| 1 | Seasonal variations and sources of mass and chemical composition for PM_(10) aerosol in Hangzhou,China显示文摘Aerosol observation was conducted for four seasons from September 2001 to August 2002 at five sampling sites in Hangzhou, South China, on PM10 mass, 22 elements (Na, Mg, Al, Si, P, S, K, Ca, Ti, V, Cr, Mn, Fe, Ni, Cu, Zn, As, Se, Br, Cd, Ba, and Pb), 5 major ions (F-, Cl-, NO3-, SO42-, and NH4+), and organic and elemental carbon (OC and EC), showing that PM10 mass ranged from 46.7 to 270.8 g/m3, with an annual average of 119.2 g/m3. Na, Al, Si, S, K, Ca, and Fe were the most abundant elements in PM10, most of S being in the form of SO42-. SO42-, NO3-, and NH4+ were the major ions, which contributed to about 20% of the PM10 mass. The mean seasonal concentrations for SO42-, averaged over all sites, were found to be 18.0, 18.5, 24.7, and 21.4 g/m3, for spring, summer, autumn, and winter, respectively, while the corresponding loadings for NO3- were 7.2, 4.7, 7.1, and 11.2 g/m3, and for NH4+ were 6.0, 5.9, 8.2, and 9.3 g/m3, in the form mostly of NH4NO3 in spring, autumn, and winter, and mostly of (NH4)2SO4 in summer. The low NO3-/SO42-ratio found indicates coal combustion as the major source throughout the year. The mean annual concentrations of OC and EC in PM10 were found to be 21.4, and 4.1 g/m3, respectively. Material balance calculation indicated that fugitive dust, the secondary aerosol, and carbonaceous matter were the most abundant species in PM10 for the four seasons, as is characteristic for cities in South China. | Junji Cao Zhenxing Shen Judith C. | 2 | Synthesis and evaluation of a paclitaxel-binding polymeric micelle for efficient breast cancer therapy显示文摘Paclitaxel(PTX) is one of the most effective anticancer drugs for the treatment of various solid tumors, but its clinical use is limited by its poor solubility, low bioavailability, and severe systemic toxicity. Encapsulation of PTX in polymeric nanoparticles is used to overcome these problems but these micelles still need improvements in stability, pharmacokinetics, therapeutic efficacy, and safety profiles. In this study, we demonstrate a facile fabrication of a stable PTX-binding micelle made from poly(ethylene glycol)-block-dendritic polylysine, whose primary amines were reacted with phenethyl isothiocyanate(PEITC), a hydrophobic anticancer agent under clinical study. The amphiphilic conjugate(PEG-Gx-PEITC; Gx, the generation of the polylysine dendron) formed well-defined micelles whose core was composed of phenyl groups and thiourea groups binding PTX via π-π stacking and hydrogen bonding. Compared with the PTX-loaded poly(ethylene glycol)-block-poly(D,L-lactide)(PEGPDLLA/PTX) micelles in clinical use, PTX-loaded PEG-Gx-PEITC third-generation(PEG-G3-PEITC/PTX) micelles showed slowed blood clearance, enhanced tumor accumulation, and thus much improved in vivo therapeutic efficacy in both subcutaneous and orthotopic human breast cancer xenografts. | 3 | Microblog Burst Keywords Detection Based on Social Trust and Dynamics Model显示文摘Microblog has emerged as a popular medium for providing new sources of information and rapid communications, particularly during burst topics. Burst keywords detection from real-time microblog streams is important for burst topics detection. The exiting algorithms may detect fake burst keywords without taking into account the trustworthiness of the users and human's daily timetable. Our work is the first to combine the trustworthiness of the users with burst keywords detection. We propose a novel approach to detect burst keywords based on social trust and dynamics model. We adapt basic notions of dynamics from physics and model keywords bursts as momentum change of the keywords. On the analogy of physical dynamics model, this approach defines mass as the trustworthiness of user and position as the frequency of keywords. We compute each keyword's burst value by using Moving average convergence/divergence(MACD) and determine whether it is a burst keyword in a given