New molecular method to detect denitrifying anaerobic methane oxidation bacteria from different environmental niches显示文摘The denitrifying anaerobic methane oxidation is an ecologically important process for reducing the potential methane emission into the atmosphere. The responsible bacterium for this process was Candidatus Methylomirabilis oxyfera belonging to the bacterial phylum of NC10.In this study,a new pair of primers targeting all the five groups of NC10 bacteria was designed to amplify NC10 bacteria from different environmental niches. The results showed that the group A was the dominant NC10 phylum bacteria from the sludges and food waste digestate while in paddy soil samples, group A and group B had nearly the same proportion. Our results also indicated that NC10 bacteria could exist in a high p H environment(pH 9.24)from the food waste treatment facility. The Pearson relationship analysis showed that the p H had a significant positive relationship with the NC10 bacterial diversity(p<0.05).The redundancy analysis further revealed that the p H,volatile solid and nitrite nitrogen were the most important factors in shaping the NC10 bacterial structure(p=0.01)based on the variation inflation factors selection and Monte Carlo test(999 times). Results of this study extended the existing molecular tools for studying the NC10 bacterial community structures and provided new information on the ecological distributions of NC10 bacteria. | Sai Xu Wenjing Lu Farooq Mustafa Muhammad Yanting Liu Hanwen Guo Ruihong Meng Hongtao Wang | 2018 | Journal of Environmental Sciences2018,30,3: | 4

Concentrations, sources, and influential factors of water-soluble ions of atmospheric particles in Dunhuang Mogao Grottoes,a world heritage site in China显示文摘Atmospheric particle pollution is one of the major factors leading to degradation of ancient wall paintings, particularly heritage sites in arid and semi-arid regions. However, current systematic research on the changes, sources, and influential factors of atmospheric particulate matter and its water-soluble ion concentrations is not sufficient. Thus, the major water-soluble ion concentrations, sources, and influential factors of atmospheric particles PM_(2.5) and PM_(10)(particulate matter with an aerodynamic equivalent diameter≤2.5 and 10.0μm,respectively,in ambient air)were collected from Cave 16 and its ambient exterior environment in the Dunhuang Mogao Grottoes,China, between April 2015 and March 2016.Results showed that the concentrations of PM_(2.5) and PM_(10) inside and outside the cave were the highest in March 2016 and the lowest in December 2015.The higher particle concentration from March to May was related to the frequent occurrence of sand and dust events, and the lower particle concentration from June to September was associated with good diffusion conditions, increased precipitation, and an established cave shelterbelt. The concentration of particulate matter inside the cave was affected by the concentration of particles in the air outside the cave.Ca2+,NH+4,Na+,Cl-,and SO2-4were the main components of the total ions of PM_(2.5) and PM_(10) both inside and outside the cave. The total ions inside the cave were frequently affected by the disturbance of tourists'activities during the peak tourist season from May to August.Under the influence of dust, the total concentrations of Cl-,SO2-4,Na+,NH+4,and Ca2+in particles of different sizes inside and outside the cave increased, and the concentrations of Cl-,SO2-4,Na+,and Ca2+decreased during precipitation period.Backward air mass trajectory analysis suggested that the pollutants were mainly from Xinjiang,China. The pollutant sources of air particulates are straw burning, secondary pollution sources, soil dust, dry spring rivers, and tourist activities. | YANG Xiaoju WU Fasi XU Ruihong LI Na ZHANG Zhengmo XUE Ping WANG Wanfu ZHAO Xueyong | 2022 | Journal of Arid Land2022,14,12: | 1

