| 2 | Detection and identification of Vibrio parahaemolyticus by multiplex PCR and DNA-DNA hybridization on a microarray显示文摘In this paper,we developed a rapid and accurate method for the detection of Vibrio parahaemalyticus strains,using multiplex PCR and DNA—DNA hybridization.Multiplex PCR was used to simultaneously amplify three diagnostic genes(tlh,tdh and fla) that serve as molecular markers of V.parahaemolyticus.Biotinylated PCR products were hybridized to primers immobilized on a microarray,and detected by chemiluminesce with avidin—conjugated alkaline phosphatase.With this method,forty-five samples were tested.Eight known virulent strains (tlh^+/tdh^+/fla^+) and four known avirulent strains(tlh^+/tdh^-/fla^+) of the V.parahaemolyticus were successfully detected,and no non-specific hybridization and cross-hybridization reaction were found from fifteen closely-related strains(tlh^-/tdh^-/fla^+) of the Vibrio spp.In addition,all the other eighteen strains of non-Vibrio bacteria(tlh^-/tdh^-/fla^-) gave negative results.The DNA microarray successfully distinguished V. parahaemolyticus from other Vibrio spp.The results demonstrated that this was an efficient and robust method for identifying virulent strains of V.parahaemolyticus. | Rongzhi Wang Jiadong Huang Wei Zhang Guangmei Lin Junwei Lian Libin Jiang Hongcong Lin Songfa Wang Shihua Wang | 2011 | Journal of Genetics and Genomics2011,38,3: | 11 |

| 4 | Finite-time Feedback Stabilization of a Class of Input-delay Systems With Saturating Actuators via Digital Control显示文摘In this paper, the problem of making an input-delay system with saturating actuators finite-time stable by virtue of digital control is investigated. A digital state feedback controller and digital observer-controller compensator are designed for two cases: when the state of the input-delay system are available or when it is unavailable. Sufficient conditions which guarantee finite-time stability of a closed-loop input-delay system are given and the proof procedure is presented in a heuristic way by constructing appropriate comparison functions. The condition can be transformed into the intersection of two curves satisfying some constraints, which reveals the relationship between designed parameters clearly. | 13 | Global Land Surface Climate Analysis Based on the Calculation of a Modified Bowen Ratio显示文摘A modified Bowen ratio(BRm),the sign of which is determined by the direction of the surface sensible heat flux,was used to represent the major divisions in climate across the globe,and the usefulness of this approach was evaluated. Five reanalysis datasets and the results of an offline land surface model were investigated. We divided the global continents into five major BRm zones using the climatological means of the sensible and latent heat fluxes during the period 1980–2010:extremely cold,extremely wet,semi-wet,semi-arid and extremely arid. These zones had BRm ranges of(-∞,0),(0,0.5),(0.5,2),(2,10) and(10,+∞),respectively. The climatological mean distribution of the Bowen ratio zones corresponded well with the K ¨oppen-like climate classification,and it reflected well the seasonal variation for each subdivision of climate classification. The features of climate change over the mean climatological BRm zones were also investigated. In addition to giving a map-like classification of climate,the BRm also reflects temporal variations in different climatic zones based on land surface processes. An investigation of the coverage of the BRm zones showed that the extremely wet and extremely arid regions expanded,whereas a reduction in area was seen for the semi-wet and semi-arid regions in boreal spring during the period 1980–2010. | 14 | Dataset of Comparative Observations for Land Surface Processes over the Semi-Arid Alpine Grassland against Alpine Lakes in the Source Region of the Yellow River显示文摘Thousands of lakes on the Tibetan Plateau(TP) play a critical role in the regional water cycle, weather, and climate. In recent years, the areas of TP lakes underwent drastic changes and have become a research hotspot. However, the characteristics of the lake-atmosphere interaction over the high-altitude lakes are still unclear, which inhibits model development and the accurate simulation of lake climate effects. The source region of the Yellow River(SRYR) has the largest outflow lake and freshwater lake on the TP and is one of the most densely distributed lakes on the TP. Since 2011,three observation sites have been set up in the Ngoring Lake basin in the SRYR to monitor the lake-atmosphere interaction and the differences among water-heat exchanges over the land and lake