1Establishment of a novel corneal endothelial cell line from domestic rabbit,Oryctolagus curiculus显示文摘To develop a rabbit corneal endothelial (RCE) cell line, in vitro culture of RCE cells was initiated from Oryctolagus curiculus corneas and a novel RCE cell line was established in this study. To initiate the primary culture of RCE cells, corneas from rabbit eyes were sliced and attached into glutin-coated wells with endothelial cell surface down. After being cultured at a time-gradient interval from 48 to 6 h, the corneal slices were detached and reattached into new wells, respectively. Cells in the wells containing only a pure population of RCE cells were collected and cultured in 20% FBS-DMEM/F12 medium con- taining chondroitin sulfate, ocular extract, epidermal growth factor (EGF), basic fibroblast growth factor (bFGF), carboxymethyl-chitosan, N-acetylglucosamine hydrochloride, glucosamine hydrochloride, culture medium of rabbit corneal stromal cells and oxidation-degradation products of chondroitin sul- fate at 37℃, 5% CO2. The cultured RCE cells, in quadrangle and polygonal shapes, proliferated to con- fluence 3 weeks later. During the subsequent subculture, the shape of RCE cells changed gradually from polygonal to more fibroblastic. A novel RCE cell line, growing at a steady rate, with a population doubling time of 53.8 h, has been established and subcultured to passage 67. Chromosome analysis showed that the RCE cells exhibited chromosomal aneuploidy with the modal chromosome number of 44. The results of immuno-cytochemical staining with neuron specific enolase (NSE) confirmed that the RCE cells were in neuroectodermal origin. Combined with the results of vascular endothelial growth factor (VEGF) treatment and endothelial cell morphology recovery, it can be concluded that the cell line established here is an RCE cell line. 2Major advances in studies of the physical geography and living environment of China during the past 70 years and future prospects显示文摘The natural environment provides material essentials for human survival and development. The characteristics,processes, regional differentiation and forcing mechanisms of the elements of the natural environment(e.g. geomorphology,climate, hydrology, soil, etc.) are the main objects of research in physical geography. China has a complex natural environment and huge regional differentiation and therefore it provides outstanding reserach opportunities in physical geography. This review summarizes the most important developments and the main contributions of research in the physical geography and human living environment in China during the past 70 years. The major topics addressed are the uplift of the Tibetan Plateau and the evolution of its cryosphere, the development of fluvial systems, the acidification of the vast arid region of the Asian interior, variations in the monsoon and westerly climate systems on multiple timescales, the development of lakes and wetlands, the watershed system model, soil erosion, past human-environment interactions, biogeography, and physical geographic zonality. After briefly introducing international research developments, we review the history of research in physical geography in China, focusing on the major achievements and major academic debates, and finally we summarize the status of current research and the future prospects. 3MicroRNA-324-3p Plays A Protective Role Against Coxsackievirus B3-Induced Viral Myocarditis显示文摘Viral myocarditis(VM) is an inflammatory disease of the myocardium associated with heart failure, which is caused by common viral infections. A majority of the infections are initiated by coxsackievirus B3(CVB3). Micro RNAs(mi RNAs)have a major role in various biological processes, including gene expression, cell growth, proliferation, and apoptosis, as well as viral infection and antiviral immune responses. Although, mi RNAs have been found to regulate viral infections,their role in CVB3 infection remains poorly understood. In the previous study, mi RNA microarray results showed that mi R-324-3 p expression levels were significantly increased when cells and mice were infected with CVB3. It was also found that miR-324-3p downregulated TRIM27 and decreased CVB3 replication in vitro and in vivo. In vitro, analysis of downstream signaling of TRIM27 revealed that, miR-324-3p inhibited CVB3 infection, and reduced cytopathic effect and viral plaque formation by reducing the expression of TRIM27. In vivo, miR-324-3p decreased the expression of TRIM27,reduced cardiac viral replication and load, thereby strongly attenuating cardiac injury and inflammation. 