2The Genesis Mechanism of the Mantle Fluid Action and Evolution in the Ore-Forming Process: A Case Study of the Laowangzhai Gold Deposit in Yunnan, China显示文摘Based on petrological studies of the wall rocks, mineralizing rocks, ores and veins from the Laowangzhai gold deposit, it is discovered that along with the development of silication, carbonation and sulfidation, a kind of black opaque ultra-microlite material runs through the spaces between grains, fissures and cleavages. Under observations of the electron microprobe, scanning electronic microscopy and energy spectrum, this kind of ultra-microlite material is confirmed to consist of ultra microcrystalline quartz, silicate, sulfides and carbonates, as well as rutile, scheelite and specularite (magnetite), showing characters of liquation by the analyses of SEM and energy spectrum. The coexistence of immiscibility and precipitating co-crystallization strongly suggests that the mineralizing fluid changed from the melt to the hydrothermal fluid. Combined with the element geochemical researches, it is realized that the ultra-microlite aggregate is the direct relics of the mantle fluid behaving like a melt and supercritical fluid, which goes along with the mantle-derived magma and will escape from the magma body at a proper time. 3MicroRNA-24 promotes pancreatic beta cells toward dedifferentiation to avoid endoplasmic reticulum stress-induced apoptosis显示文摘Current research indicates that beta cell loss in type 2 diabetes may be attributed to beta cell dedifferentiation rather than apoptosis;however,the mechanisms by which this occurs remain poorly understood.Our previous study demonstrated that elevation of microRNA-24(miR-24)in a diabetic setting caused beta cell dysfunction and replicative deficiency.In this study,we focused on the role of miR-24 in beta cell apoptosis and dedifferentiation under endoplasmic reticulum(ER)stress conditions.We found that miR-24 overabundance protected beta cells from thapsigargin-induced apoptosis at the cost of accelerating the impairment of glucosestimulated insulin secretion(GSIS)and enhancing the presence of dedifferentiation markers.Ingenuity?Pathway Analysis(IPA)revealed that elevation of miR-24 had an inhibitory effect on XBP1 and ATF4,which are downstream effectors of two key branches of ER stress,by inhibiting its direct target,Irela.Notably,elevated miR-24 initiated another pathway that targeted Mafa and decreased GSIS function in surviving beta cells,thus guiding their dedifferentiation under ER stress conditions.Our results demonstrated that the elevated miR-24,to the utmost extent,preserves beta cell mass by inhibiting apoptosis and inducing dedifFerentiation.This study not only provides a novel mechanism by which miR-24 dominates beta cell turnover under persistent metabolic stress but also offers a therapeutic consideration for treating diabetes by inducing dedifferentiated beta cells to re-differentiation.Yunxia Zhu Yi Sun Yuncai Zhou Yan Zhang Tao Zhang Yating Li Weiyan You Xiaoai Chang Li Yuan Xiao Han 2019Journal of Molecular Cell Biology2019,11,9:9
4The effect of Fe-Mn minerals and seawater interface and enrich-ment mechanism of ore-forming elements of polymetallic crusts and nodules from the South China Sea显示文摘Ferromanganese crusts and nodules are important submarine mineral resources that contain various metal elements with significant economic value. In this study, polymetallic crusts and nodules obtained from the South China Sea(SCS) were determined by using X-ray power diffraction(XRD), Raman spectroscopy(RS), Fourier transform infrared spectroscopy(FTIR), and X-ray photoelectron spectroscopy(XPS) to systematically investigate and analyze the mineralogical and spectral characteristics of the Fe-Mn minerals. XRD measurements revealed that the SCS polymetallic crusts and nodules were composed of vernadite, quartz, and plagioclase. The nodules also contained todorokite. The Fe-phase minerals of the SCS crusts and nodules were composed of amorphous Fe oxide/hydroxide, and the Mn-and Fe-phases minerals exhibited relatively poor degrees of crystallization. FTIR results showed that the Fe-Mn minerals in the crusts and nodules included a large number of surface hydroxyl groups. These surface hydroxyl groups contained protons that could provide reactive sites for complexation of ore-forming elements in seawater. XPS results indicated that the surfaces of the Fe-Mn minerals mainly contained Fe, Mn, and O. Fe was present in the trivalent oxidation state, while Mn, which may contain several bivalent oxidation state, was present in the tetravalent and trivalent oxidation states. The SCS polymetallic crusts and nodules were compared with Pacific seamount crusts, and results showed that the surface hydroxyl(-OH) groups of the SCS crusts and nodules numbered more than the lattice oxygen(O^(2-)). But the lattice oxygen of Pacific seamount crusts