| 2 | Anti-SARS-CoV-2 activities in vitro of Shuanghuanglian preparations and bioactive ingredients显示文摘Human infection with severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)causes coronavirus disease 2019(COVID-19)and there is no cure currently.The 3CL protease(3CLpro)is a highly conserved protease which is indispensable for CoVs replication,and is a promising target for development of broad-spectrum antiviral drugs.In this study we investigated the anti-SARS-CoV-2 potential of Shuanghuanglian preparation,a Chinese traditional patent medicine with a long history for treating respiratory tract infection in China.We showed that either the oral liquid of Shuanghuanglian,the lyophilized powder of Shuanghuanglian for injection or their bioactive components dose-dependently inhibited SARS-CoV-23CLpro as well as the replication of SARS-CoV-2 in Vero E6 cells.Baicalin and baicalein,two ingredients of Shuanghuanglian,were characterized as the first noncovalent,nonpeptidomimetic inhibitors of SARS-CoV-23CLpro and exhibited potent antiviral activities in a cell-based system.Remarkably,the binding mode of baicalein with SARS-CoV-23CLpro determined by X-ray protein crystallography was distinctly different from those of known 3CLpro inhibitors.Baicalein was productively ensconced in the core of the substrate-binding pocket by interacting with two catalytic residues,the crucial S1/S2 subsites and the oxyanion loop,acting as a“shield”in front of the catalytic dyad to effectively prevent substrate access to the catalytic dyad within the active site.Overall,this study provides an example for exploring the in vitro potency of Chinese traditional patent medicines and effectively identifying bioactive ingredients toward a specific target,and gains evidence supporting the in vivo studies of Shuanghuanglian oral liquid as well as two natural products for COVID-19 treatment. | Hai-xia Su Sheng Yao Wen-feng Zhao Min-jun Li Jia Liu Wei-juan Shang Hang Xie Chang-qiang Ke Hang-chen Hu Mei-na Gao Kun-qian Yu Hong Liu Jing-shan Shen Wei Tang Lei-ke Zhang Geng-fu Xiao Li Ni Dao-wen Wang Jian-ping Zuo Hua-liang Jiang Fang Bai Yan Wu Yang Ye Ye-chun Xu | 2020 | Acta Pharmacologica Sinica2020,41,9: | 32 |

| 5 | Inhibitive effects of anti-oxidative vitamins on mannitol-induced apoptosis of vascular endothelial cells显示文摘Objective: Study blood vessel injury and gene expression indicating vascular endothelial cell apoptosis induced by mannitol with and without administration of anti-oxidative vitamins. Methods: Healthy rabbits were randomly divided into four groups. Mannitol was injected into the vein of the rabbit ear in each animal. Pre-treatment prior to mannitol injection was per- formed with normal saline (group B), vitamin C (group C) and vitamin E (group D). Blood vessel injury was assessed under electron and light microscopy. In a second experiment, cell culture specimen of human umbilical vein endothelial cells were treated with mannitol. Pre-treatment was done with normal saline (sample B), vitamin C (sample C) and vitamin E (sample D). Total RNA was extracted with the original single step procedure, followed by hybridisation and analysis of gene expression. Results: In the animal experiment, serious blood vessel injury was seen in group A and group B. Group D showed light injury only, and normal tissue without pathological changes was seen in group C. Of all 330 apoptosis-related genes analysed in human cell culture specimen, no significant difference was seen after pre-treatment with normal saline, compared with the gene chip without pre-treatment. On the gene chip pre-treated with vitamin C, 45 apoptosis genes were down-regulated and 34 anti-apoptosis genes were up-regulated. Pre-treatment with vitamin E resulted in the down-regulation of 3 apoptosis genes. | 6 | Randomized Latent Factor Model for High-dimensional and Sparse Matrices from Industrial Applications显示文摘Latent factor(LF) models are highly effective in extracting useful knowledge from High-Dimensional and Sparse(HiDS) matrices which are commonly seen in various industrial applications. An LF model usually adopts iterative optimizers,which may consume many iterations to achieve a local optima,resulting in considerable time cost. Hence, determining