3Methylation Modifications in Eukaryotic Messenger RNA显示文摘RNA methylation modifications have been found for decades of years, which occur at different RNA types of numerous species, and their distribution is species-specific. However, people rarely know their biological functions. There are several identified methylation modifications in eukaryotic messenger RNA(mRNA), such as N7-methylguanosine(m7G) at the cap, N6-methyl-20-O-methyladenosine(m6Am), 20-O-methylation(Nm) within the cap and the internal positions, and internal N6-methyladenosine(m6A) and 5-methylcytosine(m5C). Among them, m7G cap was studied more clearly and found to have vital roles in several important mRNA processes like mRNA translation, stability and nuclear export. m6A as the most abundant modification in mRNA was found in the 1970s and has been proposed to function in mRNA splicing, translation, stability, transport and so on. m6A has been discovered as the first RNA reversible modification which is demethylated directly by human fat mass and obesity associated protein(FTO) and its homolog protein, alkylation repair homolog 5(ALKBH5). FTO has a special demethylation mechanism that demethylases m6A to A through two over-oxidative intermediate states: N6-hydroxymethyladenosine(hm6A) and N6-formyladenosine(f6A). The two newly discovered m6A demethylases, FTO and ALKBH5, significantly control energy homeostasis and spermatogenesis, respectively, indicating that the dynamic and reversible m6A,analogous to DNA and histone modifications, plays broad roles in biological kingdoms and brings us an emerging field 'RNA Epigenetics'. 5-methylcytosine(5mC) as an epigenetic mark in DNA has been studied widely, but m5C in mRNA is seldom explored. 4Segmentation of Hematoxylin-Eosin stained breast cancer histopathological images based on pixel-wise SVM classifier显示文摘Hematoxylin-Eosin(HE) staining is the routine diagnostic method for breast cancer(BC), and large amounts of HE stained histopathological images are available for analysis. It is emergent to develop computational methods to efficiently and objectively analyze these images, with the aim of providing potentially better diagnostic and prognostic information for BC. This work focuses on analyzing our in-house HE stained histopathological images of breast cancer tissues. Since tumor nests(TNs) and stroma morphological characteristics can reflect the biological behaviors of breast invasive ductal carcinoma(IDC), accurate segmentation of TNs and the stroma is the first step towards the subsequent quantitative analysis. We first propose a method based on the pixel-wise support vector machine(SVM) classifier for segmenting TNs and the stroma, then extract four morphological characters related to the TNs from the images and investigate their relationships with the patients' 8-year disease free survival(8-DFS). The evaluation result shows that the classification based segmentation method is able to distinguish between TNs and stroma with 87.1% accuracy and 80.2% precision,suggesting that the proposed method is promising in segmenting HE stained IDC histopathological images. 5Arab/c/opste N^(6)-methyladenosine reader CPSF30-L recognizes FUE signals to control polyadenylation site choice in liquid-like nuclear bodies显示文摘The biological functions of the epitranscriptomic modification N^(6)-methyladenosine(m^(6)A)in plants are not fully understood.CPSF30-L is a predominant isoform of the polyadenylation factor CPSF30 and consists of CPSF30-S and an m^(6)A-binding YTH domain.Little is known about the biological roles of CPSF30-L and the molecular mechanism underlying its m^(6)A-binding function in alternative polyadenylation.Here,we charac-terized CPSF30-L as an Arabidopsis m^(6)A reader whose m^(6)A-binding function is required for the floral tran-sition and abscisic acid(ABA)response.We found that the m^(6)A-binding activity of CPSF30-L enhances the formation of liquid-like nuclear bodies,where CPSF30-L mainly recognizes m*A-modified far-upstream elements to control polyadenylation site choice.Deficiency of CPSF30-L lengthens the 3'untranslated region of three phenotypes-related transcripts,thereby accelerating their mRNA degradation and leading to late flowering and ABA hypersensitivity.Collectively,this study uncovers a new molecular mechanism for m^(6)A-driven phase separation and polyadenylation in plants.Peizhe Song Junbo Yang Chunling Wang Qiang Lu Linqing Shi Subiding Tayier Guifang Jia 2021Molecular Plant2021,14,4:12
