| 1 | Seafloor geodetic network establishment and key technologies显示文摘Seafloor geodetic network construction involves the development of geodetic station shelter, network configuration design, location selection and layout, surveying strategy, observation model establishment and optimization, data processing strategy and so on. This paper tries to present main technological problems involved in the seafloor geodetic network construction, and seek the technically feasible solutions. Basic conceptions of developing seafloor geodetic station shelters for shallow sea and deep-sea are described respectively. The overall criteria of seafloor geodetic network construction for submarine navigation and those of network design for crustal motion monitoring are both proposed. In order to enhance application performances of the seafloor geodetic network, the seafloor network configuration should prefer a symmetrical network structure. The sea surface tracking line measurements for determining the seafloor geodetic station position should also adopt an approximately symmetrical configuration, and we recommend circle tracking line observations combined with cross-shaped line(or double cross-shape line) observations for the seafloor positioning mode. As to the offset correction between the Global Navigation Satellite System antenna phase center and the acoustic transducer, it is recommended to combine the calibration through external measurements and model parameter estimation. Besides, it is suggested to correct the sound speed error with a combination of observation value correction and parameterized model correction, and to mainly use the model correction to reduce the influence of acoustic ray error on the seafloor positioning. Following the proposed basic designs, experiments are performed in shallow sea area and deep-sea area respectively. | 3 | Expression of SoxC Transcription Factors during Zebrafish Retinal and Optic Nerve Regeneration显示文摘The SoxC transcription factors(Sox4,Sox11,and Sox12) play important roles in the development of the vertebrate eye and retina. However,their expression and function during retinal and optic nerve regeneration remain elusive. In this study,we investigated the expression and possible functions of the SoxC genes after retinal and optic nerve injury in adult zebrafish. We found that among the five SoxC members,Sox11b was strongly induced in BrdU-positive cells in the inner nuclear layer(INL) after retinal injury,and morpholino-mediated Sox11b-knockdown significantly reduced the number of proliferating cells in the INL at 4 days post-injury. After optic nerve lesion,both Sox11a and Sox11b were strongly expressed in retinal ganglion cells(RGCs),and knockdown of both Sox11a and Sox11b inhibited RGC axon regrowth in retinal explants. | 5 | Dynamic blood single-cell immune responses in patients with COVID-19显示文摘The 2019 coronavirus disease(COVID-19)outbreak caused by the SARS-CoV-2 virus is an ongoing global health emergency.However,the virus'pathogenesis remains unclear,and there is no cure for the disease.We investigated the dynamic changes of blood immune response in patients with COVID-19 at different stages by using 5'gene expression,T cell receptor(TCR),and B cell receptors(BCR)V(D)J transcriptome analysis at a single-cell resolution.We obtained single-cell mRNA sequencing(scRNA-seq)data of 341,420 peripheral blood mononuclear cells(PBMCs)and 185,430 donotypic T cells and 28,802 donotypic B cells from 25 samples of 16 patients with COVID-19 for dynamic studies.In addition,we used three control samples.We found expansion of dendritic cells(DCs),CD14+monocytes,and megakaryocytes progenitor cells(MP)/platelets and a reduction of naive CD4+T lymphocytes in patients with COVID-19,along with a significant decrease of CD8+T lymphocytes,and natural killer cells(NKs)in patients in critical condition.The type I interferon(IFN-I),mitogen-activated protein kinase(MAPK),and ferroptosis pathways were activated while the disease was active,and recovered gradually after patient conditions improved.Consistent with this finding,the mRNA level of IFN-I signal-induced gene IFI27 was significantly increased in patients with COVID-19 compared with that of the controls in a validation cohort that included 38 patients and 35 controls.The concentration of interferon-a(IFN-a)in the serum of patients with COVID-19 increased significantly compared with that of the controls in an additional cohort of 215 patients with COVID-19 and 106 controls,further suggesting the important role of the IFN-I pathway in the immune response of COVID-19.TCR and BCR sequences analyses indicated that patients with COVID-19 developed specific immune responses against SARS-CoV-2 antigens.Our study reveals a dynamic landscape of human blood immune responses to SARS-CoV-2 infection,providing clues for therapeutic potentials in treating COVID-19. | Lulin Huang Yi Shi Bo Gong Li Jiang Zhixin Zhang Xiaoqi Liu Jialiang Yang Yongquan He Zhilin Jiang Ling Zhong Juan Tang Chunfang You Qi Jiang Bo Long Tao Zeng Mei Luo Fanwei Zeng Fanxin Zeng Shuqiang Wang Xingxiang Yang Zhenglin Yang | 2021 | Signal Transduction and Targeted Therapy2021,6,4: | 4 |

| 17 | Genome-wide alternative splicing variation and its potential contribution to maize immature-ear heterosis显示文摘Heterosis is a well-known phenomenon widely applied in agriculture.Recent studies have suggested that differential gene and protein expression between hybrids and their parents play important roles in heterosis.Alternative splicing(AS)is an essential posttranscriptional mechanism that can greatly affect the transcriptome and proteome diversity in plants.However,genome-wide AS divergence in hybrids compared to their parents and its potential contribution to heterosis have not been comprehensively investigated.We report the direct profiling of the AS landscape using RNA sequencing data from immature ears of the maize hybrid ZD808 and its parents NG5 and CL11.Our results revealed a large number of significant differential AS(DAS)events in ZD808 relative to its parents,which can be further classified into parental-dominant and novel DAS patterns.Parental-dominant,especially NG5-dominant,events were prevalent in the hybrid,accounting for 42%of all analyzed DAS events.Functional enrichment analysis revealed that the NG5-dominant AS events were involved mainly in regulating the expression of genes associated with carbon/nitrogen metabolism and cell division processes and contributed greatly to maize ear heterosis.Among ZD808,CL11,and NG5,32.5%of DAS contained or lacked binding sites of at least one annotated maize microRNA(miRNA)and may be involved in miRNA-mediated posttranscriptional regulation.Cis regulation was the predominant contributor to AS variation and participates in many important biological processes associated with immature ear development.This study provides a comprehensive view of genome-wide alternative splicing variation in a maize hybrid. | Xiaojiao Hu Hongwu Wang Kun Li Xiaogang Liu Zhifang Liu Yujin Wu Shuqiang Li Changling Huang | 2021 | The Crop Journal2021,9,2: | 0 |
