1Distribution of high-risk human papillomavirus genotype prevalence and attribution to cervical precancerous lesions in rural North China显示文摘Objective: Precise prevention is more desired for cervical cancer due to the huge population, high prevalence of human papillomavirus(HPV) infection in China and the vision of screen-and-treat strategies in low-and middleincome countries(LMICs). Considerations of combining type-specific prevalence and attribution proportion to high-grade cervical intraepithelial neoplasia are informative to more precise and effective region-specific cervical cancer prevention and control programs. The aim of the current study was to determine the genotype distribution of HPV and attribution to cervical precancerous lesions among women from rural areas in North China.Methods: A total of 9,526 women participated in the cervical cancer screening project in rural China. The samples of women who tested positive for HPV were retested with a polymerase chain reaction(PCR)-based HPV genotyping test. The attribution proportion of specific high-risk human papillomavirus(HR-HPV) types for different grades of cervical lesions was calculated by using the type contribution weighting method.Results: A total of 22.2%(2,112/9,526) of women were HR-HPV positive and HPV52(21.7%) was the most common HR-HPV genotype, followed by HPV58(18.2%), HPV53(18.2%) and HPV16(16.2%). The top three genotypes detected in HR-HPV-positive cervical intraepithelial neoplasia(CIN)1 were HPV16(36.7%), HPV58(20.4%), HPV56(15.3%). Among CIN2+, the most frequent genotypes were HPV16(75.6%), HPV52(17.8%),HPV58(16.7%). HPV16, 56, 58, 53, 52, 59, 68, and 18 combined were attributed to 84.17% of all CIN1 lesions,and HPV16, 58, and 52 combined were attributed to 86.98% of all CIN2+ lesions.Conclusions: The prevalence of HR-HPV infection among women from rural areas in North China was high and HPV16, HPV58, HPV52 had paramount attributable fraction in CIN2+. 2Effects of dietary Clostridium butyricum supplementation on growth performance,intestinal development,and immune response of weaned piglets challenged with lipopolysaccharide显示文摘Background: Weanling pigs, with immature immune system and physiological function, usually experience postweaning diarrhea. This study determined the effects of dietary Clostridium butyricum supplementation on growth performance, diarrhea, and immunity of weaned pigs challenged with lipopolysaccharide(LPS).Methods: In Experiment(Exp.) 1,144 weaned piglets were weaned at 21 d and randomly assigned to six groups,with six replicates per group and four pigs per replicate, receiving a control diet(CON) or diet supplemented with antibiotics(AB) or C. butyricum(CB)(0.1%, 0.2%, 0.4%, or 0.8%), respectively. All diets in Exp. 1 were a highly digestible basal diet, with 3,000 mg/kg zinc oxide supplied in the first 2 wk only. In Exp. 2, 180 piglets were weaned at 21 d and randomly assigned to five groups, with six replicates per group and six pigs per replicate, receiving CON, AB, or CB(0.2%, 0.4%, or 0.6%) diets. The digestibility of diets was lower than those in Exp. 1, and did not include zinc oxide. At 36 d of Exp. 2, 12 piglets were selected from each of the CON and 0.4% CB groups, six piglets were intraperitoneally injected with LPS(50 μg/kg body weight) and the other six piglets with normal saline;animals were killed at 4 h after injection to collect blood, intestine, and digesta samples for biochemical analysis.Results: In Exp. 1, CB and AB diets had no effect on growth performance of piglets. In Exp. 2, 0.4% CB decreased feed-gain ratio(P < 0.1), diarrhea score(P < 0.05), and increased duodenal, jejunal, and ileal villus height and jejunal villus height/crypt depth(P < 0.05). The 0.4% CB decreased the plasma tumor necrosis factor(TNF) α(P < 0.05) but increased ileal mucosa IL-10 and TLR2 mRNA expression(P < 0.05). Furthermore, 0.4% CB altered the microbial profile, with Bacillus and Ruminococcaceae UGG-003 at genus level and Lactobacillus casei and Parasutterella secunda at species level were higher than CON in colonic content(P < 0.05).Conclusions: Dietary C. butyricum supplementation had positive effects on growth of weaned piglets with less digestible diets. There was a tendency to reduce the feed-gain ratio, which could reduce feed costs in pig production. 