surfaces. This study presents an eight-year(2012–19), half-hourly, observation-based dataset related to lake–atmosphere interactions composed of three sites. The three sites represent the lake surface, the lakeside, and the land. The observations contain the basic meteorological elements,surface radiation, eddy covariance system, soil temperature, and moisture(for land). Information related to the sites and instruments, the continuity and completeness of data, and the differences among the observational results at different sites are described in this study. These data have been used in the previous study to reveal a few energy and water exchange characteristics of TP lakes and to validate and improve the lake and land surface model. | 15 | iHypoxia:An Integrative Database of Protein Expression Dynamics in Response to Hypoxia in Animals显示文摘Mammals have evolved mechanisms to sense hypoxia and induce hypoxic responses.Recently,high-throughput techniques have greatly promoted global studies of protein expression changes during hypoxia and the identification of candidate genes associated with hypoxiaadaptive evolution,which have contributed to the understanding of the complex regulatory networks of hypoxia.In this study,we developed an integrated resource for the expression dynamics of proteins in response to hypoxia(iHypoxia),and this database contains 2589 expression events of 1944 proteins identified by low-throughput experiments(LTEs)and 422,553 quantitative expression events of 33,559 proteins identified by high-throughput experiments from five mammals that exhibit a response to hypoxia.Various experimental details,such as the hypoxic experimental conditions,expression patterns,and sample types,were carefully collected and integrated.Furthermore,8788 candidate genes from diverse species inhabiting low-oxygen environments were also integrated.In addition,we conducted an orthologous search and computationally identified 394,141 proteins that may respond to hypoxia among 48 animals.An enrichment analysis of human proteins identified from LTEs shows that these proteins are enriched in certain drug targets and cancer genes.Annotation of known posttranslational modification(PTM)sites in the proteins identified by LTEs reveals that these proteins undergo extensive PTMs,particularly phosphorylation,ubiquitination,and acetylation.iHypoxia provides a convenient and user-friendly method for users to obtain hypoxia-related information of interest. | Ze-Xian Liu Panqin Wang Qingfeng Zhang Shihua Li Yuxin Zhang Yutong Guo Chongchong Jia Tian Shao Lin Li Han Cheng Zhenlong Wang | 2023 | Genomics, Proteomics & Bioinformatics2023,21,2: | 0 |
| 16 | Numerical simulation of the climate effect of high-altitude lakes on the Tibetan Plateau显示文摘Lakes regulate the water and heat exchange between the ground and the atmosphere on different temporal and spatial scales. However, studies of the lake effect in the high-altitude Tibetan Plateau(TP) rarely have been performed until recently, and little attention has been paid to modelling of frozen lakes. In this study, the Weather Research and Forecasting Model(WRF v. 3.6.1) is employed to conduct three numerical experiments in the Ngoring Lake Basin(the original experiment, an experiment with a tuned model, and a no-lake experiment) to investigate the influences of parameter optimization on the lake simulation and of the high-altitude lake on the regional climate. After the lake depth, the roughness lengths, and initial surface temperature are corrected in the model, the simulation of the air temperature is distinctly improved. In the experiment using a tuned model, the simulated sensible-heat flux(H) is clearly improved, especially during periods of ice melting(from late spring to early summer) and freezing(late fall). The improvement of latent-heat flux(LE) is mainly manifested by the sharp increase in the correlation coefficient between simulation and observation, whereas the improvement in the average value is small. The optimization of initial surface temperature shows the most prominent effect in the first year and distinctly weakens after a freezing period. After the lakes become grassland in the model, the daytime temperature clearly increases during the freezing and melting periods; but the nocturnal cooling appears in other stages, especially from September to October. The annual mean H increases by 6.4 times in the regions of the Ngoring Lake and the Gyaring Lake, and the LE declines by 56.2%. The sum of H and LE increases from 71.2 W/m2(with lake) to 84.6 W/m2(no lake). For the entire simulation region, the sum of H and LE also increases slightly.
