| 1 | Antibacterial effects of platelet-rich fibrin produced by horizontal centrifugation显示文摘Platelet-rich fibrin(PRF)has been widely used owing to its ability to stimulate tissue regeneration.To date,few studies have described the antibacterial properties of PRF.Previously,PRF prepared by horizontal centrifugation(H-PRF)was shown to contain more immune cells than leukocyte-and platelet-rich fibrin(L-PRF).This study aimed to compare the antimicrobial effects of PRFs against Staphylococcus aureus and Escherichia coli in vitro and to determine whether the antibacterial effects correlated with the number of immune cells.Blood samples were obtained from eight healthy donors to prepare L-PRF and H-PRF.The sizes and weights of L-PRF and H-PRF were first evaluated,and their antibacterial effects against S.aureus and E.coli were then tested in vitro using the inhibition ring and plate-counting test methods.Flow-cytometric analysis of the cell components of L-PRF and H-PRF was also performed.No significant differences in size or weight were observed between the L-PRF and H-PRF groups.The H-PRF group contained more leukocytes than the L-PRF group.While both PRFs had notable antimicrobial activity against S.aureus and E.coli,H-PRF demonstrated a significantly better antibacterial effect than L-PRF.Furthermore,the antimicrobial ability of the PRF solid was less efficient than that of wet PRF.In conclusion,H-PRF exhibited better antibacterial activity than L-PRF,which might have been attributed to having more immune cells. | Mengge Feng Yulan Wang Peng Zhang Qin Zhao Shimin Yu Kailun Shen Richard J.Miron Yufeng Zhang | 2020 | International Journal of Oral Science2020,12,4: | 12 |
| 2 | Rapid generation of genetic diversity by multiplex CRISPR/Cas9 genome editing in rice显示文摘The clustered regularly interspaced short palindromic repeats(CRISPR)-associated endonuclease 9(CRISPR/Cas9) system has emerged as a promising technology for specific genome editing in many species. Here we constructed one vector targeting eight agronomic genes in rice using the CRISPR/Cas9 multiplex genome editing system. By subsequent genetic transformation and DNA sequencing, we found that the eight target genes have high mutation efficiencies in the T_0 generation. Both heterozygous and homozygous mutations of all editing genes were obtained in T_0 plants. In addition, homozygous sextuple, septuple, and octuple mutants were identified. As the abundant genotypes in T_0 transgenic plants, various phenotypes related to the editing genes were observed. | 3 | Regulation of extracellular bioactive cations in bone tissue microenvironment induces favorable osteoimmune conditions to accelerate in situ bone regeneration显示文摘The design of orthopedic biomaterials has gradually shifted from“immune-friendly”to“immunomodulatory,”in which the biomaterials are able to modulate the inflammatory response via macrophage polarization in a local immune microenvironment that favors osteogenesis and implant-to-bone osseointegration.Despite the well-known effects of bioactive metallic ions on osteogenesis,how extracellular metallic ions manipulate immune cells in bone tissue microenvironments toward osteogenesis and subsequent bone formation has rarely been studied.Herein,we investigate the osteoimmunomodulatory effect of an extracellular bioactive cation(Mg^(2+))in the bone tissue microenvironment using custom-made poly lactic-co-glycolic acid(PLGA)/MgO-alendronate microspheres that endow controllable release of magnesium ions.The results suggest that the Mg^(2+)-controlled tissue microenvironment can effectively induce macrophage polarization from the M0 to M2 phenotype via the enhancement of anti-inflammatory(IL-10)and pro-osteogenic(BMP-2 and TGF-β1)cytokines production.It also generates a favorable osteoimmune microenvironment that facilitates the proliferation and osteogenic differentiation of bone marrow mesenchymal stem cells.The in vivo results further verify that a large amount of bony tissue,with comparable bone mineral density and mechanical properties,has been generated at an early post-surgical stage in rat intramedullary bone defect models.This study demonstrates that the concept of in situ immunomodulated osteogenesis can be realized in a controlled magnesium tissue microenvironment. | Zhengjie Lin Danni Shen Weixiao Zhou Yufeng Zheng Tiantian Kong Xuanyong Liu Shuilin Wu Paul K.Chu Ying Zhao Jun Wu Kenneth M.C.Cheung Kelvin W.K.Yeung | 2021 | Bioactive Materials2021,6,8: | 5 |

