1MdGSTF6,activated by MdMYB1,plays an essential role in anthocyanin accumulation in apple显示文摘Anthocyanins are biosynthesized on the cytosolic surface of the endoplasmic reticulum and then transported into the vacuole for storage.Glutathione S-transferases(GSTs)are considered to be responsible for the transport of anthocyanins into the vacuole.However,the regulatory mechanisms of GSTs in plants are still unclear.Here,we performed a genome-wide analysis and identified 69 GST genes in apple.The expression of MdGSTF6 was positively correlated with the anthocyanin content(r=0.949)during‘Yanfu 8’fruit development.The overexpression of MdGSTF6 in the Arabidopsis thaliana tt19 mutant resulted in seedlings of 35S::MdGSTF6-GFP/tt19 that could accumulate anthocyanin and rescue its phenotype,suggesting that MdGSTF6 was an anthocyanin transporter.The silencing of MdGSTF6 affected anthocyanin accumulation in apple fruit.Moreover,the knockdown of MdGSTF6 by RNA interference in cultured‘Gala’seedlings inhibited anthocyanin accumulation.The interaction experiments showed that MdMYB1 could bind directly to the MdGSTF6 promoter to transcriptionally activate its expression.Collectively,our results demonstrate that MdGSTF6 encodes an important GST transporter of anthocyanins in apple fruit and provide evidence for the associated regulatory mechanisms.Therefore,MdMYB1 can not only regulate anthocyanin synthesis,but also control the transport of anthocyanin in apples.This information may be useful for further clarifying the regulation of anthocyanin transport in apple.Shenghui Jiang Min Chen Naibo He Xiaoliu Chen Nan Wang Qingguo Sun Tianliang Zhang Haifeng Xu Hongcheng Fang Yicheng Wang Zongying Zhang Shujing Wu Xuesen Chen 2019Horticulture Research2019,6,1:16
2Malus sieversii: the origin, flavonoid synthesis mechanism, and breeding of red-skinned and red-fleshed apples显示文摘Flavonoids play essential roles in human health.Apple(Malus domestica Borkh.),one of the most widely produced and economically important fruit crops in temperate regions,is a significant source of flavonoids in the human diet and is among the top nutritionally rated and most widely consumed fruits worldwide.Epidemiological studies have shown that the consumption of apples,which are rich in a variety of free and easily absorbable flavonoids,is associated with a decreased risk of various diseases.However,apple production is challenged by serious inbreeding problems.The narrowing of the hereditary base has resulted in apples with poor nutritional quality and low flavonoid contents.Recently,there have been advances in our understanding of the roles that Malus sieversii(Ledeb.)M.Roem has played in the process of apple domestication and breeding.In this study,we review the origin of cultivated apples and red-fleshed apples,and discuss the genetic diversity and construction of the core collections of M.sieversii.We also discuss current research progress and breeding programs on red-skinned and red-fleshed apples and summarize the exploitation and utilization of M.sieversii in the breeding of high-flavonoid,and red-fleshed apples.This study highlights a valuable pattern of horticultural crop breeding using wild germplasm resources.The future challenges and directions of research on the molecular mechanisms of flavonoid accumulation and high-flavonoid apple breeding are discussed.Nan Wang Shenghui Jiang Zongying Zhang Hongcheng Fang Haifeng Xu Yicheng Wang Xuesen Chen 2018Horticulture Research2018,5,1:15
3Ciliary parathyroid hormone signaling activates transforming growth factor-βto maintain intervertebral disc homeostasis during aging显示文摘Degenerative disc disease(DDD) is associated with intervertebral disc degeneration of spinal instability. Here, we report that the cilia of nucleus pulposus(NP) cells mediate mechanotransduction to maintain anabolic activity in the discs. We found that mechanical stress promotes transport of parathyroid hormone 1 receptor(PTH1 R) to the cilia and enhances parathyroid hormone(PTH) signaling in NP cells. PTH induces transcription of integrin α_vβ_6 to activate the transforming growth factor(TGF)-β-connective tissue growth factor(CCN2)-matrix proteins signaling cascade. Intermittent injection of PTH(iPTH) effectively attenuates disc degeneration of aged mice by direct signaling through NP cells, specifically improving intervertebral disc height and volume by increasing levels of TGF-β activity, CCN2, and aggrecan. PTH1 R is expressed in both mouse and human NP cells. Importantly,knockout PTH1 R or cilia in the NP cells results in significant disc degeneration and blunts the effect of PTH on attenuation of aged discs. 