1MiR-122-5p suppresses the proliferation, migration, and invasion of gastric cancer cells by targeting LYN显示文摘Gastric cancer(GC)is one of malignant tumors with high mortality and morbidity in the world.MicroRNA-122(miR-122)acts as a tumor suppressor in a variety of cancers and has been found to be dominant in gastric adenocarcinoma.However,the specific biological function of miR-122-5p in GC is not completely clear.In this study,we found that miR-122-5p was low-expressed in GC tissues and cell lines by using qRT-PCR.Overexpression of miR-122-5p inhibited the proliferation,migration,and invasion of GC cells by using CCK-8 and transwell assays.On the contrary,downregulation of miR-122-5p promoted the proliferation,migration,and invasion of GC cells.In addition,we found that the expression of LYN,an Src family tyrosine kinase,was inversely correlated with miR-122-5p expression in GC tissues by using western blot analysis,immunohistochemistry,and qRT-PCR assays.Meanwhile,luciferase assay results indicated that LYN is a direct target of miR-122-5p in GC cells.Moreover,silencing LYN expression by its siRNA inhibited the proliferation,migration,and invasion of GC cells.Importantly,overexpression of LYN restored miR-122-5p-mediated inhibition of the proliferation,migration,and invasion of GC cells.Taken together,our results indicated miR-122-5p inhibited the proliferation,migration,and invasion by targeting LYN in GC.Lei Meng Zhangming Chen Zhe Jiang Ting Huang Jie Hu Panquan Luo Hongli Zhang Mengqi Huang Lei Huang Yu Chen Ming Lu A-man Xu Songcheng Ying 2020Acta Biochimica et Biophysica Sinica2020,52,1:12

3Breast cancer-specific serum peptide profiles显示文摘Early detection of breast cancer is paramount to successful clinical therapy. Yet, early-stage breast cancer lacks specific symptoms or biomarkers. With the emerging of the mass spectrometric (MS) -based signatures as biomarkers, we investigated breast cancer-related serum profile pattern through class prediction and independent validation, and used Fourier transfer MS to identify breast cancer signature. We now show a distinctive serum peptide pattern that discriminates breast cancer from healthy controls with 93.2% sensitivity and 95.4% specificity. m/z 5901.70 and 4465.74 of ion fragment of FPA and alpha1-antichymotrypsin are found in the signatures that predominantly discriminate breast cancer from healthy individuals. 4Development of a rapid and sensitivity magnetic chemiluminescence immunoassay for DNA methyltransferase 1 in human serum显示文摘DNA methyltransferase 1(DNMT1)is a useful biomarker for lung cancer in early clinical diagnosis.A rapid magnetic chemiluminescence immunoassay(MCLIA)for DNMT1 in human serum has been developed.Horseradish peroxidase(HRP)-second-Ab was used to labeled polyclonal antibodies of anti-DNMT1.DNMT1 in sample integrates with specific immunomagnetic beads and can constitute a supersandwiched immunoreaction.In magnetic field,nonspecific materials can be separated.After luminescent substrate luminol-H2O2-BIP was added,the relative light unit(RLU)of HRP was detected and was discovered to be directly proportional to the content of DNMT1 in sample.The correlative variables involved in the MCLIA value were optimized and the methodological evaluation was carried out.After optimization,in the range of0.5–128 ng/mL,the linear regression equation was y=0.5014 x+1.769(x was logCDNMT1,y was relative luminescence units(RLU)/RLU0),and the limit of detection was 0.01 ng/mL.The RSD of intra-and interassays were 15.8%–16.9%and 14.3%–18.1%,respectively.The recovery was from 70.0%to 106.2%.Furthermore,paralleled with purchasable enzyme-linked immunosorbent assay(ELISA)kits,MCLEIA had lower detection limit,wider linear range and shorter detection time.Therefore,the MCLEIA established in this study could be used for the sensitive detection of DNMT1 in serum sample.Sitian He Leiliang He Beibei Liu Songcheng Yu Li'e Liu Yongmei Tian Jia Wang Lihua Ding Yilin Wang Lingbo Qu Fei Yu Yongjun Wu 2019Chinese Chemical Letters2019,30,5:4
