The RALF1-FERONIA Complex Phosphorylates eIF4E1 to Promote Protein Synthesis and Polar Root Hair Growth显示文摘The molecular links between extracellular signals and the regulation of localized protein synthesis in plant cells are poorly understood. Here,we show that in Arabidopsis thaliana, the extracellular peptide RALF1 and its receptor, the FERONIA receptor kinase, promote root hair(RH)tip growth by modulating protein synthesis. We found that RALF1 promotes FERONIA-mediated phosphorylation of elF4E1,a eukaryotic translation initiation factor that plays a crucial role in the control of mRNA translation rate.Phosphorylated elF4E1 increases mRNA affinity and modulates mRNA translation and, thus, protein synthesis. The mRNAs targeted by the RALF1-FERONIA-elF4E1 module include ROP2 and RSL4,which are important regulators of RH cell polarity and growth.RALF1 and FERONIA are expressed in a polar manner in RHs,which facilitate elF4E1 polar丨ocalization and thus may control local f?OP2 translation. Moreover,we demonstrated that high-level accumulation of RSL4 exerts negative-feedback regulation of RALF1 expression by directly binding the RALF1 gene promoter, determining the final RH size. Our study reveals that the link between RALF1-FERONIA signaling and protein synthesis constitutes a novel component regulating cell expansion in these polar growing cells. | Sirui Zhu Jose Manuel Estevez Hongdong Liao Yonghua Zhu Tao Yang Chiyu Li Yichuan Wang Lan Li Xuanming Liu Javier Martinez Pacheco Hongwei Guo Feng Yu | 2020 | Molecular Plant2020,13,5: | 3
Nematode-Encoded RALF Peptide Mimics Facilitate Parasitism of Plants through the FERONIA Receptor Kinase显示文摘The molecular mechanism by which plants defend against plant root-knot nematodes(RKNs)is largely unknown. The plant receptor kinase FERONIA and its peptide ligands, rapid alkalinization factors(RALFs),regulate plant immune responses and cell expansion, which are two important factors for successful RKN parasitism. In this study,we found that mutation of FERONIA in Arabidopsis thaliana resulted in plants showing low susceptibility to the RKN Meloidogyne incognita. To identify the underlying mechanisms associated with this phenomenon,we identified 18 novel RALF-likes from multiple species of RKNs and showed that two RALF-likes(i.e.,MiRALF1 and MiRALF3)from M.incognita were expressed in the esophageal gland with high expression during the parasitic stages of nematode development. These nematode RALF-likes also possess the typical activities of plant RALFs and can directly bind to the extracellular domain of FERONIA to modulate specific steps of nematode parasitism-related immune responses and cell expansion.Genetically, both MiRALF1/3 and FERONIA are required for RKN parasitism in Arabidopsis and rice.Collectively, our study suggests that nematode-encoded RALFs facilitate parasitism via plant-encoded FERONIA and provides a novel paradigm for studying host-pathogen interactions. | Xin Zhang Huan Peng Sirui Zhu Junjie Xing Xin Li Zhaozhong Zhu Jingyuan Zheng Long Wang Bingqian Wang Jia Chen Zhenhua Ming Ke Yao Jinzhuo Jian Sheng Luan Devin Coleman-Derr Hongdong Liao Yousong Peng Deliang Peng Feng Yu | 2020 | Molecular Plant2020,13,10: | 3
Isolation of a novel characterized Issatchenkia terricola from red raspberry fruits on the degradation of citric acid and enrichment of flavonoid and volatile profiles in fermented red raspberry juice显示文摘High content of citric acid in red raspberry juice leads to poor sensory experience. This study developed a feasible method to degrade citric acid in red raspberry juice using a novel characterized Issatchenkia terricola WJL-G4 isolated from red raspberry fruits. I. terricola WJL-G4 exhibited a potent capability of reducing the citric acid contents from(22.8 ± 0.08) g/L to(6.2 ± 0.02) g/L within 36 h fermentation, then completely depleted after 48 h. Furthermore, the contents of phenolic compound, including neochlorogenic acid, p-coumaric acid, raspberry ketone, and