4Beclin 1 cleavage by caspase-3 inactivates autophagy and promotes apoptosis显示文摘Autophagy and apoptosis are both highly regulated biological processes that play essential roles in tissue homeostasis,development and diseases.Autophagy is also described as a mechanism of death pathways,however,the precise mechanism of how autophagy links to cell death remains to be fully understood.Beclin 1 is a dual regulator for both autophagy and apoptosis.In this study we found that Beclin 1 was a substrate of caspase-3 with two cleavage sites at positions 124 and 149,respectively.Furthermore,the autophagosome formation occurred,followed by the appearance of morphological hallmarks of apoptosis after staurosporine treatment.The cleavage products of Beclin 1 reduced autophagy and promoted apoptosis in HeLa cells and the cells in which Beclin 1 was stably knocked down by specific shRNA.In addition,the cleavage of Beclin 1 resulted in abrogating the interaction between Bcl-2 with Beclin 1,which could be blocked by z-VAD-fmk.Thus,our results suggest that the cleavage of Beclin 1 by caspase-3 may contribute to inactivate autophagy leading towards augmented apoptosis.Yushan Zhu Lixia Zhao Lei Liu Ping Gao Weili Tian Xiaohui Wang Haijing Jin Haidong Xu Quan Chen 2010Protein & Cell2010,1,5:17

9Allele-aware chromosome-scale assembly of the allopolyploid genome of hexaploid Ma bamboo(Dendrocalamus latiflorus Munro)显示文摘Dendrocalamus latiflorus Munro is a woody clumping bamboo with rapid shoot growth.Both genetic transformation and clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated protein 9(Cas9)gene editing techniques are available for D.latiflorus,enabling reverse genetic approaches.Thus,D.latiflorus has the potential to be a model bamboo species.However,the genome sequence of D.latiflorus has remained unreported due to its polyploidy and large genome size.Here,we sequenced the D.latiflorus genome and assembled it into three allele-aware subgenomes(AABBCC),representingthe largest genome of a major bamboo species.We assembled 70 allelic chromosomes(2,737 Mb)for hexaploid D.latiflorus using both singlemolecule sequencing from the Pacific Biosciences(Pac Bio)Sequel platform and chromosome conformation capture sequencing(Hi-C).Repetitive sequences comprised 52.65%of the D.latiflorus genome.We annotated 135231 protein-coding genes in the genome based on transcriptomes from eight different tissues.Transcriptome sequencing using RNA-Seq and Pac Bio singlemolecule real-time long-read isoform sequencing revealed highly differential alternative splicing(AS)between non-abortive and abortive shoots,suggesting that AS regulates the abortion rate of bamboo shoots.This high-quality hexaploid genome and comprehensive strand-specific transcriptome datasets for this Poaceae family member will pave the way for bamboo research using D.latiflorus as a model species.Yushan Zheng Deming Yang Jundong Rong Liguang Chen Qiang Zhu Tianyou He Lingyan Chen Jing Ye Lili Fan Yubang Gao Hangxiao Zhang Lianfeng Gu 2022Journal of Integrative Plant Biology2022,64,3:6
10Porcine growth differentiation factor 9 gene polymorphisms and their associations with litter size显示文摘Growth differentiation factor 9 (GDF9) is expressed in oocytes and is thought to be required for ovarian folliculogenesis. Given this function, GDF9 may be considered as a candidate gene controlling pig ovulate rate. In this study, the complete coding sequence was cloned (encoding a 444 amino acid), intron sequence and partial 5′-UTR of pig GDF9. RT-PCR results showed that GDF9 mRNA is expressed in a wide range of tissues of the ruttish Erhualian pig. The expression levels of GDF9 mRNA in pituitary, ovary, uterus and ovi-duct are higher in the Erhualian pigs than those in Duroc pigs, especially in pituitary with a significant difference (P< 0.05). Comparative sequencing revealed 12 polymorphisms, including 8 single nucleotide polymorphisms (SNPs) and one 314 bp indel in noncoding regions, and the other 3 SNPs in coding regions. Four polymorphisms, G359C, C1801T, T1806C and 314 bp indel, were developed as markers for further use in population variation and association studies. The G359C polymorphism segregates only in Chinese native pigs, Erhualian and Dahuabai, on the contrary, 314 bp indel segregates only in Duroc and Landrace. C1801T and T1806C sites seem to be completely linked and segregate in Erhualian, Dahuabai and Landrace. 