1CRISPR/Cas9-mediated PINK1 deletion leads to neurodegeneration in rhesus monkeys显示文摘Dear Editor,PINK1 mutations cause autosomal recessive and early-onset Parkinson’s disease(PD)with selective neurodegeneration.Unfortunately,current PINK1 knockout(KO)mouse1,2 and pig models3,4 are unable to recapitulate the selective and overt neurodegeneration seen in PD patients.Furthermore,endogenous Pink1 in the mouse brain is expressed at very low levels and can only be detected via immunoprecipitation,5 meaning that PINK1’s function in the mammalian brain needs to be assessed using larger animals that are closer to humans.We previously used CRISPR/Cas9 to target the monkey gene in one-cell stage embryos.6 Using the same approach,we designed two gRNAs to target exon 2(T1)and exon 4(T2),which encode a kinase domain in the PINK1 gene of rhesus monkeys(Fig.1a).CRISPR/Cas9 and PINK1 gRNAs were injected into one-cell stage rhesus monkey embryos.A T7E1 assay and sequencing of PCR products from the injected embryos showed high efficiency(61.5%)in targeting PINK1(Fig.1b and Supplementary information,Fig.S1).Transfer of 87 embryos to 28 surrogate rhesus monkeys resulted in 11 pregnancies(39.2%)(Fig.1b).Eleven fetuses developed to term and were born naturally.Of these live monkeys,eight carried PINK1 mutations(M),and three were wild type(WT).However,three mutant monkeys(M1,M3 and M4)were newborn triplets that struggled to survive and died 3-4 days after birth.One WT newborn monkey also died after a difficult labor.Another mutant monkey(M2)died 7 days after birth without noticeable warning signs or symptoms.The other three mutant monkeys(M6,M7 and M8)have lived for three years;M5,however,reduced its food intake and showed weakness at the age of 1.5 years,and died 30 days after anesthesia for MRI examination.Weili Yang Yunbo Liu Zhuchi Tu Chong Xiao Sen Yan Xishan Ma Xiangyu Guo Xiusheng Chen Peng Yin Zhengyi Yang Su Yang Tianzi Jiang Shihua Li Chuan Qin Xiao-Jiang Li 2019Cell Research2019,29,4:12
2Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. 4SnO_2 hollow spheres:Polymer bead-templated hydrothermal synthesis and their electrochemical properties for lithium storage显示文摘SnO2 hollow spheres have been synthesized via a facile hydrothermal method using sulfonated polystyrene beads as a template followed by a calcination process in air.X-ray diffraction,scanning electron microscopy,and transmission electron microscopy show that the as-obtained SnO2 hollow spheres have a wall thickness of about 50 nm,and consist of nanosized SnO2 particles with a mean diameter of about 15 nm.Electrochemical measurements indicate that the SnO2 hollow spheres exhibit improved electrochemical performance in terms of specific capacity and rate capability in comparison with commercial SnO2 when used as anode materials for lithium-ion batteries.The enhanced performance may be attributed to the spherical and hollow structure,as well as the building blocks of SnO2 nanoparticles.YIN YaXia XIN Sen WAN LiJun LI CongJu GUO YuGuo 2012Science China Chemistry2012,55,7:6

6New taxa of Berberis (Berberidaceae) with greenish flowers from a biodiversity hotspot in Sichuan Province, China显示文摘A new species,Berberis viridiflora X.H.Li,and a new variety,Berberis sanguinea Franch.var.viridisepala X.H.Li,L.C.Zhang & W.H.Li are described and illustrated from Baoxing County,a biodiversity hotspot located on the eastern edge of Hengduan Mountains in Sichuan Province,Southwest China,Both new taxa resemble B.sanguinea Franch.var.sanguinea,but B.viridiflora differs by the greenish flowers,and the petals being truncate,obtuse,or undulate at apex;while B.sanguinea var.viridisepala differs by the greenish or yellowish green flowers.Morphological features of the pollen grains of B.sanguinea and the two new taxa are revealed by scanning election microscope.B.sanguinea var.sanguinea displays obvious similarities with a sympatric congener,Berberis multiovula T.S.Ying in the morphology of flowers,stems and leaves,especially its ovule number varies greatly from 2 to 9,thus,B.multiovula characterized by the5-ovuled ovary is reduced to a synonymy of B.sanguinea var.sanguinea.On the basis of field surveys and study of herbarium specimens,a total of 16 species and varieties of Berberis are recorded from Baoxing County,and a key is provided.Among the 16 taxa,most phenotypic variations in Chinese Berberis can be displayed,including the habit of plants,most morphological variations of stems,branches and leaves,all types of inflorescences,all color types of flowers,and nearly all types of the shape and color of fruits.Baoxing County and its adjacent Sichuan Giant Panda Sanctuaries possibly constitute an active diversification center of Berberis in eastern Hengduan Mountains of China.Xinhua Li Licun Zhang Wenhui Li Xiaoming Yin Sen Yuan 2017Plant Diversity2017,39,2:3