rutin significantly increased after 36 h fermentation. Fermentation increased total flavonoid contents in red raspberry juice, compared to that in control group. Volatile profiles exhibited to be enriched after fermentation, which contributed to the improvement of the juice taste. Our findings showed that I. terricola WJL-G4 can be applied in deacidification, enrichment of flavonoid compounds and volatile profiles in fermented red raspberry juice.. | Ying Jiang Ting Luo Ying Tang Sirui Chen Hui Ni Qihe Chen Xingshun Song Yihong Bao Zeyuan Deng Jinling Wang | 2022 | Food Science and Human Wellness2022,11,4: | 3
Treated dentin matrix induces odontogenic differentiation of dental pulp stem cells via regulation of Wnt/β-catenin signaling显示文摘Treated dentin matrix(TDM)is an ideal scaffold material containing multiple extracellular matrix factors. The canonical Wnt signaling pathway is necessary for tooth regeneration. Thus, this study investigated whether the TDM can promote the odontogenic differentiation of human dental pulp stem cells(hDPSCs)and determined the potential role of Wnt/β-catenin signaling in this process.Different concentrations of TDM promoted the dental differentiation of the hDPSCs and meanwhile, the expression of GSK3βwas decreased.Of note, the expression of the Wnt/β-catenin pathway-related genes changed significantly in the context of TDM induction,as per RNA sequencing(RNA seq)data. In addition, the experiment showed that new dentin was visible in rat mandible cultured with TDM,and the thickness was significantly thicker than that of the control group. In addition, immunohistochemical staining showed lower GSK3βexpression in new dentin.Consistently, the GSK3βknockdown hDPSCs performed enhanced odotogenesis compared with the control groups.However,GSK3βoverexpressing could decrease odotogenesis of TDM-induced hDPSCs.These results were confirmed in immunodeficient mice and Wistar rats. These suggest that TDM promotes odontogenic differentiation of hDPSCs by directly targeting GSK3βand activating the canonical Wnt/β-catenin signaling pathway and provide a theoretical basis for tooth regeneration engineering. | Sirui Liu Jingjing Sun Shuai Yuan Yanyu Yang Yuping Gong Ying Wang Runying Guo Xue Zhang Yiming Liu Hongyan Mi Meiyue Wang Mengzhe Liu Rui Li | 2022 | Bioactive Materials2022,7,1: | 2
The RALF1-FERONIA complex interacts with and activates TOR signaling in response to low nutrients显示文摘Target of rapamycin(TOR)kinase is an evolutionarily conserved major regulator of nutrient metabolism and organismal growth in eukaryotes. In plants, nutrients are remobilized and reallocated between shoots and roots under low-nutrient conditions, and nitrogen and nitrogen-related nutrients(e.g.,amino acids)are key upstream signals leading to TOR activation in shoots under low-nutrient conditions. However, how these forms of nitrogen can be sensed to activate TOR in plants is still poorly understood. Here we report that the Arabidopsis receptor kinase FERON IA(FER)interacts with the TOR pathway to regulate nutrient(nitrogen and amino acid)signaling under low-nutrient conditions and exerts similar metabolic effects in response to nitrogen deficiency. We found that FER and its partner,RPM1-induced protein kinase(RIPK),interact with the TOR/RAPTOR complex to positively modulate TOR signaling activity.During this process, the receptor complex FER/RIPK phosphorylates the TOR complex component RAPTOR1B.The RALF1 peptide,a ligand of the FER/RIPK receptor complex, increases TOR activation in the young leaf by enhancing FER-TOR interactions, leading to promotion of true leaf growth in Arabidopsis under lownutrient conditions.Furthermore,we showed that specific amino acids(e.g.,Gin,Asp,and Gly)promote true leaf growth under nitrogen-deficient conditions via the FER-TOR axis.Collectively, our study reveals a mechanism by which the RALF1-FER pathway activates TOR in the plant adaptive response to low nutrients and suggests that plants prioritize nutritional stress response over RALF1-mediated inhibition of cell growth under low-nutrient conditions. | Limei Song Guoyun Xu Tingting Li Huina Zhou Qinlu Lin Jia Chen Long Wang Dousheng Wu Xiaoxu Li Lifeng Wang Sirui Zhu Feng Yu | 2022 | Molecular Plant2022,15,7: | 2
