1Relationship between plasma D(-)-lactate and intestinal damage after severe injuries in rats显示文摘AIM To explore the kinetic changes in plasma D (-)lactate and lipopolysaccharide ( LPS ) levels, and investigate whether D (-)-lactate could be used as a marker of intestinal injury in rats following gut ischemia/reperfusion, burn, and acute necrotizing pancreatitis (ANP).METHODS Three models were developed in rats:gut ischemia/reperfusion obtained by one hour of superior severe burn injury created by 30% of total body surface induced by continuous inverse infusion of sodium taurocholate and trypsin into main pancreatic duct.Plasma levels of D( - )-lactate in systemic circulation and LPS in portal circulation were measured by enzymaticspectrophotometric method and limulus amebocyte lysate (LAL) test kit, respectively. Tissue samples of intestine were taken for histological analysis.RESULTS One hour gut ischemia followed by reperfusion injuries resulted in a significant elevation in plasma D( - )lactate and LPS levels, and there was a significant correlation between the plasma D( - )-lactate and LPS (r =0.719, P<0.05). The plasma concentrations of D(-)lactate and LPS increased significantly at 6 h postburn,and there was also a remarkable correlation between them (r = 0.877, P < 0.01). D ( - )-lactate and LPS levels elevated significantly at 2 h after ANP, with a similar significant correlation between the two levels (r-0.798,P < 0.01 ). The desquamation of intestine villi and infiltration of inflammatory cells in the lamina propria were observed in all groups.CONCLUSION The changes of plasma D( - )-lactate levels in systemic blood paralleled with LPS levels in the portal vein blood. 2Composition, source, mass closure of PM_(2.5) aerosols for four forests in eastern China显示文摘PM2.5 aerosols were collected in forests along north latitude in boreal-temperate, temperate, subtropical and tropical climatic zones in eastern China, i.e., Changbai Mountain Nature Reserve (CB), Dongping National Forest Park in Chongming Island (CM), Dinghu Mountain Nature Reserve (DH), Jianfengling Nature Reserve in Hainan Island (HN). The mass concentrations of PM2.5, organic carbon (OC), elemental carbon (EC), water soluble organic carbon (WSOC) as well as concentrations of ten inorganic ions (F-, Cl-, NO3-, SO42-, C2O42-, NH4+, Na+, K+, Ca2+, Mg2+) were determined. Aerosol chemical mass closures were achieved. The 24-hr average concentrations of PM2.5 were 38.8, 89.2, 30.4, 18 μg/m3 at CB, CM, DH and HN, respectively. Organic matter and EC accounted for 21%-33% and 1.3%-2.3% of PM2.5 mass, respectively. The sum of three dominant secondary ions (SO42-, NO3-, NH4+) accounted for 44%, 50%, 45% and 16% of local PM2.5 mass at CB, CM, DH and HN, respectively. WSOC comprised 35%-65% of OC. 6Biological character of human adipose-derived adult stem cells and influence of donor age on cell replication in culture显示文摘To investigate the biological character of human adipose-derived adult stem cells (hADAS cells) when cultured in vitro and the relationship between hADAS cell’s replication activity and the donor’s age factor, and to assess the stem cells as a new source for tissue engineering. hADAS cells are isolated from human adipose tissue of different age groups (from adolescents to olds: <20 years old, 21―40 years old, 41―60 years old and >61 years old groups). The protein markers (CD29, CD34, CD44, CD45, CD49d, HLA-DR, CD106) of hADAS cells were detected by flow cytometry (FCM) to identify the stem cell, and the cell cycle was examined for P20 hADAS cells to evaluate the safety of the subculture in vitro. The generative activity of hADAS cells in different age groups was also examined by MTT method. The formula “ log2T D = t logN t ? logN 0” was used to get the time doubling (TD) of the cells. The results showed that the cells kept heredity stabilization by chromosome analysis for at least 20 passages. The TD of these cells increased progressively by ageing, and the TD of the <20 years old group was lower than that of the >61 years old group (statistical analysis of variance (ANOVA), P=0.002, P<0.05). 