3Expression changes and bioinformatic analysis of Wallerian degeneration after sciatic nerve injury in rat显示文摘Wallerian degeneration (WD) remains an important research topic. Many genes are differentially expressed during the process of WD, but the precise mechanisms responsible for these differentiations are not completely understood. In this study, we used microarrays to analyze the expression changes of the distal nerve stump at 0, 1, 4, 7, 14, 21 and 28 days after sciatic nerve injury in rats. The data revealed 6 076 differentially-expressed genes, with 23 types of expression, specifically enriched in genes associated with nerve development and axonogenesis, cytokine biosynthesis, cell differentiation, cytokine/chemokine production, neuron differentiation, cytokinesis, phosphorylation and axon regeneration. Kyoto Encyclopedia of Genes and Genomes pathway analysis gave findings related mainly to the MAPK signaling pathway, the Jak-STAT signaling pathway, the cell cycle, cytokine-cytokine receptor interaction, the p53 signaling pathway and the Wnt signaling pathway. Some key factors were NGF, MAG, CNTF, CTNNA2, p53, JAK2, PLCB1, STAT3, BDNF, PRKC, collagen II, FGF, THBS4, TNC and c-Src, which were further validated by real-time quantitative PCR, Western blot, and immunohistochemistry. 4Ultrathin Al2O3-coated reduced graphene oxide membrane for stable lithium metal anode显示文摘Lithium(Li) metal has been considered as the most attractive anode materials for Li-ion batteries(LIBs)due to its high theoretic specific capacity. The formation of unstable solid electrolyte interphase(SEI) and dendritic Li on the metal anode, however, hindered its practical application. Herein, to address the issues, a Li-free electrode with ultrathin Al_2O_3 coated on reduced graphene oxide(rGO) membrane that covers a Cu foil current collector was developed. The composite electrode exhibits excellent interfacial protection of lithium metal deposited between Cu foil and rGO electrochemically. Firstly, it affords good Li^+ permeability from the electrolyte. Secondly, the ultrathin Al_2O_3 has sufficient mechanical strength to inhibit the penetration of Li dendrite. 5Prevalence and risk factors of active pulmonary tuberculosis among elderly people in China:a population based cross-sectional study显示文摘Background:The problem of population aging is a critical public health concern in modern China,and more tuberculosis(TB)control efforts are needed to reach elderly people at high priority.In this study,we aim to determine the prevalence and identify the risk factors of TB among elderly people in China.Methods:A multistage cluster-sampled cross-sectional survey was conducted in 2013,and 27 clusters were selected from 10 counties of 10 provinces in China.All consenting participants greater than or equal to 65 years of age were screened for pulmonary TB with a chest X-ray(CXR)and a symptom questionnaire.Three sputum specimens for bacteriological examination by microscopy and culture were collected from those whose screening was positive.Prevalence was calculated,a multiple logistic regression model was performed to confirm the risk factors,and population attributable fraction(PAF)of each risk factor was calculated to indicate the public health significance.Results:Of 38888 eligible people from 27 clusters,34269 participants finished both questionnaire and physical examination.There were 193 active pulmonary TB cases,62 of which were bacteriologically confirmed.The estimated prevalence of active pulmonary TB and bacteriologically confirmed TB in those 65 years of age and older was 563.19 per 100000(95%CI:483.73-642.65)and 180.92 per 100000(95%CI:135.89-225.96),respectively.Male sex,older age,living in rural areas,underweight,diabetes,close contact of pulmonary TB(PTB)and previous TB history are all risk factors for TB.The risk of TB increased with increasing age and decreasing body mass index(BMI)after adjusting for other factors,and there is a positive dose-response relationship.Conclusions:In China,active case finding(ACF)could be implemented among elderly people aged 65 and above with underweight,diabetes,close contact history and previous TB history as a priority,which will get significant yields and be cost-effective.Can-You Zhang Fei Zhao Yin-Yin Xia Yan-Ling Yu Xin Shen Wei Lu Xiao-Meng Wang Jin Xing Jian-Jun Ye Jian-Wei Li Fei-Ying Liu Jian-Lin Wu Lin Xu Hui Zhang Jun Cheng Li-Xia Wang 2019Infectious Diseases of Poverty2019,8,1:9
