1Association of assisted reproductive technology, germline de novo mutations and congenital heart defects in a prospective birth cohort study显示文摘Emerging evidence suggests that children conceived through assisted reproductive technology(ART)have a higher risk of congenital heart defects(CHDs)even when there is no family history.De novo mutation(DNM)is a well-known cause of sporadic congenital diseases;however,whether ART procedures increase the number of germline DNM(gDNM)has not yet been well studied.Here,we performed whole-genome sequencing of 1137 individuals from 160 families conceived through ART and 205 families conceived spontaneously.Children conceived via ART carried 4.59 more gDNMs than children conceived spontaneously,including 332 paternal and 1.26 maternal DNMs,after correcting for parental age at conception,cigarette smoking,alcohol drinking,and exercise behaviors.Paternal DNMs in offspring conceived via ART are characterized by C>T substitutions at CpG sites,which potentially affect protein-coding genes and are significantly associated with the increased risk of CHD.In addition,the accumulation of non-coding functional mutations was independently associated with CHD and 87.9% of the mutations were originated from the father.Among ART offspring,infertility of the father was associated with elevated paternal DNMs;usage of both recombinant and urinary follicle-stimulating hormone and high-dosage human chorionic gonadotropin trigger was associated with an increase of maternal DNMs.In sum,the increased gDNMs in offspring conceived by ART were primarily originated from fathers,indicating that ART itself may not be a major reason for the accumulation of gDNMs.Our findings emphasize the importance of evaluating the germline status of the fathers in families with the use of ART.Cheng Wang Hong Lv Xiufeng Ling Hong Li Feiyang Diao Juncheng Dai Jiangbo Du Ting Chen Qi Xi Yang Zhao Kun Zhou Bo Xu Xiumei Han Xiaoyu Liu Meijuan Peng Congcong Chen Shiyao Tao Lei Huang Cong Liu Mingyang Wen Yangqian Jiang Tao Jiang Chuncheng Lu Wei Wu Di Wu Minjian Chen Yuan Lin Xuejiang Guo Ran Huo Jiayin Liu Hongxia Ma Guangfu Jin Yankai Xia Jiahao Sha Hongbing Shen Zhibin Hu 2021Cell Research2021,31,8:2
2Isolation and Identification of Root Rot Fungus of Astragalus membranaceus显示文摘The perennial root of Astragalus membranaceus is used as a medicine,while root rot is a main factor causing reduction of quality and commodity value of A. membranaceus. The screening and research of the pathogenic species and their characteristics could provide theoretical and practical basis for the control of this disease. A pathogenic strain was isolated and purified from the root part of four-year-old A. membranaceus,and identified by morphological and molecular biological methods as Fusarium oxysporum. 3A nomogram predicting clinical pregnancy in the first fresh embryo transfer for women undergoing in vitro fertilization and intracytoplasmic sperm injection(IVF/ICSI) treatments显示文摘The extent to which factors affect the probability of clinical pregnancy in the first fresh embryo transfer after assisted conception is unknown.In order to examine the predictors of clinical pregnancy,a retrospective cohort study was launched between January 1,2013 and December 31,2016 in four infertility clinics including 19837 in vitro fertilization and intracytoplasmic sperm injection(IVF/ICSI)fresh cycles with known outcomes and relevant records.A multivariable logistic regression was used to select the most significant predictors in the final nomogram for predicting clinical pregnancy.Furthermore,the model was validated by an independent validation set and the performance of the model was evaluated by the receiver operating characteristic(ROC)curves along with the area under the ROC curve(AUC)and calibration plots.In a training set including 17854 participants,we identified that female age,tubal factor,number of embryos transferred,endometrial thickness and number of good-quality embryos were independent predictors for clinical pregnancy.We developed a nomogram using these five factors and the predictive ability was 0.66 for AUC(95%CI=0.64−0.68),which was independently validated in the validation set(AUC=0.66,95%CI=0.65−0.68).Our results show that some specific factors can be used to provide infertile couples with an accurate assessment of clinical pregnancy following assisted conception and facilitate to guide couples and clinicians.Fang Wu Feng Liu Yichun Guan Jiangbo Du Jichun Tan Hong Lv Qun Lu Shiyao Tao Lei Huang Kun Zhou Yankai Xia Xinru Wang Hongbing Shen Xiufeng Ling Feiyang Diao Zhibin Hu Guangfu Jin 2019The Journal of Biomedical Research2019,33,6:1
