1Ligustrazine monomer against cerebral ischemia/reperfusion injury显示文摘Ligustrazine(2,3,5,6-tetramethylpyrazine) is a major active ingredient of the Szechwan lovage rhizome and is extensively used in treatment of ischemic cerebrovascular disease.The mechanism of action of ligustrazine use against ischemic cerebrovascular diseases remains unclear at present.This study summarizes its protective effect,the optimum time window of administration,and the most effective mode of administration for clinical treatment of cerebral ischemia/reperfusion injury.We examine the effects of ligustrazine on suppressing excitatory amino acid release,promoting migration,differentiation and proliferation of endogenous neural stem cells.We also looked at its effects on angiogenesis and how it inhibits thrombosis,the inflammatory response,and apoptosis after cerebral ischemia.We consider that ligustrazine gives noticeable protection from cerebral ischemia/reperfusion injury.The time window of ligustrazine administration is limited.The protective effect and time window of a series of derivative monomers of ligustrazine such as 2-[(1,1-dimethylethyl)oxidoimino]methyl]-3,5,6-trimethylpyrazine,CXC137 and CXC195 after cerebral ischemia were better than ligustrazine.Hai-jun Gao Peng-fei Liu Pei-wen Li Zhuo-yan Huang Feng-bo Yu Ting Lei Yong Chen Ye Cheng Qing-chun Mu Hai-yan Huang 2015Neural Regeneration Research2015,10,5:48
2Bone marrow mesenchymal stem cell therapy in ischemic stroke: mechanisms of action and treatment optimization strategies显示文摘Animal and clinical studies have confirmed the therapeutic effect of bone marrow mesenchymal stem cells on cerebral ischemia, but their mechanisms of action remain poorly understood. Here, we summarize the transplantation approaches, directional migration, differentiation, replacement, neural circuit reconstruction,angiogenesis, neurotrophic factor secretion, apoptosis, immunomodulation, multiple mechanisms of action,and optimization strategies for bone marrow mesenchymal stem cells in the treatment of ischemic stroke.We also explore the safety of bone marrow mesenchymal stem cell transplantation and conclude that bone marrow mesenchymal stem cell transplantation is an important direction for future treatment of cerebral ischemia. 4An HBV-encoded miRNA activates innate immunity to restrict HBV replication显示文摘We previously identified that hepatitis B virus(HBV)encodes a microRNA(HBV-miR-3)that restrains HBV replication by targeting the HBV transcript.However,whether HBV-miR-3 affects host innate immunity to modulate HBV replication remains unclear.Here,we examined the vital functions of HBV-miR-3 in the innate immune response after HBV infection.We found that HBV-miR-3 expression gradually increased in a dose-and time-dependent manner in HBV-infected HepG2-NTCP cells.HBV-miR-3 activated the JAK/STAT signaling pathway by downregulating SOCS5 in hepatocytes,thereby enhancing the IFN-induced anti-HBV effect.In addition,HBVmiR-3 in exosomes facilitated the Ml polarization of macrophages.Furthermore,exosomes containing HBV-miR-3 enhanced the secretion of IL-6 via inhibiting the SOCS5-mediated ubiquitination of EGFR.In short,these results demonstrate that HBV-miR-3 activates the innate immune response to restrain HBV replication by multiple pathways,which may suppress HBV-induced acute liver cell injury and affect the progression of persistent HBV infection.Xiaoqing Zhao Lu Sun Ting Mu Jianying Yi Chaoqun Ma Hong Xie Min Liu Hua Tang 2020Journal of Molecular Cell Biology2020,12,4:17
5SARS-Coronavirus-2 Nsp13 Possesses NTPase and RNA Helicase Activities That Can Be Inhibited by Bismuth Salts显示文摘The ongoing outbreak of Coronavirus Disease 2019(COVID-19)has become a global public health emergency.SARScoronavirus-2(SARS-CoV-2),the causative pathogen of COVID-19,is a positive-sense single-stranded RNA virus belonging to the family Coronaviridae.For RNA viruses,virus-encoded RNA helicases have long been recognized to play pivotal roles during viral life cycles by facilitating the correct folding and replication of viral RNAs.Here,our studies show that SARS-CoV-2-encoded nonstructural protein 13(nsp13)possesses the nucleoside triphosphate hydrolase(NTPase)and RNA helicase activities that can hydrolyze all types of NTPs and unwind RNA helices dependently of the presence of NTP,and further characterize the biochemical characteristics of these two enzymatic activities associated with SARS-CoV-2 nsp13.Moreover,we found that some bismuth salts could effectively inhibit both the NTPase and RNA helicase activities of SARS-CoV-2 nsp13 in a dose-dependent manner.Thus,our findings demonstrate the NTPase and helicase activities of SARS-CoV-2 nsp13,which may play an important role in SARS-CoV-2 replication and serve as a target for antivirals.Ting Shu Muhan Huang Di Wu Yujie Ren Xueyi Zhang Yang Han Jingfang Mu Ruibing Wang Yang Qiu Ding-Yu Zhang Xi Zhou 2020Virologica Sinica2020,35,3:11