8Polychlorinated biphenyls in the atmosphere of Taizhou,a major e-waste dismantling area in China显示文摘PM2.5,total suspended particles(TSP) and gas phase samples were collected at two sites of Taizhou,a major e-waste dismantling area in China.Concentrations,seasonal variations,congener profiles,gas-particle partitioning and size distribution of the atmospheric polychlorinated biphenyls(PCBs) were studied to assess the current state of atmospheric PCBs after the phase out of massive historical dismantling of PCBs containing e-wastes.The average 38PCBs concentration in the ambient air(TSP plus gas phase) near the e-waste dismantling area was(12,407 ± 9592) pg/m3 in winter,which was substantially lower than that found one decade ago.However,the atmospheric PCBs level near the e-waste dismantling area was 54 times of the reference urban site,indicating that the impact of the historical dismantling of PCBs containing e-wastes was still significant.Tri-Penta-CBs were dominant homologues,consisting with their dominant global production.Size distribution of particle-bound PCBs showed that higher chlorinated CBs tended to partition more to the fine particles,facilitating its long range air transportation.Wenliang Han Jialiang Feng Zeping Gu Minghong Wu Guoying Sheng Jiamo Fu 2010Journal of Environmental Sciences2010,22,4:18

10Long-chain acyl-CoA synthetase in fatty acid metabolism involved in liver and other diseases:An update显示文摘Long-chain acyl-Co A synthetase(ACSL) family members include five different ACSL isoforms, each encoded by a separate gene and have multiple spliced variants. ACSLs on endoplasmic reticulum and mitochondrial outer membrance catalyze fatty acids with chain lengths from 12 to 20 carbon atoms to form acyl-Co As, which are lipid metabolic intermediates and involved in fatty acid metabolism, membrane modifications and various physiological processes. Gain- or lossof-function studies have shown that the expression of individual ACSL isoforms can alter the distribution and amount of intracellular fatty acids. Changes in the types and amounts of fatty acids, in turn, can alter the expression of intracellular ACSLs. ACSL family members affect not only the proliferation of normal cells, but the proliferation of malignant tumor cells. They also regulate cell apoptosis through different signaling pathways and molecular mechanisms. ACSL members have individual functions in fatty acid metabolism in different types of cells depending on substrate preferences, subcellular location and tissue specificity, thus contributing to liver diseases and metabolic diseases, such as fatty liver disease, obesity, atherosclerosis and diabetes. They are also linked to neurological disorders and other diseases. However, the mechanisms are unclear. 12Molecular organic geochemical evidence for paleoenvironmental changes at 11.87—12.28 m in GS-1 sedimentary core,Gucheng Lake,East China显示文摘Organic lipids with different occurrence modes in a sedimentary core in a subtropicalfreshwater lake, Gucheng Lake, were sequentially extracted, separated and analyzed. Thedown-core variations of OEP index of alkanes, C18:2/C18:0 ratios of fatty acids, ratios of boundfatty acids to free fatty acids, thermo-maturity indices of hopanoid hydrocarbons and relativeconcentrations of sedimentary minerals revealed an evident increasing in allochthonous inputfrom rock detritus below 12.28 m in the sedimentary profile. The distribution pattern of tightly-bound β-hydroxy fatty acids indicated a difference in bacterial communities above and below theboundary, as well as the gammacerane index may assign a significant change in lake watersalinity. 13Long-chain alkenones in Hotong Qagan Nur Lake sediments and its paleoclimatic implications显示文摘The long-chain alkenones present in the shallow sediment of Hotong Qagan Nur Lake in China are reported. According to the similarity of compositions and distributions of the alkenones determined commonly in the sediment section to those of marine sediment, it is reasonably referred that the alkenones might also originate from Prymnesiophyceae. ln addition, the parameter U37k'of alkenone is in positive correlation to δ18O values of authigenic calcium and magnesium minerals. 