1A mussel-inspired supramolecular hydrogel with robust tissue anchor for rapid hemostasis of arterial and visceral bleedings显示文摘In recent years,the developed hemostatic technologies are still difficult to be applied to the hemostasis of massive arterial and visceral hemorrhage,owing to their weak hemostatic function,inferior wet tissue adhesion,and low mechanical properties.Herein,a mussel-inspired supramolecular interaction-cross-linked hydrogel with robust mechanical property(308.47±29.20 kPa)and excellent hemostatic efficiency(96.5%±2.1%)was constructed as a hemostatic sealant.Typically,we combined chitosan(CS)with silk fibroin(SF)by cross-linking them through tannic acid(TA)to maintain the structural stability of the hydrogel,especially for wet tissue adhesion ability(shear adhesive strength=29.66±0.36 kPa).Compared with other materials reported previously,the obtained CS/TA/SF hydrogel yielded a lower amount of blood loss and shorter time to hemostasis in various arterial and visceral bleeding models,which could be ascribed to the synergistic effect of wound closure under wet state as well as intrinsic hemostatic activity of CS.As a superior hemostatic sealant,the unique hydrogel proposed in this work can be exploited to offer significant advantages in the acute wound and massive hemorrhage with the restrictive access of therapeutic moieties.Ziwen Qiao Xueli Lv Shaohua He Shumeng Bai Xiaochen Liu Linxi Hou Jingjing He Dongmei Tong Renjie Ruan Jin Zhang Jianxun Ding Huanghao Yang 2021Bioactive Materials2021,6,9:14
2Melatonin enhances radiofrequency-induced NK antitumor immunity,causing cancer metabolism reprogramming and inhibition of multiple pulmonary tumor development显示文摘Surgery is the comm on treatme nt for early lung cancer with multiple pulm onary no dules,but it is often accompanied by the problem of significant malignancy of other nodules in non-therapeutic areas.In this study,we found that a combined treatment of local rad iofreq ue ncy ablati on(RFA)and melatonin(MLT)greatly improved clinical outcomes for early lung cancer patie nts with multiple pulmonary nodules by minimizing lung function injury and reducing the probability of malignant transformation or enlargement of nodules in non-ablated areas.Mechanically,as demonstrated in an associated mouse lung tumor model,RFA not only effectively remove treated tumors but also stimulate antitumor immunity,which could inhibit tumor growth in non-ablated areas.MLT enhanced RFA-stimulated NK activity and exerted synergistic antitumor effects with RFA.Transcriptomics and proteomics analyses of residual tumor tissues revealed enhanced oxidative phosphorylation and reduced acidification as well as hypoxia in the tumor microenvironment,which suggests reprogrammed tumor metabolism after combined treatment with RFA and MLT.Analysis of residual tumor further revealed the depressed activity of MAPK,NF-kappa B,Wnt,and Hedgehog pathways and upregulated P53 pathway in tumors,which was in line with the inhibited tumor growth.Combined RFA and MLT treatment also reversed the Warburg effect and decreased tumor malignancy.These findings thus demonstrated that combined treatment of RFA and MLT effectively inhibited the malignancy of non-ablated nodules and provided an innovative non-invasive strategy for treating early lung tumors with multiple pulmonary nodules.Trial registration:www.chictr.org.cn,identifier ChiCTR2100042695,http://gffzz62a03ee25e6f4a77h5b9f6k9nv56x6u96.ffgz.tsg.suse.edu.cn/showproj.aspx?proj=120931.Ming Li Bingjie Hao Menghuan Zhang Russel J.Reiter Shumeng Lin Tiansheng Zheng Xiangyun Chen Yanbei Ren Liduo Yue Baigenzhin Abay Guojie Chen Xiao Xu Yufeng Shi Lihong Fan 2021Signal Transduction and Targeted Therapy2021,6,10:6

