3Identification of HSC/MPP expansion units in fetal liver by single-cell spatiotemporal transcriptomics显示文摘Limited knowledge of cellular and molecular mechanisms underlying hematopoietic stem cell and multipotent progenitor(HSC/MPP)expansion within their native niche has impeded the application of stem cell-based therapies for hematological malignancies.Here,we constructed a spatiotemporal transcriptome map of mouse fetal liver(FL)as a platform for hypothesis generation and subsequent experimental validation of novel regulatory mechanisms.Single-cell transcriptomics revealed three transcriptionally heterogeneous HSC/MPP subsets,among which a CD93-enriched subset exhibited enhanced stem cell properties.Moreover,by employing integrative analysis of single-cell and spatial transcriptomics,we identified novel HSC/MPP‘pocket-like’units(HSC PLUS),composed of niche cells(hepatoblasts,stromal cells,endothelial cells,and macrophages)and enriched with growth factors.Unexpectedly,macrophages showed an 11-fold enrichment in the HSC PLUS.Functionally,macrophage-HSC/MPP co-culture assay and candidate molecule testing,respectively,validated the supportive role of macrophages and growth factors(MDK,PTN,and IGFBP5)in HSC/MPP expansion.Finally,cross-species analysis and functional validation showed conserved cell-cell interactions and expansion mechanisms but divergent transcriptome signatures between mouse and human FL HSCs/MPPs.Taken together,these results provide an essential resource for understanding HSC/MPP development in FL,and novel insight into functional HSC/MPP expansion ex vivo.Suwei Gao Qiang Shi Yifan Zhang Guixian Liang Zhixin Kang Baofeng Huang Dongyuan Ma Lu Wang Jianwei Jiao Xiangdong Fang Cheng-Ran Xu Longqi Liu Xun Xu Berthold Gottgens Cheng Li Feng Liu 2022Cell Research2022,32,1:4
4Particulate matter pollution in Kunshan High-Tech zone: Source apportionment with trace elements, plume evolution and its monitoring显示文摘Particulate matter(PM) in the Kunshan High-Tech zone is studied during a three-month campaign. PM and trace elements are measured by the online pollution monitoring, forecastwarning and source term retrieval system AS3. Hourly measured concentrations of PM_(10), PM_(2.5) and 16 trace elements in the PM_(2.5) section(Ca, Pb, Cu, Cl, V, Cr, Fe, Ti, Mn, Ni, Zn, Ga, As, Se, Sr, Ba)are focused. Source apportionment of trace elements by Positive Matrix Factorization modeling indicates that there are five major sources, including dust, industrial processing, traffic,combustion, and sea salt with contribution rate of 23.68%, 21.66%, 14.30%, 22.03%, and 6.89%,respectively. Prediction of plume dispersion from concrete plant and traffic emissions shows that PM_(10) pollution of concrete plant is three orders of magnitude more than that of the traffic. The influence range can extend to more than 3 km in 1 hr. Because the footprint of the industrial plumes is constantly moving according to the local meteorological conditions, the fixed monitoring sites scattered in a few hundred meters haven't captured the heaviest pollution plume at the local scale of a few km^2. As a more intensive monitoring network is not operationally possible, the use of online modeling gives accurate and quantitative information of plume location, which increases the spatial pollution monitoring capacity and improves the understanding of measurement data. 5X-ray and Magnetoresistance Measurements of Electrodeposited Cu-Co Granular Films显示文摘Granular CoxCu1-x alloy films were prepared by electrodeposition at room temperature directly onto semiconducting Si substrate. X-ray diffraction (XRD) revealed that the as-deposited films formed single phase metastable fcc alloy structure. The fcc lattice parameter α was found to decrease linearly with increasing Co concentration x in the studied range. The giant magnetoresistance (GMR) of the films was improved after annealing. Pure Co fcc diffraction peaks were observed in the diffractogram of the annealed sample, indicating phase separation occurred upon annealing.The optimal annealing temperature was 450℃. The maximum of magnetoresistance (MR) ratio 8.21% was obtained for the Co20Cu80 thin film after annealing at 450℃ for 1 h. 7Gradual Evolution from Fluvial Dominated to Tide Dominated Deltas and Channel Type Transformation: A Case Study of MPE3 Block in the Orinoco Heavy Oil Belt of the Eastern Venezuelan Basin显示文摘Based on the data of core description and sporopollen analysis,the gradual evolution of deltas in vertical direction and transition of channel types in the MPE3 block of the eastern Venezuela Basin have been surveyed by seismic phase and well logging facies interpretation.The results show that due to the great sea level rise,the sedimentary system of the Miocene Oficina Formation in the MPE3 block shifted from the distal-source sandy braided river delta to tide-affected delta,and eventually to tide-dominated delta.Vertically,during the early stage of sedimentation of Oficina Formation,the distributary channels of the delta were dominated by braided river channels.While in the later stage,as the tidal effect was gradually intensified,the channel changed from braided channel to meandering channel.On plane,as a result of differential transgression,sedimentary framework and distribution of sand bodies vary across the study area.Compared with the eastern part,the western part has more braided channels,larger channel bars,less developments of distributary bay and higher ratio of sand to mud.Whereas the braided channels in the south are larger than those in the north.It is the first time we pointed out the impact of marine transgression differences on the sedimentary facies distribution and river type transition in the study area.Factors like the structural and paleogeomorphological change,sea level variation,supply of sediments have strong influence on the evolution of sedimentary system and distribution of sandbodies.It is predicted that the major sandbody is more developed in the central south,which can guide the subsequent horizontal well development.HUANG Wensong CHEN Heping XU Fang MENG Zheng ZHANG Fanqin WU Suwei 2019Acta Geologica Sinica(English Edition)2019,93,6:1

