| 1 | MdGSTF6,activated by MdMYB1,plays an essential role in anthocyanin accumulation in apple显示文摘Anthocyanins are biosynthesized on the cytosolic surface of the endoplasmic reticulum and then transported into the vacuole for storage.Glutathione S-transferases(GSTs)are considered to be responsible for the transport of anthocyanins into the vacuole.However,the regulatory mechanisms of GSTs in plants are still unclear.Here,we performed a genome-wide analysis and identified 69 GST genes in apple.The expression of MdGSTF6 was positively correlated with the anthocyanin content(r=0.949)during‘Yanfu 8’fruit development.The overexpression of MdGSTF6 in the Arabidopsis thaliana tt19 mutant resulted in seedlings of 35S::MdGSTF6-GFP/tt19 that could accumulate anthocyanin and rescue its phenotype,suggesting that MdGSTF6 was an anthocyanin transporter.The silencing of MdGSTF6 affected anthocyanin accumulation in apple fruit.Moreover,the knockdown of MdGSTF6 by RNA interference in cultured‘Gala’seedlings inhibited anthocyanin accumulation.The interaction experiments showed that MdMYB1 could bind directly to the MdGSTF6 promoter to transcriptionally activate its expression.Collectively,our results demonstrate that MdGSTF6 encodes an important GST transporter of anthocyanins in apple fruit and provide evidence for the associated regulatory mechanisms.Therefore,MdMYB1 can not only regulate anthocyanin synthesis,but also control the transport of anthocyanin in apples.This information may be useful for further clarifying the regulation of anthocyanin transport in apple. | Shenghui Jiang Min Chen Naibo He Xiaoliu Chen Nan Wang Qingguo Sun Tianliang Zhang Haifeng Xu Hongcheng Fang Yicheng Wang Zongying Zhang Shujing Wu Xuesen Chen | 2019 | Horticulture Research2019,6,1: | 16 |
| 2 | Viral MIPα homologous with human MIP-1α acts on HIV co-receptor CCR5显示文摘The function and usage of vMIPα encoded by K6 gene of herpesvirus 8 (HHV8) which has homology with human macrophage protein (MIP) have not been clearly known. In the present note the K6 gene of HHV8 was cloned and transfected into NIH3T3 cells and E. coli cells. Conditional media from the 3T3-transfected cells and K6 product vMIPa from E. coli. Cells were used to perform the experiments of ligand-receptor binding and cellular adhesion with peripheral blood macrophages. The conditional media and the purified vMIPa from E. coli could compete to bind to CCR5 located on macrophages from peripheral blood with I125-hMIP-1α chemokine of human. Cellular adhesion showed that the conditional media from transfected cells and the purified vMIPa did not induce the adhesion of macrophages from peripheral blood to ICAM-1. In conclusion, vMIPα encoded by K6 gene of HHV8 can bind to CCR5 of peripheral blood macrophage cells and does not induce their adhesion. | 4 | Neutrophil-derived heparin binding protein triggers vascular leakage and synergizes with myeloperoxidase at the early stage of severe burns(With video)显示文摘Background:Burn shock caused by vascular leakage is one of the main causes of high mortality in severe burn injury.However,the pathophysiological mechanism of vascular leakage is still unclear.The purpose of this study was to explore the molecular mechanism of vascular leakage in the early stage of severe burn and provide a new target for the treatment of severe burns.Methods:Neutrophils were isolated from human peripheral blood by magnetic beads sorting.ELISA was used to detect neutrophil-derived granule proteins and glycocalyx injury products in plasma.The vascular leakage and neutrophil movement were assessed by in vivo laser confocal imaging in mice,and high-quality video were provided.Adhesion-related molecules were investigated by qRT-PCR.The damage to glycocalyx of mice vascular endothelial cellswas observed by transmission electron microscope and scanning electron microscope.Proteomic analysis,flow cytometry and immunofluorescence were used to further study the relationship between human peripheral blood neutrophil-derived hypochlorite(HOCl)and CD44 of human vascular endothelial cells.Results:In this study,we found that rapidly increasing activated neutrophils secrete heparin binding protein(HBP)andmyeloperoxidase(MPO)after severe