6Association of HER2 status with prognosis in gastric cancer patients undergoing R0 resection: A large-scale multicenter study in China显示文摘AIM: To determine whether the positive status of human epidermal growth receptor 2(HER2) can be regarded as an effective prognostic factor for patients with gastric cancer(GC) undergoing R0 resection.METHODS: A total of 1562 GC patients treated by R0 resection were recruited. HER2 status was evaluated in surgically resected samples of all the patients using immunohistochemical(IHC) staining. Correlations between HER2 status and clinicopathological characteristics were retrospective analyzed. Hazard ratios(HRs) and 95% confidence intervals(CIs) were estimated using Cox proportional hazard model, stratified by age, gender, tumor location and tumor-nodemetastasis(TNM) stage, with additional adjustment for potential prognostic factors.RESULTS: Among 1562 patients, 548(positive rate = 35.08%, 95%CI: 32.72%-37.45%) were HER2 positive. Positive status of HER2 was significantly correlated with gender(P = 0.004), minority(P < 0.001), tumor location(P = 0.001), pathological grade(P < 0.001), TNM stage(P < 0.001) and adjuvant radiotherapy(74.67% vs 23.53%, P = 0.011). No significant associations were observed between HER2 status and disease free survival(HR = 0.19, 95%CI: 0.96-1.46, P = 0.105) or overall survival(HR = 1.19, 95%CI: 0.96-1.48, P = 0.118) using multivariate analysis, although stratified analyses showed marginally statistically significant associations both in disease free survival and overall survival, especially among patients aged < 60 years or with early TNM stages(Ⅰ and Ⅱ). Categorical age, TNM stage, neural invasion, and adjuvant chemotherapy were, as expected, independent prognostic factors for both disease free survival and overall survival. 7Receptome profiling identifies KREMEN1 and ASGR1 as alternative functional receptors of SARS-CoV-2显示文摘Host cellular receptors play key roles in the determination of virus tropism and pathogenesis.However,little is known about SARS-CoV-2 host receptors with the exception of ACE2.Furthermore,ACE2 alone cannot explain the multi-organ tropism of SARS-CoV-2 nor the clinical differences between SARS-CoV-2 and SARS-CoV,suggesting the involvement of other receptor(s).Here,we performed genomic receptor profiling to screen 5054 human membrane proteins individually for interaction with the SARS-CoV-2 capsid spike(S)protein.Twelve proteins,including ACE2,ASGR1,and KREMEN1,were identified with diverse S-binding affinities and patterns.ASGR1 or KREMEN1 is sufficient for the entry of SARS-CoV-2 but not SARS-CoV in vitro and in vivo.SARS-CoV-2 utilizes distinct ACE2/ASGR1/KREMEN1(ASK)receptor combinations to enter different cell types,and the expression of ASK together displays a markedly stronger correlation with virus susceptibility than that of any individual receptor at both the cell and tissue levels.The cocktail of ASK-related neutralizing antibodies provides the most substantial blockage of SARS-CoV-2 infection in human lung organoids when compared to individual antibodies.Our study revealed an interacting host receptome of SARS-CoV-2,and identified ASGR1 and KREMEN1 as alternative functional receptors that play essential roles in ACE2-independent virus entry,providing insight into SARS-CoV-2 tropism and pathogenesis,as well as a community resource and potential therapeutic strategies for further COVID-19 investigations.Yunqing Gu Jun Cao Xinyu Zhang Hai Gao Yuyan Wang Jia Wang Juan He Xiaoyi Jiang Jinlan Zhang Guanghui Shen Jie Yang Xichen Zheng Gaowei Hu Yuanfei Zhu Shujuan Du Yunkai Zhu Rong Zhang Jianqing Xu Fei Lan Di Qu Guoliang Xu Yun Zhao Dong Gao Youhua Xie Min Luo Zhigang Lu 2022Cell Research2022,32,1:8

9Nitazoxanide,an anti-parasitic drug,efficiently ameliorates learning and memory impairments in AD model mice显示文摘The pathogenesis of Alzheimer's disease(AD)is characterized by both accumulation ofβ-amyloid(AB)plaque and formation of neurofirillary tangles in the brain.Recent evidence shows that autophagy activation may potently promote intraellular Aβclearance.Thus targeting autophagy becomes a promising strategy for discovery of drug leads against AD.In the present study,we established a platform to discover autophagy stimulator and screened the lab in-house FDA-approved drug library.We found thatanti-parasitic drug nitazoxanide(NTZ)was an autophagy activator and could efficiently improve learning and memory impairmentsin APP/PS1 transgenic mice.In BV2 cells and primary cortical astrocytes,NTZ stimulated autophagy and promoted Aβclearance by inhibiting both P12K/AKT/mTOR/ULK1 and NQO1/mTOR/ULK1 signaling pathways;NTZ treatment attenuated LPS-induced inflammation by inhibiting PI3K/AKT/IKB/NFKB signaling.In SH-SY5Y cells and