how to accelerate the training process for LF models has become a significant issue. To address this, this work proposes a randomized latent factor(RLF) model. It incorporates the principle of randomized learning techniques from neural networks into the LF analysis of HiDS matrices, thereby greatly alleviating computational burden. | 9 | Origin, Flow of Formation Water and Hydrocarbon Accumulation in the Zhenwu Area of the North Jiangsu Basin, China显示文摘In order to understand the origin and flow of formation water and to evaluate the hydrocarbon accumulation and preservation conditions, the properties of formation water chemistry and dynamics of the Zhenwu area in the southern Gaoyou Sag, North Jiangsu Basin, China, have been investigated. The results show that Xuzhuang oilfield is infiltrated discontinuously by meteoric water under gravity, which consequently leads to the desalination of formation water. Formation water in the Zhenwu and Caozhuang oilfields is less influenced by meteoric water infiltration, and the origin is interpreted to be connate water. Hydrocarbon migration, accumulation and preservation are closely related to the hydrodynamic field of formation water. Formation water concentrates gradually during the process of centrifugal flow released by mudstone compaction and the centripetal flow of meteoric water infiltration, leading to the high salinity of the central part. The geological conditions of the southern fault-terrace belt are poor for hydrocarbon accumulation and preservation as meteoric water infiltration, leaching and oxidation, while the central part, i.e., northern Zhenwu and Caozhuang oilfields is beneficial for an abundance of hydrocarbon accumulation. | 10 | Microbial diversity accumulates in a downstream direction in the Three Gorges Reservoir显示文摘Organic and inorganic materials migrate downstream and have important roles in regulating environmental health in the river networks.However,it remains unclear whether and how a mixture of materials(i.e.,microbial species)from various upstream habitats contribute to microbial community coalescence upstream of a dam.Here we track the spatial variation in microbial abundance and diversity in the Three Gorges Reservoir based on quantitative PCR and 16 S rRNA gene high-throughput sequencing data.We further quantitatively assess the relative contributions of microbial species from mainstem,its tributaries,and the surrounding riverbank soils to the area immediately upstream of the Three Gorges Dam(TGD).We found an increase of microbial diversity and the convergent microbial distribution pattern in areas immediately upstream of TGD,suggesting this area become a new confluence for microbial diversity immigrating from upstream.Indeed,the number of shared species increased from upstream to TGD but unique species decreased,indicating immigration of various sources of microbial species overwhelms local environmental conditions in structuring microbial community close to TGD.By quantifying the sources of microbial species close to TGD,we found little contribution from soils as compared to tributaries,especially for sites closer to TGD,suggesting tributary microbes have greater influence on microbial diversity and environmental health in the Three Gorges Reservoir.Collectively,our results suggest that tracking microbial geographic origin and evaluating accumulating effects of microbial diversity shed light on the ecological processes in microbial communities and provide information for regulating aquatic ecological health. | Shang Wang Weiguo Hou Hongchen Jiang Liuqin Huang Hailiang Dong Shu Chen Bin Wang Yongcan Chen Binliang Lin Ye Deng | 2021 | Journal of Environmental Sciences2021,33,3: | 7 |