An ultrapotent pan-β-coronavirus lineage B(β-CoV-B)neutralizing antibody locks the receptor-binding domain in closed conformation by targeting its conserved epitope显示文摘New threats posed by the emerging circulating variants of SARS-CoV-2 highlight the need to find conserved neutralizing epitopes for therapeutic antibodies and efficient vaccine design. Here,we identified a receptorbinding domain(RBD)-binding antibody,XG014,which potently neutralizesβ-coronavirus lineage B(β-CoV-B),including SARS-CoV-2,its circulating variants,SARSCoV and bat SARSr-CoV WIV1.Interestingly,antibody family members competing with XG014 binding show reduced levels of cross-reactivity and induce antibodydependent SARS-CoV-2 spike(S)protein-mediated cellcell fusion, suggesting a unique mode of recognition by XG014.Structural analyses reveal that XG014 recognizes a conserved epitope outside the ACE2 binding site and completely locks RBD in the non-functional“down”conformation,while its family member XG005 directly competes with ACE2 binding and position the RBD“up”.Single administration of XG014 is effective in protection against and therapy of SARS-CoV-2 infection in vivo. Our findings suggest the potential to develop XG014 as pan-β-CoV-B therapeutics and the importance of the XG014 conserved antigenic epitope for designing broadly protective vaccines againstβ-CoV-B and newly emerging SARS-CoV-2 variants of concern. | Zezhong Liu Wei Xu Zhenguo Chen Wangjun Fu Wuqiang Zhan Yidan Gao Jie Zhou Yunjiao Zhou Jianbo Wu Qian Wang Xiang Zhang Aihua Hao Wei Wu Qianqian Zhang Yaming Li Kaiyue Fan Ruihong Chen Qiaochu Jiang Christian TMayer Till Schoofs Youhua Xie Shibo Jiang Yumei Wen Zhenghong Yuan Kang Wang Lu Lu Lei Sun Qiao Wang | 2022 | Protein & Cell2022,13,9: | 1
Seasonal variation of airborne fungi of the Tiantishan Grottoes andWestern Xia Museum,Wuwei,China显示文摘In this study,a systematic survey of cultural airborne fungi was carried out in the occurrence environments of wall paintings that are preserved in the Tiantishan Grottoes and the Western Xia Museum,China.A bio-aerosol sampler was used for sampling in four seasons in 2016.Culture-dependent and-independent methods were taken to acquire airborne fungal concentration and purified strains;by the extraction of genomic DNA,amplification of fungal ITS rRNA gene region, sequencing, and phylogenetic analysis, thereafter the fungal community composition and distribution characteristics of different study sites were clarified. We disclosure the main environmental factors which may be responsible for dynamic changes of airborne fungi at the sampling sites. The concentration of cultural airborne fungi was in a range from 13 to 1,576 CFU/m^(3),no significant difference between the two sites at the Tiantishan Grottoes, with obvious characteristics of seasonal variation,in winter and spring were higher than in summer and autumn. Also, there was a significant difference in fungal concentration between the inside and outside of the Western Xia Museum, the outside of the museum was far more than the inside of the museum in the four seasons, particularly in the winter.Eight fungal genera were detected, including Cladosporium,Penicillium,Alternaria, and Filobasidium as the dominant groups. The airborne fungal community structures of the Tiantishan Grottoes show a distinct characteristic of seasonal variation and spatial distribution.Relative humidity, temperature and seasonal rainfall influence airborne fungal distribution. Some of the isolated strains have the potential to cause biodeterioration of ancient wall paintings. This study provides supporting information for the pre-warning conservation of cultural relics that are preserved at local sites and inside museums. | YuLong Duan FaSi Wu DongPeng He RuiHong Xu HuYuan Feng Tuo Chen GuangXiu Liu WanFu Wang | 2021 | Research in Cold and Arid Regions2021,13,6: | 0
