4Generation of B Cell-Deficient Pigs by Highly Efficient CRISPR/Cas9-Mediated Gene Targeting显示文摘Generating B cell-deficient mutant is the first step to produce human antibody repertoires in large animal models. In this study, we applied the clustered regularly interspaced short palindromic repeat(CRISPR)/CRISPR-associated(Cas) system to target the JHregion of the pig Ig M heavy chain gene which is crucial for B cell development and differentiation. Transfection of Ig M-targeting Cas9 plasmid in primary porcine fetal fibroblasts(PFFs) enabled inducing gene knock out(KO) in up to 53.3% of colonies analyzed, a quarter of which harbored biallelic modification, which was much higher than that of the traditional homologous recombination(HR). With the aid of somatic cell nuclear transfer(SCNT) technology, three piglets with the biallelic Ig M heavy chain gene mutation were produced. The piglets showed no antibody-producing B cells which indicated that the biallelic mutation of the Ig M heavy chain gene effectively knocked out the function of the Ig M and resulted in a B cell-deficient phenotype. 6Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 9Gold nanoflowers for 3D volumetric molecular imaging of tumors by photoacoustic tomography显示文摘By binding molecular probes that target tumor cells, gold nanoparticles (AuNPs) with superior characteristics have shown great potential in tumor molecular imaging studies. The non-invasive, high-resolution, and three-dimensional imaging of the targeted AuNPs within the tumor is desirable for both diagnosis and therapy. In this study, gold nanoflowers (AuNFs) are presented as a novel contrast agent for photoacoustic tomography (PAT). By binding to folic acid, the molecular probe, the tail-vein injected AuNFs concentrated within the tumor site in mice; this was clearly visualized by three-dimensional (3D) PAT imaging. In addition, toxicity assay proved that AuNFs were harmless to living cells and animals. 10In vitro thyroid disrupting effects of organic extracts from WWTPs in Beijing显示文摘It is generally known that there are many endocrine disrupting compounds (EDCs) in the effluents from wastewater treatment plants (WWTPs). Most research has focused on the occurrence of estrogenic or androgenic activities, while ignoring that there are environmental chemicals disrupting thyroid system, which is essential for growth and development in both humans and animals. In the present work, a two-hybrid yeast assay was conducted to evaluate the removal efficiencies of agonistic or antagonistic thyroid receptor (TR) mediated effects in different treatment processes of three WWTPs located in Beijing. We found no TR agonistic, but TR antagonistic activities in all processes from the WWTPs. The TR antagonistic activities in organic extracts of water samples were then calibrated regarding to a known TR-inhibitor, amiodarone hydrochloride (AH). The observed concentration of TR disrupting substances ranged from 2.35 × 10 8 to 6.19 × 10 7 mol/L AH in Gaobeidian WWTP, 3.76 × 10 8 to 8.75 × 10 8 mol/L AH in Lugouqiao WWTP, and 4.80 × 10 9 to 2.55 × 10 8 mol/L AH in Beixiaohe WWTP. Of the three WWTPs, the removal rates were 92.7%, 42.2%, and 23.1% respectively. Industrial sewage may contain more TR disrupting substances compared with domestic sewage. 12Determination of estrogens and estrogenic activities in water from three rivers in Tianjin, China显示文摘Studies on estrogenic disrupting compounds(EDCs) occurrence and identification of main responsible compounds in river water discharged into the sea are of significance.In the present research,we screened estrogenic activities of 10 river water samples from 3 main rivers discharged into Bohai Sea in Tianjin using a recombinant two-hybrid yeast assay and chemical analysis by gas chromatography-mass spectrometry.All sample extracts induced significant estrogenic activity,with 17β-estradiol equivalents(EEQ) of raw water ranging from 5.72 to 59.06 ng/L.Six most concerned EDCs in the river water samples including estrone,17β-estradiol,17α-ethinylestradiol,estriol,diethylstilbestrol and estradiol valerate were determined,with their concentrations up to 50.70,31.40,24.40,37.20,2.56,and 8.47 ng/L,respectively.Through causality analysis by comparing the EEQ values of yeast assay and chemical analysis,17α-ethinylestradiol and 17β-estradiol were identified as the main contributors to the estrogenic effects of the river samples,accounting for the whole estrogenic activities(62.99% to 185.66%),and estrogen antagonistic compounds might presented in the heavy polluted water samples.The proposed approach using both chemical analysis and bioassay could be used for identification and evaluation of the estrogenic activity of EDCs in river water.Kaifeng Rao Bingli Lei Na Li Mei Ma Zijian Wang 2013Journal of Environmental Sciences2013,25,6:6