4Effects of purified zearalenone on selected immunological and histopathologic measurements of spleen in post-weanling gilts显示文摘The present study was aimed at investigating the adverse effects of dietary zearalenone(ZEA) on the lymphocyte proliferation rate(LPR), interleukin-2(IL-2), mRNA expressions of pro-inflammatory cytokines, and histopathologic changes of spleen in post-weanling gilts. A total of 20 crossbred piglets(Yorkshire × Landrace × Duroc) with an initial BW of 10.36 ± 1.21 kg(21 d of age) were used in the study.Piglets were fed a basal diet with an addition of 0.1.1,2.0, or 3.2 mg/kg purified ZEA for 18 d ad libitum.The results showed that LPR and IL-2 production of spleen decreased linearly(P < 0.05) as dietary ZEA increased. Splenic mRNA expressions of interleukin-1β(IL-1β) and interleukin-6(IL-6) were linearly upregulated(P < 0.05) as dietary ZEA increased. On the contrary, linear down-regulation(P<0.05) of mRNA expression of interferon-γ(IFN-γ) was observed as dietary ZEA increased. Swelling splenocyte in1.1 mg/kg ZEA treatments, atrophy of white pulp and swelling of red pulp in 2.0 and 3.2 mg/kg ZEA treatments were observed. 5Engineering osteoarthritic cartilage model through differentiating senescent human mesenchymal stem cells for testing disease-modifying drugs显示文摘Significant cellular senescence has been observed in cartilage harvested from patients with osteoarthritis(OA).In this study,we aim to develop a senescence-relevant OA-like cartilage model for developing disease-modifying OA drugs(DMOADs).Spe-cifically,human bone marrow-derived mesenchymal stromal cells(MSCs)were expanded in vitro up to passage 10(P10-MSCs).Following their senescent phenotype formation,P10-MSCs were subjected to pellet culture in chondrogenic medium.Results from qRT-PCR,histology,and immunostaining indicated that cartilage generated from P10-MSCs displayed both senescent and OA-like phenotypes without using other OA-inducing agents,when compared to that from normal passage 4(P4)-MSCs.Interestingly,the same gene expression differences observed between P4-MSCs and P10-MSC-derived cartilage tissues were also observed between the preserved and damaged OA cartilage regions taken from human samples,as demonstrated by RNA sequencing data and other analysis methods.Lastly,the utility of this senescence-initiated OA-like cartilage model in drug development was assessed by testing several potential DMOADs and senolytics.The results suggest that pre-existing cellular senescence can induce the generation of OA-like changes in cartilage.The P4-and P10-MSCs derived cartilage models also represent a novel platform for predicting the efficacy and toxicity of potential DMOADs on both preserved and damaged cartilage in humans.Ning Wang Yuchen He Silvia Liu Meagan J.Makarcyzk Guanghua Lei Alexander Chang Peter G Alexander Tingjun Hao Anne-Marie Padget Nuria de Pedro Tsapekos Menelaos Hang Lin 2022Science China(Life Sciences)2022,65,2:5
6A small molecule inhibitor of the UBE2F-CRL5 axis induces apoptosis and radiosensitization in lung cancer显示文摘Protein neddylation is catalyzed by a neddylation activating enzyme(NAE,E1),an E2 conjugating enzyme,and an E3 ligase.In various types of human cancers,the neddylation pathway is abnormally activated.Our previous study validated that the neddylation E2 UBE2F is a promising therapeutic target in lung cancer.Although the NAE inhibitor MLN4924/pevonedistat is currently under clinical investigation as an anti-cancer agent,there are no small molecules available that selectively target UBE2F.Here,we report,for the first time,the discovery,via structure-based virtual screen and chemical optimization,of such a small molecule,designated as HA-9104.HA-9104 binds to UBE2F,reduces its protein levels,and consequently inhibits cullin-5 neddylation.Blockage of cullin-5 neddylation inactivates cullin-RING ligase-5(CRL5)activity,leading to accumulation of the CRL5 substrate,NOXA,to induce apoptosis.Moreover,HA-9104 appears to form the DNA adduct via its 7-azaindole group to induce DNA damage and G2/M arrest.Biologically,HA-9104 effectively suppresses the growth and survival of lung cancer cells and confers radiosensitization in both in vitro cell culture and in vivo xenograft tumor models.In summary,we discovered a small molecule,designated HA-9104,that targets the UBE2F-CRL5 axis with anti-cancer activity alone or in combination with radiation.Tiantian Xu Qisheng Ma Yanan Li Qing Yu Peichen Pan Yawen Zheng Zhijian Li Xiufang Xiong Tingjun Hou Bin Yu Hongmin Liu Yi Sun 2022Signal Transduction and Targeted Therapy2022,7,11:2
7Estimating interaction between surface water and groundwater in a permafrost region of the northern Tibetan Plateau using heat tracing method显示文摘Understanding the interaction between groundwater and surface water in permafrost regions is essential to study flood frequencies and river water quality, especially in the high latitude/altitude basins. The application of heat tracing method,based on oscillating streambed temperature signals, is a promising geophysical method for identifying and quantifying the interaction between groundwater and surface water. Analytical analysis based on a one-dimensional convective-conductive heat transport equation combined with the fiber-optic distributed temperature sensing method was applied on a streambed of a mountainous permafrost region in the Yeniugou Basin, located in the upper Heihe River on the northern Tibetan Plateau. The results indicated that low connectivity existed between the stream and groundwater in permafrost regions.The interaction between surface water and groundwater increased with the thawing of the active layer. 