A New Indicator for Global Food Security Assessment: Harvested Area Rather Than Cropland Area显示文摘Cropland area has long been used as a key indicator of food security. However, grain yield is not solely controlled by the area of the cropland. Therefore,we proposed a new indicator to assess food security. Results show that from 1992 to 2004,the global cropland area increased by 840200 km^(2)(99.4%),but the grain yield increased only by 310 million t(29.1%);and from 2004 to 2015,the cropland area decreased by 39000 km^(2)(4.64%),but the grain yield increased by 370 million t(70.84%).This result showed that grain yield was not linearly correlated with cropland area, and delimiting the threshold of cropland protection may not guarantee food security.Combined with further correlation analysis,we found that the increase in the global grain yield was more closely related to the harvested area(R^(2)=0.94),which indicated that the harvested area is a more scientific and accurate indicator than cropland area in terms of guaranteeing food security. Therefore,if governments want to ensure the food security, they should choose a new and more accurate indicator:harvested area rather than cropland area. | SONG Fengjiao WANG Shijie BAI Xiaoyong WU Luhua WANG Jinfeng LI Chaojun CHEN Huan LUO Xuling XI Huipeng ZHANG Sirui LUO Guofeng YAN Mengqi ZHEN Qingqing | 2022 | Chinese Geographical Science2022,32,2: | 2
MYOD1 inhibits avian adipocyte differentiation via miRNA-206/KLF4 axis显示文摘Background:A considerable number of muscle development-related genes were differentially expressed in the early stage of avian adipocyte differentiation. However, the functions of them in adipocyte differentiation remain largely known. In this study, the myoblast determination protein 1(MYOD1)was selected as a representative of muscle development. We investigated its expression, function, and regulation in avian adipocyte differentiation. Results:The expression of MYOD1 decreased significantly in the early stage of avian adipocyte differentiation.CRIS PR/Cas9-mediated deletion of MYOD1 induced adipocyte differentiation, whereas over-expression of MYOD1 inhibited adipogenesis. The mRNA-seq data showed that MYOD1 could perturb the lipid biosynthetic process during differentiation. Our results showed that MYOD1 directly up-regulates the miR-206 expression by binding the upstream 1200 bp region of miR-206.Then,over-expression of miR-206 can inhibit the adipogenesis.Furthermore,MYOD1 affected the expression of endogenous miR-206 and its target gene Kruppel-like factor 4(KLF4),which is an important activator of adipogenesis.Accordingly, the inhibition of miR-206 or over-expression of KLF4 could counteract the inhibitory effect of MYOD1 on adipocyte differentiation. Conclusions:Our results establish that MYOD1 inhibits adipocyte differentiation by up-regulating miR-206 to suppress the KLF4 expression. These findings identify a novel function of MYOD1 in adipocyte differentiation, suggesting a potential role in body-fat distribution regulation. | Zheng Wang Qiangsen Zhao Xiaoqin Li Zhongtao Yin Sirui Chen Sen Wu Ning Yang Zhuocheng Hou | 2021 | Journal of Animal Science and Biotechnology2021,12,4: | 2
