| 1 | Sodium oligomannate therapeutically remodels gut microbiota and suppresses gut bacterial amino acids-shaped neuroinflammation to inhibit Alzheimer's disease progression显示文摘Recently,increasing evidence has suggested the association between gut dysbiosis and Alzheimer's disease(AD)progression,yet the role of gut microbiota in AD pathogenesis remains obscure.Herein,we provide a potential mechanistic link between gut microbiota dysbiosis and neuroinflammation in AD progression.Using AD mouse models,we discovered that,during AD progression,the alteration of gut microbiota composition leads to the peripheral accumulation of phenylalanine and isoleucine,which stimulates the differentiation and proliferation of pro-inflammatory T helper 1(Thl)cells.The brain-infiltrated peripheral Th1 immune cells are associated with the Ml microglia aaivation,contributing to AD-associated neuroinflammation.Importantly,the elevation of phenylalanine and isoleucine concentrations and the increase of Th1 cell frequency in the blood were also observed in two small independent cohorts of patients with mild cognitive impairment(MCI)due to AD.Furthermore,GV-971,a sodium oligomannate that has demonstrated solid and consistent cognition improvement in a phase 3 clinical trial in China,suppresses gut dysbiosis and the associated phenylalanine/isoleucine accumulation,harnesses neuroinflammation and reverses the cognition impairment.Together,our findings highlight the role of gut dysbiosis-promoted neuroinflammation in AD progression and suggest a novel strategy for AD therapy by remodelling the gut microbiota. | Xinyi Wang Guangqiang Sun Teng Feng Jing Zhang Xun Huang Tao Wang Zuoquan Xie Xingkun Chu Jun Yang Huan Wang Shuaishuai Chang Yanxue Gong Lingfei Ruan Guanqun Zhang Siyuan Yan Wen Lian Chen Du Dabing Yang Qingli Zhang Feifei Lin Jia Liu Haiyan Zhang Changrong Ge Shifu Xiao Jian Ding Meiyu Geng | 2019 | Cell Research2019,29,10: | 192 |
| 2 | Inhibition of SARS-CoV-2 (previously 2019-nCoV) infection by a highly potent pan-coronavirus fusion inhibitor targeting its spike protein that harbors a high capacity to mediate membrane fusion显示文摘The recent outbreak of coronavirus disease(COVID-19)caused by SARS-CoV-2 infection in Wuhan,China has posed a serious threat to global public health.To develop specific anti-coronavirus therapeutics and prophylactics,the molecular mechanism that underlies viral infection must first be defined.Therefore,we herein established a SARS-CoV-2 spike(S)protein-mediated cell-cell fusion assay and found that SARS-CoV-2 showed a superior plasma membrane fusion capacity compared to that of SARS-CoV.We solved the X-ray crystal structure of six-helical bundle(6-HB)core of the HR1 and HR2 domains in the SARS-CoV-2 S protein S2 subunit revealing that several mutated amino acid residues in the HR1 domain may be associated with enhanced interactions with the HR2 domain.We previously developed a pan-coronavirus fusion inhibitor,EK1,which targeted the HR!domain and could inhibit infection by divergent human coronaviruses tested,including SARS-CoV and MERS-CoV.Here we generated a series of lipopeptides derived from EK1 and found that EK1C4 was the most potent fusion inhibitor against SARS-CoV-2 S protein-mediated membrane fusion and pseudovirus infection with IC50s of 1.3 and 15.8 nM,about 241-and 149-fold more potent than the original EK1 peptide,respectively.EK1C4 was also highly effective against membrane fusion and infection of other human coronavirus pseudoviruses tested,including SARS-CoV and MERS-CoV,as well as SARSr-CoVs,and potently inhibited the replication of 5 live human coronaviruses examined,including SARS-CoV-2.Intranasal application of EK1C4 before or after challenge with HCoV-OC43 protected mice from infection,suggesting that EK1C4 could be used for prevention and treatment of infection by the currently circulating SARS-CoV-2 and other emerging SARSr-CoVs. | Shuai Xia Meiqin Liu Chao Wang Wei Xu Qiaoshuai Lan Siliang Feng Feifei Qi Linlin Bao Lanying Du Shuwen Liu Chuan Qin Fei Sun Zhengli Shi Yun Zhu Shibo Jiang Lu Lu | 2020 | Cell Research2020,30,4: | 81 |
| 3 | Transplantation of UC-MSCs on collagen scaffold activates follicles in dormant ovaries of POF patients with long history of infertility显示文摘Premature ovarian failure(POF) is a refractory disease for clinical treatment with the goal of restoring fertility. In this study,umbilical cord mesenchymal stem cells on a collagen scaffold(collagen/UC-MSCs) can activate primordial follicles in vitro via phosphorylation of FOXO3 a and FOXO1. Transplantation of collagen/UC-MSCs to the ovaries of POF patients rescued overall ovarian function, evidenced by elevated estradiol concentrations, improved follicular development, and increased number of antral follicles. Successful clinical pregnancy was achieved in women with POF after transplantation of collagen/UC-MSCs or UC-MSCs. | 4 | Targeting ERK,an Achilles' Heel of the MAPK pathway,in cancer therapy显示文摘The mitogen-activated protein kinases(MAPK) pathway, often known