6Effects of Laser Shock Processing on Mechanical Properties of Laser Welded ANSI 304 Stainless Steel Joint显示文摘With the rapid development of engineering component with integration,high-speed and multi-parameter,traditional techniques haven't met practical needs in extreme service environment.Laser welding,a new welding technology,has been widely used.However,it would generate the drop of mechanical properties for laser welded joint due to its thermal effect.Laser shock processing(LSP) is one of the most effective methods to improve the mechanical properties of laser welded ANSI 304 stainless steel joint.In this paper,the effects of LSP on the mechanical properties of laser welded ANSI 304 stainless steel joint have been investigated.The welded joint on the front of the tensile samples is treated by LSP impacts,and the overlapping rate of the laser spot is 50%.The tensile test of the laser welded joint with and without LSP impacts is carried out,and the fracture morphology of the tensile samples is analyzed by scanning electron microscope(SEM).Compared with the yield strength of 11.70 kN,the tensile strength of 37.66 kN,the yield-to-tensile strength ratio of 0.310 7,the elongation of 25.20%,the area reduction of 32.68% and the elastic modulus of 13 063.876 MPa,the corresponding values after LSP impacts are 14.25 kN,38.74 kN,0.367 8,26.58%,42.29% and 14 754.394 MPa,respectively.Through LSP impacts,the increasing ratio of the yield strength and tensile strength are 121.79% and 102.87%,respectively;the elongation and area reduction are improved by 5.48% and 29.38%,respectively.By comparing with coarse fracture surface of the welded joint,the delamination splitting with some cracks in the sharp corner of the welded joint and asymmetric dimples,LSP can cause brighter fracture surface,and finer and more uniform dimples.Finally,the schematic illustration of dimple formation with LSP is clearly described.The proposed research ensures that the LSP technology can clearly improve the yield strength,tensile strength,yield-to-tensile strength ratio,elongation,area reduction and elastic modulus of the welded joint.The enhancement mechanism of LSP on laser welded ANSI 304 stainless steel joint is mainly due to the fact that the refined and uniform dimples effectively delay the fracture of laser welded joints.ZHANG Yongkang ZHANG Lei LUO Kaiyu SUN Guifang LU Jinzhong DAI Fengze ZHONG Junwei 2012Chinese Journal of Mechanical Engineering2012,25,2:11

8Measurement Uncertainty Evaluation of Conicity Error Inspected on CMM显示文摘The cone is widely used in mechanical design for rotation, centering and fixing. Whether the conicity error can be measured and evaluated accurately will directly influence its assembly accuracy and working performance. According to the new generation geometrical product specification(GPS), the error and its measurement uncertainty should be evaluated together. The mathematical model of the minimum zone conicity error is established and an improved immune evolutionary algorithm(IIEA) is proposed to search for the conicity error. In the IIEA, initial antibodies are firstly generated by using quasi-random sequences and two kinds of affinities are calculated. Then, each antibody clone is generated and they are self-adaptively mutated so as to maintain diversity. Similar antibody is suppressed and new random antibody is generated. Because the mathematical model of conicity error is strongly nonlinear and the input quantities are not independent, it is difficult to use Guide to the expression of uncertainty in the measurement(GUM) method to evaluate measurement uncertainty. Adaptive Monte Carlo method(AMCM) is proposed to estimate measurement uncertainty in which the number of Monte Carlo trials is selected adaptively and the quality of the numerical results is directly controlled. The cone parts was machined on lathe CK6140 and measured on Miracle NC 454 Coordinate Measuring Machine(CMM). The experiment results confirm that the proposed method not only can search for the approximate solution of the minimum zone conicity error(MZCE) rapidly and precisely, but also can evaluate measurement uncertainty and give control variables with an expected numerical tolerance. 