8Start-up of the anammox process from the conventional activated sludge in a hybrid bioreactor显示文摘The anaerobic ammonium oxidation (anammox) process was successfully started up from conventional activated sludge using a hybrid bioreactor within 2 months.The average removal efficiencies of ammonia and nitrite were both over 80%,and the maximum total nitrogen removal rate of 1.85 kg N/(m 3 ·day) was obtained on day 362 with the initial sludge concentration of 0.7 g mixed liquor suspended solids (MLSS)/L.Scanning electron microscope (SEM) observation of the granular sludge in the hybrid reactor clearly showed a high degree of compactness and cell sphericity,and the cell size was quite uniform.Transmission electron microscope photos showed that cells were round or oval,the cellular diameter was 0.6-1.0 μm,and the percentage of the anammoxosome compartment was 51%-85% of the whole cell volume.Fluorescence in situ hybridization analysis (FISH) indicated that anammox bacteria became the dominant population in the community (accounting for more than 51% of total bacteria on day 250).Seven planctomycete 16S rRNA gene sequences were present in the 16S rRNA gene clone library generated from the biomass and affiliated to Candidatus Kuenenia stuttgartiensis and Candidatus Brocadia sp.,a new anammox species.In addition,the average effluent suspended solid (MLSS) concentrations of outlets I (above the non-woven carrier) and II (below the non-woven carrier) were 0.0009 and 0.0035 g/L,respectively.This showed that the non-woven carrier could catch the biomass effectively,which increased biomass and improved the nitrogen removal rate in the reactor.Xiumei Duan Jiti Zhou Sen Qiao Xin Yin Tian Tian Fangdi Xu 2012Journal of Environmental Sciences2012,24,6:3
9Clinical experience of percutaneous m itralballoon valvuloplastyin 350 casesin Chinese and long term follow-up显示文摘Objectives: This study is to introduce of the clinical experience of percutaneous mitral balloon valvuloplasty 350 cases in Chinese and the long term follow up. Method: The modfied Inoue method was performed. Results: Effective PBMV was performed in 344 cases, the success rate was 98.3%:mitral area assessed by 2 dimenrional echocardiography (1.11±0.29 to 2.19 ±0.40cm 2, P <0.01 ). One hundrad and five patients were followed at a mean (46.7± 26.3 ) months (range 9 months to 8.5 years). after procedure restenosis was 11.4%(12/15), death 2.9 %(3/105, cerebral embolism in 2, congestive heart failure in 1; mitral valve replacement in 3.8%(4/105). 10Hole quality in longitudinal–torsional coupled ultrasonic vibration assisted drilling of carbon fiber reinforced plastics显示文摘Carbon fiber reinforced plastic (CFRP) composites are extremely attractive in the manufacturing of structural and functional components in the aircraft manufacturing field due to their outstanding properties, such as good fatigue resistance, high specific stiffness/strength, and good shock absorption. However, because of their inherent anisotropy, low interlamination strength, and abrasive characteristics, CFRP composites are considered difficult-to-cut materials and are prone to generating serious hole defects, such as delamination, tearing, and burrs. The advanced longitudinal–torsional coupled ultrasonic vibration assisted drilling (LTC-UAD) method has a potential application for drilling CFRP composites. At present, LTC-UAD is mainly adopted for drilling metal materials and rarely for CFRP. Therefore, this study analyzes the kinematic characteristics and the influence of feed rate on the drilling performance of LTC-UAD. Experimental results indicate that LTC-UAD can reduce the thrust force by 39% compared to conventional drilling. Furthermore, LTC-UAD can decrease the delamination and burr factors and improve the surface quality of the hole wall. 11Water model experiments of multiphase mixing in the top-blown smelting process of copper concentrate显示文摘We constructed a 1:10 cold water experimental model by geometrically scaling down an Isa smelting furnace. The mixing processes at different liquid heights, lance diameters, lance submersion depths, and gas flow rates were subsequently measured using the conductivity method. A new criterion was proposed to determine the mixing time. On this basis, the quasi-equations of the mixing time as a function of different parameters were established. The parameters of the top-blown smelting process were optimized using high-speed photography. An excessively high gas flow rate or excessively low liquid height would enhance the fluctuation and splashing of liquid in the bath, which is unfavorable for material mixing. 