5Voltage-gated sodium channel Nav1.5 promotes proliferation, migration and invasion of oral squamous cell carcinoma显示文摘The protein voltage-gated sodium channel Nav1.5 is highly upregulated in various types of cancer and, in general, promotes cancer cell invasiveness and metastatic progression. A previous study found that Nav1.5 was highly expressed in poorly differentiated oral squamous cell carci noma (OSCC). However, whether Nav1.5 enhances invasiveness and metastasis of OSCC are still unknown. In this study, we found that Nav1.5 was highly expressed in OSCC cell lines compared with normal oral keratinocyte HOK cell line by using western blot analysis. CCK-8 assay results revealed that downregulation of Nav1.5 expression by its specific siRNA reduced proliferation of OSCC HSC-3 cells. Moreover, transwell assay results showed Nav1.5 knockdown significantly inhibited migration and invasion of HSC-3 cells. Meanwhile, qRT-PCR and western blot analysis results showed that epidermal growth factor (EGF) induced Nav1.5 expression in a time-and dosedependent manner. In addition, EGF promoted proliferation, migration and invasion of HSC-3 cells. Importantly, the Nav1.5 inhibitor tetrodotoxin significantly inhibited the proliferation of HSC-3 cells and impeded the migration and invasion of HSC-3 cells. Furthermore, it was found that siRNAmediated knockdown of Nav1.5 also lessened the proliferation of HSC-3 cells and blocked the migration and invasion of HSC-3 cells. 14Load Balancing Based on Anycast and Pseudo-Mobility in IPv6显示文摘We proposed a new Load balancing mechanism, called LBAM (Load balancing based on anycast and pseudo-mobility in IPv6), by which sessions can be distributed across a pool of servers for maximum performance. Load balancing is needed in many fields, such as Web server, FTP (File transfer protocol) server and NAT-PT (Network address translation-protocol translation) gateway, and there exist several approaches for load balancing. However, current approaches have problems such as poor scalability, inability to balance session flow,long latency time, and topological constraint on server pools. In this paper, we describe how LBAM works in principle without the above problems. We also present our simulation work using NS-2 and discuss performance comparison between LBAM and other load balancing mechanisms. The results show that LBAM has very advanced performance and scalability. 16Self-assembly of two- dimensional DNA crystals显示文摘Self-assembly of synthetic oligonucleotides into two-dimensional lattices presents a 慴ottom-up?approach to the fabrication of devices on nanometer scale. We report the design and observation of two-dimensional crystalline forms of DNAs that are composed of twenty-one plane oligonucleo-tides and one phosphate-modified oligonucleotide. These synthetic sequences are designed to self-assemble into four double-crossover (DX) DNA tiles. The 憇ticky ends?of these tiles that associate according to WatsonCrick抯 base pairing are programmed to build up specific periodic patterns upto tens of microns. 18Enhanced energy-storage density and temperature stability of Pb_(0.89)La_(0.06)Sr_(0.05)(Zr_(0.95)Ti_(0.05))O_(3) anti-ferroelectric thin film capacitor显示文摘As the fundamental energy storage components in electronic systems,dielectric capacitors with high power densities were demanded.In this work,the anti-ferroelectric Pb_(0.89)La_(0.06)Sr_(0.05)(Zr_(0.95)Ti_(0.05))O_(3)(PLSZT)ceramics and thin film capacitor were successfully fabricated by a solid-state reaction route and pulsed laser deposition method,respectively.The ferroelectric,dielectric,energy-storage properties,and temperature stability of anti-ferroelectric PLSZT capacitor were investigated in detail.By compared with the PLSZT ceramic(energy storage density is 1.29 J/cm^(3) with an efficiency of 78.7%under 75 kV/cm),the anti-ferroelectric PLSZT thin film capacitors exhibited the enhanced energy storage density of 52.6 J/cm^(3) with efficiency of 67.7%under an electric field as high as 2068.9 kV/cm,and the enhanced energy-storage temperature stabilities from room temperature(RT)to more than 200℃ were demonstrated,and the oxygen defects mechanism and size effect were discussed.Moreover,the fast charging(~0.05 μs)and discharging(~0.15 μs)time were certified for the anti-ferroelectric PLSZT film capacitor.These findings broaden the horizon for PLSZT anti-ferroelectrics in high energy storage properties and show promising for manufacturing pulse power capacitor.Zhenhua Tang Songcheng Hu Dijie Yao Zeli Li Zhigang Liu Xiaobin Guo Biao Lu Jingmin Fan Xin-Gui Tang Sheng-Guo Lu Ju Gao 2022Journal of Materiomics2022,8,1:0