primary cortical neurons,NTZ treatment restrained tau hyperphosphorylation through inhibition of PI3K/AKT/GSK3βpathway.The beneficial effects and related signaling mechanisms from the in vitro studies were also observed in APP/PS1 transgenic mice following administration of NTZ(90 mg·kg^-1d,ig)for100 days.Furthermore,NTZ administration decreased Aβlevel and senile plaque formation in the hippocampus and cerebral cortexof APP/PS1 transgenic mice,and improved learning and memory impairments in Morris water maze assay.In conclusion,our results highlight the potential of NTZ in the treatment of AD.Lei Fan Xiao-xia Qiu Zhi-yuan Zhu Jian-lu Lv Jian Lu Fei Mao Jin Zhu Jia-ying Wang Xiao-wei Guan Jing Chen Jin Ren Ji-ming Ye Yong-hua Zhao Jian Li Xu Shen 2019Acta Pharmacologica Sinica2019,40,10:6

11Gene expression profiling of the rat sciatic nerve in early Wallerian degeneration after injury显示文摘Wallerian degeneration is an important area of research in modern neuroscience.A large number of genes are differentially regulated in the various stages of Wallerian degeneration,especially during the early response.In this study,we analyzed gene expression in early Wallerian degeneration of the distal nerve stump at 0,0.5,1,6,12 and 24 hours after rat sciatic nerve injury using gene chip microarrays.We screened for differentially-expressed genes and gene expression patterns.We examined the data for Gene Ontology,and explored the Kyoto Encyclopedia of Genes and Genomes Pathway.This allowed us to identify key regulatory factors and recurrent network motifs.We identified 1 546 differentially-expressed genes and 21 distinct patterns of gene expression in early Wallerian degeneration,and an enrichment of genes associated with the immune response,acute inflammation,apoptosis,cell adhesion,ion transport and the extracellular matrix.Kyoto Encyclopedia of Genes and Genomes pathway analysis revealed components involved in the Jak-STAT,ErbB,transforming growth factor-β,T cell receptor and calcium signaling pathways.Key factors included interleukin-6,interleukin-1,integrin,c-sarcoma,carcinoembryonic antigen-related cell adhesion molecules,chemokine(C-C motif) ligand,matrix metalloproteinase,BH3 interacting domain death agonist,baculoviral IAP repeat-containing 3 and Rac.The data were validated with real-time quantitative PCR.This study provides a global view of gene expression profiles in early Wallerian degeneration of the rat sciatic nerve.Our findings provide insight into the molecular mechanisms underlying early Wallerian degeneration,and the regulation of nerve degeneration and regeneration.Dengbing Yao Meiyuan Li Dingding Shen Fei Ding Shibi Lu Qin Zhao Xiaosong Gu 2012Neural Regeneration Research2012,7,17:5

15Geniposide improves insulin resistance through AMPK-mediated Txnip protein degradation in 3T3-L1 adipocytes显示文摘Thioredoxin-interacting protein(Txnip)has emerged as a key regulator of insulin resistance.In this study,we investigated the roles of geniposide and Txnip in insulin resistance in differentiated 3T3-L1 adipocytes.Our results revealed that geniposide markedly enhanced glucose uptake,increased the protein levels of insulin receptor substrate(IRS)-1 and GLUT-1,and prevented the phosphorylation of IRS-1 and Akt Thr308 induced by insulin resistance in 3T3-L1 adipocytes.We also observed that geniposide accelerated protein degradation of Txnip through proteasome pathway,and knockdown of Txnip with small interfering RNA attenuated the effect of geniposide on insulin signaling molecules,implying that Txnip played a pivotal role in the regulation of insulin signaling molecules by geniposide in 3T3-L1 adipocytes.Furthermore,geniposide induced the phosphorylation of adenosine monophosphate-activated protein kinase(AMPK)in the presence of high glucose in differentiated 3T3-L1 adipocytes,while compound C,an inhibitor of AMPK,prevented the effect of geniposide on Txnip degradation and the regulation of glucose uptake and insulin signaling molecules including p-IRS-1,IRS-1,and GLUT-1 in differentiated 3T3-L1 adipocytes.Taken together,all these findings suggest that geniposide improves the insulin signaling defect possibly by AMPK-mediated Txnip degradation in 3T3-L1 adipocytes.Wanjun Zhao Mengru Pu Shenli Shen Fei Yin 2021Acta Biochimica et Biophysica Sinica2021,53,2:4