Photoacoustic detection of follicular thyroid carcinoma using targeted Nano-Au-Tripods显示文摘Follicular thyroid carcinoma(FTC)is the second most common form of thyroid malignancy, and it is associated with more aggressive growth and worse long-term survival outcomes relative to papillary thyroid carcinoma(PTC).Reliable approaches to preoperative FTC detection, however, remain to be established.Herein,a targeted Affibody-Au-Tripod nanoprobe was developed and successfully utilized to facilitate the targeted photoacoustic imaging(PAI)of epidermal growth factor receptor(EGFR)-positive cells and tumors. These Affibody-Au-Tripods were found to be highly sensitive and specific for cells expressing EGFR when used as a PA contrast agent in vitro, and studies conducted in an FTC-133 subcutaneous tumor model system in mice further revealed that these Affibody-Au-Tripods were able to specifically target these EGFR-expressing tumors while providing a strong photoacoustic signal in vivo.Importantly, these nanoprobes exhibited negligible cytotoxicity and robust chemical and physical stability, making Affibody-Au-Tripods promising candidates for targeted PAI-based FTC diagnosis. In addition, these nanoprobes have the potential to facilitate the individualized treatment of patients harboring EGFRpositive tumors. | Yang Gui Kai Cheng Ruojiao Wang Sirui Liu Chenyang Zhao Rui Zhang Ming Wang Zhen Cheng Meng Yang | 2022 | Chinese Journal of Chemical Engineering2022,35,4: | 2
FERONIA is involved in phototropin 1-mediated blue light phototropic growth in Arabidopsis显示文摘Plant shoot phototropism is triggered by the formation of a light-driven auxin gradient leading to bending growth. The blue light receptor phototropin 1(phot1)senses light direction, but how this leads to auxin gradient formation and growth regulation remains poorly understood.Previous studies have suggested phot1’s role for regulated apoplastic acidification, but its relation to phototropin and hypocotyl phototropism is unclear.Herein,we show that blue light can cause phot1 to interact with and phosphorylate FERONIA(FER),a known cell growth regulator, and trigger downstream phototropic bending growth in Arabidopsis hypocotyls.fer mutants showed defects in phototropic growth, similar to phot1/2 mutant.FER also interacts with and phosphorylates phytochrome kinase substrates, the phot1 downstream substrates. The phot1-FER pathway acts upstream of apoplastic acidification and the auxin gradient formation in hypocotyl under lateral blue light, both of which are critical for phototropic bending growth in hypocotyls. Our study highlights a pivotal role of FER in the phot1-mediated phototropic cell growth regulation in plants. | Chiyu Li Jia Chen Xiaoyan Li Xin Zhang Ying Liu Sirui Zhu Long Wang Heping Zheng Sheng Luan Jiada Li Feng Yu | 2022 | Journal of Integrative Plant Biology2022,64,10: | 1

Cyclic di-adenosine monophosphate regulates the osteogenic and adipogenic differentiation of hPDLSCs via MAPK and NF-κB signaling显示文摘Cyclic di-adenosine monophosphate(c-di-AMP)is a bacterial second messenger that can be recognized by infected host cells and activate the immunoinflammatory response. The purpose of this study is to demonstrate the effect of c-di-AMP on the differentiation of human periodontal ligament stem cells(hPDLSCs)and its underlying mechanisms. In the present study,we find that the gingival crevicular fluid(GCF)of patients with chronic periodontitis has a higher expression level of c-di-AMP than that of healthy people. In vitro,c-di-AMP influences the differentiation of hPDLSCs by upregulating Toll-like receptors(TLRs);specifically,it inhibits osteogenic differentiation by activating NF-κB and ERK/MAPK and promotes adipogenic differentiation through the NF-κB and p38/MAPK signaling pathways.Inhibitors of TLRs or activated pathways reduce the changes induced by c-di-AMP.Our results establish the potential correlation among bacterial c-di-AMP,periodontal tissue homeostasis and chronic periodontitis pathogenesis. | Sirui Chen Zuping Wu Yuying He Li Zhu Jiahe Wang Hengyi Lin Jing Xie Chenchen Zhou Shujuan Zou | 2023 | Acta Biochimica et Biophysica Sinica2023,55,3: | 1