as the RAS-RAFMEK-ERK signal cascade, functions to transmit upstream signals to its downstream effectors to regulate physiological process such as cell proliferation, differentiation, survival and death. As the most frequently mutated signaling pathway in human cancer, targeting the MAPK pathway has long been considered a promising strategy for cancer therapy. Substantial efforts in the past decades have led to the clinical success of BRAF and MEK inhibitors. However, the clinical benefits of these inhibitors are compromised by the frequently occurring acquired resistance due to cancer heterogeneity and genomic instability. This review briefly introduces the key protein kinases involved in this pathway as well as their activation mechanisms. We also generalize the correlations between mutations of MAPK members and human cancers, followed by a summarization of progress made on the development of small molecule MAPK kinases inhibitors. | 5 | Progress and recent trend in MILD combustion显示文摘Moderate or intense low oxygen dilution(MILD)combustion plays a significant role in the mitigation of combustion-generated pollutants and greenhouse gases whilst meeting thermal efficiency needs.However,due to the lack of the fundamental knowledge on this combustion,there is a misconception that MILD combustion should be established by high preheating of the air,which has limited its application.Our research and development on this combustion has been performed for several years. | 8 | Characterization of drug responses of mini patient-derived xenografts in mice for predicting cancer patient clinical therapeutic response显示文摘Background:Patient-derived organoids and xenografts(PDXs)have emerged as powerful models in functional diag-nostics with high predictive power for anticancer drug response.However,limitations such as engraftment failure and time-consuming for establishing and expanding PDX models followed by testing drug efficacy,and inability to subject to systemic drug administration for ex vivo organoid culture hinder realistic and fast decision-making in selecting the right therapeutics in the clinic.The present study aimed to develop an advanced PDX model,namely MiniPDX,for rapidly testing drug efficacy to strengthen its value in personalized cancer treatment.Methods:We developed a rapid in vivo drug sensitivity assay,OncoVee®MiniPDX,for screening clinically relevant regimens for cancer.In this model,patient-derived tumor cells were arrayed within hollow fiber capsules,implanted subcutaneously into mice and cultured for 7 days.The cellular activity morphology and pharmacokinetics were systematically evaluated.MiniPDX performance(sensitivity,specificity,positive and negative predictive values)was examined using PDX as the reference.Drug responses were examined by tumor cell growth inhibition rate and tumor growth inhibition rate in PDX models and MiniPDX assays respectively.The results from MiniPDX were also used to evaluate its predictive power for clinical outcomes.Results:Morphological and histopathological features of tumor cells within the MiniPDX capsules matched those both in PDX models and in original tumors.Drug responses in the PDX tumor graft assays correlated well with those in the corresponding MiniPDX assays using 26 PDX models generated from patients,including 14 gastric cancer,10 lung cancer and 2 pancreatic cancer.The positive predictive value of MiniPDX was 92%,and the negative predictive value was 81%with a sensitivity of 80%and a specificity of 93%.Through expanding to clinical tumor samples,Min-iPDX assay showed potential of wide clinical application.Conclusions:Fast in vivo MiniPDX assay based on capsule implantation was developed-to assess drug responses of both PDX tumor grafts and clinical cancer specimens.The high correlation between drug responses of paired MiniPDX and PDX tumor graft assay,as well as translational data suggest that MiniPDX assay is an advanced tool for personalized cancer treatment. | Feifei Zhang Wenjie Wang Yuan Long Hui Liu Jijun Cheng Lin Guo Rongyu Li Chao Meng Shan Yu Qingchuan Zhao Shun Lu Lili Wang Haitao Wang Danyi Wen | 2018 | Cancer Communications2018,38,1: | 20 |
| 9 | Generation of B Cell-Deficient Pigs by Highly Efficient CRISPR/Cas9-Mediated Gene Targeting显示文摘Generating B cell-deficient mutant is the first step to produce human antibody repertoires in large animal models. In this study, we applied the clustered regularly interspaced short palindromic repeat(CRISPR)/CRISPR-associated(Cas) system to target the JHregion of the pig Ig M heavy chain gene which is crucial for B cell development and differentiation. Transfection of Ig M-targeting Cas9 plasmid in primary porcine fetal fibroblasts(PFFs) enabled inducing gene knock out(KO) in up to 53.3% of colonies analyzed, a quarter of which harbored biallelic modification, which was much higher than that of the traditional homologous recombination(HR). With the aid of somatic cell nuclear transfer(SCNT) technology, three piglets with the biallelic Ig M heavy chain gene