5MdMYB6 regulates anthocyanin formation in apple both through direct inhibition of the biosynthesis pathway and through substrate removal显示文摘Anthocyanin biosynthesis and sugar metabolism are important processes during plant growth,but the molecular interactions underlying these pathways are still unclear.In this work,we analyzed the anthocyanin and soluble sugar contents,as well as the transcript levels of transcription factors that are known to be related to the biosynthesis of anthocyanin in‘Hongcui 1’apple flesh during fruit development.Overexpression of MdMYB6 in red-fleshed calli was found to reduce anthocyanin content and result in downregulated expression of the MdANS and MdGSTF12 proteins.Yeast one-hybrid and electrophoretic mobility shift analyses showed that MdMYB6 could directly bind to the promoters of MdANS and MdGSTF12,indicating that MdMYB6 could inhibit anthocyanin biosynthesis by regulating MdANS and MdGSTF12.Overexpression of MdTMT1 in the Arabidopsis tmt1 mutant restored the glucose and fructose contents to the wild-type levels,while overexpression of MdTMT1 in red-fleshed calli increased the contents of glucose and fructose but reduced the contents of UDP-glucose,UDP-galactose,and anthocyanin.Using a GUS reporter system,yeast one-hybrid,chromatin immunoprecipitation-PCR and electrophoretic mobility shift analyses,we found that MdMYB6 could bind to the promoter of MdTMT1,resulting in increased promoter activity.Overexpression of MdMYB6 in calli overexpressing MdTMT1 increased the expression of MdTMT1,which led to reduced contents of UDP-glucose and UDP-galactose and decreased anthocyanin content compared to those of the calli that overexpressed MdTMT1.This finding suggested that MdMYB6 could also inhibit anthocyanin biosynthesis by regulating MdTMT1 to decrease the contents of UDP-glucose and UDP-galactose.Taken together,these results showed that MdMYB6 and MdTMT1 play key roles in both anthocyanin biosynthesis and sugar transport.Haifeng Xu Qi Zou Guanxian Yang Shenghui Jiang Hongcheng Fang Yicheng Wang Jing Zhang Zongying Zhang Nan Wang Xuesen Chen 2020Horticulture Research2020,7,1:6
6Characteristics of Vibration and Sound Radiation of Metro Resilient Wheel显示文摘Resilient wheels are extensively used in urban rail transit, especially for tramway systems, owing to its advantages in noise reduction. A new type of resilient wheel for a metro is designed, and its characteristics of vibration and sound radiation, including the rolling noise of a resilient single wheel coupled with a track, are studied in this paper. A two-step research is presented. Firstly, laboratory experiments were conducted to obtain the vibration response of the designed resilient wheel under the radial excitation on its tread. Secondly, the rolling noise model of the resilient wheel coupled with a slab track used in a metro line is developed. The wheel model is based on the 3 D finite elementand boundary element methods and verified by using the experimental results obtained from the laboratory. The track vibration model is based on the wavenumber finite element method, and the track sound radiation is calculatedby using an e cient frequency-domain Rayleigh method. The interaction of the resilient wheel and the slab track is analyzed considering the measured wheel/rail roughness of the metro. The contribution of the resilient wheel to the reduction of wheel/rail system noise is analyzed. The results show that the resilient wheel can e ectively reduce the wheel/rail rolling noise by approximately 2 dB(A) to 3 dB(A), mainly because the radiated noise by the rail is reduced. 7Sediment Transport in the Nearshore Area of Phoenix Island显示文摘Based on the measured data, suspended sediment concentration, surface sediment grain size, current and waves, the sediment transport mechanisms and pathways in the Phoenix Island area were analyzed using methods of flux decomposition and Grain Size Trend Analysis(GSTA). The results show that net suspended sediment is mainly transported by average current, Stokes drift, and gravitational circulation. The transport direction of suspended sediment is varying and basically following the direction of residual tidal currents. Surface sediment transport pathways are primarily parallel to the coastline along with two convergent centers. Waves and longshore currents have a significant influence on sediment transport, but the influence is limited due to a steep and deep underwater bank. Tidal current is the main controlling factor for sediment transport, especially in the deep water area. Neither suspended nor surface sediment is transported towards the southwest. 