9Ground Surface Ruptures and Near-Fault,Large-Scale Displacements Caused by the Wenchuan Ms8.0 Earthquake Derived from Pixel Offset Tracking on Synthetic Aperture Radar Images显示文摘The 12 May 2008 Wenchuan Ms8.0 earthquake produced surface displacements along the causative fault,the Yingxiu-Beichuan Fault,which are up to several meters near the fault.Because of the large gradient,satellite synthetic aperture radar(SAR) interferometric data are strongly incoherent;the usual SAR interferometry method does not allow such displacements to be measured.In the present study,we employed another approach,the technique based on pixel offset tracking,to solve this problem.The used image data of six tracks are from the Advanced Land Observing Satellite,Phased Array type L-band Synthetic Aperture Radar(ALOS/PALSAR) dataset of Japan.The results show that the entire surface rupture belt is 238 km long,extending almost linearly in a direction of 42° north-east.It is offset left laterally by a north-west-striking fault at Xiaoyudong,and turns at Gaochuan,where the rupture belt shifts toward the south by 5 km,largely keeping the original trend.In terms of the features of the rupture traces,the rupture belt can be divided into five sections and three types.Among them,the Beichuan-Chaping and Hongkou-Yingxiu sections are relatively complex,with large widths and variable traces along the trend.The Pingtong-Nanba and Qingping-Jingtang sections appear uniform,characterized by straight traces and small widths.West of Yingxiu,the rupture traces are not clear.North of the rupture belt,surface displacements are 2.95 m on average,mostly 2-3.5 m,with 7-9 m the maximum near Beichuan.South of the rupture belt,the average displacement is 1.75 m,dominated by 1-2 m,with 3-4 m at a few sites.In the north,the displacements in the radar line of sight are of subsidence,and in the south,they are uplifted,in accordance with a right-slip motion that moves the northern wall of the fault to the east,and the southern wall to the west,respectively.Along the Guanxian-Jiangyou Fault,there is a uplift zone in the radar line of sight,which is 66 km long,1.5-6 km wide,and has vertical displacements of approximately 2 m,but no observable rupture traces.QU Chunyan SHAN Xinjian LIU Yunhua ZHANG Guohong SONG Xiaogang ZHANG Guifang GUO Liming HAN Yufei 2012Acta Geologica Sinica(English Edition)2012,86,2:10

11Reversible RNA Modification N^1-methyladenosine(m^1A) in mRNA and tRNA显示文摘More than 100 modifications have been found in RNA. Analogous to epigenetic DNA methylation, epitranscriptomic modifications can be written, read, and erased by a complex network of proteins. Apart from N^6-methyladenosine(m^6A), N^1-methyladenosine(m^1A) has been found as a reversible modification in tRNA and mRNA. m^1A occurs at positions 9, 14, and 58 of tRNA, with m^1A58 being critical for t RNA stability. Other than the hundreds of m^1A sites in mRNA and long non-coding RNA transcripts, transcriptome-wide mapping of m^1A also identifies>20 m^1A sites in mitochondrial genes. m^1A in the coding region of mitochondrial transcripts can inhibit the translation of the corresponding proteins. 13More than Carbon Stocks:A Case Study of Ecosystem-based Benefits of REDD+ in Indonesia显示文摘During the 15th Conference of the Parties(COP 15),Parties agreed that reducing emissions from deforestation and forest degradation and enhancing 'removals of greenhouse gas emission by forests'(REDD+) in developing countries through positive incentives under the United Nations Framework Convention on Climate Change(UNFCCC) was capable of dealing with global emissions.As REDD+ seeks to lower emissions by stopping deforestation and forest degradation with an international payment tier according to baseline scenarios,opportunities for ecosystem benefits such as slowing habitat fragmentation,conservation of forest biodiversity,soil conservation may be also part of this effort.The primary objective of this study is to evaluate ecosystem-based benefits of REDD+,and to identify the relationships with carbon stock changes.To achieve this goal,high resolution satellite images are combined with Normalized Difference Vegetation Index(NDVI) to identify historical deforestation in study area of Central Kalimantan,Indonesia.The carbon emissions for the period of 2000-2005 and 2005-2009 are 2.73 × 10 5 t CO 2 and 1.47 × 10 6 t CO 2 respectively,showing an increasing trend in recent years.Dring 2005-2009,number of patches(NP),patch density(PD),mean shape index distribution(SHAPE_MN) increased 30.8%,30.7% and 7.6%.Meanwhile,largest patch index(LPI),mean area(AREA_MN),area-weighted mean of shape index distribution(SHAPE_AM),neighbor distance(ENN_MN) and interspersion and juxtaposition index(IJI) decreased by 55.3%,29.7%,15.8%,53.4% and 21.5% respectively.The area regarding as positive correlation between carbon emissions and soil erosion was approximately 8.9 × 10 3 ha corresponding to 96.0% of the changing forest.These results support the view that there are strong synergies among carbon loss,forest fragmentation and soil erosion in tropical forests.Such mechanism of REDD+ is likely to present opportunities for multiple benefits that fall outside the scope of carbon stocks.LU Heli YAN Weiyang QINYaochen LIU Guifang 2012Chinese Geographical Science2012,22,4:6
