| 1 | High-Resolution Genome-wide Association Study Identifies Genomic Regions and Candidate Genes for Important Agronomic Traits in Wheat显示文摘Wheat(Triticum aestivum)is a major staple food crop worldwide.Genetic dissection of important agronomic traits is essential for continuous improvement of wheat yield to meet the demand of the world's growing population.We conducted a large-scale genome-wide association study(GWAS)using a panel of 768 wheat cultivars that were genotyped with 327609 single-nucleotide polymorphisms generated by genotyping-by-sequencing and detected 395 quantitative trait loci(QTLs)for 12 traits under 7 environments.Among them,273 QTLs were delimited to≤1.0-Mb intervals and 7 of them are either known genes(Rht-D,Vrn-B1,and Vrn-D1)that have been cloned or known QTLs(TaGA2ox8,APO1,TaSus1-7B,and Rht12)that were previously mapped.Eight putative candidate genes were identified for three QTLs that enhance spike seed setting and grain size using gene expression data and were validated in three bi-parental populations.Protein sequence analysis identified 33 putative wheat orthologs that have high identity with rice genes in QTLs affecting similar traits.Large r^2 values for additive effects observed among the QTLs for most traits indicated that the phenotypes of these identified QTLs were highly predictable.Results from this study demonstrated that significantly increasing GWAS population size and marker density greatly improves detection and identification of candidate genes underlying a QTL,solidifying the foundation for large-scale QTL fine mapping,candidate gene validation,and developing functional markers for genomics-based breeding in wheat. | Yunlong Pang Chunxia Liu Danfeng Wang Paul St.Amand Amy Bernardo Wenhui Li Fang He Linzhi Li Liming Wang Xiufang Yuan Lei Dong Yu Su Huirui Zhang Meng Zhao Yunlong Liangi Hongze Jia Xitong Shen Yue Lu Hongming Jiang Yuye Wu Anfei Li Honggang Wang Lingrang Kong Guihua Bai Shubing Liu | 2020 | Molecular Plant2020,13,9: | 8 |
| 2 | Breeding and molecular cytogenetic identification of wheat-Thinopyrum intermedium addition lines resistant to powdery mildew显示文摘Wheat-related species Th. intermedium was used to cross with common wheat Yannong 15. In the self progenies of the hybrid, two addition lines, II -1-7-1 and II -3-3-2, stable in cytology, were developed by cytology and powdery mildew resistance identification. Their chromosome number were 2n = 44 and formed 22 bivalents at PMC MI. In F1 of the two addition lines crossing with Yannong 15, there appeared about one univalent at PMC MI, respectively. Resistance identification in greenhouse and field using the No. 15 and mixed strains of E. gramnis f. sp. tritici showed that they were immune to powdery mildew. Chromosome number and resistance identification using the F2 single plants of the addition line crossing with Yannong 15 indicated that the resistant gene was located on the alien chromosomes. | 3 | Geochemical Characteristics of Eolian Deposits in the Chengdu Plain of Sichuan Province and the Implications for Provenance显示文摘In this study Chengdu Red Earth(CRE)from the Chengdu Plain(CP),Sichuan province,was analyzed for its elemental(major and trace elements)and isotopic(Sm-Nd)geochemistry and compared with Pleistocene loess and paleosol samples from the Chinese Loess Plateau(CLP)in Northern China.The geochemical composition of CRE is similar to north China loess,and also resembles the average UCC.This indicates that CRE,as loess deposits in Northern China,was derived from well-mixed sedimentary protoliths that have undergone numerous upper crustal recycling processes.However,obvious differences in the geochemical characteristics of CRE and the north China loess are also revealed in our results.For chemically stable elements,CRE has higher Ti,Zr,Hf and lowerΣREE,Ba contents in comparison with loess samples from the CLP.Further analysis shows that CRE has higher TiO2/Al2O3,SiO2/Al2O3,Ba/Rb and lower Ce/Yb,Eu/Yb,LaN/YbN andΣLREE/ΣHREE ratios.In Sm-Nd isotopic geochemistry,Sm and Nd content and theεNd(0)value in CRE are significantly higher than those in north China loess.The higher TiO2content in CRE coincided with a high background concentration of Ti in the Sichuan Basin and the surrounding regions.The lowerΣREE and higher Sm,Nd,εNd(0)values are related to the wide distribution of basalt in the southwest Sichuan Basin.The elemental and isotopic geochemistry of CRE indicates that eolian materials in the CP predominantly come from the Sichuan Basin and the surrounding regions,which differs from loess deposits in the CLP. | TAN Yuanlong QIAO Yansong ZHAO Zhizhong WANG Yan QI Lin FU Jianli LIU Zongxiu YAO Haitao WANG Shubing JIANG Fuchu | 2013 | Acta Geologica Sinica(English Edition)2013,87,6: | 3 |

| 12 | Histone Deacetylase Inhibitor SAHA Induces Expression of Fatty Acid-Binding Protein 4 and Inhibits Replication of Human Cytomegalovirus显示文摘Suberoylanilide hydroxamic acid(SAHA) is a histone deacetylase inhibitor that shows marked efficacy against many types of cancers and is approved to treat severe metastatic cutaneous T-cell lymphomas. In addition to its anticancer activity,SAHA has significant effects on the growth of many viruses. The effect of SAHA on replication of human cytomegalovirus(HCMV) has not, however, been investigated. Here, we showed that the replication of HCMV was significantly suppressed by treatment with SAHA at concentrations that did not show appreciable cytotoxicity. SAHA reduced transcription and protein levels of HCMV immediate early genes, showing that SAHA acts at an early stage in the viral life-cycle. RNAsequencing data mining showed that numerous pathways and molecules were affected by SAHA. Interferon-mediated immunity was one of the most relevant pathways in the RNA-sequencing data, and we confirmed that SAHA inhibits HCMV-induced IFN-mediated immune responses using quantitative Real-time PCR(qRT-PCR). Fatty acid-binding protein 4(FABP4), which plays a role in lipid metabolism, was identified by RNA-sequencing. We found that FABP4 expression was reduced by HCMV infection but increased by treatment with SAHA. We then showed that knockdown of FABP4 partially rescued the effect of SAHA on HCMV replication.
