| 1 | Metallurgical challenges in microelectronic 3D IC packaging technology for future consumer electronic products显示文摘Metallurgical challenges in controlling the microstructural stability of Cu and solder microbumps in 3D IC packaging technol-ogy are discussed. Using uni-directional <111> oriented nanotwinned Cu, the controlled growth of oriented Cu6Sn5 on the nanotwinned Cu and its transformation to Cu3Sn without Kirkendall voids have been achieved. In order to join a stack of Si chips into a 3D device, multiple reflows of solder microbumps may be required; we consider localized heating to do so by the use of self-sustained explosive reaction in multi-layered Al/Ni thin films of nano thickness. | 3 | Bioengineered miR-27b-3p and miR-328-3p modulate drug metabolism and disposition via the regulation of target ADME gene expression显示文摘Drug-metabolizing enzymes, transporters, and nuclear receptors are essential for the absorption, distribution, metabolism, and excretion(ADME) of drugs and xenobiotics. MicroRNAs participate in the regulation of ADME gene expression via imperfect complementary Watson–Crick base pairings with target transcripts. We have previously reported that Cytochrome P450 3A4(CYP3A4) and ATP-binding cassette sub-family G member 2(ABCG2) are regulated by miR-27b-3p and miR-328-3p,respectively. Here we employed our newly established RNA bioengineering technology to produce bioengineered RNA agents(BERA), namely BERA/miR-27b-3p and BERA/miR-328-3p, via fermentation. When introduced into human cells, BERA/miR-27b-3p and BERA/miR-328-3p were selectively processed to target miRNAs and thus knock down CYP3A4 and ABCG2 mRNA and their protein levels,respectively, as compared to cells treated with vehicle or control RNA. Consequently, BERA/miR-27b-3p led to a lower midazolam 10-hydroxylase activity, indicating the reduction of CYP3A4 activity. Likewise,BERA/miR-328-3p treatment elevated the intracellular accumulation of anticancer drug mitoxantrone, a classic substrate of ABCG2, hence sensitized the cells to chemotherapy. | 6 | Discrete energetic(~50-200 keV) electron events in the high-altitude cusp/polar cap/lobe显示文摘We identified 28 discrete electron events(DEEs) with enhanced fluxes at ~50-200 keV in the high-altitude cusp/polar cap/lobe,using the electron measurements by the BeiDa Image Electron Spectrometer(BD-IES) instrument onboard an inclined(55°)geosynchronous orbit(IGSO) satellite from October 2015 to January 2016. We find that among the 28 DEEs, 22 occur in the nightside and mostly in the northern cusp/polar cap/lobe, while 6 occur in the dayside and all in the southern cusp; 24 events correspond to an average interplanetary magnetic field(IMF) component B_z>0, 3 correspond to an average IMF B_z<0, and 1 has no OMNI IMF data. In these DEEs, the observed average omnidirectional electron differential flux generally fits well to a power-law spectrum, J^E^(-γ), with the spectral index y ranging from 2.6 to 4.6, while the average electron flux varies over three orders of magnitude from event to event. The spectral index of these cusp DEEs are(strongly) larger than the spectral index of solar wind superhalo electrons(radiation belt electrons) observed by the WIND 3 D Plasma & Energetic Particle instrument(the BD-IES). At^110 keV,the electron flux of DEEs in the cusp/polar cap/lobe shows a weak positive correlation with the solar wind superhalo electron flux but no obvious correlation with the radiation belt electron flux. | 8 | A Versatile Surface Bioengineering Strategy Based on Mussel-Inspired and Bioclickable Peptide Mimic显示文摘In this work,we present a versatile surface engineering strategy by the combination of mussel adhesive peptide mimicking and bioorthogonal click chemistry.The main idea reflected in this work derived from a novel mussel-inspired peptide mimic with a bioclickable azide group(i.e.,DOPA_(4)-azide).Similar to the adhesion mechanism of the mussel foot protein(i.e.,covalent/noncovalent comediated surface adhesion),the bioinspired and bioclickable peptide mimic DOPA_(4)-azide enables stable binding on a broad range of materials,such as metallic,inorganic,and organic polymer substrates.In addition to the material universality,the azide residues of DOPA_(4)-azide are also capable of a specific conjugation of dibenzylcyclooctyne-(DBCO-)modified bioactive ligands through bioorthogonal click reaction in a second step.To demonstrate the applicability of this strategy for diversified biofunctionalization,we bioorthogonally conjugated several typical bioactive molecules with DBCO functionalization on different