| 3 | Epithelial-Mesenchymal Transitions and the Expression of Twist in MCF-7/ADR, Human Multidrug-Resistant Breast Cancer Cells显示文摘0BJECTIVE To study the expression levels of Twist and epithelialmesenchymal transitions in multidrug-resistant MCF-7/ADR breast cancer cells, and to study the relationship between multidrug resistance (MDR) and metastatic potential of the cells. METHODS RT-PCR, immunohistochemical and Western blotting methods were used to examine the changes of expression levels of the transcription factor Twist, E-cadherin and N-cadherin in the MCF-7 breast cancer cell line and its multidrug-resistant variant, MCF-7/ADR. RESULTS In MCF-7 cells, the expression of E-cadherin can be detected, but there is no expression of Twist or N-cadherin. In MCF-7/ADR cells, E-cadherin expression is lost, but the expression of two other genes was significantly positive. | 8 | Tunable magneto-Seebeck effect in Co_(2)FeSi/MgO/Co_(2)FeSi heterostructure via optimized interfacial engineering显示文摘Enhancing the tunneling magneto-Seebeck(TMS) ratio and uncovering its underlying mechanism are greatly demanded in spin caloritronics.The magnitude and sign of the TMS effect depend on the type of atom and the stacking order of atoms at the interfaces.Herein,we demonstrate that TMS ratios can be effectively manipulated by altering heterogonous or homogeneous interface through decoration on the CoFeSi(001) surface inserted on the MgO insulating layers.The maximum TMS ratio of pure Co_(2)/O termination is 4565% at 800 K.Notably,the TMS ratio of the FeSi/O termination has two peak values,of which the maximum could reach up to-3290% at 650 K.By comparing two different atom arrangements at the interface,we reveal that the sign and symbol of the TMS ratio can be controlled by the temperature and different atomic configurations at the Co_(2)FeSi/MgO interface.Furthermore,the spin-Seebeck coefficient up to ~150 μV/K is also possible when we select suitable terminations and temperatures.These findings will provide useful insights into how to control the sign and symbol of the TMS ratio and accordingly stimulate the development field of magneto-thermoelectric power and spin caloritronic devices based on the magneto-Seebeck effect in Heusler-based metallic multilayers. | Jingyu Li Xianbiao Shi Yurong Jin Le Ma Liuming Wei Chi Zhang Hang Li Peng-Fei Liu | 2024 | Science China(Physics,Mechanics & Astronomy)2024,67,3: | 0 |

| 13 | Down-Regulation of CXCR4 Expression by siRNA Inhibits Invasive Ability of Breast Cancer Cells显示文摘OBJECTIVE To investigate the efficiency of gene silencing by CXCR4- siRNAs (small interfering RNA), and to examine the invasive ability and the expression of other metastatic-associated genes in siRNA-treated breast cancer cells. METHODS Three siRNAs were designed and cloned into the pSilenc ? 3.1-H1 neo vector. The reconstructed plasmids were purified and trans- fected into the T47D breast cancer cell line, which highly expressed CXCR4. The amount of CXCR4 expression in the transfected cells was measured by flow cytometry and Real-time PCR. Cell invasive ability was evaluated us- ing 24-well Matrigel invasion chambers. In addition, the expression of other metastatic-associated genes, such as E-cad, IGFBP-5, FN and MMP-2, was assessed by Real-time PCR. RESULTS The suppression rates of CXCR4 mRNA expression reached 95.7%, 85.9% and 98.3%compared with control-siRNA cells in the 3 CXCR4- siRNA T47D cells respectively. FCM assays for CXCR4 protein expression showed a similar inhibitory effect. The invasion indexes of these CXCR4- siRNA cells were 0.037, 0.290 and 0.188 respectively compared with control- siRNA cells. After treatment of the cells with CXCR4-siRNA, the expression of E-cad showed an upward tendency and that of IGFBP-5 had a downward trend, while alteration in expression of FN and MMP2 varied without a con- sistant effect. CONCLUSION CXCR4 plays an important role in modulating migra- tion of human breast cancer cells. Small interfering RNA can significantly silence the CXCR4 gene in the human T47D breast cancer cell line. | 14 | The Effect of Twist Expression on Angiogenesis in Hepatocellular Carcinoma显示文摘OBJECTIVE Hepatocellular carcinoma (HCC) is a hypervascular tumor for which angiogenesis plays an important role in its progression. The aim of this study was to investigate the expression of TWIST and VEGF and determine their roles in angiogenesis of HCC. METHODS Expression Twist and VEGF mRNA was determined by real-time RT-PCR in 30 pairs of hepatocellular carcinoma and matched non-cancerous tissues. Immunohistochemistry was carried