numbered more than the surface hydroxyl groups. This characteristic indicated that the degree of crystallization of Fe-Mn minerals from the Pacific Ocean was higher than that of minerals from the South China Sea. 9Placement of dual capsular tension rings for the combined management of traumatic cyclodialysis cleft and zonular dialysis显示文摘Background:To evaluate the efficacy and safety of placing dual capsular tension rings for the combined management of traumatic cyclodialysis cleft and zonular dialysis.Methods:A modified capsular tension ring was inserted into the ciliary sulcus and a capsular tension ring or modified capsular tension ring was inserted into the capsular bag in 20 eyes in 20 consecutive patients showing signs of ocular hypotony and ectopia lentis.Outcome measures included intraocular pressure,best-corrected visual acuity,and postoperative complications.Results:Dual capsular tension ring placement was performed in 20 patients with a mean age of 48.7 years.The cyclodialysis cleft extended over 2.9 clock hours(range 0.5-6.5).The modified capsular tension ring was successfully inserted into the ciliary sulcus and a capsular tension ring or modified capsular tension ring in the capsular bag in all eyes.At the last follow-up,the cyclodialysis cleft was closed in 16/20(80.0%)eyes.The intraocular lens was stable in all patients postoperatively.Best-corrected visual acuity,in terms of the logarithm of the minimal angle of resolution,improved from 1.3±0.8 before surgery to 0.4±0.3 after surgery(P<0.001).Intraocular pressure increased significantly from 10.6±3.2 mmHg before surgery to 13.0±4.8 mmHg after surgery(P=0.040).Postoperative complications included a painful reversible intraocular pressure spike in four patients(20.0%).Logistic regression revealed no significant factors associated with successful cleft closure and a stable final intraocular pressure of≥10 mmHg.Conclusions:The placement of two capsular tension rings into the ciliary sulcus and the capsular bag is a safe,successful procedure combined for repairing a traumatic cyclodialysis cleft and managing zonular dialysis.Jiahui Chen Lina Lan Yating Tang Yi Lu Yongxiang Jiang 2020Eye and Vision2020,7,1:1

11Mapping of individual sensory nerve axons from digits to spinal cord with the transparent embedding solvent system显示文摘Achieving uniform optical resolution for a large tissue sample is a major challenge for deep imaging.For conventional tissue clearing methods,loss of resolution and quality in deep regions is inevitable due to limited transparency.Here we describe the Transparent Embedding Solvent System(TESOS)method,which combines tissue clearing,transparent embedding,sectioning and block-face imaging.We used TESOS to acquire volumetric images of uniform resolution for an adult mouse whole-body sample.The TESOS method is highly versatile and can be combined with different microscopy systems to achieve uniformly high resolution.With a light sheet microscope,we imaged the whole body of an adult mouse,including skin,at a uniform 0.8×0.8×3.5μm^(3) voxel resolution within 120 h.With a confocal microscope and a 40×/1.3 numerical aperture objective,we achieved a uniform sub-micron resolution in the whole sample to reveal a complete projection of individual nerve axons within the central or peripheral nervous system.Furthermore,TESOS allowed the first mesoscale connectome mapping of individual sensory neuron axons spanning 5 cm from adult mouse digits to the spinal cord at a uniform sub-micron resolution.Yating Yi Youqi Li Shiwen Zhang Yi Men Yuhong Wang Dian Jing Jiayi Ding Qingjie Zhu Zexi Chen Xingjun Chen Jun-Liszt Li Yilong Wang Jun Wang Hanchuan Peng Li Zhang Wenjing Luo Jian Q.Feng Yongwen He Woo-Ping Ge Hu Zhao 2024Cell Research2024,34,2:0
12Engineering the thermostability of D-lyxose isomerase from Caldanaerobius polysaccharolyticus via multiple computer-aided rational design for efficient synthesis of D-mannose显示文摘D-Mannose is an attractive functional sugar that exhibits many physiological benefits on human health.The demand for low-calorie sugars and sweeteners in foods are increasingly available on the market.Some sugar isomerases,such as D-lyxose isomerase(D-LIase),can achieve an isomerization reaction between D-mannose and D-fructose.However,the weak thermostability of D-LIase limits its efficient conversion from D-fructose to D-mannose.Nonetheless,few studies are available that have investigated the molecular modification of D-LIase to improve its thermal stability.In this study,computer-aided tools including FireProt,PROSS,and Consensus Finder were employed to jointly design D-LIase mutants with improved thermostability for the first time.Finally,the obtained five-point mutant M5(N21G/E78P/V58Y/C119Y/K170P)showed high thermal stability and cat-alytic activity.The half-life of M5 at 65◦C was 10.22 fold,and the catalytic efficiency towards 600 g/L of D-fructose was 2.6 times to that of the wild type enzyme,respectively.Molecular dynamics simulation and intramolecular forces analysis revealed a thermostability mechanism of highly rigidity conformation,newly formed hydrogen bonds andπ-cation interaction between and within protein domains,and redistributed surface electrostatic charges for the mutant M5.This research provided a promising D-LIase mutant for the industrial production of D-mannose from D-fructose.Hao Wu Ming Yi Xiaoyi Wu Yating Ding Minghui Pu Li Wen Yunhui Cheng Wenli Zhang Wanmeng Mu 2023Synthetic and Systems Biotechnology2023,8,2:0