time window. | 4 | Patient-derived non-small cell lung cancer xenograft mirrors complex tumor heterogeneity显示文摘Objective:Patient-derived xenograft(PDX)models have shown great promise in preclinical and translational applications,but their consistency with primary tumors in phenotypic,genetic,and pharmacodynamic heterogeneity has not been well-studied.This study aimed to establish a PDX repository for non-small cell lung cancer(NSCLC)and to further elucidate whether it could preserve the heterogeneity within and between tumors in patients.Methods:A total of 75 surgically resected NSCLC specimens were implanted into immunodeficient NOD/SCID mice.Based on the successful establishment of the NSCLC PDX model,we compared the expressions of vimentin,Ki67,EGFR,and PD-L1 proteins between cancer tissues and PDX models using hematoxylin and eosin staining and immunohistochemical staining.In addition,we detected whole gene expression profiling between primary tumors and PDX generations.We also performed whole exome sequencing(WES)analysis in 17 first generation xenografts to further assess whether PDXs retained the patient heterogeneities.Finally,paclitaxel,cisplatin,doxorubicin,atezolizumab,afatininb,and AZD4547 were used to evaluate the responses of PDX models to the standard-of-care agents.Results:A large collection of serially transplantable PDX models for NSCLC were successfully developed.The histology and pathological immunohistochemistry of PDX xenografts were consistent with the patients’tumor samples.WES and RNA-seq further confirmed that PDX accurately replicated the molecular heterogeneities of primary tumors.Similar to clinical patients,PDX models responded differentially to the standard-of-care treatment,including chemo-,targeted-and immuno-therapeutics.Conclusions:Our established PDX models of NSCLC faithfully reproduced the molecular,histopathological,and therapeutic characteristics,as well as the corresponding tumor heterogeneities,which provides a clinically relevant platform for drug screening,biomarker discovery,and translational research. | Xuanming Chen Cheng Shen Zhe Wei Rui Zhang Yongsheng Wang Lili Jiang Ke Chen Shuang Qiu Yuanli Zhang Ting Zhang Bin Chen Yanjun Xu Qiyi Feng Jinxing Huang Zhihui Zhong Hongxia Li Guowei Che Kai Xiao | 2021 | Cancer Biology & Medicine2021,18,1: | 5 |
| 5 | Environmental correlates of sedentary behaviors and physical activity in Chinese preschool children:A cross-sectional study显示文摘Objective:This cross-sectional study examined environmental correlates of sedentary behavior(SB) and physical activity(PA) in preschool children in the urban area of Tianjin,China.Methods:Data were collected from the Physical Activity and Health in Tianjin Chinese Children study,involving healthy children 3-6 years old and their families.In all children(n=980),leisure-time SB(LTSB) and leisure-time PA(LTPA) were reported in min/day by parents.In a subgroup(n=134),overall sedentary time,light PA,and moderate-to-vigorous PA(MVP A) were objectively measured using ActiGraph accelerometry(>3 days,>10 h/day).Environmental correlates were collected using a questionnaire that included home and neighborhood characteristics(e.g.,traffic safety,presence of physical activity facilities) and children’s behaviors.Potential correlates were identified using linear regression analysis.Results:Multiple linear regression analysis showed that 'having grandparents as primary caregivers'(βs and 95% confidence intervals(95%CIs)for overall sedentary time:29.7(2.1-57.2);LTSB(In):0.19(0.11-0.28)) and 'having a television(for LTSB(In):0.13(0.00-0.25)) or computer(for LTSB(ln):0.13(0.03-0.23) in the child’ s bedroom' were both associated with higher SB.Furthermore, 'having grandparents as primary caregivers' was associated with less MVPA(β(95%CI):-7.6(-14.1 to-1.2)),and 'active commuting to school by walking' correlated with more MVPA(β(95%CI):9.8(2.2-17.4)).The path model showed that 'more neighborhood PA facilities close to home' was indirectly related to higher LTPA(In),which was partly mediated by 'outdoor play'(path coefficients(95%CI):0.005(0.002-0.008)) and 'going to these facilities more often'(path coefficients(95%CI):0.013(0.008-0.01 8)).Traffic safety was not a correlate.Conclusion:Family structure and media exposure in the home maybe important factors in shaping preschoolers’ PA patterns.Built environmental correlates could indirectly influence preschoolers’ LTPA through parental help with engaging in active behaviors. | Congchao Lu Tong Shen Guowei Huang Eva Corpeleijn | 2022 | Journal of Sport and Health Science2022,11,5: | 3 |