| 11 | Pore throat characteristics of tight sandstone of Yanchang Formation in eastern Gansu,Ordos Basin显示文摘An important factor to evaluate reservoir quality is the pore-throat size.However,the strong heterogeneity makes it difficult to characterize the pore-throat distribution in tight reservoirs.The field emission scanning electron microscope(FESEM),high pressure mercury injection and rate-controlled mercury injection are used to investigate the pore-throat size distribution in tight sandstone reservoirs of Member 7 of the Yanchang Formation in eastern Gansu,Ordos Basin,and studies of the pore throat size controlling on physical property of the tight sandstone reservoirs are also carried out.The result shows that the pore type is mainly dominated by the residual intergranular pore,dissolution pore,micropore and a few micro-fractures;the high-pressure mercury injection experiment indicates that the pore-throat size ranges from 0.0148 μm to 40mm,the pore throat more than 1 mm is less;the ratecontrolled mercury injection experiment reveals that for samples with different physical properties,the pore radius mainly varies from 80 μm to 350 μm;the throat radius exhibits the strong heterogeneity,and is from 0.12 μm to 30μm;the pore-throat size can be effectively characterized by combination of high-pressure and rate-controlled mercury injections,and it varies from 0.0148 μm to 350 μm.The permeability is mainly controlled by the large pore throat(>R_(50))which accounts for a small proportion;in the tight sandstone with the permeability greater than 0.1 mD,the permeability is mainly controlled by the micropore and mesopore;in the tight sandstone with the permeability smaller than 0.1 mD,the permeability is mainly controlled by the nanopore and micropore;the proportion of small pore throat increases with reduction of permeability,it is important that the small pore throat influences the reservoir storage property though its effect on permeability are small. | Hao Wu Chunlin Zhang Youliang Ji Rui'e Liu Shang Cao Sheng Chen Yunzhao Zhang Ye Wang Wei Du Gang Liu | 2018 | Petroleum Research2018,3,1: | 7 |

| 14 | A more consistent intraluminal rhesus monkey model of ischemic stroke显示文摘Endovascular surgery is advantageous in experimentally induced ischemic stroke because it causes fewer cranial traumatic lesions than invasive surgery and can closely mimic the pathophysiology in stroke patients. However, the outcomes are highly variable, which limits the accuracy of evaluations of ischemic stroke studies. In this study, eight healthy adult rhesus monkeys were randomized into two groups with four monkeys in each group: middle cerebral artery occlusion at origin segment(M1) and middle cerebral artery occlusion at M2 segment. The blood flow in the middle cerebral artery was blocked completely for 2 hours using the endovascular microcoil placement technique(1 mm × 10 cm)(undetachable), to establish a model of cerebral ischemia. The microcoil was withdrawn and the middle cerebral artery blood flow was restored. A reversible middle cerebral artery occlusion model was identified by hematoxylin-eosin staining, digital subtraction angiography, magnetic resonance angiography, magnetic resonance imaging, and neurological evaluation. The results showed that the middle cerebral artery occlusion model was successfully established in eight adult healthy rhesus monkeys, and ischemic lesions were apparent in the brain tissue of rhesus monkeys at 24 hours after occlusion. The rhesus monkeys had symptoms of neurological deficits. Compared with the M1 occlusion group, the M2 occlusion group had lower infarction volume and higher neurological scores. These experimental findings indicate that reversible middle cerebral artery occlusion can be produced with the endovascular microcoil technique in rhesus monkeys. In vitro transcription RNA from pSVOK12 transfected IBRS-2 cells and the re- covered virus RNA were isolated and sequenced, then indirect hemagglutination test, indirect im- munofluorescence assays, eleectron microscope test, 50% tissue culture infecting dose (TCID50) assays and mouse virulence studies were performed to study the antigenicity and virulence of the recovered virus. The result showed that the infectious clones we ob- tained and the virus derived from pSVOK12 had the same biological properties as the parental strain HK/70. | 16 | Prevalence of late-onset hypogonadism among middle-aged and elderly males in China:results from a national survey显示文摘This study aimed to propose an operational definition of late-onset hypogonadism(LOH)that incorporates both clinical symptoms and serum testosterone measurements to evaluate the prevalence of LOH in aging males in China.A population-based sample of 6296 men aged 40 years-79 years old was