1U-Pb zircon dating of Early Paleozoic gabbro from the Nantinghe ophiolite in the Changning-Menglian suture zone and its geological implication显示文摘The Nantinghe ophiolite is located in the northern part of the Changning-Menglian suture zone in southeast Tibet. It is composed of meta-peridotite, cumulative gabbro, meta-gabbro, plagioclase amphibolite and meta-basalt. Zircon U-Pb dating of the cumulative gabbro gives concordant ages of 473.0±3.8 Ma and 443.6±4.0 Ma respectively, indicating the early and late episodes of mafic magmatisms during the Paleo-Tethys oceanic rifting. The 16 LA-ICPMS zircon U-Pb analyses of meta-gabbro yield a weight mean age of 439±2.4Ma. The gabbro shows relatively low contents of SiO2 (46.46%-52.11%), TiO2 (0.96%-1.14%) and K2O (0.48%-0.75%). Its trace element distribution patterns are partly similar to those of the mid-ocean ridge basalts, and part is depleted in high field strength elements such as Nb, Ta, Zr, Hf and Ti. These features suggest that the mafic rocks were probably formed in a MORB-like or backarc rift basin setting. The zircon U-Pb age of gabbro is consistent with a late crystallization age of the cumulative gabbro from the Nantinghe ophiolite, suggesting that the Paleo-Tethys oceanic basin was opened during 444-439 Ma, possibly as a backarc basin. It is the first precise age which defines the formation time of the early Paleozoic ophiolite in the Changning-Menglian suture zone. These geochronological and geochemical characteristics of the Nantinghe ophiolite are consistent with those from the Guoganjianianshan and Taoxinghu of the Longmu Co-Shuanghu suture in the Qiangtang region. 2Washed microbiota transplantation vs.manual fecal microbiota transplantation:clinical findings,animal studies and in vitro screening显示文摘Fecal microbiota transplantation(FMT)by manual preparation has been applied to treat diseases for thousands of years.However,this method still endures safety risks and challenges the psychological endurance and acceptance of doctors,patients and donors.Population evidence showed the washed microbiota preparation with microfiltration based on an automatic purification system followed by repeated centrifugation plus suspension for three times significantly reduced FMT-related adverse events.This washing preparation makes delivering a precise dose of the enriched microbiota feasible,instead of using the weight of stool.Intraperitoneal injection in mice with the fecal microbiota supernatant obtained after repeated centrifugation plus suspension for three times induced less toxic reaction than that by the first centrifugation following the microfiltration.The toxic reactions that include death,the change in the level of peripheral white blood cells,and the proliferation of germinal center in secondary lymphoid follicles in spleen were noted.The metagenomic next-generation sequencing(NGS)indicated the increasing types and amount of viruses could be washed out during the washing process.Metabolomics analysis indicated metabolites with pro-inflammatory effects in the fecal microbiota supernatant such as leukotriene B4,corticosterone,and prostaglandin G2 could be removed by repeated washing.Near-infrared absorption spectroscopy could be served as a rapid detection method to control the quality of the washingprocess.In conclusion,this study for the first time provides evidence linking clinical findings and animal experiments to support that washed microbiota transplantation(WMT)is safer,more precise and more quality-controllable than the crude FMT by manual.Ting Zhang Gaochen Lu Zhe Zhao Yafei Liu Quan Shen Pan Li Yaoyao Chen Haoran Yin Huiquan Wang Cicilia Marcella Bota Cui Lei Cheng Guozhong Ji Faming Zhang 2020Protein & Cell2020,11,4:44

5Caspase-11/4 and gasdermin D-mediated pyroptosis contributes to podocyte injury in mouse diabetic nephropathy显示文摘Diabetic nephropathy(DN)is characterized by sterile inflammation with continuous injury and loss of renal inherent parenchyma cells.Podocyte is an essential early injury target in DN.The injury and loss of podocytes are closely associated with proteinuria,the early symptom of renal injury in DN.However,the exact mechanism for podocyte injury and death in DN remains ambiguous.In this study we investigated whether pyroptosis,a newly discovered cell death pathway was involved in DN.Diabetic mice were generated by high-fat diet/STZ injections.We showed that the expression levels of caspase-11 and cleavage of gasdermin D(GSDMD-N)in podocytes were significantly elevated,accompanied by reduced expression of podocyte makers nephrin and podocin,loss and fusion in podocyte foot processes,increased inflammatory cytokines NF-κB,IL-1β,and IL-18,macrophage infiltration,glomerular matrix expansion and increased urinary albumin to creatinine ratio(UACR).All these changes in diabetic mice were blunted by knockout of caspase-11 or GSDMD.Cultured human and mouse podocytes were treated with high glucose(30 mM),which significantly increased the expression levels of caspase-11 or caspase-4(the homolog of caspase-11 in human),GSDMD-N,NF-κB,IL-1β,and IL-18,and decreased the expression of nephrin and podocin.Either caspase-4 or GSDMD knockdown by siRNA significantly blunted these changes.In summary,our results demonstrate that caspase-11/4 and GSDMD-mediated pyroptosis is activated and involved in podocyte loss under hyperglycemia condition and the development of DN.Qian Cheng Jing Pan Zhuan-li Zhou Fan Yin Hong-yan Xie Pan-pan Chen Jing-yao Li Pei-qing Zheng Li Zhou Wei Zhang Jun Liu Li-min Lu 2021Acta Pharmacologica Sinica2021,42,6:33
