3Microwave Synthesis of Large Few-Layer Graphene Sheets in Aqueous Solution of Ammonia显示文摘Few-layer graphene (FLG) sheets with sizes exceeding several micrometers have been synthesized by exfoliation of expanded graphite in aqueous solution of ammonia under microwave irradiation, with an overall yield approaching 8 wt.%. Transmission electron microscopy (in bright-field and dark-field modes) together with electron diffraction patterns and atomic force microscopy confirmed that this graphene material consisted mostly of mono-, bi- or few-layer graphene (less than ten layers). The high degree of surface reduction was confirmed by X-ray photoelectron and infrared spectroscopies. 5Association of colorectal cancer with pathogenic Escherichia coli: Focus on mechanisms using optical imaging显示文摘AIM: To investigate the molecular or cellular mechanisms related to the infection of epithelial colonic mucosa by pks-positive Escherichia coli(E. coli) using optical imaging.METHODS: We choose to evaluate the tumor metabolic activity using a fluorodeoxyglucose analogue as 2-deoxyglucosone fluorescent probes and to correlate it with tumoral volume(mm^3). Inflammation measuring myeloperoxidase(MPO) activity and reactive oxygen species production was monitored by a bioluminescent(BLI) inflammation probe and related to histological examination and MPO levels by enzyme-linked immunosorbent assay(ELISA) on tumor specimens. The detection and quantitation of these two signals were validated on a xenograft model of human colon adenocarcinoma epithelial cells(HCT116) in nude mice infected with a pks-positive E. coli. The inflammatory BLI signal was validated intra-digestively in the colitisCEABAC10 DSS models, which mimicked Crohn's disease. RESULTS: Using a 2-deoxyglucosone fluorescent probe, we observed a high and specific HCT116 tumor uptake in correlation with tumoral volume(P = 0.0036). Using the inflammation probe targeting MPO, we detected a rapid systemic elimination and a significant increase of the BLI signal in the pks-positive E. coli-infected HCT116 xenograft group(P < 0.005). ELISA confirmed that MPO levels were significantly higher(1556 ± 313.6 vs 234.6 ± 121.6 ng/m L P = 0.001) in xenografts infected with the pathogenic E. coli strain. Moreover, histological examination of tumor samples confirmed massive infiltration of pks-positive E. coli-infected HCT116 tumors by inflammatory cells compared to the uninfected group. These data showed that infection with the pathogenic E. coli strain enhanced inflammation and ROS production in tumors before tumor growth. 6Organic matter produced by algae and cyanobacteria: Quantitative and qualitative characterization显示文摘This work aims at characterizing organic matter produced by an alga Euglena gracilis and a cyanobacteria Microcystis aeruginosa and assessing the evolution of its characteristics during growth.A culture medium was optimized.The species growth phases were monitored using both visible spectrophotometry and flow cytometry cell counting.Organic matter fractionation according to hydrophobicity and specific UV absorbance(SUVA) index were used to specifically characterize the produced algal organic matter(AOM).The AOM characteristics were both growth phase and species dependent.However,a similar evolution was observed.The hydrophilic fraction(HPI) was the major fraction whatever the growth phases and was almost the only one produced during lag and exponential phases.It represented around 75% of AOM during exponential phase and then decreased when the stationary phase appeared.It represented 46% and 60% of the AOM during late decline phase for the cyanobacteria and the alga respectively.The hydrophobic(HPO) and transphilic(TPH) fractions started to appear from the beginning of the stationary phase with more hydrophobic compounds coming from intracellular organic material of dying cells.HPO and TPH percentages still increased during the decline phase probably because of two additional processes:photo-dissolution and leaching of particulate organic matter from cells fragments.A comparison of AOM during late decline phase and natural organic matter(NOM) from Glane River(France) underlined that AOM was more hydrophilic and presented a lower SUVA for each fractions than NOM.However,the difference between NOM and AOM hydrophobicity narrowed during decline phase.Maud Leloup Rudy Nicolau Virginie Pallier Claude Yprmian Geneviève Feuillade-Cathalifaud 2013Journal of Environmental Sciences2013,25,6:10

