1Thrombolysis with alteplase 3-4.5 hours after acute ischaemic stroke:the first multicentre,phase III trial in China显示文摘background and purpose Data on the efficacy and safety of alteplase for acute ischaemic stroke(AIS)administered 3-4.5 hours after the onset of stroke symptoms in Chinese patients is limited.We sought to determine whether AIS patients would benefit from thrombolysis with alteplase between 3 and 4.5 hours after the onset of stroke symptoms in a prospective,multicentre,single-arm trial in China.Materials and methods Eligible AIS patients were given 0.9 mg/kg alteplase intravenously.The primary efficacy endpoint was a favourable outcome at 3 months,defined as a score of 0 or 1 on the modified Rankin Scale.Thresholds for the primary efficacy endpoint were determined to be 40%based on the literature review.The primary safety endpoint was symptomatic intracranial haemorrhage(sICH)according to the European Cooperative Acute Stroke Study III(ECASS III)trial definition.Post hoc analysis between this study and the ECASS III trial were compared using the propensity score matching(PSM)method.results A total of 120 eligible AIS patients from 11 sites in China received thrombolysis therapy in this study.The median time from onset of symptoms to needle was 3 hours 54 min.The percentage of patients with a favourable outcome was 63.3%(95%CI 54.4 to 71.4),significantly higher than the predefined threshold(p<0.0001).Three patients(2.5%,95%CI 0.5 to 7.1)had sICH,including two fatal sICH.Six patients died within 3 months after treatment.The post hoc PSM analysis showed a numerically higher rate of the primary efficacy endpoint in this study(63.3%)than the matched placebo arm(56.7%)in the ECASS III trial.Conclusions Intravenous alteplase with a standard dose administered between 3 and 4.5 hours after onset of symptoms is effective and safe for Chinese AIS patients.Huaguang Zheng Yi Yang Huisheng Chen Chuanling Li Yangkun Chen Fu-Dong Shi Li Yang Xiaoping Cui Zuneng Lu Yanling Liang Songbiao Cui Anding Xu Yiqing Wu Yaqing Sun Yongjun Wang 2020Stroke & Vascular Neurology2020,5,3:4

3High VHL Expression Reverses Warburg Phenotype and Enhances Immunogenicity in Kidney Tumor Cells显示文摘Clear cell renal cell carcinoma(ccRCC)is a frequently occurring renal cancer.The Von Hippel-Lindau disease tumor suppressor VHL,a known tumor suppressor gene,is frequently mutated in about 50%of patients with ccRCC.However,it is unclear whether VHL influences the progression of ccRCC tumors expressing wild-type VHL.In the present study,we found that higher expression of VHL was correlated with the better disease-free survival(DFS)in ccRCC patients using The Cancer Genome Atlas(TCGA)datasets.We revealed that VHL overexpression in ccRCC cells inhibited epithelial-mesenchymal transition(EMT),sterol regulatory elementbinding protein 1(SREBP1)-regulated triglyceride synthesis,and cell proliferation.Proteomic analysis provided us a global view that VHL regulated four biological processes,including metabolism,immune regulation,apoptosis,and cell movement.Importantly,we found that VHL overexpression led to up-regulated expression of proteins associated with antigen processing and interferonresponsive proteins,thus rendering ccRCC cells more sensitive to interferon treatment.We defined an interferon-responsive signature(IRS)composed of ten interferon-responsive proteins,whose m RNA expression levels were positively correlated with DFS in ccRCC patients.Taken together,our results propose that the subset of ccRCC patients with high VHL expression benefit from immunotherapy.Songbiao Zhu Wenxi Ding Yuling Chen Weixuan Wang Renhua Xu Chongdong Liu Xiaohui Liu Haiteng Deng 2022Genomics, Proteomics & Bioinformatics2022,20,4:1

5Changes of Aroma Components in 'Keitt' Mango during the Postharvest Period显示文摘[Objective]This study aimed to investigate the changes of aroma components in mango fruit at different post-harvest stages.[Method]The aroma components in‘Keitt'mango fruit at 3,6,9 and 13 d postharvest were extracted using head-space solid phase microextraction( HS-SPME) and analyzed by gas chromatograph-mass spectrophotometer( GC-MS).[Result]The results showed that a total of 42 aroma compounds were identified from the mango samples at four stages. To be specific,monoterpene and sesquiterpene were the major components; at 3,6,9 and 13 d postharvest,the relative contents of terpene were 94. 24%,98. 91%,55. 32% and 95. 97%,respectively.[Conclusion]1S-( +)-3-carene is the characteristic aroma component in ripe fruit of‘Keitt'mango.