enrolled from six representative provinces in China.Serum total testosterone(TT),sex hormone-binding globulin(SHBG),and luteinizing hormone(LH)were measured and free testosterone(cFT)was calculated.The Aging Males’Symptoms(AMS)scale was used to evaluate the LOH symptoms.Finally,5078 men were included in this analysis.The TT levels did not decrease with age(P=0.59),and had no relationship with AMS symptoms(P=0.87 for AMS total score,P=0.74 for≥3 sexual symptoms).The cFT levels decreased significantly with age(P<0.01)and showed a negative association with the presence of≥3 sexual symptoms(P=0.03).The overall estimated prevalence of LOH was 7.8%(395/5078)if a cFT level<210 pmol l−1 combined with the presence of≥3 sexual symptoms was used as the criterion of LOH.Among them,26.1%(103/395)and 73.9%(292/395)had primary and secondary hypogonadism,respectively.After adjustment for confounding factors,primary and secondary hypogonadism was positively related to age and comorbidities.Body mass index was an independent risk factor for secondary hypogonadism.The results suggest that the AMS total score is not an appropriate indicator for decreased testosterone,and that the cFT level is more reliable than TT for LOH diagnosis.Secondary hypogonadism is the most common form of LOH. | Yi-Jun Liu Xu-Bo Shen Na Yu Xue-Jun Shang Yi-Qun Gu Lian-Dong Zuo Cheng-Liang Xiong Zhen Ye Yuan-Zhong Zhou | 2021 | Asian Journal of Andrology2021,23,2: | 6 |
| 17 | Creation of Transgenic Bananas Expressing Human Lysozyme Gene for Panama Wilt Resistance显示文摘Human lysozyme (HL) inhibits Fusarium oxysporum (FocR4) growth in vitro. To obtaintransgenic bananas (Musa spp.) that are resistant to Panama wilt (F. oxysporum), we introduced an HL genethat is driven by a constitutive cauliflower mosaic virus 35S promoter into the banana via Agrobacterium-mediated transformation. PCR confirmed that 51 transgenic plants were obtained. The development ofPanama wilt symptoms were examined after the plants had been grown in pots. The non-transgenic plantsdeveloped typical fusarium symptoms 60 d after FocR4 inoculation, whereas 24 of 51 transgenic plants remained healthy. The transgenic banana plants that showed resistance to FocR4 in the pots were then planted in a field that was heavily infected with FocR4 for further investigation. Eleven of 24 plants developed symptoms before bud emergence; another 11 plants showed symptoms after bud emergence and the remaining two plants, H-67 and H-144, remained healthy and were able to fruit. Northern blotting analysisdemonstrated that H-67 and H-144, bearing the strongest resistance to Panama wilt, had the highest level ofHL expression and that the expression of HL was well correlated with the FocR4 resistance of transgenicplants. | 20 | Polycystic kidney and hepatic disease 1 gene mutations in von Meyenburg complexes: Case report显示文摘Von Meyenburg complexes(VMCs) are a rare type of ductal plate malformation. We herein report two Chinese families with VMCs, and the suspicious gene mutation of this disease. Proband A was a 62-year-old woman with abnormal echographic presentation of the liver. She received magnetic resonance imaging(MRI) examination and liver biopsy, and the results showed she had VMCs. Histologically proved hepatocellular carcinoma was found 1 year after the diagnosis of VMCs. Proband B was a 57-year-old woman with intrahepatic diffuselesions displayed by abdominal ultrasonography. Her final diagnoses were VMCs, congenital hepatic fibrosis, and hepatitis B surface e antigen-negative chronic hepatitis B after a series of examinations. Then, all the family members of both proband A and proband B were screened for VMCs by MRI or ultrasonography. The results showed that four of the 11 family members from two families, including two males and two females, were diagnosed with VMCs. DNA samples were extracted from the peripheral blood of those 11 individuals of two VMCs pedigrees and subjected to polymerase chain reaction amplification of the polycystic kidney and hepatic disease 1(PKHD1) gene. Two different mutation loci were identified. Heterozygous mutations located in exon 32(c.4280 delG, p.Gly1427 ValfsX 6) in family A and exon 28(c.3118 C>T, p.Arg1040 Ter) in family B were detected.