4Additive and Over-dominant Effects Resulting from Epistatic Loci Are the Primary Genetic Basis of Heterosis in Rice显示文摘A set of 148 F9 recombinant inbred lines(RILs) was developed from the cross of an indica cultivar 93-11 and japonica cultivar DT713,showing strong F1 heterosis.Subsequently,two backcross F1(BCF1) populations were constructed by backcrossing these 148 RILs to two parents,93-11 and DT713.These three related populations(281BCF1 lines,148 RILs) were phenotyped for six yield-related traits in two locations.Significant inbreeding depression was detected in the population of RILS and a high level of heterosis was observed in the two BCF1 populations.A total of 42 main-effect quantitative trait loci(M-QTLs) and 109 epistatic effect QTL pairs(E-QTLs) were detected in the three related populations using the mixed model approach.By comparing the genetic effects of these QTLs detected in the RILs,BCF1 performance and mid-parental heterosis(HMP),we found that,in both BCF1 populations,the QTLs detected could be classified into two predominant types:additive and over-dominant loci,which indicated that the additive and over-dominant effect were more important than complete or partially dominance for M-QTLs and E-QTLs.Further,we found that the E-QTLs detected collectively explained a larger portion of the total phenotypic variation than the M-QTLs in both RILs and BCF1 populations.All of these results suggest that additive and over-dominance resulting from epistatic loci might be the primary genetic basis of heterosis in rice.Xiaojin Luo Yongcai Fu Peijiang Zhang Shuang Wu Feng Tian Jiayong Liu Zuofeng Zhu Jinshui Yang Chuanqing Sun 2009Journal of Integrative Plant Biology2009,51,4:20
5Flavonoids from Heartwood of Dalbergia cochinchinensis显示文摘Objective To study the flavonoids from the heartwood of Dalbergia cochinchinensis. Methods The chemical constituents were isolated and purified by combination of silica gel, macroporous resin, Sephadex LH-20, and ODS column chromatography. Their structures were identified by means of spectral analysis. Results Fifteen flavonoids were isolated and identified as pinocembrin(1), liquiritigenin(2), galangin(3), 7-hydroxy-6-methoxyflavone(4), naringenin(5), alpinetin(6), 2,3-dimethoxyxanthone(7), 6,4′-dihydroxy-7-methoxy-flavan(8), mucronulatol(9), 7,8-dihydroxyflavanone(10), 5,7,3′,5′-tetrahydroxyflavanone(11), 4,2′,5′-trihydroxy-4′-methoxychalcone(12), isoliquiritigenin(13), butein(14), and 3′,5′,5,7-tetrahydroxy-6-C-β-D-glucopyranosylflavanone(15), respectively. 7Endothelium-specific SIRT1 overexpression inhibits hyperglycemia-induced upregulation of vascular cell senescence显示文摘The rapidly increasing prevalence of diabetes mellitus worldwide is one of the most serious and challenging health problems in the 21st century.Mammalian sirtuin 1(SIRT1) has been shown to decrease high-glucose-induced endothelial cell senescence in vitro and prevent hyperglycemia-induced vascular dysfunction.However,a role for SIRT1 in prevention of hyperglycemia-induced vascular cell senescence in vivo remains unclear.We used endothelium-specific SIRT1 transgenic(SIRT1-Tg) mice and wild-type(WT) mice to construct a 40-week streptozotocin(STZ)-induced diabetic mouse model.In this mode,42.9% of wild-type(WT) mice and 38.5% of SIRT1-Tg mice were successfully