1Punicalagin ameliorates collagen-induced arthritis by downregulating M1 macrophage and pyroptosis via NF-κB signaling pathway显示文摘Rheumatoid arthritis(RA)is a chronic inflammatory disease that eventually leads to disability.Inflammatory cell infiltration,severe joint breaking and systemic bone loss are the main clinical symptoms.In this study,we established a collagen-induced arthritis(CIA)model and found a large number of M1 macrophages and pyroptosis,which are important sources of proinflammatory cytokines.Punicalagin(PUN)is an active substance extracted from pomegranate peel.We found that it inhibited joint inflammation,cartilage damage and systemic bone destruction in CIA mice.PUN effectively alleviated the high expression of inflammatory cytokines in synovial tissue in vivo.PUN treatment shifted macrophages from the M1 phenotype to the M2 phenotype after stimulation with lipopolysaccharide(LPS)and interferon(IFN)-γ.The expression of inducible nitric oxide synthase(i NOS)and other proinflammatory cytokines released by M1 macrophages was decreased in the PUN treatment group.However,simultaneously,the expression of markers of anti-inflammatory M2 macrophages,such as arginase(Arg)-1 and interleukin(IL)-10,was increased.In addition,PUN treatment attenuated pyroptosis by downregulating the expression of NLRP3 and caspase-1,thereby preventing inflammatory cell death resulting from the release of IL-1βand IL-18.Mechanistically,PUN inhibited the activation of receptor activators of the nuclear factor-κB(NF-κB)signaling pathway,which contributes to M1 polarization and pyroptosis of macrophages.We concluded that PUN ameliorated pathological inflammation by inhibiting M1 phenotype polarization and pyroptosis and has great potential as a therapeutic treatment for human RA.Gaoran Ge Jiaxiang Bai Qing Wang Xiaolong Liang Huaqiang Tao Hao Chen Minggang Wei Junjie Niu Huilin Yang Yaozeng Xu Yuefeng Hao Yi Xue Dechun Geng 2022Science China(Life Sciences)2022,65,3:14
2Impact of diabetes on prognosis of gastric cancer patients performed with gastrectomy显示文摘Objective: This study aimed to determine the impact of type 2 diabetes mellitus(T2 DM) on clinical outcomes of gastric cancer(GC) patients and explore whether metformin use and good glycemic control could reverse it.Methods: Clinicopathologic data of consecutive GC patients who underwent gastrectomy at Nanfang Hospital between October 2004 and December 2015 were included. Propensity score matching(PSM) was performed to balance the important factors of the disease status between non-T2 DM and T2 DM group. The last follow-up time was January 2019.Results: A total of 1,692 eligible patients(1,621 non-T2 DM vs. 71 T2 DM) were included. After PSM, nonT2 DM group(n=139) and T2 DM group(n=71) were more balanced in baseline variables. The 5-year cancerspecific survival(CSS) rate in T2 DM group(47.0%) was inferior to that in non-T2 DM group(58.0%), but did not reach statistical significance [hazard ratio(HR)=1.319, 95% confidence interval(95% CI): 0.868-2.005, P=0.192].While the 5-year progress-free survival(PFS) rate of T2 DM group(40.6%) is significantly worse than that in nonT2 DM group(56.3%)(HR=1.516, 95% CI: 1.004-2.290, P=0.045). Univariate and multivariate analyses showed that T2 DM was an independent risk factor for PFS but not for CSS. In T2 DM group, metformin use subgroup was associated with superior 5-year CSS and PFS in compared with non-metformin use subgroup, although the difference was not statistically significant(5-year CSS: 48.0% vs. 45.4%, HR=0.680, 95% CI: 0.352-1.313,P=0.246;5-year PFS: 43.5% vs. 35.7%, HR=0.763, 95% CI: 0.400-1.454, P=0.406). The 5-year CSS rate was47.5% in good glycemic control subgroup and 44.1% in poor glycemic control subgroup(HR=0.826, 95% CI:0.398-1.713, P=0.605). And both two subgroups yielded a similar 5-year PFS rate(42.2% vs. 36.3%, HR=0.908,95% CI: 0.441-1.871, P=0.792).Conclusions: DM promoted disease progress of GC after gastrectomy but had not yet led to the significant discrepancy of CSS. 4Rational integration of defense and repair synergy on PEEK osteoimplants via biomimetic peptide clicking strategy显示文摘Polyetheretherketone(PEEK)has been widely used as orthopedic and dental materials due to excellent mechanical and physicochemical tolerance.However,its biological inertness,poor osteoinduction,and weak antibacterial activity make the clinical applications in a dilemma.Inspired by the mussel adhesion mechanism,here we reported a biomimetic surface strategy for rational integration and optimization of anti-infectivity and osteo-inductivity onto PEEK surfaces using a mussel foot proteins(Mfps)-mimic peptide with clickable azido terminal.The peptide enables mussel-like adhesion on PEEK biomaterial surfaces,leaving azido groups for the further steps of biofunctionalizations.In this study,antimicrobial peptide(AMP)and osteogenic growth peptide(OGP)were bioorthogonally clicked on the azido-modified PEEK biomaterials to obtain a dual-effect of host defense and tissue repair.Since bioorthogonal clicking allows precise collocation between AMP and OGP through changing their feeding molar ratios,an optimal PEEK surface was finally obtained in this research,which could long-term inhibit bacterial growth,stabilize bone homeostasis and facilitate interfacial bone regeneration.In a word,this upgraded mussel surface strategy proposed in this study is promising for the surface bioengineering of inert medical implants,in particular,achieving rational integration of multiple biofunctions to match clinical requirements.Meng Li Jiaxiang Bai Huaqiang Tao Li Hao Weiling Yin Xiaoxue Ren Ang Gao Ning Li Miao Wang Shiyuan Fang Yaozeng Xu Liang Chen Huilin Yang Huaiyu Wang Guoqing Pan Dechun Geng 2022Bioactive Materials2022,7,2:2
5Silver nanoparticle labeled immunoresonance scattering spectral assay for trace fibrinogen显示文摘Silver nanoparticles in size of 8.0 nm was prepared by the trisodium citrate and used to label goat anti-human fibrinogen. In the pH 5.8 Na2HPO4-NaH2O4 buffer solution (PBS) and in the presence of polyethylene glycol (PEG) and KCl, the immune reaction between silver-labeled goat anti-human fibrinogen and fibrinogen took place and led the resonance scattering intensity at 465 nm (I465) to decreasing. The I465 decreased intensity was linear to the fibrinogen concentration in the range from 0.067 to 1.67 μg/mL, with a detection limit of 0.024 μg/mL. 9Drying characteristics of biological porous media during convective drying显示文摘Experiments with potatoes were carried out in order to analyze the variation of the temperature and the mean dry basis moisture content over time,the effect of the drying conditions on the drying rate,and the relationship between deformation and dry basis moisture content.A two-way sequentially coupled thermo-hydro-mechanical math model was developed on the basis of Fickian diffusion theory,Fourier’s law of heat conduction and thermoelasticity mechanics in order to analyze the spatio-temporal distributions of moisture,temperature and drying stresses in the potatoes.The transient mathematical model,composed of a system of partial differential equations,was solved by finite difference methods.The numerical results obtained by using the mathematical model were in good agreement with the experimental data.The variations in temperature and moisture distributions,drying curves and stresses within potatoes over time were simulated,and the ways in which these are affected by the drying conditions were discussed.This work could help in developing an understanding of the relationship between mass and heat transfer,shrinkage,stress,and physical degradation of biological materials.Wang Huilin Lu Tao Zhang Quanguo 2016International Journal of Agricultural and Biological Engineering2016,9,5:1