substrates to fabricate functional surfaces which fulfil essential requirements of biomedically used implants.For instance,antibiofouling,antibacterial,and antithrombogenic properties could be easily applied to the relevant biomaterial surfaces,by grafting antifouling polymer,antibacterial peptide,and NO-generating catalyst,respectively.Overall,the novel surface bioengineering strategy has shown broad applicability for both the types of substrate materials and the expected biofunctionalities.Conceivably,the“clean”molecular modification of bioorthogonal chemistry and the universality of mussel-inspired surface adhesion may synergically provide a versatile surface bioengineering strategy for a wide range of biomedical materials. | Yu Xiao Wenxuan Wang Xiaohua Tian Xing Tan Tong Yang Peng Gao Kaiqing Xiong Qiufen Tu Miao Wang Manfred FMaitz Nan Huang Guoqing Pan Zhilu Yang | 2020 | Research2020,,1: | 3 |

| 11 | Optimization and Control of Extractive Distillation with Heat Integration for Separating Benzene/Cyclohexane Mixtures显示文摘In this work, the extractive distillation with heat integration process is extended to separate the pressure-insensitive benzene-cyclohexane azeotrope by using furfural as the entrainer. The optimal design of extractive distillation process is established to achieve minimum energy requirement using the multi-objective genetic algorithm, and the results show that energy saving for this heat integration process is 15.7%. Finally, the control design is performed to investigate the system's dynamic performance, and three control structures are studied. Methods:Human hepatoma HepG2 cells were transfected with pcDNA3.1(-)-AEG1 plasmid or psilencer2.0-AEG1-shRNA1 plasmid to up/down-regulate AEG1 expression, pcDNA3.1(-) and psilencer 2.0 empty vector plasmids were transfected respectively as control. Real-time RT-PCR was carried out to measure the expression levels of AEG1 and COX-2 mRNA. The expression levels of AEG1 and COX-2 protein were detected by Western blot. NF-κB signaling was blocked by PDTC, and AP-1 signaling was blocked by curcumin. Results:AEG1 mRNA and protein levels were increased after pcDNA3.1(-)-AEG1 transfection, and decreased after psilencer2.0-AEG1-shRNAs transfection. COX-2 mRNA and protein levels were increased in AEG1-overexpressing cells and decreased in AEG1-knockdown cells. Phosphorylations of p65 and c-jun were up-regulated in AEG1-overexpressing cells. Both PDTC and curcumin reduced COX-2 expression in HepG2 cells with AEG1 overexpression. Conclusion:AEG1-overexpressing and-knockdown HepG2 cells are established successfully. | 18 | Quantitative proteomics revealed extensive microenvironmental changes after stem cell transplantation in ischemic stroke显示文摘The local microenvironment is essential to stem cell-based therapy for ischemic stroke,and spatiotemporal changes of the microenvironment in the pathological process provide vital clues for understanding the therapeutic mechanisms.However,relevant studies on microenvironmental changes were mainly confined in the acute phase of stroke,and long-term changes remain unclear.This study aimed to investigate the microenvironmental changes in the subacute and chronic phases of ischemic stroke after stem cell transplantation.Herein,induced pluripotent stem cells(iPSCs)and neural stem cells(NSCs)were transplanted into the ischemic brain established by middle cerebral artery occlusion surgery.Positron emission tomography imaging and neurological tests were applied to evaluate the metabolic and neurofunctional alterations of rats transplanted with stem cells.Quantitative proteomics was employed to investigate the protein expression profiles in iPSCs-transplanted brain in the subacute and chronic phases of stroke.Compared with NSCs-transplanted rats,significantly increased glucose metabolism and neurofunctional scores were observed in iPSCs-transplanted rats.Subsequent proteomic data of iPSCs-transplanted rats identified a total of 39 differentially expressed proteins in the subacute and chronic phases,which are involved in various ischemic stroke-related biological processes,including neuronal survival,axonal remodeling,antioxidative stress,and mitochondrial function restoration.Taken together,our study indicated that iPSCs have a positive therapeutic effect in ischemic stroke and emphasized the wide-ranging microenvironmental changes in the subacute and chronic phases. | Yao Chen Fahuan Song Mengjiao Tu Shuang Wu Xiao He Hao Liu Caiyun Xu Kai Zhang Yuankai Zhu Rui Zhou Chentao Jin Ping Wang Hong Zhang Mei Tian | 2022 | Frontiers of Medicine2022,16,3: | 1 |
