1Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29

3Chemical characterization of size-resolved aerosols in four seasons and hazy days in the megacity Beijing of China显示文摘Size-resolved aerosol samples were collected by MOUDI in four seasons in 2007 in Beijing. The PM10 and PM1.8mass concentrations were 166.0 ± 120.5 and 91.6 ± 69.7 μg/m^3, respectively,throughout the measurement, with seasonal variation: nearly two times higher in autumn than in summer and spring. Serious fine particle pollution occurred in winter with the PM1.8/PM10 ratio of 0.63, which was higher than other seasons. The size distribution of PM showed obvious seasonal and diurnal variation, with a smaller fine mode peak in spring and in the daytime. OM(organic matter = 1.6 × OC(organic carbon)) and SIA(secondary inorganic aerosol) were major components of fine particles, while OM, SIA and Ca_2+were major components in coarse particles. Moreover, secondary components, mainly SOA(secondary organic aerosol) and SIA,accounted for 46%–96% of each size bin in fine particles, which meant that secondary pollution existed all year. Sulfates and nitrates, primarily in the form of(NH_4)_2SO_4, NH_4NO_3, Ca SO_4, Na_2SO_4 and K_2SO_4, calculated by the model ISORROPIA II, were major components of the solid phase in fine particles. The PM concentration and size distribution were similar in the four seasons on non-haze days, while large differences occurred on haze days, which indicated seasonal variation of PM concentration and size distribution were dominated by haze days. 6Soil respiration in tropical seasonal rain forest in Xishuangbanna, SW China显示文摘With the static opaque chamber and gas chromatography technique, from January 2003 to January 2004 soil respiration was investigated in a tropical seasonal rain forest in Xishuangbanna, SW China. In this study three treatments were applied, each with three replicates: A (bare soil), B (soil+litter), and C (soil+litter+seedling). The results showed that soil respiration varied seasonally, low from December 2003 to February 2004, and high from June to July 2004. The annual average values of CO2 efflux from soil respiration differed among the treatments at 1% level, with the rank of C (14642 mgCO2· m-2. h-1)>B (12807 mgCO2· m-2. h-1)>A (9532 mgCO2· m-2. h-1). Diurnal variation in soil respiration was not apparent due to little diurnal temperate change in Xishuangbanna. There was a parabola relationship between soil respiration and soil moisture at 1% level. Soil respiration rates were higher when soil moisture ranged from 35% to 45%. There was an exponential relationship between soil respiration and soil temperature (at a depth of 5cm in mineral soil) at 1% level. The calculated Q1o values in this study,ranging from 2.03 to 2.36, were very near to those of tropical soil reported. 8Chemical characteristics of size-resolved aerosols in winter in Beijing显示文摘Size-resolved aerosols were continuously collected by a Nano Sampler for 13 days at an urban site in Beijing during winter 2012 to measure the chemical composition of ambient aerosol particles. Data collected by the Nano Sampler and an ACSM(Aerodyne Aerosol Chemical Speciation Monitor) were compared. Between the data sets,similar trends and strong correlations were observed,demonstrating the validity of the Nano Sampler. PM10 and PM2.5concentrations during the measurement were 150.5 ± 96.0 μg/m3(mean ± standard variation)and 106.9 ± 71.6 μg/m3,respectively. The PM2.5/PM10 ratio was 0.70 ± 0.10,indicating that PM2.5dominated PM10. The aerosol size distributions showed that three size bins of 0.5–1,1–2.5 and 2.5–10 μm contributed 21.8%,23.3% and 26.0% to the total mass concentration(TMC),respectively. OM(organic matter) and SIA(secondary ionic aerosol,mainly SO42-,NO3-and NH4+) were major components of PM2.5. Secondary compounds(SIA and secondary organic carbon) accounted for half of TMC(about 49.8%) in PM2.5,and suggested that secondary aerosols significantly contributed to the serious particulate matter pollution observed in winter. Coal burning,biomass combustion,vehicle emissions and SIA were found to be the main sources of PM2.5. 10Famitinib in combination with concurrent chemoradiotherapy in patients with locoregionally advanced nasopharyngeal carcinoma: a phase 1, open-label, dose-escalation Study显示文摘Background:Famitinib is a tyrosine kinase inhibitor against multiple targets,including vascular endothelial growth factor receptor 2/3,platelet-derived growth factor receptor,and stem cell factor receptor(c-kit).Previous studies have demonstrated anti-tumour activities of famitinib against a wide variety of advanced-stage solid cancers.We aimed to determine the safety and efficacy of famitinib with concurrent chemoradiotherapy(CCRT)in patients with locoregionally advanced nasopharyngeal carcinoma(NPC).We also evaluated the feasibility of contrast-enhanced ultrasound(D-CEUS)as a