13Preoperative Characteristics of Ocular Biometry in Children with Unilateral Congenital Cataracts显示文摘The ocular biometry characteristics are clinically significant for children with unilateral congenital cataracts,but there is a lack of data analysis concerning the preoperative measurements.The axial length(AL),mean keratometry(Km),cor-neal astigmatism(CA),and the anterior chamber depth(ACD)from both eyes before cataract surgery were obtained from 205 patients(410 eyes,3-15 years of age)with unilateral congenital cataracts.In the congenital cataract eyes,shorter AL(22.44±1.52 mm vs.22.57±1.04 mm,p=0.036)and higher CA(-1.89±0.91 D vs.-1.24±0.67 D,p<0.001)were found,and no significant difference was found in the Km and the ACD measurements compared to the contralateral normal eyes.Females had shorter AL and shallower ACD compared to males.However,the Km and CA in the females were significantly larger than that in males.Shorter AL,larger Km,higher CA,and shallower ACD were also found in females who had a binocular axial difference(the value obtained by subtraction of the contralateral normal eye from the congenital cataract eye)that less than zero.The preoperative ocular biometry of shorter AL,larger Km,higher CA,and shallower ACD should be considered in females with unilateral congenital cataracts.The age and the binocular axial differences had a statistically significant correlation(r=-0.192,p=0.006).Therefore,changes in the binocular axial differences associated with aging may enhance the guidelines for intraocular lens selection and the management of congenital cataracts.Jiahui Chen Yating Tang Yongxiang Jiang Yi Lu 2022Phenomics2022,2,2:0

15A Genome‑Wide Association Study for Susceptibility to Axial Length in Highly Myopic Eyes显示文摘High myopia has long been highly prevalent worldwide with a largely yet unexplained genetic contribution.To identify novel susceptibility genes for axial length(AL)in highly myopic eyes,a genome-wide association study(GWAS)was performed using the genomic dataset of 350 deep whole-genome sequencing data from highly myopic patients.Top single nucleotide polymorphisms(SNPs)were functionally annotated.Immunofluorescence staining,quantitative polymerase chain reaction,and western blot were performed using neural retina of form-deprived myopic mice.Enrichment analyses were further performed.We identified the four top SNPs and found that ADAM Metallopeptidase With Thrombospondin Type 1 Motif 16(ADAMTS16)and Phosphatidylinositol Glycan Anchor Biosynthesis Class Z(PIGZ)had the potential of clinical signifi-cance.Animal experiments confirmed that PIGZ expression could be observed and showed higher expression level in form-deprived mice,especially in the ganglion cell layer.The messenger RNA(mRNA)levels of both ADAMTS16 and PIGZ were significantly higher in the neural retina of form-deprived eyes(p=0.005 and 0.007 respectively),and both proteins showed significantly upregulated expression in the neural retina of deprived eyes(p=0.004 and 0.042,respectively).Enrichment analysis revealed a significant role of cellular adhesion and signal transduction in AL,and also several AL-related pathways including circadian entrainment and inflammatory mediator regulation of transient receptor potential channels were proposed.In conclusion,the current study identified four novel SNPs associated with AL in highly myopic eyes and confirmed that the expression of ADAMTS16 and PIGZ was significantly upregulated in neural retina of deprived eyes.Enrichment analyses provided novel insight into the etiology of high myopia and opened avenues for future research interest.Qiang Lu Yu Du Ye Zhang Yuxi Chen Hao Li Wenwen He Yating Tang Zhennan Zhao Yinglei Zhang Jihong Wu Xiangjia Zhu Yi Lu 2023Phenomics2023,3,3:0
