1Texture analysis on gadoxetic acid enhanced-MRI for predicting Ki-67 status in hepatocellular carcinoma: A prospective study显示文摘Objective: To investigate the value of whole-lesion texture analysis on preoperative gadoxetic acid enhanced magnetic resonance imaging(MRI) for predicting tumor Ki-67 status after curative resection in patients with hepatocellular carcinoma(HCC).Methods: This study consisted of 89 consecutive patients with surgically confirmed HCC. Texture features were extracted from multiparametric MRI based on whole-lesion regions of interest. The Ki-67 status was immunohistochemical determined and classified into low Ki-67(labeling index ≤15%) and high Ki-67(labeling index >15%) groups. Least absolute shrinkage and selection operator(LASSO) and multivariate logistic regression were applied for generating the texture signature, clinical nomogram and combined nomogram. The discrimination power, calibration and clinical usefulness of the three models were evaluated accordingly. Recurrence-free survival(RFS) rates after curative hepatectomy were also compared between groups.Results: A total of 13 texture features were selected to construct a texture signature for predicting Ki-67 status in HCC patients(C-index: 0.878, 95% confidence interval: 0.791-0.937). After incorporating texture signature to the clinical nomogram which included significant clinical variates(AFP, BCLC-stage, capsule integrity, tumor margin,enhancing capsule), the combined nomogram showed higher discrimination ability(C-index: 0.936 vs. 0.795,P<0.001), good calibration(P>0.05 in Hosmer-Lemeshow test) and higher clinical usefulness by decision curve analysis. RFS rate was significantly lower in the high Ki-67 group compared with the low Ki-67 group after curative surgery(63.27% vs. 85.00%, P<0.05).Conclusions: Texture analysis on gadoxetic acid enhanced MRI can serve as a noninvasive approach to preoperatively predict Ki-67 status of HCC after curative resection. 2Imaging evaluation of sorafenib for treatment of advanced hepatocellular carcinoma显示文摘Sorafenib,which is a novel targeted agent,plays an important role in treating advanced hepatocellular carcinoma(HCC)through its antiangiogenic and antiproliferative effects.However,conventional morphology-based radiographic evaluation systems may underestimate the efficacy of sorafenib in HCC due to a lack of apparent tumor shrinkage or altered tumor morphology in many cases.This calls for the development of more accurate imaging methods for evaluating sorafenib.The introduction of tumor burden measurements based on viability and other evolving imaging approaches for assessing therapeutic effects are promising for overcoming some of the limitations of the morphology-based criteria.In this review,we summarize various imaging methods that are used to assess treatment responses of advanced HCC to sorafenib.Imaging markers predictive of prognosis in advanced HCC after treatment with sorafenib are also included and discussed.Tianying Zheng Hanyu Jiang Yi Wei Zixing Huang Jie Chen Ting Duan Bin Song 2018Chinese Journal of Cancer Research2018,30,3:0
3Harnessing decellularised extracellular matrix microgels into modular bioinks for extrusion-based bioprinting with good printability and high post-printing cell viability显示文摘The printability of bioink and post-printing cell viability is crucial for extrusion-based bioprinting.A proper bioink not only provides mechanical support for structural fidelity,but also serves as suitable three-dimensional(3D)microenvironment for cell encapsulation and protection.In this study,a hydrogel-based composite bioink was developed consisting of gelatin methacryloyl(GelMA)as the continuous phase and decellularised extracellular matrix microgels(DMs)as the discrete phase.A flow-focusing microfluidic system was employed for the fabrication of cell-laden DMs in a high-throughput manner.After gentle mixing of the DMs and GelMA,both rheological characterisations and 3D printing tests showed that the resulting DM-GelMA hydrogel preserved the shear-thinning nature,mechanical properties,and good printability from GelMA.The integration of DMs not only provided an extracellular matrix-like microenvironment for cell encapsulation,but also considerable shear-resistance for high post-printing cell viability.The DM sizes and inner diameters of the 3D printer needles were correlated and optimised for nozzle-based extrusion.Furthermore,a proof-of-concept bioink composedg of RSC96 Schwann cells encapsulated DMs and human umbilical vein endothelial cell-laden GelMA was successfully bioprinted into 3D constructs,resulting in a modular co-culture system with distinct cells/materials distribution.Overall,the modular DM-GelMA bioink provides a springboard for future precision biofabrication and will serve in numerous biomedical applications such as tissue engineering and drug screening.Hanyu Chu Kexin Zhang Zilong Rao Panpan Song Zudong Lin Jing Zhou Liqun Yang Daping Quan Ying Bai 2023Biomaterials Translational2023,4,2:0
