1Indoleamine 2,3-dioxygenase (IDO) is essential for dendritic cell activation and chemotactic responsiveness to chemokines显示文摘Indoleamine 2, 3-dioxygenase (IDO) is a rate-limiting enzyme for the tryptophan catabolism. In human and murine cells, IDO inhibits antigen-specific T cell proliferation in vitro and suppresses T cell responses to fetal alloantigens during murine pregnancy. In mice, IDO expression is an inducible feature of specific subsets of dendritic cells (DCs), and is important for T cell regulatory properties. However, the effect of IDO and tryptophan deprivation on DC func- tions remains unknown. We report here that when tryptophan utilization was prevented by a pharmacological inhibitor of IDO, 1-methyl tryptophan (1MT), DC activation induced by pathogenic stimulus lipopolysaccharide (LPS) or inflam- matory cytokine TNF-α was inhibited both phenotypically and functionally. Such an effect was less remarkable when DC was stimulated by a physiological stimulus, CD40 ligand. Tryptophan deprivation during DC activation also regu- lated the expression of CCR5 and CXCR4, as well as DC responsiveness to chemokines. 2Nutrient supplemented serum-free medium increases cardiomyogenesis efficiency of human pluripotent stem cells显示文摘AIM: To development of an improved p38 MAPK inhibitor-based serum-free medium for embryoid body cardiomyocyte differentiation of human pluripotent stem cells. METHODS: Human embryonic stem cells (hESC) differentiated to cardiomyocytes (CM) using a p38 MAPK inhibitor (SB203580) based serum-free medium (SB media). Nutrient supplements known to increase cell viability were added to SB medium. The ability of these supplements to improve cardiomyogenesis was evaluated by measurements of cell viability, total cell count, and the expression of cardiac markers via flow cytometry. An improved medium containing Soy hydrolysate (HySoy) and bovine serum albumin (BSA) (SupSB media) was developed and tested on 2 additional cell lines (H1 and Siu-hiPSC). Characterization of the cardiomyocytes was done by immunohistochemistry, electrophysiology and quantitative real-time reverse transcriptionpolymerase chain reaction. RESULTS: hESC cell line, HES-3, differentiating in SB medium for 16 d resulted in a cardiomyocyte yield of 0.07 ± 0.03 CM/hESC. A new medium (SupSB media) was developed with the addition of HySoy and BSA to SB medium. This medium resulted in 2.6 fold increase in cardiomyocyte yield (0.21 ± 0.08 CM/hESC). The robustness of SupSB medium was further demonstrated using two additional pluripotent cell lines (H1, hESC and Siu1, hiPSC), showing a 15 and 9 fold increase in cardiomyocyte yield respectively. The age (passage number) of the pluripotent cells did not affect the cardiomyocyte yields. Embryoid body (EB) cardiomyocytes formed in SupSB medium expressed canonical cardiac markers (sarcomeric α-actinin, myosin heavy chain and troponin-T) and demonstrated all three major phenotypes: nodal-, atrial- and ventricular-like. Electrophysiological characteristics (maximum diastolic potentials and action potential durations) of cardiomyocytes derived from SB and SupSB media were similar. 4Outcome and prognostic factors in 110 consecutive patients with primary uterine leiomyosarcoma:A Rare Cancer Network study显示文摘Objective: Primary uterine leiomyosarcomas(ULMS) are rare,and the optimal treatment is controversial.We aimed to assess the outcome and prognostic factors in a multicenter population of women treated for primary ULMS.Methods: We retrospectively collected data of 110 women treated in 19 institutions of the Rare Cancer Network(RCN).Inclusion criteria consisted of a pathology report confirming the diagnosis of ULMS,aged 18–80 years,complete International Federation of Gynecology and Obstetrics(FIGO) stage information,complete information on treatment,and a minimum follow-up of 6 months.Local control(LC) and locoregional control(LRC),overall survival(OS) and disease-free survival(DFS) rates were computed using the Kaplan-Meier method.Univariate analysis was implemented using the log rank test,and multivariate analysis using the Cox model.Results: All patients underwent surgery.Seventy-five patients(68%) received adjuvant radiotherapy(RT),including brachytherapy in 18(16%).Seventeen patients(15%) received adjuvant chemotherapy.Median follow-up was 58(range,6–240) months.Five-year OS and DFS rates were 50% and 34%,and LC and LRC rates were 88%and 72%,respectively.On multivariate analysis,independent favorable prognostic factors were younger age,FIGO stage I,small tumor size,previous uterine disease,and no vascular invasion for OS and DFS.FIGO stage was the only favorable factor influencing LRC.Adjuvant local or systemic treatments did not improve the outcomes.Eight patients treated with RT presented a grade 3 acute toxicity,and only one patient with grade 3 late toxicity.Conclusions: In this large population of primary ULMS patients,we found good results in terms of LC and LRC.Nevertheless,OS remains poor,mainly due to the occurrence of distant metastases.An early diagnosis seemed to improve the prognosis of the patients.Adjuvant local or systemic treatments,or more aggressive surgicalprocedures such as the Wertheim procedure,did not seem to impact the outcome.Alessandra Franzetti Pellanda Berardino De Bari Elisabeth Deniaud-Alexandre MarcoKrengli Paul Van Houtte Antonella Riehetti Salvador Villa Hadassah Goldberg EwaSzutowiez-Zielifiska Michel Bolla Heidi Rutten Marc Van Eijkeren Philip Poortmans Guido Henke Yavuz Anaeak Steve Chan Christine Landmann Carine Kirkove LueianoScandolaro Jacques Bernier Rene-Olivier Mirimanof Mahmut Ozsahin 2017Chinese Journal of Cancer Research2017,29,6:2

19A Systematic Approach to Pulmonary Valve Replacement in the Current Era显示文摘Background:Pulmonary valve replacement(PVR)can be accomplished via surgical,transcatheter,or hybrid approaches.There are inherent advantages to transcatheter PVR and hybrid PVR without cardiopulmonary bypass.We review the methods and results of a standardized institutional approach to PVR.Methods:Retrospective review of all PVR cases between February 2017 and February 2020.Hybrid PVR entailed off-pump RVOT plication with percutaneous transcatheter PVR.Results:Primary transcatheter PVR was attempted in 37,hybrid PVR was performed in 11,and on-pump surgical PVR was performed in 9.Median age at PVR was 27 years(6–65).Primary transcatheter PVR was successful in 35/37(2 converted to surgical).Standard surgical PVR was utilized for positive coronary compression testing(n=4),stent/valve system migration(n=2),or patient preference(n=3).In the hybrid group mean RVOT diameter was 34 mm(32–38).Median length of stay was 1 day for transcatheter PVR,5 for surgical,and 3 for hybrid(p=0.02).Median follow-up was 1.5 years.Re-interventions were one balloon valve dilation in a transcatheter PVR,and one valve dilation with subsequent transcatheter valve-in-valve PVR in the surgical cohort.One hybrid patient expired 11 months post procedure.Conclusions:A systematic approach to PVR utilizing all approaches in pre-defined order of preference leads to consistent outcomes in a wide variety of anatomic configurations.Transcatheter PVR may be accomplished in the majority of patients.When necessary,hybrid off-pump RVOT plication with transcatheter PVR avoids the need for cardiopulmonary bypass.R.Allen Ligon Larry A.Latson Mark M.Ruzmetov Kak-Chen Chan Todd Roth Immanuel I.Turner Frank G.Scholl Steve Bibevski 2021Congenital Heart Disease2021,16,3:0