predictor of early tumour response to famitinib and to correlate functional parameters with clinical efficacy.Methods:The trial was conducted in subjects with stage III or IVa-b NPC using a 3+3 design of escalating fami-tinib doses.Briefly,subjects received 2 weeks of famitinib monotherapy followed by 7 weeks of famitinib plus CCRT.D-CEUS of the neck lymph nodes was performed at day 0,8 and 15 after famitinib was administered before starting concurrent chemoradiotherapy.End points included safety,tolerability and anti-tumour activity.Results:Twenty patients were enrolled(six each for 12.5,16.5 and 20 mg and two for 25 mg).Two patients in the 25 mg cohort developed dose-limiting toxicities,including grade 4 thrombocytopenia and grade 3 hypertension.The most common grade 3/4 adverse events were leukopenia,neutropenia and radiation mucositis.D-CEUS tests showed that more than 60%of patients achieved a perfusion parameter response after 2 weeks taking famitinib alone,and the parameter response was associated with disease improvement.In the famitinib monotherapy stage,three patients(15%)showed partial responses.The complete response rate was 65%at the completion of treatment and 95%3 months after the treatment ended.After a median follow-up of 44 months,the 3-year progression-free survival(PFS)and distant metastasis-free survival were 70%and 75%,respectively.Subjects with a decrease of perfusion parameter response,such as peak intensity decreased at least 30%after 1 week of famitinib treatment,had higher 3-year PFS(90.9%vs.44.4%,95%CI 73.7%-100%vs.11.9%-76.9%,P<0.001)than those with an increase or a reduction of less than 30%.Conclusions:The recommended famitinib dose for phase II trial is 20 mg with CCRT for patients with local advanced NPC.D-CEUS is a reliable and early measure of efficacy for famitinib therapies.Further investigation is required to confirm the effects of famitinib plus chemoradiotherapy.Qiuyan Chen Linquan Tang Na Liu Feng Han Ling Guo Shanshan Guo Jianwei Wang Huai Liu Yanfang Ye Lu Zhang Liting Liu Pan Wang Yingqin Li Qingmei He Xiaoqun Yang Qingnan Tang Yang Li YuJing Liang XueSong Sun Chuanmiao Xie Yunxian Mo Ying Guo Rui Sun Haoyuan Mo Kajia Cao Xiang Guo Musheng Zeng Haiqiang Mai Jun Ma 2018Cancer Communications2018,38,1:6
11Achieving three-layered Al/Mg/Al sheet via combining porthole die co-extrusion and hot forging显示文摘Al/Mg/Al sheet with good bonding quality and mechanical properties was fabricated based on the proposed porthole die co-extrusion and forging(PCE-F)process.There were no voids,cracks or other defects on the Al/Mg interface.A continuous diffusion zone with two-sub-layer structure was formed across the Al/Mg interface,and its width increased with higher temperature or reduction ratio.The sub-layers formed at low and high temperature were identified to be solid solutions and intermetallic compounds(IMCs)including y-MgpAl^and^-Al3Mg.In Al layer,the welding zone mainly consisted of fine equiaxed grains with several coarse elongated grains,while the majority of matrix zone is coarse elongated grains.The rolling textures were dominated in both welding and matrix zones.In Mg layer,the welding zone exhibited complete DRXed grain structure,while several unDRXed coarse grains were observed in the matrix zone.With the increasing temperature,the grain size of Al and Mg layer firstly decreased and then increased.High reduction ratio strongly refined the grain structure of Al layer,while slightly affected the Mg layer.The Al/Mg/AI sheet experienced stress-drops twice during the tensile test.The first stress-drop was determined by the IMCs and microstructure of Mg layer,while the second stress-drop was closely related to the microstructure of Al layer.Al/Mg/Al sheet forged at the lowest temperature without the formation IMCs exhibited the highest stress for the first stress-drop,and that forged under the highest reduction ratio with the smallest grain size in Al layer had the highest stress for the second stress-drop.Jianwei Tang Liang Chen Guoqun Zhao Cunsheng Zhang Lu Sun 2020Journal of Magnesium and Alloys2020,8,3:6
12Investigation on the spatial distribution characteristics of behind-armor debris formed by the perforation of EFP through steel target显示文摘The behind-armor debris(BAD) formed by the perforation of an EFP is the main damage factor for the secondary destruction to the behind-armor components.Aiming at investigating the BAD caused by EFP,flash X-ray radiography combined with an experimental witness plate test method was used,and the FEM-SPH adaptive conversion algorithm in LS-DYNA software was erployed to model the perforation process.The simulation results of the debris cloud shape and number of debris were in good agreement with the flash X-ray radiographs and perforated holes on the witness plate,respectlvely.Threedimensional numerical simulations of EFP's penetration under various impact conditions were