were studied by scanning electron microscopy.Drug release profiles of both MS-S and D-S were determined under cyclic loading and shaking water bath,respectively.The cumulative release of Dex was measured using an ultraviolet visible spectrophotometer.The results show that the incorporation of Dex and microspheres had little effect on the overall morphology of the porous PLGA scaffolds.Cyclic loading significantly accelerated the release of Dex from the drug-containing scaffolds.Compared with D-S,MS-S reduced the drug release rate.The controlled drug delivery of tissue engineering scaffolds under cyclic loading is a key factor to mimic the in vivo mechanical environments and achieve optical clinical efficacy.Tang GongWen Yang YanFang Sun AiPing Song TianTian Zhao YunHui Yuan XuBo Yuan XiaoYan Fan YuBo Wang Min 2010Science China Chemistry2010,53,3:0