14A phaseⅠtrial of an oral subtype-selective histone deacetylase inhibitor,chidamide,in combination with paclitaxel and carboplatin in patients with advanced non-small cell lung cancer显示文摘Objective: This phase Ⅰ study was to evaluate safety, maximum tolerated dose, pharmacokinetics and preliminary antitumor activity of chidamide, a novel subtype-selective histone deacetylase(HDAC) inhibitor, in combination with paclitaxel and carboplatin in patients with advanced non-small cell lung cancer(NSCLC).Methods: Ten patients received oral chidamide 20, 25, or 30 mg twice per week continuously with paclitaxel(175 mg/m^2) and carboplatin [area under the curve(AUC) 5 mg/m L/min] administered in a 3-week cycle. Patients with response and stable disease after four cycles maintained chidamide monotherapy until disease progression or unacceptable toxicity. Blood samples were collected for pharmacokinetic analysis after the first single oral of chidamide and first combination treatment in cycle 1 from all patients.Results: Two dose-limiting toxicities were recorded in the 30 mg cohort, including thrombocytopenia and prolonged neutropenia in the first cycle. Grade 3/4 neutropenia in any cycle was observed in all patients, but was not associated with significant complications. Other grade 3/4 hematologic toxicities included thrombocytopenia and leucopenia. No significant changes were observed in pharmacokinetic parameters for both chidamide and paclitaxel. One patient in the 20 mg cohort had confirmed partial response(PR). Two out of 5 patients with brain metastases had intracranial complete remission after 4-cycle treatment.Conclusions: Chidamide combined with paclitaxel and carboplatin was generally tolerated without unanticipated toxicities or clinically relevant pharmacokinetic interactions. 15A novel mechanism controls anther opening and closing in Paris polyphylla var. Yunnanensis显示文摘The phenomena of anther opening and closing in Paris polyphylla var. yunnanensis (Franch) Hand.-Mazz were described in detail, and the effects of ecological factors on those phenomena related to anther opening, closing and the fly-pollination mechanism were discussed. Anthers of P. polyphylla var. yunnanensis open in the morning and close in the evening every day during an over 20 days’ period of anthesis. Light was detected as the main factor controlling this daily anther opening and closing. Anther opening can be reversed in response to rain and re-opens when rain stops. Relative humidity (RH) has a slight effect on anther opening and closing. Flower spans can be prolonged under laboratory conditions. 16Preparation and characteristics of flexible all-organic thin-film field-effect transistor显示文摘All-organic thin-film field-effect transistor was prepared on flexible poly(ethylene-terephthalate) (PET) substrate. Poly(methyl-methacrylate) (PMMA) and pentacene are used as a dielectric layer and a semiconductor layer, respectively. The hole mobility of the transistor can reach 2.10×10-2 cm2/Vs, and the on/off current ratio was larger than 105. The performances of the transistor, when the substrate is cured under different radius, were also measured. 17Application of a new feature extraction and optimization method to surface defect recognition of cold rolled strips显示文摘Considering that the surface defects of cold rolled strips are hard to be recognized by human eyes under high-speed cir- cumstances, an automatic recognition technique was discussed. Spectrum images of defects can be got by fast Fourier transform (FFT) and sum of valid pixels (SVP), and its optimized center region, which concentrates nearly all energies, are extracted as an original feature set. Using genetic algorithm to optimize the feature set, an optimized feature set with 51 features can be achieved. Using the optimized feature set as an input vector of neural networks, the recognition effects of LVQ neural networks have been studied. 