6Roles of Chemokine Receptor 4 (CXCR4) and Chemokine Ligand 12 (CXCL12) in Metastasis of Hepatocellular Carcinoma Cells显示文摘Chemokines are involved in human hepatocellular carcinoma (HCC) carcinogenesis. However, the exact mechanism of chemokines in HCC carcinogenesis remains unknown. Here we investigated the roles of chemokine receptor 4 (CXCR4) and chemokine ligand 12 (CXCL12) in the metastasis of HCC. We found that the expression levels of CXCR4 mRNA in HCC tissues, MHCC97 cells, and HUVEC cells were 2.52 ± 1.13, 2.34 ± 1.16 and 1.63 ± 1.26, respectively and that the CXCR4 protein levels were 1.38 ± 0.13, 1.96 ± 0.32 and 1.86 ± 0.21, respectively. In contrast, CXCR4 was not detected in normal hepatic tissues. In 78 HCC patients, we also found that the concentration of CXCL12 in cancerous ascitic fluid was 783-8,364 pg/ml and that CXCL12 mRNA level in HCC metastasis portal lymph nodes was 1.21 ± 0.87 but undetectable in normal hepatic tissues. Finally we discovered that recombinant human CXCL12 could induce MHCC97 cells and HUVEC cells to migrate with chemotactic indexes (CI) of 3.9 ± 1.1 and 4.1 ± 1.6, respectively. Cancerous ascitic fluid could also induce the migration of MHCC97 cells with a CI of 1.9 ± 0.8. 10Analysis of the Fungal Community in Apple Replanted Soil Around Bohai Gulf显示文摘Apple replant disease(ARD) is a frequently occurring plant disease in replanted orchards around Bohai Gulf, which causes growth inhibition and even death of plants. The aim of this study was to investigate the etiology of ARD around Bohai Gulf. In this study, the primary growth inhibition of apple seedlings was evaluated in ten replanted soils, sampled around Bohai Gulf. A fungal clone library was used to identify changes in the structure and composition of the soil fungal community. The results revealed that the Simpson diversity indices of Laizhou and Pulandian orchards were higher than others, presenting severe ARD. Ascomycota dominated around Bohai Gulf at the phyla level. Fusarium and Saccharomyces were abundant in all replanted soils. In addition, correlations between the relative abundance of fungal genera in soils and the severity of ARD were analyzed. The results showed that Fusarium was correlated positively with the severity of ARD, but Mortierella was negatively correlated. Furthermore, the quantitative PCR of Fusarium oxysporum, which was regarded as a factor of ARD, was performed. 11Soil Erosion Changes over the Past Five Decades in the Red Soil Region of Southern China显示文摘This paper reports the dynamic changes of soil and water loss in the red soil region of Southern China since the 1950s. The red soil region covers eight provinces: Jiangxi, Zhejiang, Fujian, Anhui, Hubei, Hunan, Guangdong and Hainan. From the 1950s to 1986, the annual rate of soil erosion increased by 3.4%. From 1986 to 1996 and from 1996 to 2000, the annual rates of soil erosion decreased by 2.0% and 0.32%, respectively. Field surveys showed that from 2000 to 2005, the area of soil and water loss decreased annually by 1.2%. This decrease was a result of large-scale erosion control activities across China. Although the eroded soil has been restored, the restoration process is very slow and full restoration will take a long time. 14Prospect of the QTL-qSB-9^(Tq) utilized in molecular breeding program of japonica rice against sheath blight显示文摘The major QTL-qSB-9Tq conferring partial resistance to rice (Oryza sativa L.) sheath blight (Rhizoctonia solani Kühn) has been verified on chromosome 9 of the indica rice cultivar, Teqing. In this study, the prospect of this QTL utilized in molecular breeding program of japonica rice for sheath blight resistance was investigated. Most of the japonica rice cultivars showed lower level of sheath blight resistance than the indica rice cultivars. At the corresponding site of qSB-9Tq, nine typical japonica rice cultivars from different ecological regions or countries proved to possess the susceptible allele(s). Introgression of qSB-9Tq into these cultivars enhanced their resistance level by decreasing sheath blight score of 1.0 (0.5–1.3), which indicated that qSB-9Tq had a large potential in strengthening the resistance of japonica rice to sheath blight. 15Cropland redistribution to marginal lands undermines environmental sustainability显示文摘Cropland redistribution to marginal land has been reported worldwide;however,the resulting impacts on environmental sustainability have not been investigated sufficiently.Here we investigated the environmental impacts of cropland redistribution in China.As a result of urbanization-induced loss of high-quality croplands in south China(~8.5 t ha^(-1)),croplands expanded to marginal lands in northeast(~4.5 t ha^(-1)) and northwest China(~2.9 t ha^(-1)) during 1990-2015 to pursue food security.However,the reclamation in these low-yield and ecologically vulnerable zones considerably undermined local environmental sustainability,for example increasing wind erosion(+3.47%),irrigation water consumption(+34.42%),fertilizer use(+20.02%) and decreasing natural habitats(-3.11%).Forecasts show that further reclamation in marginal lands per current policies would exacerbate environmental costs by 2050.The future cropland security risk will be remarkably intensified because of the conflict between food production and environmental sustainability.Our research suggests that globally emerging reclamation of marginal lands should be restricted and crop yield boost should be encouraged for both food security and environmental benefits.Wenhui Kuang Jiyuan Liu Hanqin Tian Hao Shi Jinwei Dong Changqing Song Xiaoyong Li Guoming Du Yali Hou Dengsheng Lu Wenfeng Chi Tao Pan Shuwen Zhang Rafiq Hamdi Zherui Yin Huimin Yan Changzhen Yan Shixin Wu Rendong Li Jiuchun Yang Yinyin Dou Wenbin Wu Liqiao Liang Bao Xiang Shiqi Yang 2022National Science Review2022,9,1:13