5Research on the Work-rest Scheduling in the Manual Order Picking Systems to Consider Human Factors显示文摘As the status of order picking in the warehousing and distribution system has been raised,the work-rest scheduling of picking becomes particularly important.Although science and technology have developed rapidly,manual picking is still essential and indispensable.However,previous researches focused on the study of the sequencing,ignoring human factors.The paper presents a work-rest schedule model in parts to picker picking system.Two objectives are proposed that include minimizing the picking time and minimizing picking error rate.And workers'fatigue,workload is taken into account in the manual order picking systems because the fatigue can have a large influence on the picking time and the picking error rate.A genetic algorithm is used to solve a multi-objective optimization problem that the model concerns and looking for a Pareto front as the most effective methods for solving this problem.Once the original data is given,the work-rest scheduling model is built and the work sequence,and the number of breaks are determined to be chosen by decision makers.In addition,a case study of the model is used to confirm that the model is effective and it is necessary to consider the human factor in the picking system.Xiaosong Zhao Na Liu Shumeng Zhao Jinhui Wu Kun Zhang Rui Zhang 2019Journal of Systems Science and Systems Engineering2019,28,3:1
6Precisely engineering a dual-drug cooperative nanoassembly for proteasome inhibition-potentiated photodynamic therapy显示文摘Photodynamic therapy(PDT) has been widely investigated for cancer therapy. The intracellular accumulation of reactive oxygen species(ROS)-damaged protein facilitates tumor cell apoptosis. However, there is growing evidence that the ubiquitin-proteasome pathway(UPP) significantly impedes PDT by preventing the enrichment of ROS-damaged proteins in tumor cells. To tackle this challenge, we report a facile dual-drug nanoassembly based on the discovery of an interesting co-assembly of bortezomib(BTZ, a proteasome inhibitor) and pyropheophorbide a(PPa) for proteasome inhibition-mediated PDT sensitization.The precisely engineered nanoassembly with the optimal dose ratio of BTZ and PPa demonstrates multiple advantages, including simple fabrication, high drug co-loading efficiency, flexible dose adjustment,good colloidal stability, long systemic circulation, favorable tumor-specific accumulation, as well as significant enrichment of ROS-damaged proteins in tumor cells. As a result, the cooperative nanoassembly exhibits potent synergistic antitumor activity in vivo. 8Customizable Ligand Exchange for Tailored Surface Property of Noble Metal Nanocrystals显示文摘It is highly desirable,while still challenging,to obtain noble metal nanocrystals with custom capping ligands,because their colloidal synthesis relies on specific capping ligands for the shape control while conventional ligand exchange processes suffer from“the strong replaces the weak”limitation,which greatly hinders their applications.Herein,we report a general and effective ligand exchange approach that can replace the native capping ligands of noble metal nanocrystals with virtually any type of ligands,producing flexibly tailored surface properties.The key is to use diethylamine with conveniently switchable binding affinity to the metal surface as an intermediate ligand.As a strong ligand,it in its original form can effectively remove the native ligands;while protonated,it loses its binding affinity and facilitates the adsorption of new ligands,especially weak ones,onto the metal surface.By this means,the irreversible order in the conventional ligand exchange processes could be overcome.The efficacy of the strategy is demonstrated by mutual exchange of the capping ligands among cetyltrimethylammonium,citrate,polyvinylpyrrolidone,and oleylamine.This novel strategy significantly expands our ability to manipulate the surface property of noble metal nanocrystals and extends their applicability to a wide range of fields,particularly biomedical applications.Qikui Fan Hao Yang Juan Ge Shumeng Zhang Zhaojun Liu Bo Lei Tao Cheng Youyong Li Yadong Yin Chuanbo Gao 2020Research2020,,1:1