7Plasma metabolomic and lipidomic alterations associated with COVID-19显示文摘The pandemic of the coronavirus disease 2019(COVID-19)has become a global public health crisis.The symptoms of COVID-19 range from mild to severe,but the physiological changes associated with COVID-19 are barely understood.In this study,we performed targeted metabolomic and lipidomic analyses of plasma from a cohort of patients with COVID-19 who had experienced different symptoms.We found that metabolite and lipid alterations exhibit apparent correlation with the course of disease in these patients,indicating that the development of COVID-19 affected their whole-body metabolism.In particular,malic acid of the TCA cycle and carbamoyl phosphate of the urea cycle result in altered energy metabolism and hepatic dysfunction,respectively.It should be noted that carbamoyl phosphate is profoundly down-regulated in patients who died compared with patients with mild symptoms.And,more importantly,guanosine monophosphate(GMP),which is mediated not only by GMP synthase but also by CD39 and CD73,is significantly changed between healthy subjects and patients with COVID-19,as well as between the mild and fatal cases.In addition,dyslipidemia was observed in patients with COVID-19.Overall,the disturbed metabolic patterns have been found to align with the progress and severity of COVID-19.This work provides valuable knowledge about plasma biomarkers associated with COVID-19 and potential therapeutic targets,as well as an important resource for further studies of the pathogenesis of COVID-19.Di Wu Ting Shu Xiaobo Yang Jian-Xin Song Mingliang Zhang Chengye Yao Wen Liu Muhan Huang Yuan Yu Qingyu Yang Tingju Zhu Jiqian Xu Jingfang Mu Yaxin Wang Hong Wang Tang Tang Yujie Ren Yongran Wu Shu-Hai Lin Yang Qiu Ding-Yu Zhang You Shang Xi Zhou 2020National Science Review2020,7,7:9
8An increase in corn resistant starch decreases protein fermentation and modulates gut microbiota during in vitro cultivation of pig large intestinal inocula显示文摘High-protein diet could cause an increase in protein fermentation in the large intestine, leading to an increased production of potentially detrimental metabolites. We hypothesized that an increase in corn resistant starch content may attenuate the protein fermentation. The aim of this study was to evaluate the effect of resistant starch on protein fermentation by inocula from large intestine of pigs using in vitro cultivation. Fermentation patterns were analyzed during a 24-h incubation of cecal and colonic digesta with varying corn resistant starch contents, using casein protein as sole nitrogen source. The results showed that the concentration of short-chain fatty acids(SCFA) and cumulative gas production were significantly increased(P < 0.05), while ammonia-nitrogen(NH_3-N) and branched-chain fatty acids(BCFA), which indicated protein fermentation, decreased when the corn resistant starch levels increased(P < 0.05). The copies of total bacteria, Bifidobacterium and Lactobacillus were significantly increased with the increased corn resistant starch levels after incubation(P < 0.05). The copies of the Bifidobacterium and Lactobacillus in cecum were significantly higher than those in colon(P < 0.05). 9Progress in ENSO prediction and predictability study显示文摘ENSO is the strongest interannual signal in the global climate system with worldwide climatic, ecological and societal impacts. Over the past decades, the research about ENSO prediction and predictability has attracted broad attention. With the development of coupled models, the improvement in initialization schemes and the progress in theoretical studies, ENSO has become the most predictable climate mode at the time scales from months to seasons. This paper reviews in detail the progress in ENSO predictions and predictability studies achieved in recent years. An emphasis is placed on two fundamental issues: the improvement in practical prediction skills and progress in the theoretical study of the intrinsic predictability limit. 10Meiofauna Distribution in Intertidal Sandy Beaches Along China Shoreline(18?–40?N)显示文摘In this study,the distribution pattern of meiofauna from nine sandy beaches at six latitudinal gradients along Chinese coast between 18 and 40?N was studied on their meiofauna abundance to examine the effect of latitudinal gradients.In general,meiofauna abundance was lower in four subtropical beaches in Xiamen(24?N) and Zhoushan(29?N) than that in other beaches.Meiofauna abundance differed little between tropical and temperate beaches.The taxonomic structure of meiofauna showed a dominance of nematode in colder area.The relative composition of turbellarians and polychaetes increased in warmer area.In addition to latitudinal gradient,salinity,oxygenation,sediment grain size affect also the meiofauna latitudinal distribution.As for horizontal distribution,the highest meiofauna abundance was found in low tidal zone at tropical beaches,and in middle tidal zone at temperate beaches.The horizontal distribution of meiofauna was controlled by both physical and biotic factors including feeding and anthropogenic activities.Although meiofauna abundance exhibited a horizontal difference,the composition of meiofaunal main taxa was unanimous horizontally at all beaches at the same sampling latitude.HUA Er ZHANG Zhinan ZHOU Hong MU Fanghong LI Jia ZHANG Ting CONG Bingqing LIU Xiaoshou 2016Journal of Ocean University of China2016,15,1:7