burn injury.Increased HBP triggers vascular leakage with synergy of MPO,results in systemic edema and burn shock.Furthermore,we found that the MPO catalytic product HOCl but not MPO triggers CD44 extracellular domain shedding from vascular endothelial cells to damage the glycocalyx.Damage to the glycocalyx results in firm adhesion of neutrophils and increases vascular leakage.However,MPO inhibitors partially protect the glycocalyx of vascular endothelial cells.The combination of HBP and MPO inhibitors markedly reduces vascular leakage and systemic edema in the early stage of severe burns.Conclusions:Taken together,these data reveal that neutrophil-derived HBP and MPO play an important synergies role in triggering vascular leakage at the early stage of severe burns.Targeted intervention in these two biomolecules may introduce new strategies for helping to reduce large amount of fluid loss and subsequent burn shock. | Lu Liu Yiming Shao Yixuan Zhang Yunxi Yang Jiamin Huang Linbin Li Ran Sun Yuying Zhou Yicheng Su Bingwei Sun | 2021 | Burns & Trauma2021,9,1: | 3 |

| 6 | Genome-wide analysis of expression quantitative trait loci(eQTLs)reveals the regulatory architecture of gene expression variation in the storage roots of sweet potato显示文摘Dissecting the genetic regulation of gene expression is critical for understanding phenotypic variation and species evolution.However,our understanding of the transcriptional variability in sweet potato remains limited.Here,we analyzed two publicly available datasets to explore the landscape of transcriptomic variations and its genetic basis in the storage roots of sweet potato.The comprehensive analysis identified a total of 724,438 high-confidence single nucleotide polymorphisms(SNPs)and 26,026 expressed genes.Expression quantitative trait locus(eQTL)analysis revealed 4408 eQTLs regulating the expression of 3646 genes,including 2261 local eQTLs and 2147 distant eQTLs.Two distant eQTL hotspots were found with target genes significantly enriched in specific functional classifications.By combining the information from regulatory network analyses,eQTLs and association mapping,we found that IbMYB1-2 acts as a master regulator and is the major gene responsible for the activation of anthocyanin biosynthesis in the storage roots of sweet potato.Our study provides the first insight into the genetic architecture of genome-wide expression variation in sweet potato and can be used to investigate the potential effects of genetic variants on key agronomic traits in sweet potato. | Lei Zhang Yicheng Yu Tianye Shi Meng Kou Jian Sun Tao Xu Qiang Li Shaoyuan Wu Qinghe Cao Wenqian Hou Zongyun Li | 2020 | Horticulture Research2020,7,1: | 2 |
| 7 | Overexpression of phosphatidylserine synthase IbPSS1 affords cellular Na+homeostasis and salt tolerance by activating plasma membrane Na^(+)/H^(+)antiport activity in sweet potato roots显示文摘Phosphatidylserine synthase(PSS)-mediated phosphatidylserine(PS)synthesis is crucial for plant development.However,little is known about the contribution of PSS to Na^(+)homeostasis regulation and salt tolerance in plants.Here,we cloned the IbPSS1 gene,which encodes an ortholog of Arabidopsis AtPSS1,from sweet potato(Ipomoea batatas(L.)Lam.).The transient expression of IbPSS1 in Nicotiana benthamiana leaves increased PS abundance.We then established an efficient Agrobacterium rhizogenes-mediated in vivo root transgenic system for sweet potato.Overexpression of IbPSS1 through this system markedly decreased cellular Na^(+)accumulation in salinized transgenic roots(TRs)compared with adventitious roots.The overexpression of IbPSS1 enhanced salt-induced Na^(+)/H^(+)antiport activity and increased plasma membrane(PM)Ca^(2+)-permeable channel sensitivity to NaCl and H2O2 in the TRs.We confirmed the important role of IbPSS1 in improving salt tolerance in transgenic sweet potato lines obtained from an Agrobacterium tumefaciens-mediated transformation system.Similarly,compared with the wild-type(WT)plants,the transgenic lines presented decreased Na^(+)accumulation,enhanced Na^(+)exclusion,and increased PM Ca^(2+)-permeable channel sensitivity to NaCl and H2O2 in the roots.Exogenous application of lysophosphatidylserine triggered similar shifts in Na^(+)accumulation and Na^(+)and Ca^(2+)fluxes in the salinized roots of WT.Overall,this study provides an efficient and reliable transgenic method for functional genomic studies of sweet potato.Our results revealed that IbPSS1 contributes to the salt tolerance of sweet potato by enabling Na^(+)homeostasis and Na^(+)exclusion in the roots,and the latter process is possibly controlled by PS reinforcing Ca^(2+)signaling in the roots. | Yicheng Yu Ying Xuan Xiaofeng Bian Lei Zhang Zhiyuan Pan Meng Kou Qinghe Cao Zhonghou Tang Qiang Li Daifu Ma Zongyun Li Jian Sun | 2020 | Horticulture Research2020,7,1: | 2 |

