| 1 | A Resource for Inactivation of MicroRNAs Using Short Tandem Target Mimic Technology in Model and Crop Plants显示文摘microRNAs (miRNAs)are endogenous small non-coding RNAs that bind to mRNAs and target them for cleavage and/or translational repression,leading to gene silencing.We previously developed short tandem target mimic (STTM)technology to deactivate endogenous miRNAs in Arabidopsis.Here,we created hundreds of STTMs that target both conserved and species-specific miRNAs in Arabidopsis,tomato,rice,and maize,providing a resource for the functional interrogation of miRNAs.We not only revealed the functions of several miRNAs in plant development,but also demonstrated that tissue-specific inactivation of a few miRNAs in rice leads to an increase in grain size without adversely affecting overall plant growth and development.RNA-seq and small RNAseq analyses of STTM156/157 and STTM165/166 transgenic plants revealed the roles of these miRNAs in plant hormone biosynthesis and activation,secondary metabolism,and ion-channel activity-associated electrophysiology,demonstrating that STTM technology is an effective approach for studying miRNA functions.To facilitate the study and application of STTM transgenic plants and to provide a useful platform for storing and sharing of information about miRNA-regulated gene networks,we have established an online Genome Browser (http://gffzzc750bc0108fa4044sbwx0wcnnfok966n0.ffgz.tsg.suse.edu.cn/designindex2.php) to display the transcriptomic and miRNAomic changes in STTMinduced miRNA knockdown plants. | Ting Peng Mengmeng Qiao Haiping Liu Sachin Teotia Zhanhui Zhang Yafan Zhao Bobo Wang Dongjie Zhao Lina Shi Cui Zhang Brandon Le Kestrel Rogers Chathura Gunasekara Haitang Duan Yiyou Gu Lei Tian Jinfu Nie Jian Qi Fanrong Meng Lan Huang Qinghui Chen Zhenlin Wang Jinshan Tang Xiaoqing Tang Ting Lan Xuemei Chen Hairong Wei Quanzhi Zhao Guiliang Tang | 2018 | Molecular Plant2018,11,11: | 7 |
| 2 | In vivo degradation and endothelialization of an iron bioresorbable scaffold显示文摘Detection of in vivo biodegradation is critical for development of next-generation medical devices such as bioresorbable stents or scaffolds(BRSs).In particular,it is urgent to establish a nondestructive approach to examine in vivo degradation of a new-generation coronary stent for interventional treatment based on mammal experiments;otherwise it is not available to semi-quantitatively monitor biodegradation in any clinical trial.Herein,we put forward a semi-quantitative approach to measure degradation of a sirolimus-eluting iron bioresorbable scaffold(IBS)based on optical coherence tomography(OCT)images;this approach was confirmed to be consistent with the present weight-loss measurements,which is,however,a destructive approach.The IBS was fabricated by a metal-polymer composite technique with a polylactide coating on an iron stent.The efficacy as a coronary stent of this new bioresorbable scaffold was compared with that of a permanent metal stent with the name of trade mark Xience,which has been widely used in clinic.The endothelial coverage on IBS was found to be greater than on Xience after implantation in a rabbit model;and our well-designed ultrathin stent exhibited less individual variation.We further examined degradation of the IBSs in both minipig coronary artery and rabbit abdominal aorta models.The present result indicated much faster iron degradation of IBS in the rabbit model than in the porcine model.The semi-quantitative approach to detect biodegradation of IBS and the finding of the species difference might be stimulating for fundamental investigation of biodegradable implants and clinical translation of the next-generation coronary stents. | Wenjiao Lin Hongjie Zhang Wanqian Zhang Haiping Qi Gui Zhang Jie Qian Xin Li Li Qin Haifeng Li Xiang Wang Hong Qiu Xiaoli Shi Wei Zheng Deyuan Zhang Runlin Gao Jiandong Ding | 2021 | Bioactive Materials2021,6,4: | 3 |
| 3 | Surface engineering of titania nanotubes incorporated with double-layered extracellular vesicles to modulate inflammation and osteogenesis显示文摘Titania nanotubes(TNT)generated on titanium implant are emerged as important modification technique to facilitate bone regeneration.Mesenchymal stem cells(MSCs)-derived exosomes are membrane bound extracellular vesicles(EVs),which play an important role in tissue regeneration.The objective of this study was to generate an EVs hybrid TNT aiming at regulating inflammation,MSCs recruitment and osteogenesis.We isolated EVs from MSCs(MSCs EVs)and 3-day osteogenically differentiated MSCs(3d EVs).MSC EVs and 3d EVs exhibited round morphology under TEM,which also showed robust internalization by human bone marrow derived MSCs(hBMSCs).Next,we fabricated 3d EVs/MSC EVs hybrid TNT.When inflammatory macrophages were co-cultured with EVs hybrid TNT,the gene and protein expression of inflammatory cytokine were significantly reduced.Macrophage morphology was also examined by confocal laser scanning microscopy(CLSM)and scanning electron microscopy(SEM).Further migratory ability study using hBMSCs indicated significant enhancement of MSCs migration in EVs hybrid TNT.In addition,we further demonstrated significant increase of osteogenic differentiation of hBMSCs in EVs hybrid TNT.This study suggests that EVs hybrid TNT may serve as a viable therapeutic approach to enhance osteogenesis and bone regeneration. | Qingyu Zhao Yi Zhang Lan Xiao Haiping Lu Yaping Ma Qi Liu Xin Wang | 2021 | Regenerative Biomaterials2021,8,3: | 3 |