mutation were produced. The piglets showed no antibody-producing B cells which indicated that the biallelic mutation of the Ig M heavy chain gene effectively knocked out the function of the Ig M and resulted in a B cell-deficient phenotype. | 10 | Bird watching in China reveals bird distribution changes显示文摘Using China Bird Report(2003-2007) as data source in combination with descriptions about bird habitats,we built up the China Bird Watching Database.We also developed spherical GIS software 'Global Analyst' to create the point-based database which contains accurate spatial-temporal information.The China Bird Watching Database can reflect the achievement of Chinese bird watchers and complement the basic knowledge of bird distribution.Now a total of 30936 records from 17 Orders,70 Families and 1078 Species of 5 years are included in the database,representing over 80% of all bird species in China.Till 2007,the geographic coverage has encompassed all provincial level administrative districts in China,with the exception of Hong Kong and Taiwan.The China Bird Watching Database also recorded a group of species which are additions at national and provincial levels,including 14 species which are additions to the national checklist and 109 species which appeared outside their original distributions.Comparing the new records with their original distributions,we found the trend that species move to higher latitude and higher elevation regions and some species of waterfowls in Xinjiang Uygur Autonomous Region,including a suite of rare seabirds in the China's Mainland.The majority of bird watchers come from the Eastern Region of China,but their covering range is spreading northwest.At the same time,we appeal to adopting a suite of new technologies for observation,and building up sharing platform of bird watching data to capture the distribution dynamics of birds in China and provide a direct foundation for bird conservation. | LI XueYan LIANG Lu GONG Peng LIU Yang LIANG FeiFei | 2013 | Chinese Science Bulletin2013,58,6: | 13 |

| 12 | Secondary Crystallization of Na_2CO_3-Modified HZSM-5 Zeolites with Tetrapropylammonium Hydroxide and Their Catalytic Performance in Thiophene Alkylation Reaction显示文摘The Na2CO3-modified HZSM-5 zeolites were further treated by tetrapropylammonium hydroxide(TPAOH) solution. The effect of TPAOH concentration on the secondary crystallization process was investigated. The resulting samples were characterized by a complementary combination of X-ray diffraction, N2 adsorption/desorption, scanning electron microscopy, X-ray fluorescence spectroscopy, XPS, 27 Al and 29 Si magic-angle spinning nuclear magnetic resonance spectroscopy, BET and temperature-programmed desorption techniques. The results showed that the secondary crystallization of the HZSM-5 zeolite could result in migration of non-framework species from the internal channels to the zeolite surface and their transformation into framework species. The catalytic activity of these modified samples for thiophene alkylation was evaluated. | 13 | Paleomagnetic results of Late Paleozoic rocks from northern Qiangtang Block in Qinghai-Tibet Plateau, China显示文摘Tectonic evolution of the Tethys and the boundary between the Gondwanaland and the Eurasia during the Carboniferous and Permian remain hotly debated. Qiangtang region in the Qinghai-Tibet Plateau may be a key place to study these problems. A paleomagnetic study was conducted on the Late Paleozoic rocks in the northern Qiangtang region (33.7°N, 86.7°W), Tibet. Two sites (21 samples) in the Upper Carboniferous, eleven sites (101 samples) in the Permian, and two sites (16 samples) in the Lower Triassic were investigated. The rock magnetic data revealed hematite and magnetite as the main magnetic carriers. In stepwise thermal demagnetization and/or combined alternating field (AC) demagnetization, two characteristic components in the majority of the samples were identified as (1) the Low-temperature Component (LTC), characterized by northerly decli- nation and moderate to steep inclination, corresponding to a pole position overlay with the present North Pole. A minority of the samples present single component, and their directions are the same as (2) the High-temperature Component (HTC) of double components. The combined single-component and HTC data of the Permian can pass the R-test at 95% level and the F-test at 99% level, as well as the BC-test. The pole position from the Late Carboniferous is at 31.8°S, 45.7°E with dp=2.1, dm=3.9, that from the Early and Middle (Late) Permian is at 31.7°S, 46.8°E with dp=9.2, dm=16.9 (34.4°N, 54.1°E with dp=6.9, dm=12.5) respectively, and that from the Early Triassic is at 16.9°S, 22.5°E with dp=4.9, dm=9.2. Starting with the definition of object-oriented remotely sensed information classification pattern and a literature review of related research progress, this paper sums up 4 developing phases of object-oriented classification pattern during the past 20 years. Then, we discuss the three aspects of method- ology in