| 2 | Alternative splicing of MaMYB16L regulates starch degradation in banana fruit during ripening显示文摘The alternative splicing of select genes is an important mechanism to regulate responses to endogenous and environmental signals in plants.However,the role of alternative splicing in regulating fruit ripening remains unclear.Here,we discovered that MaMYB16L,an R1-type MYB transcription factor,undergoes alternative splicing and generates two transcripts,the full-length isoform MaMYB16L and a truncated form MaMYB16S,in banana fruit.During banana fruit ripening,the alternative splicing process intensifies with downregulated MaMYB16L and upregulated MaMYB16S.Moreover,MaMYB16L is a transcriptional repressor that directly binds with the promoters of many genes associated with starch degradation and MaDREB2,a positive ripening regulator,and represses their expression.In contrast,MaMBY16S lacks a DNA-binding domain but competitively combines and forms non-functional heterodimers with functional MaMYB16L.MaMYB16L-MaMYB16S heterodimers decrease the binding capacity and transrepression activity of MaMYB16L.The downregulation of MaMYB16L and the upregulation of MaMYB16S,that is,a decreased ratio of active to non-active isoforms,facilitates the activation of ripening-related genes and thereby promotes fruit ripening.Furthermore,the transient overexpression of MaMYB16S promotes banana fruit ripening,whereas the overexpression of MaMYB16L delays this process.Therefore,the alternative splicing of MaMYB16L might generate a self-controlled regulatory loop to regulate banana fruit ripening. | Guoxiang Jiang Dandan Zhang Zhiwei Li Hanzhi Liang Rufang Deng Xinguo Su Yueming Jiang Xuewu Duan | 2021 | Journal of Integrative Plant Biology2021,63,7: | 2 |

| 17 | The surface activation of boron to improve ignition and combustion characteristic显示文摘Boron is a very promising and highly attractive fuel because of high calorific value. However, the practical applications in explosives and propellants of boron have been limited by long ignition delay time and low combustion efficiency. Herein, nano-Al and graphene fluoride(GF) as surface activated materials are employed to coat boron(B) particles to improve ignition and combustion performance. The reaction heat of nano-Al coated B/KNO_(3)and GF coated B/KNO_(3)are 1116.83 J/g and 862.69 J/g, respectively, which are higher than that of pure B/KNO_(3)(823.39 J/g). The ignition delay time of B/KNO_(3)could be reduced through nano-Al coating. The shortest ignition delay time is only 75 ms for B coated with nano-Al of 8 wt%, which is much shorter than that of pure B/KNO_(3)(109 ms). However, the ignition delay time of B/KNOcoated with GF has been increased from 109 to 187 ms. B coated with GF and nano-Al shown significantly influence on the pressure output and flame structure of B/KNO_(3). Furthermore, the effects of B/O ratios on the pressure output and ignition delay time have been further fully studied. For B/KNO_(3)coated with nano-Al and GF, the highest pressures are 88 KPa and 59 KPa for B/O ratio of 4:6, and the minimum ignition delay time are 94 ms and 148 ms for B/O ratio of 7:3. | 18 | Direct somatic embryogenesis and related gene expression networks in leaf explants of Hippeastrum ‘Bangkok Rose’显示文摘Hippeastrum, a highly diverse genus in the Amaryllidaceae family, is a valuable ornamental bulbous flowering plant. Somatic embryogenesis(SE) is an efficient method for mass production of Hippeastrum plantlets. Previous studies have been devoted to the in vitro propagation of Hippeastrum, but the SE and its regulatory networks are rarely reported. In this study, we established a direct SE method of Hippeastrum Bangkok Rose' using leaf bases as explants. MS supplemented with 1.00 mg·L^(-1)NAA +1.00 mg·L^(-1)KT + 0.25 mg·L^(-1)TDZ was the optimal medium for SE. Histological observations showed that the bipolar somatic embryo originated from the epidermal cell layer and underwent initiation,globular, scutellar and coleoptile stages. During SE, endogenous hormones of IAA, CTK, ABA, and SA were highly accumulated. Transcriptomic analysis revealed the genes encoding auxin biosynthesis/metabolic enzymes and efflux carriers were induced, while the auxin receptor of TIR1 and ARF transcriptional repressor of Aux/IAA were down-regulated and up-regulated, respectively, leading to suppression of auxin signaling. In contrast, cytokine signaling was promoted at the early stage of SE, as biosynthesis, transport, and signaling components were up-regulated.Various stress-related genes were up-regulated at the early or late stages of SE. Chromatin remodeling could also be dynamically regulated via distinct expression enzymes that control histone methylation and acetylation during SE. Moreover, key SE regulators, including WOXs and SERKs were highly expressed along with SE.