4Wet-chemical synthesis and applications of amorphous metalcontaining nanomaterials显示文摘In the past decades,metal-containing nanomaterials have attracted increasing interests owing to their intriguing physicochemical properties and various promising applications.Recent research has revealed that the phase of metal-containing nanomaterials could significantly affect their properties and functions.In particular,nanomaterials with amorphous phase,which possess long-range disordered atomic arrangements,and the amorphous/crystalline heterophase nanostructures comprised of both amorphous and crystalline phases,have exhibited superior performance in various applications,e.g.,catalysis and energy storage.In this review,a brief overview of the recent progress on the wet-chemical synthesis and applications of amorphous and amorphous/crystalline heterophase metal-containing nanomaterials has been provided.Subsequently,on the basis of different categories of metal-containing nanomaterials,including metals,metal alloys,and metal compounds,their synthetic routes and promising applications will be highlighted.Finally,current challenges and some personal perspectives in this emerging research field will be proposed.Jinzhe Liang Yiyao Ge Zhen He Qinbai Yun Guigao Liu Shiyao Lu Li Zhai Biao Huang Hua Zhang 2023Nano Research2023,16,4:0
5Synthesis of amorphous Pd-based nanocatalysts for efficient alcoholysis of styrene oxide and electrochemical hydrogen evolution显示文摘Amorphous nanomaterials with long-range disordered structures could possess distinct properties and promising applications,especially in catalysis,as compared with their conventional crystalline counterparts.It is imperative to achieve the controlled preparation of amorphous noble metal-based nanomaterials for the exploration of their phase-dependent applications.Here,we report a facile wet-chemical reduction strategy to synthesize various amorphous multimetallic Pd-based nanomaterials,including PdRu,PdRh,and PdRuRh.The phase-dependent catalytic performances of distinct Pd-based nanomaterials towards diverse catalytic applications have been demonstrated.Specifically,the usage of PdRu nanocatalysts with amorphous and crystalline face-centered cubic(fcc)phases can efficiently switch the ring-opening route of styrene oxide to obtain different products with high selectivity through alcoholysis reaction and hydrogenation reaction,respectively.Moreover,when used as an electrocatalyst for hydrogen evolution reaction(HER),the synthesized amorphous PdRh nanocatalyst exhibits low overpotential and high turnover frequency values,outperforming its crystalline fcc counterpart and most of the reported Pd-based HER electrocatalysts.Yiyao Ge Jingjie Ge Biao Huang Xixi Wang Guigao Liu Xiang-Huan Shan Lu Ma Bo Chen Guanghua Liu Songmo Du An Zhang Hongfei Cheng Qingbo Wa Shiyao Lu Lujiang Li Qinbai Yun Kuo Yuan Qinxin Luo Zhichuan J.Xu Yonghua Du Hua Zhang 2023Nano Research2023,16,4:0
6Neuronal K^(+)-Cl^(-) cotransporter KCC2 as a promising drug target for epilepsy treatment显示文摘Epilepsy is a prevalent neurological disorder characterized by unprovoked seizures.y-Aminobutyric acid(GABA)serves as the primary fast inhibitory neurotransmitter in the brain,and GABA binding to the GABAA receptor(GABAAR)regulates CI and bicarbonate(HCO3)influx or efflux through the channel pore,leading to GABAergic inhibition or excitation,respectively.The neuron-specific k+-cr cotransporter 2(KCC2)is essential for maintaining a low intracellular Cr concentration,ensuring GABAAR-mediated inhibition.Impaired KCC2 function results in GABAergic excitation associated with epileptic activity.Loss-offunction mutations and altered expression of KCC2 lead to elevated[CI];and compromised synaptic inhibition,contributing to epilepsy pathogenesis in human patients.KcC2 antagonism studies demonstrate the necessity of limiting neuronal hyperexcitability within the brain,as reduced KcC2 functioning leads to seizure activity.Strategies focusing on direct(enhancing KCC2 activation)and indirect KCC2 modulation(altering KCC2 phosphorylation and transcription)have proven effective in attenuating seizure severity and exhibiting anti-convulsant properties.These findings highlight KCC2 as a promising therapeutic target for treating epilepsy.Recent advances in understanding KCC2 regulatory mechanisms,particularly via signaling pathways such as WNK,PKC,BDNF,and its receptor TrkB,have led to the discovery of novel small molecules that modulate KCC2.Inhibiting WNK kinase or utilizing newly discovered KCC2 agonists has demonstrated KCC2 activation and seizure attenuation in animal models.This review discusses the role of KCC2 in epilepsy and evaluates its potential as a drug target for epilepsy treatment by exploring various strategies to regulate KCC2 activity.Erin McMoneagle Jin Zhou Shiyao Zhang Weixue Huang Sunday Solomon Josiah Ke Ding Yun Wang Jinwei Zhang 2024Acta Pharmacologica Sinica2024,45,1:0