11Structure and antibacterial activity of Ce3+ exchanged montmorillonites显示文摘Four kinds of Ce3+ exchanged montmorillonites(Ce/MMTs) were prepared by an ion-exchange reaction, and characterized with energy dispersive X-ray analysis (EDX), X-ray difference (XRD), X-ray photoelectron spectroscopy (XPS) and scanning electron micros-copy (SEM). The surface properties and antibacterial activity of Ce/MMTs were also investigated. The chemical compositions of Ce/MMTs were determined, and the cerium of Ce/MMTs was confirmed to be present as trivalent cerium state. The d001 basal spacings of Ce/MMTs were enlarged with the enhancement of the cerium contents, and the particles were formed with irregular shape. On increasing the Ce con-tents of Ce/MMTs, the special surface areas were decreased, but the total pore volumes and the average pore sizes were increased. The anti-bacterial activity of Ce/MMTs is increased with increasing the cerium contents, and 1.5g/L of Ce/MMT-3 containing 11.46wt.% of curium could remove all the Staphylococcus aureus and more than 99.9% of the Escherichia coli within 24.0 h of contact. 12Study on Electrokinetic Remediation of Cadmium-contaminated Soil显示文摘In order to improve electrokinetic remediation efficiency of cadmium-contaminated soil,the effects of electric field intensity,remediation time and electrolyte on removal rate of total Cd in Cd-contaminated soil by electrokinetic remediation were studied through the preparation of Cd-contaminated soil and the construction of electrokinetic remediation equipment. The results showed that under the same condition,with the electric field intensity increasing from 2. 5 to 3 V/cm,the total Cd removal rate increased by 10. 62%,and with the increase of the electric field intensity from 3 to 3. 5 V/cm,the removal rate increased by 1. 87%;when the remediation time was prolonged from 72 to 96 h,the removal rate of total Cd increased by 6. 68%,and with the remediation time prolonged from 96 to120 h,the removal rate of total Cd increased by 8. 75%; and with the remediation time prolonged from 120 to 144 h,the removal rate of total Cd only increased by1. 07%. Compared with citric acid as the electrolyte,the acetic acid group improved the remediation efficiency by 12. 14% and the total energy consumption by62. 13%,while the hydrochloric acid group improved the remediation efficiency by 18. 04% and the total energy consumption by 187. 9%. 13Numerical Simulation of Transient 3-D Turbulent Heated Jet into Crossflow in a Thick-Wall T-Junction Pipe显示文摘The present work is to investigate the transient three-dimensional heated turbulent jet into crossflow in a thickwall T-junction pipe using CFD package. Two cases with the jet-to-crossflow velocity ratio of 0.05 and 0.5 are computed, with a finite-volume method utilizing κ-ε model. Comparison of the steady-state computations with measured data shows good qualitative agreement. Transient process of injection is simulated to examine the thermal shock on the T-junction component. Temporal temperature of the component is acquired by thermal coupling with the fluid. Via analysis of the flow and thermal characteristics, factors causing the thermal shock are studied. 14Transmission and mortality risk assessment of severe fever with thrombocytopenia syndrome in China:results from 11-years’study显示文摘Background: The transmission and fatal risk of severe fever with thrombocytopenia syndrome (SFTS), an emerging infectious disease first discovered in China in 2009, still needed further quantification. This research aimed to analyze the SFTS clusters and assess the transmission and mortality risk for SFTS.Methods: Both epidemiological investigation and case reports regarding SFTS clusters in China during 2011-2021 were obtained from the Public Health Emergency Information Management System of the Chinese Center for Disease Control and Prevention Information System. The transmission risk was evaluated by using the secondary attack rate (SAR) and relative risk (RR). Mortality risk factors were analyzed using a logistic regression model.Results: There were 35 SFTS clusters during 2011-2021 involving 118 patients with a fatality rate of 22.0%. The number of clusters annually increased seasonally from April to September. The clusters mainly occurred in Anhui (16 clusters) and Shandong provinces (8 clusters). The SAR through contact with blood or bloody fluids was much higher than that through contact with non-bloody fluids (50.6% vs 3.0%;χ^(2) = 210.97,P < 0.05), with anRR of 16.61 [95% confidence interval (CI): 10.23-26.97]. There was a statistically significant difference in the SAR between exposure to the blood of a deceased person during burial preparation and exposure to the living patients’ blood (66.7% vs 34.5%;χ^(2)= 6.40,P < 0.05), with anRR of 1.93 (95%CI: 1.11-3.37). The mortality risk factors were a long interval from onset to diagnosis [odds ratio (OR)= 1.385), 95%CI: 1.083-1.772,P= 0.009) and advanced age (OR: 1.095, 95%CI: 1.031-1.163,P= 0.01).Conclusions: The SFTS clusters showed a high mortality rate and resulted in a high SAR. Contact with a bleeding corpse was associated with a higher infection risk, compared with contacting the blood from living patients. 