11The existence of a nonclassical TCA cycle in the nucleus that wires the metabolic-epigenetic circuitry显示文摘The scope and variety of the metabolic intermediates from the mitochondrial tricarboxylic acid(TCA)cycle that are engaged in epigenetic regulation of the chromatin function in the nucleus raise an outstanding question about how timely and precise supply/consumption of these metabolites is achieved in the nucleus.We report here the identification of a nonclassical TCA cycle in the nucleus(nTCA cycle).We found that all the TCA cycle-associated enzymes including citrate synthase(CS),aconitase 2(ACO2),isocitrate dehydrogenase 3(IDH3),oxoglutarate dehydrogenase(OGDH),succinyl-CoA synthetase(SCS),fumarate hydratase(FH),and malate dehydrogenase 2(MDH2),except for succinate dehydrogenase(SDH),a component of electron transport chain for generating ATP,exist in the nucleus.We showed that these nuclear enzymes catalyze an incomplete TCA cycle similar to that found in cyanobacteria.We propose that the nTCA cycle is implemented mainly to generate/consume metabolic intermediates,not for energy production.We demonstrated that the nTCA cycle is intrinsically linked to chromatin dynamics and transcription regulation.Together,our study uncovers the existence of a nonclassical TCA cycle in the nucleus that links the metabolic pathway to epigenetic regulation.Xujun Liu Wenzhe Si Lin He Jianguo Yang Yani Peng Jie Ren Xiaoping Liu Tong Jin Huajing Yu Zihan Zhang Xiao Cheng Wenting Zhang Lu Xia Yunchao Huang Yue Wang Shumeng Liu Lin Shan Yu Zhang Xiaohan Yang Haixia Li Jing Liang Luyang Sun Yongfeng Shang 2021Signal Transduction and Targeted Therapy2021,6,12:0
12Structural insights into photosynthetic cyclic electron transport显示文摘During photosynthesis,light energy is utilized to drive sophisticated biochemical chains of electron transfers,converting solar energy into chemical energy that feeds most life on earth.Cyclic electron transfer/flow(CET/CEF)plays an essential role in efficient photosynthesis,as it balances the ATP/NADPH ratio required in various regulatory and metabolic pathways.Photosystem I,cytochrome b6f,and NADH dehydrogenase(NDH)are large multisubunit protein complexes embedded in the thylakoid membrane of the chloroplast and key players in NDH-dependent CEF pathway.Furthermore,small mobile electron carriers serve as shuttles for electrons between these membrane protein complexes.Efficient electron transfer requires transient interactions between these electron donors and acceptors.Structural biology has been a powerful tool to advance our knowledge of this important biological process.A number of structures of the membrane-embedded complexes,soluble electron carrier proteins,and transient complexes composed of both have now been determined.These structural data reveal detailed interacting patterns of these electron donor-acceptor pairs,thus allowing us to visualize the different parts of the electron transfer process.This review summarizes the current state of structural knowledge of three membrane complexes and their interaction patterns with mobile electron carrier proteins.Shumeng Zhang Baohua Zou Peng Cao Xiaodong Su Fen Xie Xiaowei Pan Mei Li 2023Molecular Plant2023,16,1:0
13Investigating the crucial roles of aliphatic tails in disulfide bond-linked docetaxel prodrug nanoassemblies显示文摘Disulfide bond-bridging strategy has been extensively utilized to construct tumor specificity-responsive aliphatic prodrug nanoparticles(PNPs) for precise cancer therapy. Yet, there is no research shedding light on the impacts of the saturation and cis-trans configuration of aliphatic tails on the self-assembly capacity of disulfide bond-linked prodrugs and the in vivo delivery fate of PNPs. Herein, five disulfide bond-linked docetaxelfatty acid prodrugs are designed and synthesized by using stearic acid, elaidic acid, oleic acid, linoleic acid and linolenic acid as the aliphatic tails, respectively. Interestingly, the cistrans configuration of aliphatic tails significantly influences the self-assembly features of prodrugs, and elaidic acid-linked prodrug with a trans double bond show poor self-assembly capacity. Although the aliphatic tails have almost no effect on the redox-sensitive drug release and cytotoxicity, different aliphatic tails significantly influence the chemical stability of prodrugs and the colloidal stability of PNPs, thus affecting the in vivo pharmacokinetics, biodistribution and antitumor efficacy of PNPs.