15Soil microbial activity and nutrients of evergreen broad-leaf forests in mid-subtropical region of China显示文摘To better understand the effects of forest succession on soil microbial activity, a comparison of soil microbial properties and nutrients was conducted between three forest types representing a natural forest succession chronosequence. The study compared a pine(Pinus massoniana) forest(P_F), a pine and broadleaf mixed forest(M_F) and an evergreen broadleaf forest(B_F), in the Yingzuijie Biosphere Reserve, Hunan Province, China.Results showed that soil nutrients in the M_Fand BFplots were higher than in the P_Fplots. The range in microbial biomass carbon followed a similar pattern with B_Fhaving the greatest values, 522–1022 mg kg^-1, followed by M_F368–569 mg kg^-1, and finally, P_F193–449 mg kg^-1. Soil nutrients were more strongly correlated with microbial biomass carbon than basal respiration or metabolic quotient. 17Retinoid X receptor activities of source waters in China and their removal efficiencies during drinking water treatment processes显示文摘There is increasing evidence of estrogenic activities of source waters and drinking waters in China based on estrogen receptors(ERs) testing.However,relating such activities to retinoid X receptors(RXRs) in both drinking and source waters are lacking.To rectify this situation,we assessed 23 source water samples from six major river systems in China.We also collected samples at various stages of water processing from three drinking water treatment plants(DWTPs) using a two-hybrid RXR yeast assay with and without metabolism.No RXR agonistic activity was observed,but significant antagonistic activity was detected in all sample extracts.The RXR antagonistic activities of source water sample extracts ranged from 15.2% to 57.8% without metabolism and 11.5% to 68.3% with metabolism,respectively.In the drinking water treatment processes,RXR antagonistic activities without metabolism and with metabolism of up to 31.4% and 37.5% were removed,respectively.Nevertheless,the remaining RXR antagonists in treated drinking water from these source waters could still be harmful to human health.To the best of our knowledge,the occurrence of in vitro RXR disruption activities in source and drinking water has not been previously reported in China.Therefore,an attempt was made to conduct detailed studies investigating RXR disrupting activities and their possible risks in source and drinking water.JIANG WeiWei YAN Ye LI Na MA Mei WANG DongHong RAO KaiFeng WANG ZiJian SATYANARAYANAN Senthi Kumaran 2012Chinese Science Bulletin2012,57,6:5
18Proteasomal deubiquitinase UCH37 inhibits degradation of β-catenin and promotes cell proliferation and motility显示文摘The ubiquitin–proteasome system degrades most cellular proteins in eukaryotes.UCH37,also known as UCH-L5,is a deubiquitinase binding to Rpn13,a receptor for ubiquitinated substrates in the 26 S proteasome.But,it remains unclear how UCH37 influences the proteasomal degradation of the ubiquitinated substrates.Because deletion of UCH37 is embryonically lethal in mice,this study aims to investigate the role of UCH37 in proteasomal degradation by constructing the UCH37-deficient cell lines using CRISPR/Cas9 technology.Our results demonstrated that deletion of UCH37 decreased the levels of proteasomal Rpn13,implying that UCH37 might facilitate incorporation of Rpn13 into the proteasome.Meanwhile,deletion of UCH37 decreased the levels of β-catenin and the early endosomal protein Rab8.β-Catenin interacts with TCF/LEF to control transcription,and is involved in development,tissue homeostasis and tumorigenesis.We further found that deletion of UCH37 increased the levels of the ubiquitinated β-catenin and accelerated the hydrogen peroxide-stimulated degradation of β-catenin.Deletion of UCH37 also down-regulated the transcription of c-Myc,a downstream effector of β-catenin,and inhibited cell proliferation and motility.These results raise the possibility that UCH37 maintains the homeostasis of proteasomal degradation reciprocally by assisting the recruitment of the ubiquitin receptor Rpn13 into the proteasome and by reversing ubiquitination of certain critical substrates of the 26 S proteasome.Zijian Li Luming Zhou Tianxia Jiang Libin Fan Xiaoying Liu Xiaobo Qiu 2019Acta Biochimica et Biophysica Sinica2019,51,3:4

20Sustained‑Release Nanocapsules Enable Long‑Lasting Stabilization of Li Anode for Practical Li‑Metal Batteries显示文摘A robust solid-electrolyte interphase(SEI)enabled by electrolyte additive is a promising approach to stabilize Li anode and improve Li cycling efficiency.However,the self-sacrificial nature of SEI forming additives limits their capability to stabilize Li anode for long-term cycling.Herein,we demonstrate nanocapsules made from metal–organic frameworks for sustained release of LiNO3 as surface passivation additive in commercial carbonate-based electrolyte.The nanocapsules can offer over 10 times more LiNO3 than the solubility of LiNO3.Continuous supply of LiNO3 by nanocapsules forms a nitride-rich SEI layer on Li anode and persistently remedies SEI during prolonged cycling.As a result,lifespan of thin Li anode in 50μm,which experiences drastic volume change and repeated SEI formation during cycling,has been notably improved.By pairing with an industry-level thick LiCoO2 cathode,practical Li-metal full cell demonstrates a remarkable capacity retention of 90%after 240 cycles,in contrast to fast capacity drop after 60 cycles in LiNO3 saturated electrolyte.Qianqian Liu Yifei Xu Jianghao Wang Bo Zhao Zijian Li Hao Bin Wu 2020Nano-Micro Letters2020,12,12:4