14A STUDY ON UNSTEADY SEEPAGE FLOW THROUGH DAM显示文摘A numerical model is proposed to simulate the unsteady seepage flow through dam,with both saturation and water head as variables to describe the seepage domain.Experiments of seepage flow through dam with unsteady conditions were conducted in laboratory to verify the reliability and the accuracy of the numerical model.The performance of the model is also assessed by comparing with analytical solutions.The examples show that the accuracy and the reliability of the model are adequate to handle seepage flow problems with complex conditions.It is also demonstrated that the method of determining the seepage surface through the saturation description is natural,logical and practical in actual applications.The method provides a relatively simple computing scheme,as compared with traditional methods.FU Jun-feng JIN Sheng 2009Journal of Hydrodynamics2009,21,4:14
15Effects of Salvia miltiorrhiza on intestinal microflora in rats with ischemia/reperfusion liver injury显示文摘BACKGROUND: Hepatic ischemia/reperfusion injury may induce intestinal microflora imbalance. Salvia miltiorrhiza is effective in promoting blood circulation and counteracting peroxidation in tissues. The aim of the present study was to determine the effects of Salvia miltiorrhiza on intestinal mi- croflora, endotoxemia, and bacterial translocation in rats with hepatic I/R injury. METHODS: Sprague-Dawley rats in specific pathogen free grade were divided into 3 groups: group I(n =6) for sham operation: groups ( n = 7) for liver ische- mia for 20 minutes and reperfusion for 22 hours. Group was also pretreated with 4 ml/day of Salvia miltiorrhiza solu- tion (250 mg/kg) by daily gavage for 7 days. The levels of serum alanine aminotransferase (ALT), aspartate amino- transferase (AST), malondialdehyde ( MDA) and supero- xide dismutase ( SOD ) in liver tissues, serum endotoxin, intestinal bacterial counts, intestinal mucosal histology and bacterial translocation were studied. RESULTS: The levels of ALT, AST, plasma endotoxin and MDA in liver tissues were decreased more markedly in group (57.57 ± 18.08 U/L, 147.57 ±40.84 U/L, 0.42 ± 0.144 EU/ml and 0. 52 ±0.19 nmol/mg-prot respectively) in group 295.9±216.92 U/L, 0.80± 0.262 EU/ml and 0.72±0.12 nmol/mg-prot; P <0.05-0.01 respectively). Liver SOD activity was increased more sig- nificantly in group (318.47±64.62 U/mg-prot) than in group U/mg-prot, P<0.05). The counts of Bifidobacteria and Bacteroides increased more significantly in group than in group but were similar to those in group I. Bacterial translocation to the kidney in group was 50% (5/10), whereas no bacterial translocation to the kidney occurred in the other two groups (P <0. 01). 16Diurnal variations of polycyclic aromatic hydrocarbons associated with PM_(2.5) in Shanghai, China显示文摘Forty-eight daily time interval PM2.5 samples were collected from December 2006 to January 2008 in an urban site in Shanghai, China. Concentrations and compositions of polycyclic aromatic hydrocarbons (PAHs) were analyzed with GC-MS to study the diurnal and seasonal variations and to identify the main emitting sources. The diurnal variation of the PAHs concentrations was greater in the late autumn and winter sampling days, and was greatly influenced by meteorological conditions such as wind speed and ambient temperature. The concentration of PAHs in the mornings (6:30-10:00) increased distinctly, and was high in the late autumn and winter sampling days, indicating the contribution from vehicle emissions during rush hours. The diurnal variation of the high molecular weight PAHs did not seem to be controlled by the shift of gas-particle partitioning due to temperature variation, instead, it could be indicative of the variation in the source. Statistical analyses showed that the concentrations of PAHs were negatively correlated with temperature and wind speed, and positively correlated with relative humidity. 17Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 18Rapid mitogen-activated protein kinase by basic fibroblast growth factor in rat intestine after ischemia/reperfusion injury显示文摘AIM: Previous studies showed that exogenous basic fibroblast growth factor (bFGF or FGF-2) could improve physiological dysfunction after intestinal ischemia/reperfusion (I/R) injury. However, the mechanisms of this protective effect of bFGF are still unclear. The present study was to detect the effect of bFGF on the activities of mitogen-activated protein kinase (MAPK) signaling pathway in rat intestine after I/R injury, and to investigate the protective mechanisms of bFGF on intestinal ischemiain jury.METHODS: Rat intestinal I/R injury was produced by clamping the superior mesenteric artery (SMA) for 45 minutes and followed by reperfusion for 48 hours. Seventy-eight Wistar rats were used and divided randomly into sham-operated group (A), normal saline control group (B),bFGF antibody pre-treated group (C), and bFGF treated group (D). In group A, SMA was separated without occlusion. In groups B, C and D, SMA was separated and occluded for 45 minutes, then, released for reperfusion for 48 hours. After the animals were sacrificed, blood and tissue samples were taken from the intestine 45 minutes after ischemia in group A and 2, 6, 24, and 48 hours after reperfusion in the other groups. Phosphorylated forms of p42/p44 MAPK, p38 MAPK and stress activated protein kinase/C-Jun N-terminal kinase (SAPK/JNK) were measured by immunohistochemistry. Plasma levels of D-lactate were examined and histological changes were observed under the light microscope.RESULTS: Intestinal I/R injury induced the expression of p42/p44 MAPK, p38 MAPK, and SAPK/JNK pathways and exogenous bFGF stimulated the early activation of p42/p44 MAPK and p38 MAPK pathways. The expression of phosphorylated forms of p42/p44 MAPK was primarily localized in the nuclei of crypt cells and in the cytoplasm and nuclei of villus cells. The positive expression of p38 MAPK was localized mainly in the nuclei of crypt cells, very few in villus cells. The activities of p42/p44 MAPK and p38 MAPK peaked 6 hours after reperfusion in groups B and C,while SAPK/JNK peaked 24 hours after reperfusion. The activities of p42/p44 MAPK and p38 MAPK peaked 2 hours after reperfusion in group D and those of SAPK/JNK were not changed in group B. 19APC gene mutations in Chinese familial adenomatous polyposis patients显示文摘AIM:To study the characteristics of APC(adenomatous polyposis coli)gene germline mutation in Chinese patients with familial adenomatous polyposis(FAP).METHODS:APC gene from 14 FAP families was amplified by polymerase chain reaction(PCR)and underwent direct sequencing to determine the micromutation type.For the samples without micromutation,the large fragment deletion of APC gene was examined by multiplex ligation-dependent probe amplification(MLPA).RESULTS:There were gene micromutations in 9 families with a micromutation detection rate of 64.3%(9/14),including 6 frameshift mutations(66.7%),1 nonsense mutation(11.1%)and 2 splicing mutations(22.2%).Large fragment deletions were detected by MLPA in 2 families.The total mutation detection rate of micromutations and large fragment deletions was 78.6%(11/14).CONCLUSION:The detection rate of APC gene germline mutation can be improved by direct sequencing combined with MLPA large fragment deletion detection.Sheng, Jian-Qiu Cui, Wei-Jia Fu, Lei Jin, Peng Han, Ying Li, Shu-Jun Fan, Ru-Ying Li, Ai-Qin Zhang, Ming-Zhi Li, Shi-Rong 2010World Journal of Gastroenterology2010,16,12:11
20An experimental and thermodynamic equilibrium investigation of the Pb,Zn,Cr,Cu,Mn and Ni partitioning during sewage sludge incineration显示文摘The effects of different chlorides and operational conditions on the distribution and speciation of six heavy metals(Pb, Zn, Cr, Cu, Mn and Ni) during sludge incineration were investigated using a simulated laboratory tubular-furnace reactor. A thermodynamic equilibrium investigation using the Fact Sage software was performed to compare the experimental results. The results indicate that the volatility of the target metals was enhanced as the chlorine concentration increased. Inorganic-Cl influenced the volatilization of heavy metals in the order of Pb > Zn > Cr > Cu > Mn > Ni. However, the effects of organic-Cl on the volatility of Mn, Pb and Cu were greater than the effects on Zn, Cr and Ni.With increasing combustion temperature, the presence of organic-Cl(PVC) and inorganic-Cl(NaCl) improved the transfer of Pb and Zn from bottom ash to fly ash or fuse gas. However,the presence of chloride had no obvious influence on Mn, Cu and Ni. Increased retention time could increase the volatilization rate of heavy metals; however, this effect was insignificant. During the incineration process, Pb readily formed Pb SiO4 and remained in the bottom ash.