18Stem cell lineage in body layer specialization and vascular patterning of rice root and leaf显示文摘Since the first appearance of vascular plants during evolution, the plant body has become specialized for adaption to land conditions. Much of our knowledge of plant body specialization and the origins of tissues from stem cells have been obtained from studies on the dicot Arabidopsis thaliana. However, less is known about plant body specialization in monocots, another important branch of angiosperms. In this study, we analyzed stem cell lineage and differentiation during development of the root and leaf of the monocot model plant rice(Oryza sativa). Our results showed that three body layers of rice are established from stem cells accompanied by progressively reduced pluripotency. Layer 1(L1) is a single-cell layer of epidermis; L2 is the cortex/endodermis in the root and the mesophyll in the leaf; and L3 is the site of vascular initiation. At least two common steps in vascular development are shared between rice root and leaf. The preprocambium divides to form the procambium and root pericycle or leaf outer sheath. The procambium further differentiates into the xylem, phloem and circumambient cells. We found that the outer sheath of leaf vascular bundles originates not only from the preprocambium of L3,but also from the mesophyll precursor cells of L2. In addition, WUSCHEL-RELATED HOMEOBOX(WOX)genes are expressed in not only the stem cell niche but also metaxylem precursor in rice. 19Expression of human soluble TRAIL in Chlamydomonas reinhardtii chloroplast显示文摘Tumor necrosis factor-related apop-tosis-inducing ligand (TRAIL) induces selectively apoptosis in various tumor cells and virus-infected cells, but rarely in normal cells. A chloroplast expres-sion vector, p64TRAIL, containing the cDNA coding for the soluble TRAIL (sTRAIL), was constructed with clpP-trnL-petB-chlL-rpl23-rpl2 as Chlamydomonas reinhardtii plastid homologous recombinant frag-ments and spectinomycin-resistant aadA gene as a select marker. The plasmid p64TRAIL was trans-ferred into the chloroplast genome of C. reinhardtii by the biolistic method. Three independently trans-formed lines were obtained by 100 mg/L spectino-mycin selection. PCR amplification, Southern blot analysis of the sTRAIL coding region DNA and culti-vation cells in the dark all showed that the exogenous DNA had been integrated into chloroplast genome of C. reinhardtii. Western blot analysis showed that human soluble TRAIL was expressed in C. reinhardtii chloroplast. The densitometric analysis of Western blot indicated that the expressed human sTRAIL protein in the chloroplasts of C. reinhardtii accounted for about 0.43%―0.67% of the total soluble proteins. 20Underwater Laser Welding/Cladding for High-performance Repair of Marine Metal Materials:A Review显示文摘With the rapid developments of marine resource exploitation,mounts of marine engineering equipment are settled on the ocean.When it is not possible to move the damaged equipment into a dry dock,welding operations must be performed in underwater environments.The underwater laser welding/cladding technique is a promising and advanced technique which could be widely applied to the maintenance of the damaged equipment.The present review paper aims to present a critical analysis and engineering overview of the underwater laser welding/cladding technique.First,we elaborated recent advances and key issues of drainage nozzles all over the world.Next,we presented the underwater laser processing and microstructural-mechanical behavior of repaired marine materials.Then,the newly developed powder-feeding based and wire-feeding based underwater laser direct metal deposition techniques were reviewed.The differences between the convection,conduction,and the metallurgical kinetics in the melt pools during underwater laser direct metal deposition and in-air laser direct metal deposition were illustrated.After that,several challenges that need to be overcame to achieve the full potential of the underwater laser welding/cladding technique are proposed.Finally,suggestions for future directions to aid the development of underwater laser welding/cladding technology and underwater metallurgical theory are provided.The present review will not only enrich the knowledge in the underwater repair technology,but also provide important guidance for the potential applications of the technology on the marine engineering.Guifang Sun Zhandong Wang Yi Lu Mingzhi Chen Kun Yang Zhonghua Ni 2022Chinese Journal of Mechanical Engineering2022,35,1:4