| 4 | Magnetic resonance (MR) safety and compatibility of a novel iron bioresorbable scaffold显示文摘Fully bioresorbable scaffolds have been designed to overcome the limitations of traditional drug-eluting stents(DESs),which permanently cage the native vessel wall and pose possible complications.The ultrathin-strut designed sirolimus-eluting iron bioresorbable coronary scaffold system(IBS)shows comparable mechanical properties to traditional DESs and exhibits an adaptive degradation profile during target vessel healing,which makes it a promising candidate in all-comers patient population.For implanted medical devices,magnetic resonance(MR)imaging properties,including MR safety and compatibility,should be evaluated before its clinical use,especially for devices with intrinsic ferromagnetism.In this study,MR safety and compatibility of the IBS scaffold were evaluated based on a series of well-designed in-vitro,ex-vivo and in-vivo experiments,considering possible risks,including scaffold movement,over-heating,image artifact,and possible vessel injury,under typical MR condition.Traditional ASTM standards for MR safety and compatibility evaluation of intravascular devices were referred,but not only limited to that.The unique time-relevant MR properties of bioresorbable scaffolds were also discussed.Possible forces imposed on the scaffold during MR scanning and MR image artifacts gradually decreased along with scaffold degradation/absorption.Rigorous experiments designed based on a scientifically based rationale revealed that the IBS scaffold is MR conditional,though not MR compatible before complete absorption.The methodology used in the present study can give insight into the MR evaluation of magnetic scaffolds(bioresorbable)or stents(permanent). | Dong Bian Li Qin Wenjiao Lin Danni Shen Haiping Qi Xiaoli Shi Gui Zhang Hongwei Liu Han Yang Jin Wang Deyuan Zhang Yufeng Zheng | 2020 | Bioactive Materials2020,5,2: | 2 |

| 11 | A Novel Shilling Attack Detection Model Based on Particle Filter and Gravitation显示文摘With the rapid development of e-commerce, the security issues of collaborative filtering recommender systems have been widely investigated. Malicious users can benefit from injecting a great quantities of fake profiles into recommender systems to manipulate recommendation results. As one of the most important attack methods in recommender systems, the shilling attack has been paid considerable attention, especially to its model and the way to detect it. Among them, the loose version of Group Shilling Attack Generation Algorithm (GSAGenl) has outstanding performance. It can be immune to some PCC (Pearson Correlation Coefficient)-based detectors due to the nature of anti-Pearson correlation. In order to overcome the vulnerabilities caused by GSAGenl, a gravitation-based detection model (GBDM) is presented, integrated with a sophisticated gravitational detector and a decider. And meanwhile two new basic attributes and a particle filter algorithm are used for tracking prediction. And then, whether an attack occurs can be judged according to the law of universal gravitation in decision-making. The detection performances of GBDM, HHT-SVM, UnRAP, AP-UnRAP Semi-SAD,SVM-TIA and PCA-P are compared and evaluated. | 15 | Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment. | 16 | Significant expansion and red-shifting of fluorescent protein chromophore determined through computational design and genetic code expansion显示文摘Fluorescent proteins (FPs)with emission wavelengths in the far-red and infrared regions of the spectrum provide powerful tools for deep-tissue and super-resolution imaging.The development of red-shifted FPs has evoked widespread interest and continuous engineering efforts.In this article, based on a computational design and genetic code expansion,we report a rational approach to significantly expand and red-shift the chromophore of green fluorescent protein (GFP).We applied computational calculations to predict the excitation and emission wavelengths of a FP chromophore harboring unnatural amino acids (UAA)and identify in silico an appropriate UAA,2-amino-3-(6- hydroxynaphthalen-2-yl)propanoic acid (naphthol-Ala).Our methodology allowed us to formulate a GFP variant (cpsfGFP-66-Naphthol-Ala)with red-shifted absorbance and emission spectral maxima exceeding 60 and 130 nm,respectively,compared to those of GFP.The GFP chromophore is formed through autocatalytic post-translational modification to generate a planar 4-(p-hydroxybenzylidene)-5- imidazolinone chromophore.We solved the crystal structure of cpsfGFP-66-naphthol-Ala at 1.3 ■ resolution and demonstrated the formation of a much larger conjugated n-system when the phenol group is replaced by naphthol.These results explain the significant red-shifting of the excitation and emission spectra of cpsfGFP-66-naphthol-Ala. | Li Wang Xian Chen Xuzhen GUO liasong Li Qi Liu Fuying Kang Xudong Wang Cheng HU Haiping Liu Weimin Gong Wei Zhuang Xiaohong Liu Jiangyun Wang | 2018 | Biophysics Reports2018,4,5: | 0 |