| 7 | UGGreedy: Influence Maximization for User Group in Microblogging显示文摘We study a new influence maximization problem about how to find a seed set which can maximize the influence spread to a targeted user group in microblogging. To solve this problem, we propose a threestage User group greedy algorithm(UGGreedy) based on user attributes. To reduce network scale, we delete useless user nodes, and rank the rest of users based on user attributes to form a seed candidate set. We employ the seed candidate set to construct a simplified microblogging network graph. | 9 | Granitoid emplacement during syn-convergent transtension:An example from the Huamenlou pluton in North Qinling,central China显示文摘The Huamenlou pluton,is an elongated granite intrusion with high aspect ratio,emplaced within the southern margin of the North Qinling(central China).Here we investigate this pluton through multiple techniques including the fabric study,microstructural observation and zircon geochronology.Our zircon U-Pb data confirm that the granite crystallized at ca.462 Ma which is consistent with the ages of other linear plutons in North Qinling.Microstructural observations of the Huamenlou granites illustrate that the pluton has undergone superimposed deformation during its emplacement,from magmatic to hightemperature solid state conditions.The internal fabric obtained by anisotropy of magnetic susceptibility(AMS)and shape preferred orientation(SPO)show similar results.The fabrics are relatively concordant and generally vary from NE-SW to NEE-SWW which are roughly oblique to the trend of the pluton elongation and the regional structures.Meanwhile,scalar parameters reflect two completely different strain regimes for the pluton and its host rocks,i.e.,the fabrics within host rocks are mainly oblate while the central part of the intrusion displays mainly prolate fabrics.It is inferred that the structural pattern recorded in this pluton was caused by local dextral transtension in consequence of oblique convergence between the South and North China Blocks.We propose that the local transtension in convergence setting probably evolved from vertical extrusion tectonics that provided room for the magma emplacement and imparted prolate fabrics in the Huamenlou pluton. | Yang Li Wentian Liang Guowei Zhang Yazhou Ran Qi Shen Jingli Wang Chunsheng Jin | 2018 | Geoscience Frontiers2018,9,1: | 1 |

| 15 | Allergic Phenotypes and Sarcopenia:Evidence from Observational Studies and Mendelian Randomization Analysis显示文摘Commonly afected in early-life population,the impact of allergic phenotypes on mid-or late-life health is less discussed.This study is to explore the association of allergic phenotypes including atopic dermatitis(AD),asthma,eosinophils count(EC),and sarcopenia.We conducted observational studies and mendelian randomization(MR)analysis based on UK Biobank(UKB),the China Health and Retirement Longitudinal Study(CHARLS)and data from genome-wide association study(GWAS).Based on the UKB data,AD,asthma and EC were positively correlated with pre-sarcopenia and decreased skeletal muscle mass index and hand grip in fully adjusted model.Asthma and EC were signifcantly associated with sarcopenia while AD was marginally associated(p=0.095).Based on the CHARLS cohort,asthma signifcantly added 109.4%risk for pre-sarcopenia in adjusted model(relative risk=2.094;p=0.002),respectively.Both asthma(β=0.100,p=0.006)and EC(β=0.023,p=0.017)exerted