conducted.The results show that,an ellipsoidal debris cloud,with the major-to-minor axis radio(a/b)smaller than that caused by shaped charge jets,was formed behind the target.With the increase of target thickness(h) and decrease of impact velocity(v_0) and obliquity(θ),the value of a/b decreases.The number of debris ejected from target is significantly higher than that from EFP.Based on the statistical analysis of the spatial distribution of the BAD,An engineering calculation model was established considering the influence of h,v_0 and θ.The model can with reasonable accuracy predict the quantity and velocity distribution characteristics of BAD formed by EFP.Xin Wang Jianwei Jiang Shengjie Sun Jianbing Men Shuyou Wang 2020Defence Technology(防务技术)2020,16,1:6
13Identification of causative compounds and microorganisms for musty odor occurrence in the Huangpu River,China显示文摘There are regular problems of musty odor in the Huangpu River,a major source of drinking water for Shanghai,China.In this study,the musty odor and its main causative compounds in the Huangpu River source water were confirmed through a yearly investigation using flavor profile analysis combined with HSPME-GC-MS analysis.The investigation showed that 2-methylisoborneol (2-MIB) with a concentration level between 28.6 and 71.0 ng/L was responsible for the musty odor in summer from July to September.Microscopic observation confirmed with the cloning results showed that Phormidium spp.,which accounted for 80%-95% of the algal cell density,was the microorganisms responsible for the production of 2-MIB and the estimated 2-MIB yield was 0.022 pg/cell.Results from a wide-area sampling campaign in the Huangpu River watershed showed that,other than the large tributaries receiving water from Tai Lake,several small creeks close to the intake may have contributed most of the 2-MIB and the Phormidium spp.to the Huangpu River source water.This study provides methodology for the investigation of odor causing compounds and microorganisms in river-type source water,and the result will be useful for water quality control in both source water and drinking water.Daolin Sun Jianwei Yu Wei An Min Yang Guoguang Chen Shujun Zhang 2013Journal of Environmental Sciences2013,25,3:6
14Synergistic effect of W and P on ZSM-5 and its catalytic performance in the cracking of heavy oil显示文摘In order to develop the conversion of heavy oil with a high yield of propylene in the catalytic cracking process, ZSM-5 zeolite was modified by tungsten and phosphorus, which was proved to be an effective method. Characterization results show that the improvement of catalytic performance could be correlated to the interaction of phosphorus and tungsten species on ZSM-5. P inhibited the aggregation of tungsten species on ZSM-5 and was conductive to convert the tungsten species with octahedral coordination into tetrahedral coordination. And this ultimately led to that more acid sites were reserved after hydrothermal treatment in the tungsten and phosphorus co-modified ZSM-5 catalyst.Phosphorus species played an important role to restrain the dehydrogenation activity of tungsten. 15In-situ structural evolution analysis of Zr-doped Na_(3)V_(2)(PO_(4))_(2)F_(3) coated by N-doped carbon layer as high-performance cathode for sodium-ion batteries显示文摘With great superiorities in energy density,rate capability and structural stability,Na_(3)V_(2)(PO_(4))_(2) F_(3)(NVPF)has attracted much attentions as cathode of sodium ion battery(SIB),but it also faces challenges on its poor intrinsic electronic conductivity and the controversial de/sodiation mechanism.Herein,a series of Zr-doped NVPF coated by N-doped carbon layer(~5 nm in thickness,homogenously)materials are fabricated by a sol-gel method,and the optimized heteroatom-doping amounts of Zr and N doping improve intrinsic properties on enlarging lattice distance and enhancing electronic conductivity,respectively.Specifically,among all samples of Na_(3) V_(2-x)Zr_(x)(PO_(4))_(2) F_(3)/NC(NVPF-Zr-x/NC,x=0,0.01,0.02,0.05,and 0.1),the optimized electrode of NVPF-Zr-0.02/NC delivers high reversible capacities(119.2 mAh g^(-1) at0.5 C),superior rate capability(98.1 mA h g^(-1) at 20 C)and excellent cycling performance.The structural evolution of NVPF-Zr-0.02/NC electrode,in-situ monitored by X-ray diffractometer,follows a step-wise Na-extraction/intercalation mechanism with reversible multi-phase changes,not just a solid-solutionreaction one.Full cells of NVPF-Zr-0.02/NC//hard carbon demonstrate high capacity(99.8 mA h g^(-1) at 0.5 C),high out-put voltage(3.5 V)and good cycling stability.This work is favorable to accelerate the development of high-performance cathode materials and explore possible redox reaction mechanisms of SIBs.Chuan Guo Jianwei Yang Zhiyuan Cui Shuo Qi Qianqian Peng Weiwei Sun Li-Ping Lv Yi Xu Yong Wang Shuangqiang Chen 2022Journal of Energy Chemistry2022,31,2:5