Observational study of the physical and chemical characteristics of the winter radiation fog in the tropical rainforest in Xishuangbanna, China显示文摘We conducted a three-month field experiment focusing on the physical and chemical characteristics of fog in a tropical rainforest in Xishuangbanna,Southwest China,in the winter of 2019.In general, the fog would form at midnight and persist because of the increased long-wave radiative cooling combined with the high relative humidity, gentle breeze, and a relatively low aerosol number concentration in the forest;the fog would dissipate before noon due to the increasing turbulence near the surface. This diurnal cycle is typical for radiation fog.The microphysical fog properties included a relatively low number concentration of the fog droplet, large droplet size, high liquid water content, narrow droplet number-size distribution, and high supersaturation. The chemical properties showed that the fog water was slightly alkaline with low electrical conductivity, whereas the highest proportions of anions and cations therein were Cl^(−)and Ca^(2+),respectively;the chemical components were enriched in small fog droplets. In addition,we indirectly calculated the fog supersaturation according to theκ-Köhler theory. We found that condensation broadens the droplet number-size distribution at relatively low supersaturation, which is positively correlated with the fog-droplet number concentration and negatively correlated with the droplet mean-volume diameter;this affects the key microphysical processes of fog. | Yuan WANG Shengjie NIU Chunsong LU Jingjing LV Jing ZHANG Hongwei ZHANG Sirui ZHANG Naifu SHAO Wei SUN Yuchen JIN Qinghai SONG | 2021 | Science China Earth Sciences2021,64,11: | 1
A carbon-neutrality-capactiy index for evaluating carbon sink contributions显示文摘The accurate determination of the carbon-neutrality capacity(CNC)of a region is crucial for developing policies related to emissions and climate change. However,a systematic diagnostic method for determining the CNC that considers the rock chemical weathering carbon sink(RCS)is lacking. Moreover,it is challenging but indispensable to establish a fast and practical index model to determine the CNC.Here,we selected Guizhou as the study area, used the methods for different types of carbon sinks, and constructed a CNC index(CNCI)model. We found that:(1)the carbonate rock chemical weathering carbon sink flux was 30.3 t CO_(2)km^(-2)yr^(-1).Guizhou accounted for 1.8%of the land area and contributed 5.4%of the carbonate chemical weathering carbon sink;(2)the silicate rock chemical weathering carbon sink and its flux were 1.44×10^(3)t CO_(2)and 2.43 t CO_(2)km^(-2)yr^(-1),respectively;(3)the vegetation-soil ecosystem carbon sink and its flux were 1.37×10^(8)t CO_(2)and 831.70 t CO_(2)km^(-2)yr^(-1),respectively;(4)the carbon emissions(CEs)were 280 Tg CO_(2),about 2.8%of the total for China;and(5)the total carbon sinks in Guizhou were 160 Tg CO_(2),with a CNCI of 57%,which is 4.8 times of China and 2.1 times of the world. In summary,we conducted a systematic diagnosis of the CNC considering the RCS and established a CNCI model. The results of this study have a strong implication and significance for national and global CNC determination and gap analysis. | Xiaoyong Bai Sirui Zhang Chaojun Li Lian Xiong Fengjiao Song Chaochao Du Minghui Li Qing Luo Yingying Xue Shijie Wang | 2023 | Environmental Science and Ecotechnology2023,,3: | 1
Efficacy and safety of Guipi Decoction combined with conventional Western medicine in the treatment of insomnia with deficiency of heart and spleen:a meta-analysis显示文摘Objective:To systematically evaluate the efficacy and safety of Guipi Decoction combined with Western medicine in the treatment of insomnia with deficiency of heart and spleen. Methods:English databases(PubMed,Web of Science,The Cochrane Library,EMBASE)and Chinese databases(CNKI,Wanfang database,China Biomedical Literature Service System,VIP database)were searched by computer.Randomized controlled trials of Guipi Decoction on insomnia with deficiency of heart and spleen were searched from the database construction to November 2021.After the first and second authors independently screened the literature, extracted the data and evaluated the risk of bias in the included studies,meta-analysis was performed