5Difluorocarbene-derived rapid late-stage trifluoromethylation of 5-iodotriazoles for the synthesis of^(18)F-labeled radiotracers显示文摘Difluorocarbene has emerged as a valuable intermediate to synthesize fluorides.However,difluorocarbene-derived synthesis of^(19)F/^(18)F-trifluoromethyl triazoles has not been explored.Herein,we reported the Cu(I)-promoted difluorocarbene-derived^(19)F/^(18)F-trifluoromethylation of iodotriazoles using KF/K^(18)F as the fluorine source.This approach rapidly generated a wide range of 5-trifluoromethyl-1,2,3-triazoles in good yields showing high functional group compatibility.The reaction was effective for late-stage functionalization of bioactive molecules and^(18)F-trifluoromethylation of iodotriazoles.This work provides a practical synthetic methodology for the development of triazole drugs and^(18)F-radiotracers for positron emission tomography.Fang Yuan Hongbao Sun Cheng Yang Haojie Yang Lili Pan Xiaoyang Zhang Rong Tian Lingjun Li Wei Chen Xiaoai Wu Haoxing Wu 2023Chinese Chemical Letters2023,34,6:0
6Production of transgenic mice by random recombination of targeted genes in female germline stem cells显示文摘Oocyte production in most mammalian species is believed to cease before birth.However,this idea has been challenged with the finding that postnatal mouse ovaries possess mitotically active germ cells.A recent study showed that female germline stem cells(FGSCs)from adult mice were isolated,cultured long term and produced oocytes and progeny after transplantation into infertile mice.Here,we demonstrate the successful generation of transgenic or gene knock-down mice using FGSCs.The FGSCs from ovaries of 5-day-old and adult mice were isolated and either infected with recombinant viruses carrying green fluorescent protein,Oocyte-G1 or the mouse dynein axonemal intermediate chain 2 gene,or transfected with the Oocyte-G1 specific shRNA expression vector(pRS shOocyte-G1 vector),and then transplanted into infertile mice.Transplanted cells in the ovaries underwent oogenesis and produced heterozygous offspring after mating with wild-type male mice.The offspring were genetically characterized and the biological functions of the transferred or knock-down genes were investigated.Efficiency of genetransfer or gene knock-down was 29%–37%and it took 2 months to produce transgenic offspring.Gene manipulation of FGSCs is a rapid and efficient method of animal transgenesis and may serve as a powerful tool for biomedical science and biotechnology.Yong Zhang Zhaojuan Yang Yunze Yang Shuzeng Wang Lingjun Shi Wenhai Xie Kejing Sun Kang Zou Lei Wang Ji Xiong Jie Xiang Ji Wu 2011Journal of Molecular Cell Biology2011,3,2:0
7Coherent optical vortex generation with multiple topological charges based on a seeded free electron laser显示文摘To generate optical vortex with multiple topological charges,a simple scheme based on the phase mask shaping technique is proposed and applied in a seeded free electron laser.With a tailored phase mask,an extreme-ultraviolet(EUV)vortex with multiple topological charges can be produced.To prove the feasibility of this method,an eight-step phase mask is designed to shape the seed laser.The simulation results demonstrate that 100-MW,fully coherent EUV vortex pulses with topological charge 2 can be generated based on the proposed technique.We have also demonstrated the possibility of generating higher topological charges by using a phase mask with more steps.Hao Sun Xiaofan Wang Chao Feng Lingjun Tu Weijie Fan Bo Liu 2021High Power Laser Science and Engineering2021,9,4:0