10Biomimetic melatonin-loaded silk fibroin/GelMA scaffold strengthens cartilage repair through retrieval of mitochondrial functions显示文摘Articular cartilage injury induced by collision or trauma is a common sports-related condition that may progress to pain,dysfunction,and secondary osteoarthritis(OA).Limited by self-renewal potential,cartilage regeneration faces a quandary while bio-inspired novel strategies are urgently required.In this study,by a soft freezing method and surface modification technique,a multi-functional silk fibroin(SF)plus gelatin methacrylate(GelMA)scaffold laden with melatonin(MT)was prepared.SF-GelMA@MT scaffold demonstrated enhanced biomechanical characteristics and long-acting melatonin release.In vitro treatment with SF-GelMA@MT induced the synthesis of cartilage extracellular matrix(ECM)components.Mechanistically,sustained release of melatonin yielded robust chondroprotective effects via improving mitochondrial polarization and antioxidant properties.SF-GelMA@MT implantation boosted cartilage renascence in a full-thickness cartilage defect model via mitochondria-associated sirtuins 1(SIRT1)-superoxide dismutase 2(SOD2)signaling pathway in vivo.In summary,this research proposed a welldesigned bionic composite scaffold that promotes cartilage regeneration via mitochondrial function enhancement,which is of tremendous potential for cartilage tissue engineering.Mingzhuang Hou Yijian Zhang Yang Liu Xiaoyang Ge Xiayu Hu Zhijian Zhao Xin Tian Tao Liu Huilin Yang Xi Chen Fan He Xuesong Zhu 2023Journal of Materials Science & Technology2023,,15:1
11Diabetes mellitus promoted lymph node metastasis in gastric cancer: a 15-year single-institution experience显示文摘Background: Previous studies have revealed that diabetes mellitus (DM) promotes disease progress of gastric cancer (GC). This study aimed to further investigating whether DM advanced lymph nodes (LNs) metastasis in GC.Methods: The clinicopathologic data of GC patients with >15 examined LN (ELN) between October 2004 and December 2019 from a prospectively maintained database were included. The observational outcomes included the number (N3b status) and anatomical distribution (N3 stations) of metastatic LN (MLN).Results: A total of 2142 eligible patients were included in the study between October 2004 and December 2019. N3 stations metastasis (26.8% in DMvs. 19.3% in non-DM,P = 0.026) and N3b status (18.8% in DMvs. 12.8% in non-DM,P = 0.039) were more advanced in the DM group, and multivariate logistic regression analyses confirmed that DM was an independent factor of developing N3 stations metastasis (odds ratio [OR] = 1.771,P= 0.011) and N3b status (OR= 1.752,P= 0.028). Also, multivariate analyses determined DM was independently associated with more MLN (β = 1.424,P = 0.047). The preponderance of N3 stations metastasis (DMvs. non-DM, T1-2: 2.2%vs. 4.9%, T3: 29.0%vs. 20.3%, T4a: 38.9%vs. 25.8%, T4b: 50.0%vs. 36.6%;ELN16-29: 8.6%vs. 10.4%, ELN30-44: 27.9%vs. 20.5%, ELN ≥ 45: 37.7%vs. 25.3%), N3b status (DMvs. non-DM, T1-2: 0%vs. 1.7%, T3: 16.1%vs. 5.1%, T4a: 27.8%vs. 19.1%, T4b: 44.0%vs. 28.0%;ELN16-29: 8.6%vs. 7.9%, ELN30-44: 18.0%vs. 11.8%, ELN ≥ 45: 26.4%vs. 17.3%), and the number of MLN (DMvs. non-DM, T1-2: 0.4vs. 1.1, T3: 8.6vs. 5.2, T4a: 9.7vs. 8.6, T4b: 17.0vs. 12.8;ELN16-29: 3.6vs. 4.6, ELN30-44: 5.8vs. 5.5, ELN ≥ 45: 12.0vs. 7.7) of DM group increased with the advancement of primary tumor depth stage and raising of ELN.Conclusions: DM was an independent risk factor for promoting LN metastasis. 