detail, namely remotely sensed imagery segmentation, feature analysis and feature selection, and classification rule generation, through comparing them with remotely sensed information classification method based on per-pixel. | 17 | A GATA-type transcription factor AcAREB for nitrogen metabolism is involved in regulation of cephalosporin biosynthesis in Acremonium chrysogenum显示文摘In filamentous fungi,nitrogen metabolism is repressed by GATA-type zinc finger transcription factors.Nitrogen metabolite repression has been found to affect antibiotic production,but the mechanism is still poorly understood.AcareB,encoding a homologue of fungal GATA-type regulatory protein,was cloned from Acremonium chrysogenum.Gene disruption and genetic complementation demonstrated that AcareB plays a key role in utilization of ammonium,glutamine and urea.In addition,significant reduction of cephalosporin production in the AcareB disruption mutant indicated that AcareB is important for cephalosporin production.In consistence with it,the transcriptional level of cephalosporin biosynthetic genes was significantly decreased in the AcareB disruption mutant.Electrophoretic mobility shift assay showed that AcAREB directly bound to the intergenic regions of pcbAB-pcbC,cefD1-cefD2 and cefEF-cefG.Sequence analysis showed that all the AcAREB binding sites contained the consensus GATA elements.AcareB is negatively autoregulated during cephalosporin production.Moreover,another GATA zinc-finger protein encoded by AcareA positively regulates the transcription of AcareB.However,AcareB does not regulate the transcription of AcareA.These results indicated that AcAREB plays an important role in both regulation of nitrogen metabolism and cephalosporin production in A.chrysogenum. | Feifei Guan Yuanyuan Pan Jinyang Li Gang Liu | 2017 | Science China(Life Sciences)2017,60,9: | 8 |
| 18 | Formaldehyde concentration and its influencing factors in residential homes after decoration at Hangzhou,China显示文摘Air pollution surveys of formaldehyde(HCHO) were conducted in 2324 rooms decorated within one year in 2007-2009 in Hangzhou,China.The mean HCHO concentration(C HCHO) was 0.107 ± 0.095 mg/m 3,and 38.9% of samples exceeded the Chinese National Standard GB 50325-2010.Over the past 3 years,the C HCHO decreased with time(p < 0.05).Relationships of potential factors to indoor C HCHO were also evaluated.C HCHO was related to temperature(T),relative humidity(RH),time duration of the windows and doors being closed before sampling(DC),time duration from the end of decoration to sampling(DR) and source characteristics(d).A model to relate indoor C HCHO to these five factors(T,RH,DC,DR,d) was established based on 298 samples(R 2 = 0.87).Various factors contributed to C HCHO in the following order:T,43.7%;d,31.0%;DC,10.2%;DR,8.0%;RH,7.0%;specifically,meteorological conditions(i.e.,RH plus T) accounted for 50.7%.The coefficient of T and RH,R TH,was proposed to describe their combined influence on HCHO emission,which also had a linear relationship(R 2 = 0.9387) with HCHO release in a simulation chamber test.In addition,experiments confirm that it is a synergistic action as T and RH accelerate the release of HCHO,and that is a significant factor influencing indoor HCHO pollution.These achievements could lead to reference values of measures for the efficient reduction of indoor HCHO pollution. | Min Guo Xiaoqiang Pei Feifei Mo Jianlei Liu Xueyou Shen | 2013 | Journal of Environmental Sciences2013,25,5: | 7 |

| 20 | Geochronology and geochemistry of mafic dykes in the Helanshan complex: Implications for Mesozoic tectonics in the North China Craton显示文摘The Helanshan tectonic belt(HTB) is a major tectonic divide between the Alxa and Ordos blocks in the North China Craton. The geochronology and petrogenesis of the mafic dykes in the northern HTB are keys to understanding the tectonic evolution of this belt. The mafic dykes, intruded into the Neoarchean-Paleoproterozoic metamorphic basement, are mainly composed of diabase with a mineral assemblage of plagioclase(45%-60%), pyroxene(25%-35%), minor quartz and Fe-Ti oxides. The LA-ICPMS U-Pb analysis of zircon grains from representative dykes yield a weighted mean age of 206 ± 1.9 Ma, which represents the crystallization age of the dyke. The diabases show high contents of Fe_2 O_3~T(11.88-17.55 wt.%), low contents of SiO_2(45.65-50.95 wt.%) and MgO(3.31-5.50 wt.%) with low Mg#(=100×MgO/(MgO + FeO) atomic ration) of 33-44. They are characterized by enrichment of light rare earth elements(LREEs) and large ion lithophile elements(LILEs)(e.g., Rb, Ba and Pb), and slight depletion of high field strength elements(HFSEs). These features suggest that the magma has undergone extensive fractionation of olivine and pyroxene but only minor crustal contamination during its evolution. Their high Sm contents and La/Sm ratios, and low Sm/Yb ratios indicate that magma from which the dykes formed was derived from low degree(about 5%) partial melting of an enriched garnet + spinel lherzolite mantle source.