out to analyze the protein expression of Twist and VEGF in 40 hepatocellular carcinoma cases. Staining of endothelial cells for CD34 was used to evaluate the microvessel density (MVD). RESULTS We found that the HCC specimens showing positive Twist expression in tumor cells had a higher microvessel density than those without Twist expression. Furthermore, we found that overexpression of the Twist protein positively correlated with up-regulation of VEGF in the HCC tissues (r=0.479, P=0.002). | 15 | Proteasome Inhibitors Sensitize Hepatocellular Carcinoma Cells to TRAIL显示文摘OBJECTIVE To investigate the effect of proteasome inhibition on the sensitivity of carcinoma cells to TRAIL-inducing apoptosis, and to study the mechanism of the response. METHODS Human hepatocellular carcinoma cells, pretreated with the proteasome inhibitor, MG132, were cotreated with TRAIL. Western blot assays, immunoprecipitation and RT-PCR were performed to test the expression of the Bcl-2 family proteins and Bax mRNA. RESULTS We found that (i) proteasome inhibition sensitized the human hepatocellular carcinoma cells to TRAIL; and (ii) resulted in Bax accumulation before release of cytochrome C and induction of apoptosis. These results were associated with the ability of proteasome inhibitors to overcome Bcl-2-mediated antiapoptotic function; (iii) Bax is regulated by an ubiquitin/proteasome-dependent degradation pathway. | 17 | RNA sequencing-based optimization of biological lipid droplets for sonodynamic therapy to reverse tumor hypoxia and elicit robust immune response显示文摘Mitochondria-targeted sonodynamic therapy(SDT)is a promising strategy to inhibit tumor growth and activate the anti-tumor immune responses.Identifying the mechanisms underlying mitochondria-targeted SDT,further optimizing its efficacy,developing novel sonosensitizer carriers with good biocompatibility pose major challenges to the clinical practice of SDT.In this study,we investigated the mechanisms of mitochondria-targeted SDT and demonstrated that it suppressed the mitochondrial electron transport chain(ETC)in pancreatic cancer cells through RNA-sequencing analysis.Based on these findings,we constructed the functional lipid droplets(LDs)(CPI-613/IR780@LDs),which combined mitochondria-targeted SDT with the tricarboxylic acid(TCA)cycle inhibitor CPI-613.CPI-613/IR780@LDs synergistically inhibited the TCA cycle and the ETC of mitochondrial aerobic respiration to reduce oxygen consumption and increase reactive oxygen species(ROS)generation at the tumor site,thus enhancing the efficacy of SDT in hypoxic pancreatic cancer.Moreover,the combination of mitochondria-targeted SDT and anti-PD-1 antibody exhibited excellent tumor inhibition and activated anti-tumor immune responses by increasing tumorinfiltrating CD8+T cells and reducing regulatory T cells,synergistically arresting the growth of both primary and metastatic pancreatic tumors.Meanwhile,lipid droplets are cell-derived biological carriers with natural mitochondrial targeting ability and can achieve efficient hydrophobic drug loading through active phagocytosis.Therefore,the functional lipid droplet-based SDT combined with anti-PD-1 antibody holds great potential in the clinical treatment of hypoxic pancreatic cancer. | Zhan Shi Xue Wang Jiali Luo Yiqing Zeng Qing Wen Yurong Hong Tao Zhang Pintong Huang | 2023 | Nano Research2023,16,5: | 0 |

| 20 | Clonal Analysis of Peripheral Papilloma and Cancerous Cells of the Breast显示文摘OBJECTIVE Because almost al malignancies represent monoclonal proliferations,we have studied the clonal status of peripheral papil omas (peri-PM),ductal carcinomas in situ(DCIS),and normal breast tissues to explore a reliable way to distinguish benign and malignant(or pre-malignant) cases previously diagnosed morphologically. METHODS Twenty-six cases of peri-PM,25 cases of peri-PM with atypical ductal hyperplasia(ADH),27 cases of DCIS,16 cases of developed canceration and 20 specimens of normal tissue were examined in the study.The clonal status of these tissues was studied using an assay based on inactivation mosaicism of the lenth-polymorphic X-chromosomes at the androgen receptor(AR)locus. RESULTS Loss of polymorphism at the AR locus was found in all DCIS cases and 10 cases(10/25,40.0%)of peri-PM with ADH,indicating the monoclonality of the tumors.Twenty-four out of 26(92.3%)cases with peri-PM and 19 specimens of normal tissue were shown to be polyclonal.In 16 cases of developed canceration,identical X chromosome inactivation(monoclonal alterations)was observed from both the peri-PM with ADH part,and the DCIS part in each case.