| 5 | Effect of sulfur source on photocatalytic degradation performance of CdS/MoS2 prepared with one-step hydrothermal synthesis显示文摘Cd S/Mo S_2,an extremely efficient photocatalyst,has been extensively used in hydrogen photoproduction and pollutant degradation.Cd S/Mo S_2can be synthesized by a facile one-step hydrothermal process.However,the effect of the sulfur source on the synthesis of Cd S/Mo S_2via one-step hydrothermal methods has seldom been investigated.We report herein a series of one-step hydrothermal preparations of Cd S/Mo S_2using three different sulfur sources:thioacetamide,L-cysteine,and thiourea.The results revealed that the sulfur source strongly affected the crystallization,morphology,elemental composition and ultraviolet(UV)–visible-light-absorption ability of the Cd S/Mo S_2.Among the investigated sulfur sources,thioacetamide provided the highest visible-light absorption ability for Cd S/Mo S_2,with the smallest average particle size and largest surface area,resulting in the highest efficiency in Methylene Blue(MB)degradation.The photocatalytic activity of Cd S/Mo S_2synthesized from the three sulfur sources can be arranged in the following order:thioacetamide>L-cysteine>thiourea.The reaction rate constants(k)for thioacetamide,L-cysteine,and thiourea were estimated to be 0.0197,0.0140,and 0.0084 min^(-1),respectively.However,thioacetamide may be limited in practical application in terms of its price and toxicity,while L-cysteine is relatively economical,less toxic and exhibited good photocatalytic degradation performance toward MB. | Yanfeng Wang Wei Chen Xiao Chen Huajun Feng Dongsheng Shen Bin Huang Yufeng Jia Yuyang Zhou Yuxiang Liang | 2018 | Journal of Environmental Sciences2018,30,3: | 3 |
| 6 | Safety and efficacy of Yupingfeng granules in children with recurrent respiratory tract infection:A randomized clinical trial显示文摘Importance:Recurrent respiratory tract infection(RRTI)is common in children.Inappropriate RRTI treatment will lead to asthma and other diseases,thereby seriously affecting the growth and physical health of children.Immune function modulation can prevent and alleviate childhood RRTI.Yupingfeng(YPF),a patented traditional Chinese medicine(TCM),has immunomodulatory effects and is widely used in China to treat children with RRTI.Objective:To evaluate the safety and efficacy of YPF monotherapy in treating children with RRTI.Methods:This multicenter,randomized,double-blind,double-simulation,noninferiority clinical trial was conducted from January 2015 to August 2017,with an 8-week treatment period and 52-week follow-up after the drug withdrawal.Children aged 2–6 years with RRTI meeting the inclusion and exclusion criteria were enrolled in 13 hospitals in China and divided randomly into three groups(2:2:1 ratio)to receive YPF,pidotimod,or placebo.The primary outcome was the proportion of RRTI returning to normal standard level during the follow-up.The secondary outcomes were reduction in the number of RRTI recurrences,effect on clinical symptoms(in accord with TCM practice),effect per symptom,and safety.The trial was registered at the Chinese Clinical Trials Registry(www.chictr.org.cn)under the unique identifier ChiCTR-IPR-15006847.Results:Three hundred and fifty-one children were enrolled and randomly assigned to 3 groups;124,125,and 61 children in the YPF,pidotimod,and placebo groups,respectively,had completed the trial.During the follow-up,the proportion of RRTI returning to normal standard level was 73.13%,67.15%,and 38.81%with YPF,pidotimod,and placebo,respectively(P<0.0001).The proportion of cases who returned to normal standard level in the YPF group was 34.32%higher than that in the placebo group.The safety profile did not significantly differ among the groups.Interpretation:YPF granules were noninferior to the active control drug pidotimod oral solution for the treatment of RRTI in children,and were superior to placebo,with a high safety profile. | Baoping Xu Xinmin Li Siyuan Hu Yixiao Bao Fengmei Chen Zhimin Chen Yonggang Du Enmei Liu Yufeng Liu Qinghui Mou Baoling Su Bo Wang Jianwen Xu Guiping Xu Qiaozhi Yang Liwei Gao Xiaohui Liu Lei Li Rong Ma Kunling Shen | 2022 | Pediatric Investigation2022,6,2: | 3 |