17A chromosome-level genome assembly of rugged rose (Rosa rugosa) provides insights into its evolution, ecology, and floral characteristics显示文摘Rosa rugosa,commonly known as rugged rose,is a perennial ornamental shrub.It produces beautiful flowers with a mild fragrance and colorful seed pods.Unlike many other cultivated roses,R.rugosa adapts to a wide range of habitat types and harsh environmental conditions such as salinity,alkaline,shade,drought,high humidity,and frigid temperatures.Here,we produced and analyzed a high-quality genome sequence for R.rugosa to understand its ecology,floral characteristics and evolution.PacBio HiFi reads were initially used to construct the draft genome of R.rugosa,and then Hi-C sequencing was applied to assemble the contigs into 7 chromosomes.We obtained a 382.6Mb genome encoding 39,704 protein-coding genes.The genome of R.rugosa appears to be conserved with no additional whole-genome duplication after the gamma whole-genome triplication(WGT),which occurred~100 million years ago in the ancestor of core eudicots.Based on a comparative analysis of the high-quality genome assembly of R.rugosa and other high-quality Rosaceae genomes,we found a unique large inverted segment in the Chinese rose R.chinensis and a retroposition in strawberry caused by post-WGT events.We also found that floral development-and stress response signaling-related gene modules were retained after the WGT.Two MADS-box genes involved in floral development and the stress-related transcription factors DREB2A-INTERACTING PROTEIN 2(DRIP2)and PEPTIDE TRANSPORTER 3(PTR3)were found to be positively selected in evolution,which may have contributed to the unique ability of this plant to adapt to harsh environments.In summary,the high-quality genome sequence of R.rugosa provides a map for genetic studies and molecular breeding of this plant and enables comparative genomic studies of Rosa in the near future.Fei Chen Liyao Su Shuaiya Hu Jia-Yu Xue Hui Liu Guanhua Liu Yifan Jiang Jianke Du Yushan Qiao Yannan Fan Huan Liu Qi Yang Wenjie Lu Zhu-Qing Shao Jian Zhang Liangsheng Zhang Feng Chen Zong-Ming(Max)Cheng 2021Horticulture Research2021,8,1:2

19Maximal methane potential of different animal manures collected in northwest region of China显示文摘Maximum methane potential(B0)is an important parameter used in assessing suitability of a substrate for biogas production.This study examined maximum methane potential of different manures generated from three major Chinese livestock,namely chicken,hog and cattle,and evaluated the important factors that affect the maximum methane potential of a substrate.The livestock manures collected from the local farms were incubated under a thermophilic anaerobic condition(55℃).The results showed that the maximum methane potential(B0)of cattle,hog and chicken manures were 292.0 mL/g VS,272.0 mL/g VS and 266.4 mL/g VS,respectively.The B0 value decreases with increasing contents of crude protein and crude fat,while increases with increasing the contents of carbohydrates and crude fiber in manures.The content of NH4+-N in chicken manure was significantly higher during the digestion period,reached as high as 1962.5 mg/L by the end of incubation period.Heavy metals of Cu and Zn in the manure also affect the B0.Empirical relationships that describe the B0 decrease in response to increase of Zn and Cu contents in manure were developed and used as a simple tool to assess the effects of these metals on the B0.It was concluded that the protein,Cu and Zn contents of manure are most important chemical compositions that negatively affect maximum methane potential.Based on the three experimental manures,the maximum methane potential was limited by either ammonium content or Cu and Zn content in the manure.For a commercial biogas production facility using these manures as main feedstock,one should consider to add co-substrate or co-substrates to reduce concentration of these chemicals to maximize biogas production.Chen Fen Yu Gao Li Wei Liu Fenwu Zhang Wuping Bu Yushan Li Xiaomei 2017International Journal of Agricultural and Biological Engineering2017,10,1:2