| 10 | Effective ensemble learning approach for SST field prediction using attention-based PredRNN显示文摘Accurate prediction of sea surface temperature (SST) is extremely important for forecasting oceanic environmental events and for ocean studies. However, the existing SST prediction methods do not consider the seasonal periodicity and abnormal fluctuation characteristics of SST or the importance of historical SST data from different times;thus, these methods suffer from low prediction accuracy. To solve this problem, we comprehensively consider the effects of seasonal periodicity and abnormal fluctuation characteristics of SST data, as well as the influence of historical data in different periods, on prediction accuracy. We propose a novel ensemble learning approach that combines the Predictive Recurrent Neural Network(PredRNN) network and an attention mechanism for effective SST field prediction. In this approach, the XGBoost model is used to learn the long-period fluctuation law of SST and to extract seasonal periodic features from SST data. The exponential smoothing method is used to mitigate the impact of severely abnormal SST fluctuations and extract the a priori features of SST data. The outputs of the two aforementioned models and the original SST data are stacked and used as inputs for the next model, the PredRNN network. PredRNN is the most recently developed spatiotemporal deep learning network, which simulates both spatial and temporal representations and is capable of transferring memory across layers and time steps. Therefore, we used it to extract the spatiotemporal correlations of SST data and predict future SSTs. Finally, an attention mechanism is added to capture the importance of different historical SST data, weigh the output of each step of the PredRNN network, and improve the prediction accuracy. In this study, it is observed that the nitrogen fixation capacity of Klebsiella pneumoniae varies strongly when cells are grown on different carbohydrates, and this carbon effect occurs at the level of nif gene expression. Here we show that CRP can repress σ54-dependent nif promoters (nifB, nifE, nifF, nifH, nifJ, nifLA and nifU), in a cAMP dependent fashion, in closed related E. coll background. Sequence analysis of these nif promoters indicates that there is no direct correlation between the fold of CRP-cAMP-mediated inhibition and the upstream cis elements at the promoters. In addition, the crp gene of K. pneumoniae has been isolated and sequenced, which is structural and functional highly homologous to that of E. coli. Although promising, alternative heterogeneous catalysts with stable structure, strong hydrophilicity, environmental friendliness and excellent catalytic performance are highly desirable to facilitate the wide industrialization of PMS. In this work, Ni doped CoOOH catalyst was employed as PMS activator. Ni dopant had a significant influence on the morphology, structure and catalytic performance of CoOOH. NiojCoo.gOOH exhibited the best catalytic performance. Reaction rate ofNio.2Coo.8OOH was 2, 4, and 4.4 times that of CoOOH, CoFe2O4 and CO3O4, respectively. Moreover, Ni。2C00.8OOH/PMS system had potential application to organic pollutants and displayed a great catalytic activity over a broader pH value(e.g., 4-10). More importantly, Ni doping accelerated the transfonnation of Co(Ⅲ) and Co(Ⅱ) and formed active species CoOH^+ and NiOH^+ which were responsible to the enhancement of PMS activation.OH, SO4^-:O2^- and 1^O2 were detected, indicating both non-radical and radical processes in the Nio.2Coo.8OOH/PMS system.