4Comparisons of microstructure and elemental composition of eggshells among wild plover populations显示文摘Reproduction investment is a prominent trade-off in life-history theory and is subject to strong selection pressure.The avian eggshell,as a crucial barrier between the bird embryo and the surrounding environment,undergoes optimization under different environmental selection regimes to ensure the successful development of embryos,which can be linked to local adaptation.Therefore,understanding the variation in eggshell microstructure and composition in wild bird populations living in contrasting ambient environments is of great significance.In this study,we utilized electron microscope ultrastructure measurement and elemental analyses to measure and compare the microstructure and element composition of eggshells from three wild plover populations(Charadrius alexandrinus and C.dealbatus)residing in heterogeneous habitats across varied climatic zones.These populations include the high-altitude Qinghai Lake population,the temperate coastal Tangshan population,and the tropical coastal Zhanjiang population.Our findings revealed that the palisade layer was thinner in the Qinghai Lake population compared to its lowland populations.This difference might be attributed to hypoxia which facilitates the hatching process by allowing chicks to easily break through their shells.Additionally,the variations in the elemental composition of the eggshells among populations well reflected the distribution of element content in different geographical regions.The Qinghai Lake population displayed low zinc and low manganese levels but high calcium levels,while the Zhanjiang population exhibited high zinc,high iron,high manganese,and high phosphorus levels.Furthermore,these variations in elemental composition could also account for the observed microstructural differences among populations.Collectively,we propose that the dissimilarities in eggshell microstructure and elemental composition among populations could be attributed to adaptations to different environmental conditions.Our findings lay the groundwork for future research to explore the mechanisms behind the variations in eggshell characteristics among wild bird populations,and contribute to a broader understanding of biodiversity mechanisms.Langyu Gu Hanyu Yang Canwei Xia Zitan Song Yachang Cheng Chenjing Huang Yuelou Liu Yang Liu 2023Avian Research2023,14,4:0
5Preclinical and early clinical studies of a novel compound SYHA1813 that efficiently crosses the blood-brain barrier and exhibits potent activity against glioblastoma显示文摘Glioblastoma(GBM)is the most common and aggressive malignant brain tumor in adults and is poorly controlled.Previous studies have shown that both macrophages and angiogenesis play significant roles in GBM progression,and co-targeting of CSF1R and VEGFR is likely to be an effective strategy for GBM treatment.Therefore,this study developed a novel and selective inhibitor of CSFIR and VEGFR,SYHA1813,possessing potent antitumor activity against GBM.SYHA1813 inhibited VEGFR and CSFIR kinase activities with high potency and selectivity and thus blocked the cell viability of HUVECs and macrophages and exhibited anti-angiogenetic effects both in vitro and in vivo.SYHA1813 also displayed potent in vivo antitumor activity against GBM in immune-competent and immune-deficient mouse models,including temozolomide(TMZ)insensitive tumors.Notably,SYHA1813 could penetrate the blood-brain barrier(BBB)and prolong the survival time of mice bearing intracranial GBM xenografts.Moreover,SYHA1813 treatment resulted in a synergistic antitumor efficacy in combination with the PD-1 antibody.As a clinical proof of concept,SYHA1813 achieved confirmed responses in patients with recurrent GBM in an ongoing first-in-human phase I trial.The data of this study support the rationale for an ongoing phase I clinical study(ChiCTR2100045380).Yingqiang Liu Zhengsheng Zhan Zhuang Kang Mengyuan Li Yongcong Lv Shenglan Li Linjiang Tong Fang Feng Yan Li Mengge Zhang Yaping Xue Yi Chen Tao Zhang Peiran Song Yi Su Yanyan Shen Yiming Sun Xinying Yang Yi Chen Shanyan Yao Hanyu Yang Caixia Wang Meiyu Geng Wenbin Li Wenhu Duan Hua Xie Jian Ding 2023Acta Pharmaceutica Sinica B2023,13,12:0
6O-GlcNAcylation regulates phagocytosis by promoting Ezrin localization at the cell cortex显示文摘O-GlcNAcylation is a post-translational modification that serves as a cellular nutrient sensor and participates in multiple physiological and pathological processes.However,it remains uncertain whether O-GlcNAcylation is involved in the regulation of phagocytosis.Here,we demonstrate a rapid increase in protein OGlcNAcylation in response to phagocytotic stimuli.Knockout of the O-GlcNAc transferase or pharmacological inhibition of O-GlcNAcylation dramatically blocks phagocytosis,resulting in the disruption of retinal structure and function.Mechanistic studies reveal that the O-GlcNAc transferase interacts with Ezrin,a membrane-cytoskeleton linker protein,to catalyze its O-GlcNAcylation.Our data further show that Ezrin OGlcNAcylation promotes its localization to the cell cortex,thereby stimulating the membrane-cytoskeleton interaction needed for efficient phagocytosis.These findings identify a previously unrecognized role for protein O-GlcNAcylation in phagocytosis with important implications in both health and diseases.Song Yang Hanyu Liu Hua Ni Lingyu Jiang Mulin Yang Quan Chen Jun Zhou Fan Yu 2023Journal of Genetics and Genomics2023,50,7:0
7Mobileman,Construction Agile Goods Delivery Robot显示文摘The traditional automated guided vehicle(AGV) on goods delivery faces the challenges when task space expands beyond 2 D plans. 3 D environments such as uneven terrain, ramps, and staircase are typical in construction site. Thus, the key to introducing this technology into construction industry is to improve AGV’s stability and autonomous navigation ability in more complex three-dimensional environments. In this paper, mobileman, a novel tracked autonomous guide vehicle, is introduced. Compared with other construction robots, mobileman maximizes its load capacity on the basis of assuring accessibility. Furthermore, its modular designs and self-balancing platform enable it to cope with more complex challenging scenarios, such as staircase with 35-degree sloped staircase, while another modular design featured automated loading and unloading functionality.