15Oxalic acid functionalization of BaTiO_(3) nanobelts for promoting their piezo-degradation organic contaminants显示文摘Surface functionalization of piezoelectric greatly determines its piezo-catalytic activity and selectivity in that functional group affect specific reactants’surface adsorption and activation abilities.Herein,we propose chemical functionalization on the surface of nano-scaled BaTiO_(3) piezoelectric via a one-pot hydrothermal process using polyethylene glycol as surfactant,and evaluate its piezo-catalytic activity and selectivity by degrading model antibiotic and dye reactants under ultrasonic vibrations.Acetate or/and oxalic unidentate ligands on the surface of BaTiO_(3) nanobelts can be formed by controlling precursor and hydrothermal parameters.Particularly,oxalic acid functionalized BaTiO_(3) nanobelts presented a high piezo-catalytic rate constant of 0.068 min^(-1) for Rhodamine B solution and maintained>87%degradation efficiency within 30 min under the condition of ultrasonic bath with 40 kHz and 100 W,which was mainly ascribed to piezo-sensitization effect.This work provides references for the preparation of chemical functionalized catalysts and also contributes to the development of novel catalysts for special applications.Shitao Gao Hongjie Xing Jingji Zhang Yapi Liu Huiwei Du Zejie Zhu Jiangying Wang Xu Li Suwei Zhang Yaxuan Yao Lingling Ren 2021Journal of Materiomics2021,7,6:1

18Exploiting Complex-Type N-Glycan to Improve the in Vivo Stability of Bioactive Peptides显示文摘Peptides can be potentmolecules with high efficacy and selectivity in the development of biotherapeutics.However,the poor pharmacokinetic properties of peptides pose major challenges for their broader medicinal applications.Inspired by the proteinstabilizing role of natural N-glycosylation,we design and synthesize a series of parathyroid hormone(PTH)peptides(1-34),bearing either N-GlcNAc or biantennary complex-type N-glycan modification,and evaluate their serum stability and biological activities.The results indicate that an N-Asn-linked complex-type sialylundecasaccharide can increase the serum half-life and in vivo bioactivity of PTH peptides with a broad tolerance of modification sites.Further,hydrogen/deuterium exchange mass spectroscopy indicates that the larger-sized Nglycan can induce enhanced hydration dynamics in its surroundings,which may facilitate an improved resistance for the peptide against enzymatic proteolysis.This sialylundecasaccharide-based peptideengineering strategy has also been applied to glucagon-like peptide-1(7-37),leading to glycopeptides with enhanced hypoglycemic activity and acting time in vivo.Together,these results demonstrate the potential of using sialylated complextype N-glycan as a general engineering strategy for developing long-acting peptide therapeutics.Qijia Wei Jun Zhang Yuankun Dao Mengliang Ye Dangliang Liu Weidong Dong Ning Yuan Hongxing Li Chunli Song Mo Li Xiaomeng Shi Suwei Dong 2023CCS Chemistry2023,5,7:0
19Visible light cross-linking and bioactive peptides loaded integrated hydrogel with sequential release to accelerate wound healing complicated by bacterial infection显示文摘The effective management of bacterial infections that are resistant to multiple drugs remains a substantial clinical challenge.The eradication of drug-resistant bacteria and subsequent promotion of angiogenesis are imperative for the regeneration of the infected wounds.Here,a novel and facile peptide containing injectable hydrogel with sustained antibacterial and angiogenic capabilities is developed.The antibacterial peptide that consists of 11 residues(CM11,WKLFKKILKVL)is loaded onto acrylate-modified gelatin through charge interactions.A vascular endothelial growth factor mimetic peptide KLT(KLTWQELYQLKYKGI)with a GCG(Gly-Cys-Gly)modification at the N-terminal is covalently coupled through a visible light-induced thiol-ene reaction.In this reaction,the acrylate gelatin undergoes cross-linkage within seconds.Based on the physical/chemical double crosslinking strategy,the bioactive peptides achieve sustained and sequential release.The results show that the hydrogel significantly inhibits methicillin-resistant Staphylococcus aureus(MRSA)growth through the rapid release of CM11 peptides at early stage;it forms obvious growth inhibition zones against pathogenic bacterial strains.Moreover,cell counting kit-8 assay and scratch test confirm that the CM11/KLT-functionalized hydrogels promote cell proliferation and migration through the later release of KLT peptides.In a mouse skin wound infected with self-luminous MRSA,the CM11/KLT-functionalized hydrogels enhance wound healing,with rapidly bacterial infection reduction,lower expression of inflammatory factors,and neovascularization promotion.These results suggest that the rationally designed,sustained and sequential release CM11/KLT-functionalized hydrogels have huge potential in promoting the healing of multi-drug resistant bacterial infected wounds.Guiyan Wang Ning Yuan Jun Zhang Man Qin Suwei Dong Yuguang Wang 2024Nano Research2024,17,3:0
20Phase transition and piezoelectric property of (Ag,K)NbO_(3) ceramics显示文摘AgNbO_(3) is an antiferroelectric (AFE) material with double hysteresis loop. Both the antiferroelectricityand ferroelectricity can be enhanced by doping. Herein, the ferroelectricity of AgNbO_(3) ceramics wasenhanced via K-doping and the phase diagram of the (Ag_(1-x)K_(x))NbO_(3) ceramics was upgraded. In details,(Ag_(1-x)K_(x))NbO_(3) ceramics are ferrielectric (FIE) M1 phase as x=5.00-5.50 mol% and ferroelectric (FE) Ophase as x=5.75-6.00 mol% before poling, and FE O phase as x=5.00-6.00 mol% after poling at roomtemperature.