Spatial changes and driving factors of lake water quality in Inner Mongolia, China显示文摘Lakes play important roles in sustaining the ecosystem and economic development in Inner Mongolia Autonomous Region of China, but the spatial patterns and driving mechanisms of water quality in lakes so far remain unclear. This study aimed to identify the spatial changes in water quality and the driving factors of seven lakes(Juyanhai Lake,Ulansuhai Lake,Hongjiannao Lake,Daihai Lake,Chagannaoer Lake,Hulun Lake, and Wulannuoer Lake)across the longitudinal axis(from the west to the east)of Inner Mongolia.Large-scale research was conducted using the comprehensive trophic level index(TLI(Σ)),multivariate statistics, and spatial analysis methods. The results showed that most lakes in Inner Mongolia were weakly alkaline.Total dissolved solids and salinity of lake water showed obvious zonation characteristics.Nitrogen and phosphorus were identified as the main pollutants in lakes, with high average concentrations of total nitrogen and total phosphorus being of 4.05 and 0.21 mg/L,respectively.The values of TLI(Σ)ranged from 49.14 to 71.77,indicating varying degrees of lake eutrophication, and phosphorus was the main driver of lake eutrophication. The lakes of Inner Mongolia could be categorized into lakes to the west of Daihai Lake and lakes to the east of Daihai Lake in terms of salinity and TLI(Σ).The salinity levels of lakes to the west of Daihai Lake exceeded those of lakes to the east of Daihai Lake, whereas the opposite trend was observed for lake trophic level. The intensity and mode of anthropogenic activities were the driving factors of the spatial patterns of lake water quality. It is recommended to control the impact of anthropogenic activities on the water quality of lakes in Inner Mongolia to improve lake ecological environment. These findings provide a more thorough understanding of the driving mechanism of the spatial patterns of water quality in lakes of Inner Mongolia, which can be used to develop strategies for lake ecosystem protection and water resources management in this region. | REN Xiaohui YU Ruihong LIU Xinyu SUN Heyang GENG Yue QI Zhen ZHANG Zhuangzhuang LI Xiangwei WANG Jun ZHU Penghang GUO Zhiwei WANG Lixin XU Jifei | 2023 | Journal of Arid Land2023,15,2: | 0
Early Permian A-type Granites in the Zhangdaqi Area, Inner Mongolia, China and Their Tectonic Implications显示文摘There is a controversy regarding the amalgamation of Xing’an and Songnen Blocks along the Hegenshan-Heihe Suture(HHS)in the eastern Central Asian Orogenic Belt(CAOB).To solve this problem,we performed detailed study on the granites from the Zhangdaqi area, adjacent to the north of the HHS in the northern part of the Great Xing’an Range,NE China.Geochemically, the granites in the study area are metaluminous-weak peraluminous and high-K calc-alkaline series.Trace elements of the granites show that LREEs are relatively enriched, while HREEs are relatively deficient and obvious REE fractionation. The granites are characterized by obvious negative Eu anomalies, meanwhile, they are relatively enriched in Rb,K,Th and depleted in Ba,Nb,Sr,P,Ti.All the geochemical features suggest that the granites in the Zhangdaqi area are aluminum A-type granites. The zircon LA-ICP-MS U-Pb ages of these granites are 294-298 Ma,indicating that they formed in the Early Permian. These granites also have positiveεHf(t)values(8.4-14.2)and a relatively young two-stage model age between 449 Ma and 977 Ma,implying that the magma was derived from the re-melting of the Early Paleozoic-Neoproterozoic juvenile crust.Combined with geochemical characteristics(Nb/Ta ratios of 9.0-22.2,and Zr/Hf ratios of 52.3-152.0),we believe that the magmatic source area is a mixture of partial melting of the lower crust and depleted mantle.A-type granites and bimodal volcanic rocks along the Hegenshan-Heihe Suture formed during the Late Carboniferous-Early Permian, indicating that the HHS between Xing’an and Songnen Blocks closed in the late EarlyCarboniferous.Subsequently, the Zhangdaqi area was in a post-orogenic extensional environment from Late Carboniferous to Early Permian and resulted in the formation of the A-type granites. | ZHANG Li LIU Yongjiang SHAO Jun LI Weimin LIANG Chenyue CHANG Ruihong YANG Hongzhi FENG Zhiqiang ZHANG Chao XU Jia SHI Yi YANG Fan HE Pengfei | 2019 | Acta Geologica Sinica(English Edition)2019,93,5: | 0