established as diabetic.Forty weeks of hyperglycemia induced significant vascular cell senescence in aortas of mice,as indicated by upregulation of expression of senescence-associated markers including p53,p21 and plasminogen activator inhibitor-1(PAI-1).However,SIRT1-Tg diabetic mice displayed dramatically decreased expression of p53,p21 and PAI-1 compared with diabetic WT mice.Moreover,manganese superoxide dismutase expression(MnSOD) was significantly downregulated in the aortas of diabetic WT mice,but was preserved in diabetic SIRT1-Tg mice.Furthermore,expression of the oxidative stress adaptor p66Shc was significantly decreased in aortas of SIRT1-Tg diabetic mice compared with WT diabetic mice.Overall,these findings suggest that SIRT1-mediated inhibition of hyperglycemia-induced vascular cell senescence is mediated at least partly through the reduction of oxidative stress.CHEN HouZao WAN YanZhen ZHOU Shuang LU YunBiao ZHANG ZhuQin ZHANG Ran CHEN Feng HAO DeLong ZHAO Xiang GUO ZhiChen LIU DePei LIANG ChihChuan 2012Science China(Life Sciences)2012,55,6:14

9Prognostic value of M30/M65 for outcome of hepatitis B virus-related acute-on-chronic liver failure显示文摘AIM:To determine the prognostic value of circulating indicators of cell death in acute-on-chronic liver failure(ACLF)patients with chronic hepatitis B virus(HBV)infection as the single etiology.METHODS:Full length and caspase cleaved cytokeratin 18(detected as M65 and M30 antigens)represent circulating indicators of necrosis and apoptosis.M65and M30 were identified by enzyme-linked immunosorbent assay in 169 subjects including healthy controls(n=33),patients with chronic hepatitis B(CHB,n=55)and patients with ACLF(n=81).According to the 3-mo survival period,ACLF patients were defined as having spontaneous recovery(n=33)and non-spontaneous recovery which included deceased patients and those who required liver transplantation(n=48).RESULTS:Both biomarker levels significantly increased gradually as liver disease progressed(for M65:P<0.001 for all;for M30:control vs CHB,P=0.072;others:P<0.001 for all).In contrast,the M30/M65 ratio was significantly higher in controls compared with CHB patients(P=0.010)or ACLF patients(P<0.001).In addition,the area under receiver operating characteristic curve(AUC)analysis demonstrated that both biomarkers had diagnostic value(AUC≥0.80)in identifying ACLF from CHB patients.Interestingly,it is worth noting that the M30/M65 ratio was significantly different between spontaneous and non-spontaneous recovery in ACLF patients(P=0.032).The prognostic value of the M30/M65 ratio was compared with the Model for End-Stage Liver Disease(MELD)and Child-Pugh scores at the 3-mo survival period,the AUC of the M30/M65ratio was 0.66 with a sensitivity of 52.9%and the highest specificity of 92.6%(MELD:AUC=0.71;sensitivity,79.4%;specificity,63.0%;Child-Pugh:AUC=0.77;sensitivity,61.8%;specificity,88.9%).CONCLUSION:M65 and M30 are strongly associated with liver disease severity.The M30/M65 ratio may be a potential prognostic marker for spontaneous recovery in patients with HBV-related ACLF.Su-Jun Zheng Shuang Liu Mei Liu Malcolm A McCrae Jun-Feng Li Yuan-Ping Han Chun-Hui Xu Feng Ren Yu Chen Zhong-Ping Duan 2014World Journal of Gastroenterology2014,20,9:13