12Impact of preoperative therapy on surgical outcomes of laparoscopic total gastrectomy for gastric/gastroesophageal junction cancer显示文摘Objective: As laparoscopic surgery is widely applied for primarily treated gastric cancer(GC)/gastroesophageal junction cancer(GEJC) and gains many advantages, the feasibility of laparoscopic total gastrectomy(LTG) for GC/GEJC patients who have received preoperative therapy(PT) has come to the fore. This study aims to analyze the safety and feasibility of LTG after PT for GC/GEJC patients.Methods: We retrospectively analyzed the data of 511 patients with GC/GEJC undergoing LTG, of which 405received LTG(LTG group) and 106 received PT+LTG(PT-LTG group) at Nanfang Hospital between June 2018and September 2022. The surgical outcomes were compared between the two groups.Results: The surgical duration was significantly longer in the PT-LTG group(P<0.001), while the incidence of intraoperative complications(P=1.000), postoperative complications(LTG group vs. PT-LTG group: 26.2% vs.23.6%, P=0.587), the classification of complication severity(P=0.271), and postoperative recovery was similar between two groups. Notably, the incidence of anastomotic complications of esophagojejunostomy was also comparable between the two groups(LTG group vs. PT-LTG group: 5.9% vs. 5.7%, P=0.918). The univariate and multivariate analysis confirmed that positive proximal margin [positive vs. negative: odds ratio(OR)=14.094, 95%confidence interval(95% CI): 2.639-75.260, P=0.002], rather than PT, has an impact on anastomotic complications after LTG(OR=0.945, 95% CI: 0.371-2.408, P=0.905).Conclusions: PT did not increase the surgical risk of LTG for GC/GEJC. 13Study on direct point spread function measurement of capillary plate collimators with divergent X-ray beams显示文摘Purpose The Filed of View(FOV)of eXTP/LAD is limited by lead-glass capillary plate collimators placed in front of the Silicon Drift Detectors(SDDs)for reducing the background contamination caused by the photons of Cosmic X-ray Background(CXB)leaking from outside the FOV.The core quality parameters of lead-glass capillary plate collimators are Open Area Ratio(OAR)and FOV.The performance of lead-glass capillary plate collimators is determined by its design specification such as pore-to-pore misalignment,pore perpendicularity with collimator surface and pore diameter.Since the design specification is the result of a large number of micropores,we use the full width at half maximum(FWHM)and peak value of point spread function(PSF)to characterize the capillary plate collimator performance.In order to quickly evaluate the performances of collimators,we developed a direct PSF measurement method using divergent beams.Methods In this paper,the simulation package of the divergent beamsmeasurement setup is developed based on Geant4.The simulation parameters of scintillator absorption length are corrected by experimental data.We simulate the PSF of collimators with different distribution of pore diameter and misalignment by parallel beams and divergent beams.By comparing the peak value and FWHM of collimators PSF,the feasibility in replacing parallel beams measurement with divergent beams measurement is studied.Results It is verified that the influence of the geometric size of 33μm radius of the micro-focus of X-ray tube can be ignored.The results show that the FWHM of collimators is increased with error root mean square error(RMS)of the distribution of collimators pore diameter and pore inclination angle.The peak value of PSF is mainly influenced by pore non-parallelism of the pores,and it decreases with an increase in pore inclination angle.Comparison of parallel beams PSF and divergent beams PSF shows well consistency of peak value and FWHM of collimator.But,the peak value of the divergent beams PSF is mainly affected by the defects of the pores close to the optical axis and result in difference from the parallel beams results consequently.We have developed capillary plate collimators response function based on the projected area function of a cylindrical pore and the fluorescence broadening of the scintillator and fitted PSF by response function.The capillary plate collimator open area ratio is calculated by using the micropores radius and collimator frame thickness obtained by fitting parameter,and the result is close to the parallel beams PSF.Conclusion The feasibility divergent beams measurement is verified.Tianming Li Tao Luo Yupeng Xu Huilin He Liming Song 2023Radiation Detection Technology and Methods2023,7,3:0