7A non-cooperative game-based distributed optimization method for chiller plant control显示文摘The heating,ventilation,and air-conditioning(HVAC)systems account for about half of the building energy consumption.The optimization methodology access to optimal control strategies of chiller plant has always been of great concern as it significantly contributes to the energy use of the whole HVAC system.Given that conventional centralized optimization methods relying on a central operator may suffer from dimensionality and a tremendous calculation burden,and show poorer flexibility when solving complex optimization issues,in this paper,a novel distributed optimization approach is presented for chiller plant control.In the proposed distributed control scheme,both trade-offs of coupled subsystems and optimal allocation among devices of the same subsystem are considered by developing a double-layer optimization structure.Non-cooperative game is used to mathematically formulate the interaction between controlled components as well as to divide the initial system-scale nonlinear optimization problem into local-scale ones.To solve these tasks,strategy updating mechanisms(PSO and IPM)are utilized.In this way,the approximate global optimal controlled variables of devices in the chiller plant can be obtained in a distributed and local-knowledge-enabled way without neither global information nor the central workstation.Furthermore,the existence and effectiveness of the proposed distributed scheme were verified by simulation case studies.Simulation results indicate that,by using the proposed distributed optimization scheme,a significant energy saving on a typical summer day can be obtained(1809.47 kW·h).The deviation from the central optimal solution is 3.83%.Shiyao Li Yiqun Pan Qiujian Wang Zhizhong Huang 2022Building Simulation2022,15,6:0
8Mobile Phone-Based Population Flow Data for the COVID-19 Outbreak in China' Mainland显示文摘Background.Human migration is one of the driving forces for amplifying localized infectious disease outbreaks into widespread epidemics.During the outbreak of COVID-19 in China,the travels of the population from Wuhan have furthered the spread of the virus as the period coincided with the world’s largest population movement to celebrate the Chinese New Year.Methods.We have collected and made public an anonymous and aggregated mobility dataset extracted from mobile phones at the national level,describing the outflows of population travel from Wuhan.We evaluated the correlation between population movements and the virus spread by the dates when the number of diagnosed cases was documented.Results.From Jan 1 to Jan 22 of 2020,a total of 20.2 million movements of at-risk population occurred from Wuhan to other regions in China.A large proportion of these movements occurred within Hubei province(84.5%),and a substantial increase of travels was observed even before the beginning of the official Chinese Spring Festival Travel.The outbound flows from Wuhan before the lockdown were found strongly correlated with the number of diagnosed cases in the destination cities(log-transformed).Conclusions.The regions with the highest volume of receiving at-risk populations were identified.The movements of the at-risk population were strongly associated with the virus spread.These results together with province-by-province reports have been provided to governmental authorities to aid policy decisions at both the state and provincial levels.We believe that the effort in making this data available is extremely important for COVID-19 modelling and prediction.Xin Lu Jing Tan Ziqiang Cao Yiquan Xiong Shuo Qin Tong Wang Chunrong Liu Shiyao Huang Wei Zhang Laurie B.Marczak Simon I.Hay Lehana Thabane Gordon H.Guyatt Xin Sun 2021Health Data Science2021,,1:0
9Cumulative live birth rates of in vitro fertilization/intracytoplasmic sperm injection after multiple complete cycles in China显示文摘There were few studies of cumulative live birth rates(CLBRs) based on multicenter reproductive clinical data from the general Chinese population.Here we report a retrospective cohort study,including 14 311 women with17 315 cycles,in three reproductive centers to evaluate two estimated parameters of CLBRs with multiple transfer cycles of in vitro fertilization(IVF)/intracytoplasmic sperm injection(ICSI) in a Chinese population.We found that CLBRs were related to female age and endometrial thickness.By the fourth transfer cycle,the conservative and optimal estimates of CLBRs were 52.95% and 77.30% in women under 30 years of age,and 18.17% and26.51% in those 37 years of age or older,respectively.The two estimates were 44.70% and 63.15% in women with endometrial thickness more than 7 mm,and 32.05% and 46.18% in those with less than 7 mm,respectively.In addition,body mass index(BMI),duration of infertility,and infertility diagnoses may also be related to CLBRs on certain conditions.The findings from this study on CLBRs after multiple transfer cycles of IVF/ICSI treatment on different conditions in the Chinese population should be beneficial to both infertile couples and clinicians.Lei Huang Qun Lu Jiangbo Du Hong Lv Shiyao Tao Shiyao Chen Xiuzhu Li Xiumei Han Kun Zhou Bo Xu Xiaoyu Liu Hongxia Ma Yankai Xia Guangfu Jin Hongbing Shen Xiufeng Ling Zhibin Hu Jichun Tan Feiyang Diao 2020The Journal of Biomedical Research2020,34,5:0
