3Discovery of CECR2 Bromodomain Inhibitors with High Selectivities over BPTF Bromodomain显示文摘Cat's eye syndrome chromosome candidate 2 bromodomain(CECR2 BRD)and Bromodomain PHD transcription factor bromodomain(BPTF BRD)are the same subfamily proteins,both of which are highly conserved in sequence and binding pockets.Challenges remain in the development of small molecules to inhibit one of the two bromodomains(BRDs),in view of each subtype may possess unique physiological and pathological functions.There is still a lack of effective selective inhibitors of CECR2 BRD,which makes it difficult to fully understand the pathogenesis of CECR2-BRD in diseases,especially cancers.Herein,we report our efforts to discover a series of highly selective CECR2 BRD inhibitors over BPTF BRD based on TP-248.Structure-based molecular optimization led to the discovery of DC-CEi-26,whose IC_(50) for CECR2 BRD was 96.7±14.9 nmol/L and selectivity was up to 590×over BPTF BRD.DC-CEi-26 showed weak potencies for other classic BRDs in different subfamily,which may serve as a chemical probe for CECR2 BRD biological research.Haibo Lu Shijia Zu Zhe Duan Yueyao Feng Jie Wang Jingyi Ma Qi Li Dongying Chen Bo Li Kaixian Chen Cheng Luo Jin Lin Tian Lu Hua Lin 2022Chinese Journal of Chemistry2022,40,17:0
4Metabolome profiling of stratified seeds provides insight into the regulation of dormancy in Davidia involucrata显示文摘Dove tree(Davidia involucrata),a tertiary vestige species,is well-adapted to cool conditions.Dormancy in D.involucrata seed lasts for an extremely long period of time,typically between 3 and 4 years,and this characteristic makes the species an excellent model for studying the mechanisms of seed dormancy.The molecular mechanisms governing germination control in D.involucrata are still unknown.Seed stratification have been reported to enhance germination in recalcitrant seeds.We performed a widely targeted metabolome profiling to identify metabolites and associated pathways in D.involucrata seeds from six different moist sand stratification durations(0-30 months) using the ultra-high-performance liquid chromatography-Q Exactive Orbitrap-Mass spectrometry.There was an increasing germination rate with prolonged stratification durations(12-30 months).Furthermore,we detected 10,008 metabolites in the stratified seeds.We also detected 48 differentially accumulated metabolites(DAMs) between all stratification periods in the seeds,with 10 highly conserved metabolites.Most of the differentially accumulated metabolites between unstratified and stratified seeds were enriched in purine metabolism,pyrimidine metabolism,flavone and flavonol biosynthesis,phenylpropanoid biosynthesis,and arginine biosynthesis pathways.Key phytohormones,abscisic acid,indole-3 acetic acid,and sinapic acid were differentially accumulated in the seeds and are predicted to regulate dormancy in D.involucrata.We have provided extensive metabolic information useful for future works on dove tree germination study.Shiming Deng Qiang Xiao Cigui Xu Jian Hong Zhijun Deng Dan Jiang Shijia Luo 2022Plant Diversity2022,44,4:0

6Global multifaceted biodiversity patterns,centers,and conservation needs in angiosperms显示文摘The Convention on Biological Diversity seeks to conserve at least 30%of global land and water areas by 2030,which is a challenge but also an opportunity to better preserve biodiversity,including flowering plants(angiosperms).Herein,we compiled a large database on distributions of over 300,000 angiosperm species and the key functional traits of 67,024 species.Using this database,we constructed biodiversity-environment models to predict global patterns of taxonomic,phylogenetic,and functional diversity in terrestrial angiosperms and provide a comprehensive mapping of the three diversity facets.We further evaluated the current protection status of the biodiversity centers of these diversity facets.Our results showed that geographical patterns of the three facets of plant diversity exhibited substantial spatial mismatches and nonoverlapping conservation priorities.Idiosyncratic centers of functional diversity,particularly of herbaceous species,were primarily distributed in temperate regions and under weaker protection compared with other biodiversity centers of taxonomic and phylogenetic facets.Our global assessment of multifaceted biodiversity patterns and centers highlights the insufficiency and unbalanced conservation among the three diversity facets and the two growth forms(woody vs.herbaceous),thus providing directions for guiding the future conservation of global plant diversity.Ao Luo Yaoqi Li Nawal Shrestha Xiaoting Xu Xiangyan Su Yichao Li Tong Lyu Kilara Waris Zhiyao Tang Xiaojuan Liu Luxiang Lin Yongsheng Chen Kuiling Zu Wenqi Song Shijia Peng Niklaus E.Zimmermann Loïc Pellissier Zhiheng Wang 2024Science China(Life Sciences)2024,67,4:0
