| 1 | Increased liver stiffness in alcoholic liver disease:Differentiating fibrosis from steatohepatitis显示文摘AIM:To test if inflammation also interferes with liver stiffness (LS) assessment in alcoholic liver disease (ALD) and to provide a clinical algorithm for reliable fibrosis assessment in ALD by FibroScan (FS).METHODS:We first performed sequential LS analysis before and after normalization of serum transaminases in a learning cohort of 50 patients with ALD admitted for alcohol detoxification. LS decreased in almost all patients within a mean observation interval of 5.3 d. Six patients (12%) would have been misdiagnosed with F3and F4 fibrosis but LS decreased below critical cut-off values of 8 and 12.5 kPa after normalization of trans-aminases. RESULTS:Of the serum transaminases,the decrease in LS correlated best with the decrease in glutamic oxaloacetic transaminase (GOT). No significant chang-es in LS were observed below GOT levels of 100 U/L. After establishing the association between LS and GOT levels,we applied the rule of GOT < 100 U/L for reliable LS assessment in a second validation cohort of 101 patients with histologically confi rmed ALD. By ex-cluding those patients with GOT > 100 U/L at the time of LS assessment from this cohort,the area under the receiver operating characteristic (AUROC) for cirrhosis detection by FS improved from 0.921 to 0.945 while specificity increased from 80% to 90% at a sensitivity of 96%. A similar AUROC could be obtained for lower F3 fibrosis stage if LS measurements were restricted to patients with GOT < 50 U/L. Histological grading of inflammation did not further improve the diagnostic accuracy of LS.CONCLUSION:Coexisting steatohepatitis markedly increases LS in patients with ALD independent of fibrosis stage. | 3 | Bioengineering bacterial encapsulin nanocompartments as targeted drug delivery system显示文摘The development of Drug Delivery Systems(DDS)has led to increasingly efficient therapies for the treatment and detection of various diseases.DDS use a range of nanoscale delivery platforms produced from polymeric of inorganic materials,such as micelles,and metal and polymeric nanoparticles,but their variant chemical composition make alterations to their size,shape,or structures inherently complex.Genetically encoded protein nanocages are highly promising DDS candidates because of their modular composition,ease of recombinant production in a range of hosts,control over assembly and loading of cargo molecules and biodegradability.One example of naturally occurring nanocompartments are encapsulins,recently discovered bacterial organelles that have been shown to be reprogrammable as nanobioreactors and vaccine candidates.Here we report the design and application of a targeted DDS platform based on the Thermotoga maritima encapsulin reprogrammed to display an antibody mimic protein called Designed Ankyrin repeat protein(DARPin)on the outer surface and to encapsulate a cytotoxic payload.The DARPin9.29 chosen in this study specifically binds to human epidermal growth factor receptor 2(HER2)on breast cancer cells,as demonstrated in an in vitro cell culture model.The encapsulin-based DDS is assembled in one step in vivo by co-expressing the encapsulin-DARPin9.29 fusion protein with an engineered flavin-binding protein mini-singlet oxygen generator(MiniSOG),from a single plasmid in Escherichia coli.Purified encapsulin-DARPin_miniSOG nanocompartments bind specifically to HER2 positive breast cancer cells and trigger apoptosis,indicating that the system is functional and specific.The DDS is modular and has the potential to form the basis of a multi-receptor targeted system by utilising the DARPin screening libraries,allowing use of new DARPins of known specificities,and through the proven flexibility of the encapsulin cargo loading mechanism,allowing selection of cargo proteins of choice. | Alexander Van de Steen Rana Khalife Noelle Colant Hasan Mustafa Khan Matas Deveikis Saverio Charalambous Clare M.Robinson Rupali Dabas Sofia Esteban Serna Diana A.Catana Konstantin Pildish Vladimir Kalinovskiy Kenth Gustafsson Stefanie Frank | 2021 | Synthetic and Systems Biotechnology2021,6,3: | 2 |

| 6 | A new endophytic insect-associated Daldinia species, recognised from a comparison of secondary metabolite profiles and molecular phylogeny显示文摘Xylariaceous endophytes are commonly encountered in plants,and it has recently been found out that insect vectors play a major role in their life cycle,as well as in the apparent host-specificity of the fungal stromata.In this study,a new insect-associated,endophytic anamorph species of Daldinia is recognised from a comparison of (a)morphological and ecological data;(b)molecular phylogenetic data based on rDNA,alpha-actin and beta-tubulin genes;and (c)secondary metabolite profiles based on high performance liquid chromatography/diode array/mass spectrometry(HPLC-DAD/MS)and gas chromatography/mass spectrometry(GC-MS)in comparison with authentic strains of Daldinia and other Xylariaceae.The new species,Daldinia hawksworthii,is a symbiont of the willow woodwasp,Xiphydria prolongata,which is in turn preferentially associated with Salix.A comparison of HPLCDAD/MS profiles from standardised cultures revealed the presence of a specific compound,which was not encountered in any of the ca.250 Daldinia strains studied for comparison.This compound was isolated after fermentation of the fungus in 10 l scale by preparative chromatography.Structure elucidation by NMR spectroscopy revealed a novel pyrone,for which the name dalsymbiopyrone is proposed.The compound showed weak antimicrobial and cytotoxic effects and might be a signal molecule involved in the trilateral ecological interaction between the host plant,the insect,and its fungal symbiont. | Sylvie Pažoutová Stefanie Follert Jens Bitzer Matthias Keck Frank Surup PetrŠrůtka Jaroslav Holuša Marc Stadler | 2013 | Fungal Diversity2013,,3: | 1 |

| 17 | Antibacterial activity of Bixa orellana L.(achiote) against Streptococcus mutans and Streptococcus sanguinis显示文摘Objective: To evaluate the cytotoxic and antibacterial effect of Bixa orellana L.(B. orellana)(achiote) methanol extract against Streptococcus mutans(ATCC 25175)(S. mutans) and Streptococcus sanguinis(ATCC 10556)(S. sanguinis).Methods: Two methanol extracts of B. orellana were prepared in vitro, from the seeds and leaves. The antibacterial activity of extracts against S. mutans and S. sanguinis was evaluated using the cup-plate agar diffusion method. The minimum inhibitory concentration(MIC) was determined using the microdilution method and the cytotoxic activity was determinated by using the cell line MDCK.Results: A stronger antibacterial effect was observed with the leaves methanolic extract with an inhibition zone of(19.97 ± 1.31) mm against S. mutans and(19.97 ± 1.26) mm against S. sanguinis. The methanolic extract of the seeds had an activity of(15.11 ± 1.03)mm and(16.15 ± 2.15) mm against S. mutans and S. sanguinis, respectively. The MIC of the leaf and the seed extracts against S. sanguinis was 62.5 and 125 mg/m L, respectively,and the MIC of the leaf extract against S. mutans was 62.5 mg/m L, and for the seed extract it was 31.25 mg/m L. The 50% cytotoxic concentration was 366.45 and 325.05 mg/m L for the leaves and seeds extracts, respectively.Conclusions: The experimental findings demonstrated the antibacterial effect of the methanolic extract of B. orellana(achiote) on S. mutans and S. sanguinis.