13Density-functional-theory predictions of mechanical behaviour and thermal properties as well as experimental hardness of the Ga-bilayer Mo_(2)Ga_(2)C显示文摘Mo_(2)Ga_(2)C is a new MAX phase with a stacking Ga-bilayer as well as possible unusual properties.To understand this unique MAX phase structure and promote possible future applications,the structure,chemical bonding,and mechanical and thermodynamic properties of Mo_(2)Ga_(2)C were investigated by first-principles.Using the“bond stiffness”model,the strongest covalent bonding(1162 GPa)was formed between Mo and C atoms in Mo_(2)Ga_(2)C,while the weakest Ga–Ga(389 GPa)bonding was formed between two Ga-atomic layers,different from other typical MAX phases.The ratio of the bond stiffness of the weakest bond to the strongest bond(0.33)was lower than 1/2,indicating the high damage tolerance and fracture toughness of Mo_(2)Ga_(2)C,which was confirmed by indentation without any cracks.The high-temperature heat capacity and thermal expansion of Mo_(2)Ga_(2)C were calculated in the framework of quasi-harmonic approximation from 0 to 1300 K.Because of the metal-like electronic structure,the electronic excitation contribution became more significant with increasing temperature above 300 K.Xinxin QI Weilong YIN Sen JIN Aiguo ZHOU Xiaodong HE Guangping SONG Yongting ZHENG Yuelei BAI 2022Journal of Advanced Ceramics2022,11,2:2
14MYOD1 inhibits avian adipocyte differentiation via miRNA-206/KLF4 axis显示文摘Background:A considerable number of muscle development-related genes were differentially expressed in the early stage of avian adipocyte differentiation.However,the functions of them in adipocyte differentiation remain largely known.In this study,the myoblast determination protein 1(MYOD1)was selected as a representative of muscle development.We investigated its expression,function,and regulation in avian adipocyte differentiation.Results:The expression of MYOD1 decreased significantly in the early stage of avian adipocyte differentiation.CRIS PR/Cas9-mediated deletion of MYOD1 induced adipocyte differentiation,whereas over-expression of MYOD1 inhibited adipogenesis.The mRNA-seq data showed that MYOD1 could perturb the lipid biosynthetic process during differentiation.Our results showed that MYOD1 directly up-regulates the miR-206 expression by binding the upstream 1200 bp region of miR-206.Then,over-expression of miR-206 can inhibit the adipogenesis.Furthermore,MYOD1 affected the expression of endogenous miR-206 and its target gene Kruppel-like factor 4(KLF4),which is an important activator of adipogenesis.Accordingly,the inhibition of miR-206 or over-expression of KLF4 could counteract the inhibitory effect of MYOD1 on adipocyte differentiation.Conclusions:Our results establish that MYOD1 inhibits adipocyte differentiation by up-regulating miR-206 to suppress the KLF4 expression.These findings identify a novel function of MYOD1 in adipocyte differentiation,suggesting a potential role in body-fat distribution regulation.Zheng Wang Qiangsen Zhao Xiaoqin Li Zhongtao Yin Sirui Chen Sen Wu Ning Yang Zhuocheng Hou 2021Journal of Animal Science and Biotechnology2021,12,4:2

18The Inhibitory Effect of Endostatin and Doxycycline Administration on B16 Melanoma Angiogenesis and Cellular Proliferation显示文摘OBJECTIVE To investigate the effect of endostatin and doxycycline on melanoma cellular proliferation and tumor angiogenesis.METHODS The effects of endostatin and doxycycline were studied in mice transplanted with B16 melanoma cells.The mice were divided into 4 groups that were treated as follows:endostatin treatment(E group),doxycycline treatment(D group),endostatin plus doxycycline trearment(DE group),controls(C group) received no treatment.Following 9 days of treatment the tumor tissue was removed to compare the differences in the tumor necrotic rate and micro-vessel density(MVD) among the different groups.Immunohistochemical staining was conducted to detect the expression of proliferating cell nuclear antigen(PCNA)in the di?erent groups. RESULTS The MVD of the 3 experimental groups was significantly less than the control group,(F=10.888,P<0.05),indicating that doxycycline and endostatin can inhibit tumor angiogenesis by decreasing the tumor blood supply.This effect results in inhibition of tumor cellular proliferation and promotion of tumor cell necrosis.The tumor cell necrotic rate of the 3 experimental groups were all significantly higher than the C group(F=7.229,P<0.05) and the difference between the DE and C groups also was statistically significant.PCNA expression in all 3 experimental groups was statistically less than the C group(F=17.729,P<0.05).