9Synthesis and Characterization of a Silica-Based Drug Delivery System for Spinal Cord Injury Therapy显示文摘Acute inflammation is a central component in the progression of spinal cord injury(SCI).Anti-inflammatory drugs used in the clinic are often administered systemically at high doses,which can paradoxically increase inflammation and result in drug toxicity.A cluster-like mesoporous silica/arctigenin/CAQK composite(MSN-FC@ARCG)drug delivery system was designed to avoid systemic side effects of high-dose therapy by enabling site-specific drug delivery to the spinal cord.In this nanosystem,mesoporous silica was modified with the FITC fluorescent molecule and CAQK peptides that target brain injury and SCI sites.The size of the nanocarrier was kept at approximately 100 nm to enable penetration of the blood–brain barrier.Arctigenin,a Chinese herbal medicine,was loaded into the nanosystem to reduce inflammation.The in vivo results showed that MSN-FC@ARC-G could attenuate inflammation at the injury site.Behavior and morphology experiments suggested that MSN-FC@ARC-G could diminish local microenvironment damage,especially reducing the expression of interleukin-17(IL-17) and IL-17-related inflammatory factors,inhibiting the activation of astrocytes,thus protecting neurons and accelerating the recovery of SCI.Our study demonstrated that this novel,silica-based drug delivery system has promising potential for clinical application in SCI therapy.Guodong Sun Shenghui Zeng Xu Liu Haishan Shi Renwen Zhang Baocheng Wang Changren Zhou Tao Yu 2019Nano-Micro Letters2019,11,2:1
10Large eddy simulation of spray combustion using flamelet generated manifolds combined with artificial neural networks显示文摘In the present work,artificial neural networks(ANN)technique combined with flamelet generated manifolds(FGM)is proposed to mitigate the memory issue of FGM models.A set of ANN models is firstly trained using a 68-species mass fractions in mixture fraction-progress variable space.The ANN prediction accuracy is examined in large eddy simulation(LES)and Reynolds averaged Navier-Stokes(RANS)simulations of spray combustion.It is shown that the present ANN models can properly replicate the FGM table for most of the species mass fractions.The network models with relative error less than 5%are considered in RANS and LES to simulate the Engine Combustion Network(ECN)Spray H flames.Validation of the method is firstly conducted in the framework of RANS.Both non-reacting and reacting cases show the present method predicts very well the trend of spray and combustion process under different ambient temperatures.The results show that FGM-ANN can replicate the ignition delay time(IDT)and lift-off length(LOL)precisely as the conventional FGM method,and the results agree very well with the experiments.With the help of ANN,it is possible to achieve high efficiency and accuracy,with a significantly reduced memory requirement of the FGM models.LES with FGM-ANN is then applied to explore the detailed spray combustion process.Chemical explosive mode analysis(CEMA)approach is used to identify the local combustion modes.It is found that before the spray flame is developed to the steady-state,the high CH_(2)O zone is always associated with ignition mode.However,high CH_(2)O zone together with high OH zone is dominated by the burned mode after the steady-state.The lift-off position is dominated mainly by the diffusion mode.Yan Zhang Shijie Xu Shenghui Zhong Xue-Song Bai Hu Wang Mingfa Yao 2020Energy and AI2020,2,2:1

13Exosomes-loaded electroconductive nerve dressing for nerve regeneration and pain relief against diabetic peripheral nerve injury显示文摘Over the years,electroconductive hydrogels(ECHs)have been extensively applied for stimulating nerve regeneration and restoring locomotor function after peripheral nerve injury(PNI)with diabetes,given their favorable mechanical and electrical properties identical to endogenous nerve tissue.Nevertheless,PNI causes the loss of locomotor function and inflammatory pain,especially in diabetic patients.It has been established that bone marrow stem cells-derived exosomes(BMSCs-Exos)have analgesic,anti-inflammatory and tissue regeneration properties.Herein,we designed an ECH loaded with BMSCs-Exos(ECH-Exos)electroconductive nerve dressing to treat diabetic PNI to achieve functional recovery and pain relief.Given its potent adhesive and self-healing properties,this laminar dressing is convenient for the treatment of damaged nerve fibers by automatically wrapping around them to form a size-matched tube-like structure,avoiding the cumbersome implantation process.Our in vitro studies showed that ECH-Exos could facilitate the attachment and migration of Schwann cells.Meanwhile,Exos in this system could modulate M2 macrophage polarization via the NF-κB pathway,thereby attenuating inflammatory pain in diabetic PNI.Additionally,ECH-Exos enhanced myelinated axonal regeneration via the MEK/ERK pathway in vitro and in vivo,consequently ameliorating muscle denervation atrophy and further promoting functional restoration.Our findings suggest that the ECH-Exos system has huge prospects for nerve regeneration,functional restoration and pain relief in patients with diabetic PNI.Qinfeng Yang Shenghui Su Shencai Liu Sheng Yang Jing Xu Yixiu Zhong Yusheng Yang Liangjie Tian Zilin Tan Jian Wang Zhiqiang Yu Zhanjun Shi Fangguo Liang 2023Bioactive Materials2023,,8:0