| 7 | Ticagrelor inhibits the NLRP3 inflammasome to protect against inflammatory disease independent of the P2Y_(12 ) signaling pathway显示文摘Ticagrelor is the first reversibly binding oralP2Y_(12) receptor antagonist to inhibit platelet activation and has been approved by the Food and Drug Administration for the treatment of coronary artery disease. At present, the other pharmacological functions of ticagrelor remain poorly understood. The NOD-, LRR-, and pyrin domain-containing protein 3 (NLRP3) inflammasome plays a critical role in the innate immune system, but its excessive activation also contributes to the pathogenesis of complex diseases. In this study, we systematically examined the effects of ticagrelor on the NLRP3 inflammasome and found that ticagrelor inhibits NLRP3 inflammasome activation in macrophages independent of its classic inhibitory effect on theP2Y_(12) signaling pathway. Further mechanistic studies demonstrate that ticagrelor attenuates the oligomerization of apoptosis-associated speck-like protein containing a CARD (ASC) by blocking chloride efflux, an effect achieved through the degradation of chloride intracellular channel proteins (CLICs) and blockade of the translocation of CLICs to the plasma membrane. Moreover, experiments on lipopolysaccharide-induced sepsis and alum-induced peritonitis in mice confirmed that ticagrelor mitigates the severity of systemic inflammation independent ofP2Y_(12) receptor antagonism. Importantly, oral administration of ticagrelor rapidly and strongly inhibited NLRP3 inflammasome activation in peripheral blood mononuclear cells from patients with acute coronary syndrome. | 8 | Machine learning helps improve diagnostic ability of subclinical keratoconus using Scheimpflug and OCT imaging modalities显示文摘Purpose:To develop an automated classification system using a machine learning classifier to distinguish clinically unaffected eyes in patients with keratoconus from a normal control population based on a combination of Scheimpflug camera images and ultra-high-resolution optical coherence tomography(UHR-OCT)imaging data.Methods:A total of 121 eyes from 121 participants were classified by 2 cornea experts into 3 groups:normal(50 eyes),with keratoconus(38 eyes)or with subclinical keratoconus(33 eyes).All eyes were imaged with a Scheimpflug camera and UHR-OCT.Corneal morphological features were extracted from the imaging data.A neural network was used to train a model based on these features to distinguish the eyes with subclinical keratoconus from normal eyes.Fisher’s score was used to rank the differentiable power of each feature.The receiver operating characteristic(ROC)curves were calculated to obtain the area under the ROC curves(AUCs).Results:The developed classification model used to combine all features from the Scheimpflug camera and UHR-OCT dramatically improved the differentiable power to discriminate between normal eyes and eyes with subclinical keratoconus(AUC=0.93).The variation in the thickness profile within each individual in the corneal epithelium extracted from UHR-OCT imaging ranked the highest in differentiating eyes with subclinical keratoconus from normal eyes.Conclusion:The automated classification system using machine learning based on the combination of Scheimpflug camera data and UHR-OCT imaging data showed excellent performance in discriminating eyes with subclinical keratoconus from normal eyes.The epithelial features extracted from the OCT images were the most valuable in the discrimination process.This classification system has the potential to improve the differentiable power of subclinical keratoconus and the efficiency of keratoconus screening. | Ce Shi Mengyi Wang Tiantian Zhu Ying Zhang Yufeng Ye Jun Jiang Sisi Chen Fan Lu Meixiao Shen | 2020 | Eye and Vision2020,7,1: | 3 |
| 9 | Predicting the degradation behavior of magnesium alloys with a diffusion-based theoretical model and in vitro corrosion testing显示文摘Magnesium alloys have shown great potential for their use in the medical device field, due to the promising biodegradability. However, it remains a challenge to characterize the degradation behavior of the Mg alloys in a quantitative manner. As such, controlling the degradation rate of the Mg alloys as per our needs is still hard, which greatly limits the practical application of the Mg alloys as a degradable biomaterial.This paper discussed a numerical model developed based on the diffusion theory, which can capture the experimental degradation behavior of the Mg alloys precisely. The numerical model is then implemented into a finite element scheme, where the model is calibrated with the data from our previous studies on the corrosion of the as-cast Mg-1 Ca and the as-rolled Mg-3 Ge binary alloys. The degradation behavior of a pin implant is predicted using the calibrated model to demonstrate the model’s capability. A standard flow is provided in a practical framework for obtaining the degradation behavior of any biomedical Mg alloys. This methodology was further verified via the comparison with enormous available experimental results. | 10 | Magnetic resonance (MR) safety and compatibility of a novel iron bioresorbable scaffold显示文摘Fully bioresorbable scaffolds have been designed to overcome the limitations of traditional drug-eluting stents(DESs),which permanently cage the native vessel wall and pose possible complications.The ultrathin-strut designed sirolimus-eluting iron bioresorbable coronary scaffold system(IBS)shows comparable mechanical properties to traditional DESs and exhibits an adaptive degradation profile during target vessel healing,which makes it a promising candidate in all-comers patient population.For implanted medical devices,magnetic resonance(MR)imaging properties,including MR safety and compatibility,should be evaluated before its clinical use,especially for devices with intrinsic ferromagnetism.In this study,MR safety and compatibility of the IBS scaffold were evaluated based on a series of well-designed in-vitro,ex-vivo and in-vivo experiments,considering possible risks,including scaffold movement,over-heating,image artifact,and possible vessel injury,under typical MR condition.Traditional ASTM standards for MR safety and compatibility evaluation of intravascular devices were referred,but not only limited to that.The unique time-relevant MR properties of bioresorbable scaffolds were also discussed.Possible forces imposed on the scaffold during MR scanning and MR image artifacts gradually decreased along with scaffold degradation/absorption.Rigorous experiments designed based on a scientifically based rationale revealed that the IBS scaffold is MR conditional,though not MR compatible before complete absorption.The methodology used in the present study can give insight into the MR evaluation of magnetic scaffolds(bioresorbable)or stents(permanent). | Dong Bian Li Qin Wenjiao Lin Danni Shen Haiping Qi Xiaoli Shi Gui Zhang Hongwei Liu Han Yang Jin Wang Deyuan Zhang Yufeng Zheng | 2020 | Bioactive Materials2020,5,2: | 2 |
| 11 | Formalin-induced astrocyte glutamate-glutamine cycle involved in rat spinal cord central sensitization显示文摘BACKGROUND:Central sensitization,a state of increased excitability of nociceptive neurons in the spinal dorsal horn following peripheral tissue injury and/or inflammation,is an important mechanism underlying hyperalgesia and neuropathic pain.Participation of the glutamate-glutamine cycle in central sensitization of the spinal cord remains poorly understood. OBJECTIVE:To determine whether the astrocyte-neuronal glutamate-glutamine cycle is involved in formalin-induced central sensitization in the spinal cord. DESIGN,TIME AND SETTING:A randomized,controlled,animal experiment was performed at the Institute of Orthopedics,Second Hospital,Lanzhou University,China from September 2007 to August 2008. MATERIALS:Methionine sulfoximine(MSO,0.1 mmol/L),glutamine(0.25 mmol/L),and formalin were used for this study. METHODS:A total of 43 male,Sprague Dawley rats,aged 4 months,were randomly assigned to a sham operation group(n = 6) and a model group(n = 37).Rats in the model group received intrathecal infusion in the spinal cord.7 days later,37 model rats were randomly divided into PBS, MSO,glutamine,MSO + glutamine and formalin subcutaneous injection alone groups.The PBS, MSO,glutamine,MSO + glutamine groups were respectively intrathecally injected with PBS,MSO, glutamine,MSO + glutamine(50μL each),and then infused with 10μL of saline.Rats from the sham operation group were not subjected to intrathecal infusion in the spinal cord.At 15 minutes after intrathecal injection,a rat model of formalin-induced inflammatory pain was established by subcutaneous injection of 5%formalin(50μL) in the left hindpaw. MAIN OUTCOME MEASURES:Changes in spontaneous nociceptive behavior(licking/biting or flinching) were observed following formalin injection into the rat hindpaw. RESULTS:Compared with the PBS group,duration of licking/biting was significantly shortened,and flinching frequency was significantly diminished in the MSO group(P<0.05).Compared with the MSO group,duration of licking/biting was significantly prolonged,and flinching frequency was significantly increased in the MSO + glutamine group(P<0.05).There was no significant difference in inflammatory pain behaviors among the sham operation,PBS,glutamine,MSO + glutamine,and formalin subcutaneous injection alone groups(P>0.05). Time-dependent ex-situ 3D information including the number, volume fraction, distribution and morphology of micro-pores was analyzed. The results reveal that the signifi cant formation and growth of micro-pores occur at the end of secondary/beginning of tertiary creep stage. The