signifcantly casual efects on pre-sarcopenia.However,as for sarcopenia,merely EC exhibited a signifcantly casual efect(β=0.005,p=0.048).Signifcant casual efects of AD(β=–0.027,p=0.003),asthma(β=–0.029,p=0.027)and EC(β=–0.041,p<0.001)on decreased appendicular lean mass(ALM)were observed using the inverse-variance weighted method and the Mendelian randomization pleiotropy residual sum and outlier(MR-PRESSO)method.Our results revealed a contributory role of AD,asthma and EC on sarcopenia,especially in terms of decreased ALM,an indicator for sarcopenia diagnosis.The fndings of our study will raise the awareness of preventing aging-related disorders or geriatric syndromes among allergic populations. | Zhenwei Tang Guowei Zhou Yi Xiao Hong Liu Xiang Chen Minxue Shen | 2024 | Phenomics2024,4,1: | 0 |
| 16 | Long-term correction of hemorrhagic diathesis in hemophilia A mice by an AAV-delivered hybrid FⅧcomposed of the human heavy chain and the rat light chain显示文摘Conventional therapies for hemophilia A(HA)are prophylactic or on-demand intravenous FⅧinfusions.However,they are expensive and inconvenient to perform.Thus,better strategies for HA treatment must be developed.In this study,a recombinant FⅧcDNA encoding a human/rat hybrid FⅧwith an enhanced procoagulant potential for adeno-associated virus(AAV)-delivered gene therapy was developed.Plasmids containing human FⅧheavy chain(hHC),human light chain(hLC),and rat light chain(rLC)were transfected into cells and hydrodynamically injected into HA mice.Purified AAV viruses were intravenously injected into HA mice at two doses.Results showed that the hHC+rLC protein had a higher activity than the hHC+hLC protein at comparable expression levels.The specific activity of hHC+rLC was about 4-to 8-fold higher than that of their counterparts.Hydrodynamic injection experiments obtained consistent results.Notably,the HA mice undergoing the AAV-delivered hHC+rLC treatment exhibited a visibly higher activity than those treated with hHC+hLC,and the therapeutic effects lasted for up to 40 weeks.In conclusion,the application of the hybrid FⅧ(hHC+rLC)via an AAV-delivered gene therapy substantially improved the hemorrhagic diathesis of the HA mice.These data might be of help to the development of optimized FⅧexpression cassette for HA gene therapy. | Jianhua Mao Yun Wang Wei Zhang Yan Shen Guowei Zhang Wenda Xi Qiang Wang Zheng Ruan Jin Wang Xiaodong Xi | 2022 | Frontiers of Medicine2022,16,4: | 0 |
| 17 | Enhancement of a recombinase-aided amplification assay using betaine and pullulan显示文摘Background:Nucleic acid amplification enhancers suitable for use in a recombinase-aided amplification(RAA)assay were studied for the first time,and amplification of a long-fragment(509 bp)was initially explored.Methods:Using recombinant plasmids and clinical samples,RAA fluorescence and basic methods were used to evaluate the efficacy.The fluorescence method was evaluated by threshold time and fluorescence value,and the basic method was characterized by 2%agarose gel electrophoresis.Results:Taking a previously established RAA assay for HPV18 as an example,we demonstrated that the addition of 0.2 M,0.4 M,and 0.6 M betaine and 10%pullulan could enhance the RAA.The new RAA assays with betaine and pullulan were named B-RAA and P-RAA,respectively.Using the B-RAA and P-RAA fluorescence methods,the threshold time values could be shortened by 1.72-2.32 minutes and 2.60 minutes,respectively,and the fluorescence values could be enhanced by 8847.25-9094.37 mv and 5250 mv,respectively.Using the basic method,the sensitivity could be increased 10-fold.We successfully amplified a long-fragment of 509 bp using a P-RAA assay with a sensitivity of 102 copies/μL(compared with 103 copies/μL in the RAA assay).Conclusions:Thus,we concluded that betaine and pullulan are effective additives to enhance the sensitivity of RAA assays. | Jinrong Wang Guowei Song Yue Ming Jing Pan Ruiqing Zhang Guohao Fan Xinxin Shen Xuejun Ma Lixin Li | 2022 | Infectious Medicine2022,1,2: | 0 |