using RevMan5.3 software. Results:A total of 9 RCTS were included after screening, including 914 patients.Meta-analysis results showed that:Total effective rate[RR=1.22,95%CI(1.16,1.30),P<0.00001],total PSQI score[MD=-3.05,95%CI(-3.96,-2.14),P=0.008],number of night awakening times[[MD=-1.18,95%CI(-1.42,-0.94),P<0.00001],adverse reaction rate[RR=0.32,95%CI(0.21,0.51),P<0.00001]were better than the control group, and the differences were statistically significant. Conclusion:The current evidence shows that, compared with pure using conventional western medicine, belongs to the spleen decoction combined western medicine therapy, cases both deficiency type of insomnia in the total effective rate, reducing the total PSQI score(improve sleep quality, sleep efficiency, sleep disorder, daytime function, etc.),reducing frequency of nighttime awakening, security, have more advantages. However, due to the limitation of the quality and quantity of articles included in the study, more randomized,double-blind,large-sample clinical studies are needed to confirm the above conclusions. | Huixin Chen Zhogqing Dou Sirui Liu Hongchen Dong Di Zhang Shenjun Wang | 2022 | Asian Toxicology Tesearch2022,4,1: | 1
The tumor immunosuppressive microenvironment impairs the therapy of anti-HER2/neu antibody显示文摘It has been well established that immune surveillance plays critical roles in preventing the occurrence and prog-ression of tumor.More and more evidence in recent years showed the host anti-tumor immune responses also play important roles in the chemotherapy and radiotherapy of cancers. Our previous study found that tumor-targeting therapy of anti-HER2/neu mAb is mediated by CD8+T cell responses. However,we found here that enhancement of CD8+T cell responses by combination therapy with IL-15R/IL-15 fusion protein or anti-CD40,which are strong stimultors for T cell responses, failed to promote the tumor therapeutic effects of anti-HER2/neu mAb.Analysis of tumor microenviornment showed that tumor tissues were heavily infiltrated with the immunosuppressive macrophages and most tumor infiltrating T cells, especially CD8^(+)T cells, expressed high level of inhibitory co-signaling receptor PD-1.These data suggest that tumor microenvironment is dominated by the immunosuppressive strategies, which thwart anti-tumor immune responses. Therefore, the successful tumor therapy should be the removal of inhibitory signals in the tumor microenvir-onment in combination with other therapeutic strategies. | Meng Xu Xuexiang Du Mingyue Liu Sirui Li Xiaozhu Li Yang-Xin Fu Shengdian Wang | 2012 | Protein & Cell2012,3,6: | 1
Self-induced collision risk of the Starlink constellation based on long-term orbital evolution analysis显示文摘The deployment of mega constellations has had a significant effect on the compounding space debris environment, increasing the number of on-orbit objects in all conditions and damaging the stability of the space debris environment. The increased density of space objects is associated with an increased risk of on-orbit collisions.Collision risk exists not only between a mega constellation and the space debris environment but also inside a mega constellation. In this study,we used the Starlink constellation to investigate the self-induced collision risk caused by malfunctioning satellites.First,we analyzed the conjunction condition between malfunctioning and operative satellites based on long-term orbital evolution characteristics. The collision probability was then calculated based on the conjunction analysis results. The results show that malfunctioning satellites in Phase 1 cause an 86.2%self-induced collision probability based on a malfunctioning rate of 1%,which is close to the collision probability caused by objects larger than 6 cm during five years of service.Therefore,self-induced collisions are another important risk factor for the Starlink constellation. | Wei Zhang Xiuhong Wang Wen Cui Zhi Zhao Sirui Chen | 2023 | Astrodynamics2023,7,4: | 0