18Strontium doped electrospinning fiber membrane with antibacterial and osteogenic properties prepared by pulse electrochemical method显示文摘The osteogenic differentiation and new bone formation are commonly delayed by bacterial infection of orthopedic implants,which is urgent to be resolved quickly in the clinic.The current paper prepared a strontium-doped elec-trospinning fiber membrane with antibacterial and osteogenic properties by pulse electrochemical method.Poly-lactic acid/hydroxyapatite(PLLA/HA)composite fiber substrate was fabricated by electrospinning technology,and strontium doped SrHA/Cu/Polypyrrole(PPy)composite coating was constructed with pulse electrodeposi-tion method on its surface.The strontium doping technique,degradation of Sr^(2+)and Cu^(2+),cellular compatibility,and the antibacterial activity of the fiber membrane were examined.The results revealed that the deposition of phosphorus and calcium on composite fiber was the highest,indicating good biological activity.The release of Sr^(2+)and Cu^(2+)was stable and gradual due to the modulation of PPy.The composite fiber presented excellent antibacterial performance and the antibacterial rate was close to 100%against Staphylococcus aureus and Es-cherichia coli.Furthermore,it is conducive to the adhesion,spread,and proliferation of vascular endothelial cells and osteoblasts,namely outstanding osteogenesis and angiogenesis abilities.In conclusion,the multifunctional PLLA/HA@SrHA/Cu/PPy composite fiber membrane with good antibacterial and osteogenic activity by electro-spinning technology and pulsed electrochemical deposition method provides an effective strategy for the poor bone healing of infected bone defects.Yonggang Liu Bolin Zhang Feifei Liu Yani Qiu Wenbo Mu Liang Chen Chuang Ma Tingjun Ye Yingbo Wang 2022Engineered Regeneration2022,3,4:0
19Transcriptome of hepatopancreas in kuruma shrimp Marsupenaeus japonicus under low-salinity stress显示文摘The kuruma shrimp Marsupenaeus japonicus is one of the most commercially important shrimp species in the world.Low salinity would aff ect the penetration and immunity,and even led to its death of the shrimp.However,little is known about the molecular mechanism of the effect.Therefore,hepatopancreas of M.japonicus reared under low-salinity stress for 6,12,24,48,and 96 h was analyzed and the results were compared with that of the control group using transcriptomics.After removing reads containing adapters,88890960-1051300444 clean reads were generated from 10 libraries in the control group and experimental group.Compared with the control group,811,589,1095,745,and 875 diff erentially expressed genes were obtained in the five treatment groups.The N50 and N90 lengths of the transcripts were 1746 bp and 436 bp,respectively.The top 20 gene ontology terms and Kyoto Encyclopedia of Genes and Genomes pathways associated with the diff erentially expressed genes were related mainly to osmotic regulation(ion exchange,lipid metabolism and carbohydrate metabolism),immune regulation(cellular and humoral immunity),chitin metabolism,and related functions.The diff erential expression patterns of nine randomly selected genes were confi rmed by quantitative real-time PCR.This is the fi rst report of osmotic regulation-related genes that are diff erentially expressed under low-salinity stress in the hepatopancreas of M.japonicus.Furthermore,we found that M.japonicus initiated its own immune regulation under low-salinity stress.These results will help elucidating the mechanism of osmotic regulation and immune responses in this shrimp species.Tingjun CHEN Zhimin LI Jianyong LIU Caifeng LIANG Le YUAN 2022Journal of Oceanology and Limnology2022,40,2:0
20Thermal Deformation Behavior and Processing Map of a Novel CrFeNiSi_(0.15)Medium Entropy Alloy显示文摘The thermal deformation behavior of a novel CrFeNiSi_(0.15)medium entropy alloy(MEA)was studied via isothermal compression experiments,with the processing parameter range of 900–1200℃and 0.001–1 s^(−1).According to experimental data,the modified constitutive equation had been obtained,which precisely predicted the flow behavior of CrFeNiSi_(0.15)MEA during thermal deformation.At the same time,the processing map was established on the basis of the dynamic material model(DMM)theory.According to the map,the optimal processing parameters were determined at 1130–1200℃/0.06–1 s−1,under which the power dissipation efficiency could reach above 34%.The peak efficiency was above 38%,which occurred at 1200℃/1 s^(−1).In such parameter,complete dynamic recrystallization(DRX)also occurred.The flow instability of CrFeNiSi_(0.15)MEA was estimated to occur at 900–985℃/0.12–1 s^(−1),which was shown as grain boundaries cracking.Furthermore,both the continuous DRX(CDRX)and discontinuous DRX(DDRX)occurred simultaneously during thermal deformation.Meanwhile,some twins were also newly formed during DRX process,most of which were primary twins.The occurrence of twinning was beneficial to promote the development of DRX behavior.Hongbin Zhang Kang Chen Zhongwei Wang Haiping Zhou Chengcheng Shi Shengxue Qin Jie Liu Tingjun Lv Jian Xu 2023Acta Metallurgica Sinica(English Letters)2023,36,11:0