14Human Ear Image Recognition Method Using PCA and Fisherface Complementary Double Feature Extraction显示文摘Ear recognition is a new kind of biometric identification technology now.Feature extraction is a key step in pattern recognition technology,which determines the accuracy of classification results.The method of single feature extraction can achieve high recognition rate under certain conditions,but the use of double feature extraction can overcome the limitation of single feature extraction.In order to improve the accuracy of classification results,this paper proposes a new method,that is,the method of complementary double feature extraction based on Principal Component Analysis(PCA)and Fisherface,and we apply it to human ear image recognition.The experiment was carried out on the ear image library provided by the University of Science and Technology Beijing.The results show that the ear recognition rate of the proposed method is significantly higher than the single feature extraction using PCA,Fisherface,or Independent component analysis(ICA)alone.Yang Wang Ke Cheng Shenghui Zhao Xu E 2023Journal of Artificial Intelligence and Technology2023,3,1:0
15Effect of organophilic group of coupling agent on the electrical performance of Boron Nitride/meta-aramid composite paper显示文摘Organophilic groups of coupling agents can be the key factor for the electrical performance of Boron Nitride(BN)/meta-aramid composite paper.In this article,four types of triethoxy-silane coupling agents with different organophilic groups,including mercaptopropyl(MP),glycidylether-propyl(GP),amino-propyl(AP)and trideca-fluoro-octyl(TFO)were studied.The structural and electrical properties of modified papers were compared,and the molecular simulation was conducted to reveal the interface interaction of modified BN and aramid.The results show that AP,GP and MP follow the same mechanism,improving the electrical performance by modifying the interface stability and paper structure,and the ranks of improving effects are AP,MP and GP.While,TFO mainly relies on the fluorine components to implement the electrical reinforcement,which compensates for the shortcoming in the paper structure.By choosing TFO and filler content of 15 wt%,the optimal breakdown strength of the paper can be increased to 28.17 kV/mm,1.8 times that of the paper doped with the hydroxylated BN.Haoou Ruan Qing Xie Jingxuan Song Shenghui Wang Xiaobin Chang Yuqin Xu Fangcheng Lü 2023High Voltage2023,8,4:0
16Self-assembled multiepitope nanovaccine based on NoV P particles induces effective and lasting protection against H3N2 influenza virus显示文摘Current seasonal influenza vaccines confer only limited coverage of virus strains due to the frequent genetic and antigenic variability of influenza virus(IV).Epitope vaccines that accurately target conserved domains provide a promising approach to increase the breadth of protection;however,poor immunogenicity greatly hinders their application.The protruding(P)domain of the norovirus(NoV),which can self-assemble into a 24-mer particle called the NoV P particle,offers an ideal antigen presentation platform.In this study,a multiepitope nanovaccine displaying influenza epitopes(HMN-PP)was constructed based on the NoV P particle nanoplatform.Large amounts of HMN-PP were easily expressed in Escherichia coli in soluble form.Animal experiments showed that the adjuvanted HMN-PP nanovaccine induced epitope-specific antibodies and haemagglutinin(HA)-specific neutralizing antibodies,the antibodies could persist for at least three months after the last immunization.Furthermore,HMN-PP induced matrix protein 2 extracellular domain(M2e)-specific antibody-dependent cell-mediated cytotoxicity,CD4^(+)and CD8^(+)T-cell responses,a nucleoprotein(NP)-specific cytotoxic T lymphocyte(CTL)response.These results indicated that the combination of a multiepitope vaccine and self-assembled NoV P particles may be an ideal and effective vaccine strategy for highly variable viruses such as IV and SARS-CoV-2.Jiaojiao Nie Qingyu Wang Shenghui Jin Xin Yao Lipeng Xu Yaotian Chang Fan Ding Zeyu Li Lulu Sun Yuhua Shi Yaming Shan 2023Nano Research2023,16,5:0