14Marine Target Detection Using Dual-polarimetric SAR Imagery显示文摘In this study,we provide a summary of research advances in the field of maritime target detection using DP(dualpolarimetric)SAR(Synthetic Aperture Radar)imagery,accomplished during the European and China collaboration in the framework of the Dragon-4 program ID 32235.The main innovative contribution is twofold:(1)We addressed ship detection proposing an improved GP-PNF(Geometrical Perturbation-Polarimetric Notch Filter),termed as IGP-PNF,that is characterized by a new feature vector that includes three new scattering features;(2)We addressed oil platform detection by contrasting singlepolarization SAR methods with polarimetric ones in order to quantify the extra-benefit carried on polarimetric information.The proposed theoretical framework is tested against actual multi-polarization SAR data.In particular,ship detection methods are verified against a Sentinel-1 SAR scene where a large number of ships is present;while,oil platform detection is discussed using Terra SAR-X SAR data.Experimental analysis shows that:(1)The IGP-PNF method performs best in terms of clutter-to-target ratio;(2)Coherent polarimetric information significantly outperforms single-polarization SAR measurements in highlighting targets in challenging cases.Tao ZHANG Armando MARINO Ferdinando NUNZIATA Domenico VELOTTO Weizeng SHAO Xiaofeng LI Maurizio MIGLIACCIO Huilin XIONG 2021Journal of Geodesy and Geoinformation Science2021,4,1:0
15Apoptotic Signal Pathways and Regulatory Mechanisms of Cancer Cells Induced by IL-24显示文摘The melanoma differentiation-associated gene-7(mda-7),IL-24,has the specific functions that induce cancer cell apoptosis without doing harm to normal cells. We systematically review the apoptotic signal pathways and their regulatory mechanisms induced by Ad.IL-24 and IL-24 in diverse cancer cells. IL-24 can participate in varied signal transduction pathways,including JAK,p38 MAPK,Wnt/β-catenin,JNK,ER stress and mitochondria-associated signal pathways. And we review five proteins interacting with IL-24,including Bip/GRP78,S1 R,PKR,Beclin1 and soluble clusterin,which are relative to the tumor-specific effect of IL-24. It is speculated that ER stress,G-protein pathways and MAPK signal pathways may be the primary upstream effectors which activate the sequential downstream mediators resulting in apoptosis induced by IL-24 in tumor cells. 17Fe doped g-C_(3)N_(4) composited ZnIn_(2)S_(4) promoting Cr(Ⅵ) photoreduction显示文摘30% Fe CN/ZIS(30% Fe doped g-C_(3)N_(4)composited ZnIn_(2)S_(4)) was synthesized by a simple water bath method, via in-situ growth of abundant well-dispersed ZnIn_(2)S_(4)nanosheets on the Fe doped g-C_(3)N_(4)surface. Experimental results showed the optimized 30% Fe CN/ZIS achieved the best photoreduction of Cr(VI)performance within a wide p H range, which was 9.5 times and 700 times higher than that of pure ZnIn_(2)S_(4)and 30% Fe CN(Fe doped g-C_(3)N_(4)). This is due to the intense synergy between the Fe-Nxbond and close interface contact produces a high-speed charge transfer channel, thus significantly improving the efficiency of optical carrier separation and migration. Meanwhile, UV-vis diffuse reflection spectra and photoluminescence spectroscopy showed that iron doping significantly narrowed the bandgap of gC_(3)N_(4), preventing electron-hole pair recombination. Further, the microstructures and charge separation properties were analyzed by scanning electron microscope, Photoluminescence Spectroscopy and timeresolved photoluminescence, which revealed the structure-activity relationship of composite structure and the synergistic mechanism of each functional component.