17Growth and differentiation of neural stem cells in a three-dimensional collagen gel scaffold显示文摘Collagen protein is an ideal scaffold material for the transplantation of neural stem cells.In this study,rat neural stem cells were seeded into a three-dimensional collagen gel scaffold,with suspension cultured neural stem cells being used as a control group.Neural stem cells,which were cultured in medium containing epidermal growth factor and basic fibroblast growth factor,actively expanded and formed neurospheres in both culture groups.In serum-free medium conditions,the processes extended from neurospheres in the collagen gel group were much longer than those in the suspension culture group.Immunofluorescence staining showed that neurospheres cultured in collagen gels were stained positive for nestin and differentiated cells were stained positive for the neuronal marker βIII-tubulin,the astrocytic marker glial fibrillary acidic protein and the oligodendrocytic marker 2',3'-cyclic nucleotide 3'-phosphodiesterase.Compared with neurospheres cultured in suspension,the differentiation potential of neural stem cells cultured in collagen gels increased,with the formation of neurons at an early stage.Our results show that the three-dimensional collagen gel culture system is superior to suspension culture in the proliferation,differentiation and process outgrowth of neural stem cells.Fei Huang Qiang Shen Jitong Zhao 2013Neural Regeneration Research2013,8,4:3
18Three-dimensional reconstruction of Picea wilsonii Mast. pollen grains using automated electron microscopy显示文摘Three-dimensional electron microscopy(3 D-EM) has attracted considerable attention because of its ability to provide detailed information with respect to developmental analysis. However, large-scale high-resolution 3 D reconstruction of biological samples remains challenging. Herein, we present a 3 D view of a Picea wilsonii Mast. pollen grain with 100 nm axial and38.57 nm lateral resolution using AutoCUTS-SEM(automatic collector of ultrathin sections-scanning electron microscopy). We established a library of 3,127 100 nm thick serial sections of pollen grains for preservation and observation, demonstrating that the protocol can be used to analyze large-volume samples. After obtaining the SEM images, we reconstructed an entire pollen grain comprising 734 serial sections. The images produced by 3D reconstruction clearly revealed the main components of the P.wilsonii pollen grain, i.e., two sacci and pollen corpus, tube cell, generative cell, and two degenerated prothallial cells, and their internal organization. 20Sex disparity of lung cancer risk in non-smokers:a multicenter population-based prospective study based on China National Lung Cancer Screening Program显示文摘Background: Non-smokers account for a large proportion of lung cancer patients, especially in Asia, but the attention paid to them is limited compared with smokers. In non-smokers, males display a risk for lung cancer incidence distinct from the females-even after excluding the influence of smoking;but the knowledge regarding the factors causing the difference is sparse. Based on a large multicenter prospective cancer screening cohort in China, we aimed to elucidate the interpretable sex differences caused by known factors and provide clues for primary and secondary prevention.Methods: Risk factors including demographic characteristics, lifestyle factors, family history of cancer, and baseline comorbidity were obtained from 796,283 Chinese non-smoking participants by the baseline risk assessment completed in 2013 to 2018. Cox regression analysis was performed to assess the sex difference in the risk of lung cancer, and the hazard ratios (HRs) that were adjusted for different known factors were calculated and compared to determine the proportion of excess risk and to explain the existing risk factors.Results: With a median follow-up of 4.80 years, 3351 subjects who were diagnosed with lung cancer were selected in the analysis. The lung cancer risk of males was significantly higher than that of females;the HRs in all male non-smokers were 1.29 (95% confidence interval [CI]: 1.20-1.38) after adjusting for the age and 1.38 (95% CI: 1.28-1.50) after adjusting for all factors, which suggested that known factors could not explain the sex difference in the risk of lung cancer in non-smokers. Known factors were 7% (|1.29-1.38|/1.29) more harmful in women than in men. For adenocarcinoma, women showed excess risk higher than men, contrary to squamous cell carcinoma;after adjusting for all factors, 47% ([1.30-1.16]/[1.30-1]) and 4% ([7.02-6.75]/[7.02-1])) of the excess risk was explainable in adenocarcinoma and squamous cell carcinoma. The main causes of gender differences in lung cancer risk were lifestyle factors, baseline comorbidity, and family history.Conclusions: Significant gender differences in the risk of lung cancer were discovered in China non-smokers.