13MIMO SAR Waveform Separation Based on Costas-LFM Signal and Co-arrays for Maritime Surveillance显示文摘In case of bright targets over a dark background, the cross-correlation noise of Multiple input and multiple output(MIMO) Synthetic aperture radar(SAR) orthogonal waveforms in the same frequency coverage will increase which can degrade the image. An Interpulse Costas hopping and intra-pulse Slope coded linearly frequency modulated(IPC-SCLFM) waveform is designed and analyzed to decrease the autocorrelation and cross-correlation noise. Besides a novel acquisition mode of 'MIMO Coprime SAR'(MIMO-Cop SAR) is proposed with Coprime arrays(Co-arrays) in receive across the SAR line of flight based on digital beamforming. An improved processing by multiplying values at each pixel of two coarrays images is proposed to suppress the cross-correlation noise. MIMO-Cop SAR can effectively suppress the crosscorrelation noise, reduce the amount of data to be stored and get high signal-to-noise ratio in a simple way. 15Hepatitis C Virus NS2 Protein Suppresses RNA Interference in Cells显示文摘RNAi interference(RNAi)is an evolutionarily conserved post-transcriptional gene silencing mechanism and has been well recognized as an important antiviral immunity in eukaryotes.Numerous viruses have been shown to encode viral suppressors of RNAi(VSRs)to antagonize antiviral RNAi.Hepatitis C virus(HCV)is a medically important human pathogen that causes acute and chronic hepatitis.In this study,we screened all the nonstructural proteins of HCV and found that HCV NS2 could suppress RNAi induced either by small hairpin RNAs(shRNAs)or small interfering RNAs(siRNAs)in mammalian cells.Moreover,we demonstrated that NS2 could suppress RNAi via its direct interaction with doublestranded RNAs(dsRNAs)and siRNAs,and further identified that the cysteine 184 of NS2 is required for the RNAi suppression activity through a serial of point mutation analyses.Together,our findings uncovered that HCV NS2 can act as a VSR in vitro,thereby providing novel insights into the life cycle and virus-host interactions of HCV.Hui Zhou Qi Qian Ting Shu Jiuyue Xu Jing Kong Jingfang Mu Yang Qiu Xi Zhou 2020Virologica Sinica2020,35,4:3

18Nematode Community Structure and Diversity Pattern in Sandy Beaches of Qingdao,China显示文摘We investigated the diversity and structure of free-living marine nematode communities at three sandy beaches representing typical intertidal environments of a temperate zone in Qingdao,Shandong Province,China.Average nematode abundance ranged from 1006 to 2170 ind.10 cm-2,and a total of 34 nematode genera were recorded,of which only 8 were common in all the studied beaches.Pielou's evenness and Shannon-Wiener diversity index were the lowest at the second beach where nematode abundance was the highest.The highest species diversity index coincided with the lowest nematode abundance at Shilaoren beach.Sediment median grain size,sorting coefficient,and chlorophyll-a content were essential for differentiation in nematode abundance and species diversity,whereas taxonomic diversity of nematode was homogeneous across the three beaches.In 0–20 cm sediment profile,nematode abundance declined abruptly with depth,whereas nematode diversity changed gently with obvious difference in 16–20 cm layer.Sediment granulometry and chlorophyll-a content were the two foremost factors which influenced the vertical distribution pattern of nematode generic diversity.Non-selective deposit feeders constituted the most dominant trophic group,followed by epistratum feeders.Bathylaimus(family:Tripyloididae) dominated at the second and Yangkou beach,while Theristus(family:Xyalidae) prevailed at Shilaoren beach.Omnivores and predators became important at Shilaoren beach because of the high proportion of Enoplolaimus.Even though,nematode community of the studied beaches did not differ significantly from each other.HUA Er MU Fanghong ZHANG Zhinan YANG Shichao ZHANG Ting LI Jia 2016Journal of Ocean University of China2016,15,1:2
19Nr2e1 Downregulation Is Involved in Excess Retinoic Acid-induced Developmental Abnormality in the Mouse Brain显示文摘Objective This study aimed to investigate the expression pattern and function of Nuclear receptor subfamily 2 group E member 1(Nr2e1) in retinoic acid(RA)-induced brain abnormality. Methods The mouse model of brain abnormality was established by administering 28 mg/kg RA, and neural stem cells(NSCs) were isolated from the mouse embryo and cultured in vitro. Nr2e1 expression was detected by whole mount in situ hybridization, RT-PCR, and Western blotting. Nr2e1 function was determined by transducing Nr2e1 shRNA into NSCs, and the effect on the sonic hedgehog(Shh) signaling pathway was assessed in the cells. In addition, the regulation of Nr2e1 expression by RA was also determined in vitro. Results Nr2e1 expression was significantly downregulated in the brain and NSCs of RA-treated mouse embryos, and knockdown of Nr2e1 affected the proliferation of NSCs in vitro. In addition, a similar expression pattern of Nr2e1 and RA receptor(RAR) α was observed after treatment of NSCs with different concentrations of RA.