10Metamorphic Characteristics and Tectonic Implications of the Kadui Blueschist in the Central Yarlung Zangbo Suture Zone,Southern Tibet显示文摘The Kadui blueschist is located in the central section of Yarlung Zangbo suture zone(YZSZ), southern Tibet, and has been subjected to the subduction of the Neo-Tethyan Ocean below the Asian Plate and provides important clues for better understanding the evolution of the India-Asia convergence zone. In this paper, the systematical petrographic and mineral chemical studies were carried out on the Kadui blueschist, which reveal a mineral assemblage of sodic amphibole, chlorite, epidote, albite and quartz with accessory minerals of titanite, calcite and zircon. Electron microprobe analyses demonstrate that amphibole shows zoned from actinolite core to ferrowinchite/riebeckite rim composition indicating that the sodic amphibole has formed during a prograde metamorphic event. The protolith of the blueschist is an intermediate-basic pyroclastic rock. The calculated pseudosection indicates a clockwise P-T path and constrains peak metamorphic conditions of about 5.9 kbar at 345 ℃. This condition is transitional between pumpellyite-actinolite, greenschist and blueschist facies with a burial depth of 20–22 km and a thermal gradient of 15–16 ℃/km. This thermal gradient belongs to high pressure intermediate P/T facies series and is possibly related to a warm subduction setting of young oceanic slabs. 11Metamorphic Evolution and Zircon U-Pb Ages of the Nanshankou Mafic High Pressure Granulites from the Jiaobei Terrane, North China Craton显示文摘Petrological analysis and LA-ICP-MS zircon U-Pb dating were conducted on high-pressure mafic granulites, which occured as xenolith within TTG gneisses, from the Nanshankou Village of the Jiaobei terrane, Shandong Peninsula in the north-eastern part of the North China Craton(NCC). The mafic HP granulite is composed of garnet, clinopyroxene, orthopyroxene, amphibole and symplectitic clinopyroxene, orthopyroxene, plagioclase, ilmente and magnetite which were formed after the decomposition of porphyroblastic garnet and clinopyroxene. Four stages of metamorphic mineral assemblages for the mafic HP granulites were constrained by detail petrological and mineralogical investigations. The early prograde assemblage is represented by the mineral inclusions within garnet and clinopyroxene porphyroblasts(Opx_1+Pl_1+Qtz_1), recording the metamorphic conditions at ~754–757 ℃, 0.63–0.71 GPa; peak metamorphic conditions were determined at ~874–891 ℃, 1.32–1.35 GPa with the mineral assemblage of Grt_2+Cpx_2+Amp_2+Pl_2+Qtz_2. Retrograde minerals derived from symplectitic assemblage Opx_3+Cpx_3+Amp_3+Pl_3+Qtz_3+Ilm_3±Mag_3 were formed at 693–796 ℃, 0.60–0.84 GPa. A final greenschist to sub-greenschist facies event was recorded by the exsolution of actinolite and albite within a retrograded clinopyroxene, as well as the occurrence of prehnite, chlorite and calcite minerals. Accordingly, a clockwise P-T path was concluded on the basis of the different stages of mineral asseblage. Cathodoluminescence imaging, trace element and U-Pb dating of zircons from the mafic HP granulites recorded similar charactistics for three episodes of Paleo–Meso Proterozoic metamorphic events. These are the metamorphic events preserved in mafic and pelitic granulites in the Jiao-Liao-Ji belt(JLJB) with ^(207)Pb/^(206)Pb ages of 2.0-1.9 Ga for peak metamorphism and of 1.86-1.84 Ga for decomposing process, followed by a retrograde amphibolite facies metamorphic event related to the post-orogenic extension at the age of 1.76-1.74 Ga, resulting the exhumation of the granulite to the upper crust level.Shuang Chen Xu-Ping Li Fanmei Kong Qingda Feng 2018Journal of Earth Science2018,29,5:12

13QAAR exploration on pesticides with high solubility:An investigation on sulfonylurea herbicide dimers formed through π–π stacking interactions显示文摘Bioactive compounds could form aggregates that influence the bio-interactive processes. In this letter,based on p–p stacking models, quantitative aggregation–activity relationship(QAAR) studies were carried out on a series of sulfonylurea herbicides with good solubility. Four QAAR/QSAR models were constructed, which indicated that the bioactivity may strongly depend on both the characters of the dimeric aggregates and the monomer. 