16Proto-Tethys ophiolitic mélange in SW Yunnan: Constraints from zircon U-Pb geochronology and geochemistry显示文摘An early Paleozoic Proto-Tethys ocean in western Yunnan has long been postulated although no robust geological evidence has been identified.Here we investigated the recently-identified Mayidui and Wanhe ophiolitic mélanges in SW Yunnan,which occurs in a N-S trending belt east of the late Paleozoic Changning-Menglian suture zone.The ophiolites consist mainly of meta-basalts(amphibole schists),meta-(cumulate)gabbros and gabbroic diorites,and meta-chert-shale,representing ancient oceanic crust and pelagic and hemipelagic sediments,respectively.Six samples of gabbros and gabbroic diorites from 3 profiles(Mayidui,Kongjiao and Yinchanghe)yielded zircon U-Pb ages between 462±6 Ma and 447±9 Ma,constraining the formation of the Mayidui and Wanhe ophiolites to Middle Ordovician.Gabbros from the Mayidui and Kongjiao profiles share similar geochemical characteristics with affinities to tholeiitic series,and are characterized by depleted to slightly enriched LREEs relative to HREEs with(La/Sm)N=0.69-1.87,(La/Yb)N=0.66-4.72.These,along with their predominantly positive wholerock eNd(t)and zircon eHf(t)values,indicate a MORB-like magma source.By contrast,the meta-mafic rocks from the Yinchanghe profile show significantly enriched LREEs((La/Sm)N=0.97-3.33,(La/Yb)N=1.19-14.93),as well as positive whole-rock eNd(t)and positive to negative zircon eHf(t)values,indicating an E-MORB-type mantle source.These geochemical features are consistent with an intra-oceanic setting for the formation of the Mayidui-Wanhe ophiolites.Our data,integrated with available geological evidence,provide robust constraints on the timing and nature of the Mayidui-Wanhe ophiolitic mélange,and suggest that the ophiolites represent remnants of the Proto-Tethys Ocean,which opened through separation of the Indochina and Simao blocks from the northern margin of Gondwana before the Early Cambrian,and evolved through to the Silurian.Guichun Liu Zaibo Sun Jianwei Zi M.Santosh Tianyu Zhao Qinglai Feng Guangyan Chen Xiaomei Nie Jing Li Shitao Zhang 2021Geoscience Frontiers2021,12,5:5
17Comparison of viral RNA-host protein interactomes across pathogenic RNA viruses informs rapid antiviral drug discovery for SARS-CoV-2显示文摘In contrast to the extensive research about viral protein–host protein interactions that has revealed major insights about how RNA viruses engage with host cells during infection,few studies have examined interactions between host factors and viral RNAs(vRNAs).Here,we profiled vRNA–host protein interactomes for three RNA virus pathogens(SARS-CoV-2,Zika,and Ebola viruses)using ChIRP-MS.Comparative interactome analyses discovered both common and virus-specific host responses and vRNA-associated proteins that variously promote or restrict viral infection.In particular,SARS-CoV-2 binds and hijacks the host factor IGF2BP1 to stabilize vRNA and augment viral translation.Our interactome-informed drug repurposing efforts identified several FDA-approved drugs(e.g.,Cepharanthine)as broad-spectrum antivirals in cells and hACE2 transgenic mice.A co-treatment comprising Cepharanthine and Trifluoperazine was highly potent against the newly emerged SARS-CoV-2 B.1.351 variant.Thus,our study illustrates the scientific and medical discovery utility of adopting a comparative vRNA-host protein interactome perspective.Shaojun Zhang Wenze Huang Lili Ren Xiaohui Ju Mingli Gong Jian Rao Lei Sun Pan Li Qiang Ding Jianwei Wang Qiangfeng Cliff Zhang 2022Cell Research2022,32,1:5

19Estimation of thermophysical properties of solid propellants based on particle packing model显示文摘A method for the estimation of thermophysical properties of two-and multi-phase solid propellants is proposed in this paper.The theoretical solutions for thermal conductivity and specific heat of a homogeneous solid propellant cell in the transient thermal conductivity process are deduced on the condition that one boundary of the cell is heated while others are adiabatic.A homogenization theory and the finite element method are employed to compute the mean temperature and heat flux of a representative volume element(RVE).According to the mean results and the theoretical solutions,the effective thermal conductivity and specific heat of solid propellant can be estimated.A packing algorithm,considering the solid particles(ammonium perchlorate(AP)or aluminum)as spheres or discs,is used to match the size distribution and volume fraction of solid propellants,and some mesoscopic models of two-phase and three-phase solid propellants are established.According to the estimation theory proposed in this paper,the effective thermal conductivity and specific heat of solid propellants are predicted.The effect of AP or Al volume fraction is also discussed in this paper.ZHANG JianWei ZHI ShiJun SUN Bing 2013Science China(Technological Sciences)2013,56,12:4