16Analysis of the Freezing Damage to Biennial Eucalyptus dunnii and Eucalyptus smithii in Haikou Forest Farm显示文摘In winter of 2013,cold weather occurred in Kunming,and biennial E. dunnii and E. smithii in Haikou Forest Farm suffered different degrees of freezing damage. The freezing damage to them was analyzed here. The results showed that in the families of E. dunnii,families suffering severe and especially severe freezing damage accounted for 7. 7%,while 92. 3% of families suffered moderate and below freezing damage; in the families of E. smithii,56. 7% of families suffered severe and especially severe freezing damage,while families suffering moderate and below freezing damage accounted for 43. 3%. There was significant difference between two species of eucalyptuses in tree height( P <0. 05),while there was extremely significant difference in freezing damage grade and index( P <0. 01). There was significantly negative correlation between the tree height and freezing damage grade of E. dunnii and E. smithii,with the correlation coefficients of- 0. 374 and- 0. 234. 17Selective fluorescence labeling and sensitive determination of Staphylococcus aureus by microchip electrophoresis with a multiple-concentration approach显示文摘The detection of Staphylococcus aureus(S. aureus) is very important as it is responsible for bacterial infectious diseases and food poisoning. In this paper we explored the application of fluorescently labelled vancomycin to specifically bind and detect S. aureus. In view of the specificity of vancomycin towards bacterial cell surfaces, we utilized Cy5 to label vancomycin(Cy5-Van) for the identification of S. aureus.Our experiments were designed to examine in greater detail the specificity of the reaction between Cy5-Van and S. aureus. Detection parameters such as the derivatization conditions, concentrations of buffer,p H value, response performance of Cy5-Van to bacterial surface, injection time and reversed-polarity time have been investigated and optimized. To develop a simple and quick assay for the detection of S. aureus at low concentrations, we propose to use the Cy5-Van for labeling the S. aureus coupled with an on-line multiple-concentration in microchip electrophoresis. Under the optimized conditions, the detection of S. aureus was achieved within 150 s with limit of detection(S/N = 3) of 981 CFU/mL, and 350-fold enhancement was obtained for S. aureus as compared to using the no concentration step. 19Three Novel Mutations of Microphthalmos Identified in Two Chinese Families显示文摘Genetic alterations are a major cause of microphthalmos,while novel-related genes and mutations in microphthalmos have rarely been explored.To identify the underlying genetic defect responsible for microphthalmos eyes in two three-generation Chinese families,we screened 425 genes involved in common inherited non-syndromic eye diseases with next-generation sequencing-based target capture sequencing of the two probands of two three-generation Chinese families diagnosed with microphthalmos.Variants were filtered and analyzed to identify possible disease-causing variants before Sanger sequencing validation.We enrolled two families with microphthalmos(Family 1:microphthalmos with congenital ocular coloboma and Family 2:simple microphthalmos).Two novel heterozygous mutations,Peroxidasin(PXDN)c.3165C>T(p.Pro1055Pro)and PXDN c.2640C>G(p.Arg880Arg),were found in Family 1,and Crystallin Beta B2(CRYBB2)c.481G>A(p.Gly161Arg)was found in Family 2,but none of the mutations were found in the unaffected individuals,who were phenotypically nor-mal.Multiple orthologous sequence alignment(MSA)revealed that the CRYBB2 p.Gly161Arg mutation was a deleterious effect mutation.In conclusion,the three novel mutations found in our study extend our current understanding of the genetic basis of microphthalmos and provide early pre-symptomatic diagnosis and emphasize the significance of genetic diagnosis of microphthalmos.Yating Tang Jie Xu Yi Lu Tianyu Zheng 2022Phenomics2022,2,4:0
20Nuclear localization of human MEIOB requires its NLS in the OB domain and interaction with SPATA22显示文摘MEIOB is a vital protein in meiotic homologous recombination and plays an indispensable role in human gametogenesis.In mammals,MEIOB and its partner SPATA22 form a heterodimer,ensuring their effective localization on single-strand DNA(ssDNA)and proper synapsis processes.Mutations in human MEIOB(hMEIOB)cause human infertility attributed to the failure of its interaction with human SPATA22(hSPATA22)and ssDNA binding.However,the detailed mechanism is still unclear.In our study,truncated or full-length hMEIOB and hSPATA22 are traced by fused expression with fluorescent proteins(i.e.,copGFP or mCherry),and the live cell imaging system is used to observe the expression and localization of the proteins.When transfected alone,hMEIOB accumulates in the cytoplasm.Interestingly,a covered NLS in the OB domain of hMEIOB is identified,which can be exposed by hSPATA22 and is necessary for the nuclear localization of hMEIOB.When hSPATA22 loses its hMEIOB interacting domain or NLS,the nuclear localization of hMEIOB is aborted.Collectively,our results prove that the NLS in the OB domain of hMEIOB and interaction with hSPATA22 are required for hMEIOB nuclear localization.Yating Xu Zhen Chen Panfeng Wu Wei Qu Hanqing Shi Muyang Cheng Yumin Xu Tingyu Jin Cong Liu Cong Liu Yi Li Mengcheng Luo 2023Acta Biochimica et Biophysica Sinica2023,55,1:0