11Evaluation of the colorectal cancer risk conferred by rare UNC5C alleles显示文摘AIM:To evaluate the risk associated with variants of the UNC5C gene recently suspected to predispose to familial colorectal cancer(CRC).METHODS:We screened patients with familial CRC forms as well as patients with sporadic CRCs.In a first time,we analyzed exon 11 of the UNC5C gene in 120unrelated patients with suspected hereditary CRC,58patients with suspected Lynch-associated cancer or polyposis,and 132 index cases of Lynch syndrome families with a characterized mutation in a DNA mismatch repair(MMR).Next,1023 patients with sporadic CRC and1121 healthy individuals were screened for the variants identified in patients with familial cancer.RESULTS:Of 120 patients with familial CRC of unknown etiology,one carried the previously reported mis-sense mutation p.Arg603Cys(R603C)and another exhibited the unreported variant of unknown significance p.Thr617Ile(T617I).The p.Ala628Lys(A628K)mutation previously described as the main UNC5C risk variant for familial CRC was not detected in any cases of familial CRC of unknown etiology,but was present in a patient with familial gastric cancer and in two Lynch syndrome patients in co-occurrence with MMR mutations.A statistically non-significant increase in cancer risk was identified in familial CRC and/or other Lynchassociated cancers(1/178 patients vs 2/1121 healthy controls,OR=3.2,95%CI:0.29-35.05,P=0.348)and in sporadic CRCs(4/1023 patients vs 2/1121 healthy controls,OR=2.2,95%CI:0.40-12.02,P=0.364).CONCLUSION:We confirm that UNC5C mutations are very rare in familial and sporadic CRCs,but further investigations are needed to justify routine UNC5C testing for diagnostic purposes.Sébastien Küry Céline Garrec Fabrice Airaud Flora Breheret Virginie Guibert Cécile Frenard Shuo Jiao Dominique Bonneau Pascaline Berthet Céline Bossard Olivier Ingster Estelle Cauchin Stéphane Bezieau 2014World Journal of Gastroenterology2014,20,1:4
12Microscopic features of colorectal neoplasia in inflammatory bowel diseases显示文摘The risk of developing dysplasia leading to colorectal cancer(CRC)is increased in both ulcerative colitis and Crohn’s disease.The prognosis of CRC may be poorer in patients with inflammatory bowel disease(IBD)than in those without IBD.Most CRCs,in general,develop from a dysplastic precursor lesion.The interpretation by the pathologist of the biopsy will guide decision making in clinical practice:colonoscopic surveillance or surgical management.This review summarizes features of dysplasia(or intraepithelial neoplasia)with macroscopic and microscopic characteristics.From an endoscopic(gross)point of view,dysplasia may be classified as flat or elevated(raised);from a histological point of view,dysplasia is separated into 3 distinct categories:negative for dysplasia,indefinite for dysplasia,and positive for dysplasia with low-or high-grade dysplasia.The morphologic criteria for dysplasia are based on a combination of cytologic(nuclear and cytoplasmic)and architectural aberrations of the crypt epithelium.Immunohistochemical and molecular markers for dysplasia are reviewed and may help with dysplasia diagnosis,although diagnosis is essentially based on morphological criteria.The clinical,epidemiologic,and pathologic characteristics of IBD-related cancers are,in many aspects,different from those that occur sporadically in the general population.Herein,we summarize macroscopic and microscopic features of IBD-related colorectal carcinoma.Aude Bressenot Virginie Cahn Silvio Danese Laurent Peyrin-Biroulet 2014World Journal of Gastroenterology2014,20,12:4

16Functionalization of Single- and Multi-Walled Carbon Nanotubes with Cationic Amphiphiles for Plasmid DNA Complexation and Transfection显示文摘The possibility of delivering DNA effi ciently to cells represents a crucial issue for the treatment of both genetic and acquired diseases.However,even although the effi ciency of non-viral transfection systems has improved in the last decade,none have yet proven to be suffi ciently effective in vivo.We report herein our results on the functionalization of single-walled carbon nanotubes(SWNT)and multi-walled carbon nanotubes(MWNT)by two cationic amphiphiles(lipid RPR120535 and pyrenyl polyamine),their use for the complexation of plasmid DNA,and their efficiency in transfecting cells in vitro.The experiments have shown that the efficiency of transfection is higher when using SWNT instead of MWNT,and that transfection effi ciency is similar or slightly higher when using nanoplexes(SWNT/lipid RPR120535/DNA)instead of lipoplexes(lipid RPR120535/DNA)and several orders of magnitude higher than that of naked DNA.This study therefore shows both that the transfection is better when using SWNTs and that it is dependent on the nature of the amphiphilic molecules adsorbed on the nanotubes.Cyrille Richard Nathalie Mignet Céline Largeau Virginie Escriou Michel Bessodes Daniel Scherman 2009Nano Research2009,2,8:3