irregular large pores as well as high density pores located at strain concentration region are the major detrimental factors facilitating the creep damage. Fast emerging of MRSA prevalence requires special attention for strengthening the inventory of antimicrobial compounds. Silver nanoparticles(AgNPs) have been widely used to treat multi-drug resistant pathogens due to the unique antibacterial properties, meanwhile spermine has been proven to exert outstanding inhibition effect to S.aureus with not yet fully understood mechanisms. The aim of this study was to investigate the synergistic effect of AgNPs and spermine as well as to determine the antibacterial activity of their combination against MRSA strains. Several clinical MRSA isolates and ATCC BAA-1026 were used to determine minimum inhibitory concentration(MIC) and fractional inhibitory concentration indices(FICI) of AgNPs and spermine, and a synergistic effect was observed. | 19 | Developing a tool for nurses to assess risk of infection in pediatric oncology patients in China:a modified Delphi study显示文摘Infections are identified as the most common preventable cause of death in pediatric oncology patients.Assessing and stratifying risk of infections are essential to prevent infection in these patients.To date,no tool can fulfill this demand in China.This study aimed to develop a nursing work-based and Chinese-specific tool for pediatric nurses to assess risk of infection in oncology patients.This research was a modified Delphi study.Based on a literature review,a 37-item questionnaire rating on a 0-5 scale was developed.Twenty-four experts from 8 hospitals in 6 provinces of China were consulted for three rounds.Consensus for each item in the first round was defined as:the rating mean was > 3 and the coefficient of variation(CV) was < 0.5.Consensus for each item in the second round was defined as CV<0.3.Consensus among experts was defined as:P value of Kendall's coefficient of concordance(W)<0.05.After three rounds of consultation,a two-part tool was developed:the Immune Status Scale(ISS) and the Checklist of Risk Factors of Infection(CRFI).There were 5 items in the ISS and 14 in the CRFI.Based on the ISS score,nurses could stratify children into the low-risk and high-risk groups.For high-risk children,nurses should screen risk factors of infection every day by the CRFI,and twice weekly for low-risk children.Further study is needed to verify this tool's efficacy. | Yufeng Zhou Yan Cui Hong Wang Fang Wang Chao Lu Yan Shen | 2016 | The Journal of Biomedical Research2016,30,5: | 1 |
| 20 | The importance of proleptic branch traits in biomass production of poplar in high-density plantations显示文摘Branch phenotypic traits determine tree crown architecture,which in turn governs leaf display,light interception,and biomass production.Sylleptic and proleptic branches are the obviously different branch phenotypes in the poplar crown.Many studies have focused on the influence of sylleptic branch numbers(SBN)on biomass production,but the research on the influence of proleptic branch phenotypes was only a few.To explore the relationship between proleptic branch traits and biomass generation production in a high-density poplar plantation,we investigated the branch phenotypic traits of three poplar genotypes,all of which have high survival rates in forests(>95%)and significantly different crown architecture and biomass performance in the high-density plantations(1667 stems ha−1).The plantation site was established in 2007.A terrestrial laser scanner was used to measure branch characteristics such as length,angle of origin and termination,and azi-muth angle.A hierarchical cluster analysis performed on branch characteristics showed that SBN,crown depth,and proleptic branch curvature(PBC)were clustered with bio-mass production and leaf area index(LAI).Among all of the monitored traits,PBC played the second most important role in biomass production after SBN and was significantly correlated with SBN,LAI,and biomass production.The positive correlation between PBC and SBN indicated that a larger PBC was associated with more sylleptic branches within the monitored genotypes planted in the high-density plantation,providing greater leaf area and biomass produc-tion.The results of this study will improve the identification of high-production poplar varieties for cultivation in high-density plantations for biofuel production. | Changjun Ding Ningning Wang Qinjun Huang Weixi Zhang Juan Huang Suli Yan Boyi Chen Dejun Liang Yufeng Dong Yingbai Shen Xiaohua Su | 2022 | Journal of Forestry Research2022,33,2: | 1 |