15Hydrochemical Characteristics and Solute Dynamics of Meltwater Runoff of Urumqi Glacier No.1,Eastern Tianshan,Northwest China显示文摘Hydrochemical characteristics and solute dynamics of bulk meltwater draining from Urumqi Glacier No.1 were investigated in years 2006 and 2007.The glacial meltwater was slightly alkaline with the mean pH of 7.64 and 7.61 in 2006 and 2007,respectively.In the meltwater,the dominant anions were the bicarbonate and sulphate,and the dominant cation was calcium.The concentration of major cations were varied as c(Ca2+) > c(Mg2+) > c(K+) > c(Na+),while the order for the cations was c(HCO3) > c(SO42) > c(NO3) > c(Cl).The total dissolved solids(TDS) in meltwater had inverse relationships with the diurnal discharge.The major ion composition of meltwater was mainly controlled by rock weathering as inferred from the Gibbs model.Furthermore,the ion ratios and Piper diagram indicated that the main processes controlling the meltwater chemistry were carbonate weathering,pyrite weathering and feldspar weathering in rocks,and Ca2+ and HCO3 were the dominant ions during the carbonate weathering process.Solute flux calculation at Glacier No.1 station suggested that chemical denudation rates were 11.46 and 13.90 ton.km 2.yr 1 in 2006 and 2007,respectively.FENG Fang LI Zhongqin JIN Shuang DONG Zhiwen WANG Feiteng 2012Journal of Mountain Science2012,9,4:10

17Discriminated sgRNAs-Based SurroGate System Greatly Enhances the Screening Efficiency of Plant Base-Edited Cells显示文摘The development of CRISPR/Cas9-mediated base editing has made genomic modification more efficient. However, selection of genetically modified cells from millions of treated cells, especially plant cells, is still challenging. In this study, an efficient surrogate reporter system based on a defective hygromycin resistance gene was established in rice to enrich base-edited cells. After step-by-step optimization, the Discriminated sgRNAs-based SurroGate system (DisSUGs) was established by artificially differentiating the editing abilities of a wild-type single guide RNA (sgRNA) targeting the surrogate reporter gene and an enhanced sgRNA targeting endogenous sites. The DisSUGs enhanced the efficiency of screening base-edited cells by 3- to 5-fold for a PmCDA1-based cytosine-to-tyrosine base editor (PCBE), and 2.5- to 6.5-fold for an adenine base editor (ABE) at endogenous targets. These targets showed editing efficiencies of <25% in the conventional systems. The DisSUGs greatly enhanced the frequency of homozygous substitutions and expanded the activity window slightly for both a PCBE and an ABE. Analyses of the total number of single-nucleotide variants from whole-genome sequencing revealed that, compared with the no-enrichment PCBE strategy, the DisSUGs did not alter the frequency of genome-wide sgRNA-independent off-target mutations, but slightly increased the frequency of target-dependent off-target mutations. 18CIVB flashback analysis of hydrogen flame based on azimuthal vorticity at mixing zone exit显示文摘Swirl-premixed combustion systems exhibit potential to meet future regulations on pollution emissions. However, combustion induced vortex breakdown(CIVB) flashback is frequently observed in these systems, especially for high hydrogen content fuel. In this study, a swirl-premixed burner with diverging centerbody was used to investigate CIVB flashback based on azimuthal vorticity at mixing zone exit. Through 2D axisymmetric model, it was found that there was a maximal azimuthal vorticity at mixing zone exit for each equivalence ratio. The physical meaning of these maximal azimuthal vorticity values was the minimally required azimuthal vorticity to trigger CIVB flashback. At the same time, the required azimuthal vorticity declined with the increase of equivalence ratio since turbulent burning velocity started to control flashback.
