| 8 | Modelling impacts of acid deposition and groundwater level on habitat quality and plant species diversity显示文摘Introduction:We quantified the effects of the site factors pH and nitrate(NO_(3))concentration in soil solution and groundwater level on the vegetation of terrestrial ecosystems for the Netherlands in response to changes in atmospheric nitrogen(N)and sulphur(S)deposition and groundwater level over the period 1990-2030.The assessment was made with the SMART2 model,a simple one-layer model including geochemical buffer processes,element cycling by litterfall,mineralisation and uptake,nitrogen transformation processes and element input through deposition,weathering and upward seepage.Methods:To assess the effects of changes in abiotic site factors on the vegetation,we developed a simple plant diversity indicator for grassland,heathland and forest,based on the occurrence of target plant species and competing species.Species occurrence was calculated from the preferred ranges of each species for the NO_(3) concentration and pH in soil solution and mean spring groundwater level.Changes in the plant diversity indicator were assessed from effects of changes in the occurrence of target and competing plant species in response to changes in mean spring groundwater level and in pH and NO_(3) concentration,as calculated with SMART2.Calculations were made for combinations of five vegetation structure types(three forest types,semi-natural grassland and heathland)and seven soil types(three sandy soils,two clay soils,peat and loess soils)using a 250×250 m grid.We used data for atmospheric deposition and groundwater level in the past to assess trends between 1990 and 2010 and evaluated two future scenarios for the period 2010-2030:a Business as Usual and an Improved Environment scenario.Results:Comparison of model predictions on pH and NO_(3) with measured soil solution concentrations for forest showed a reasonable to good agreement for pH but rather poor for NO_(3).The largest impacts were found for the combination of the two Improved Environment scenarios.Conclusions:Reductions in N and S deposition and an increase in groundwater level between 1990 and 2030 hardly caused changes in soil pH and only relatively small reductions in NO_(3) concentration(11-13%).Nevertheless,those changes caused a significant increase in plant diversity indicator. | J.Kros J.P.Mol-Dijkstra G.W.W.Wamelink G.J.Reinds Avan Hinsberg W.de Vries | 2016 | Ecological Processes2016,5,1: | 1 |

| 13 | Transport receptor occupancy in nuclear pore complex mimics显示文摘Nuclear pore complexes(NPCs)regulate all molecular transport between the nucleus and the cytoplasm in eukaryotic cells.Intrinsically disordered Phe-Gly nucleoporins(FG-Nups)line the central conduit of NPCs to impart a selective barrier where large proteins are excluded unless bound to a transport receptor(karyopherin;Kap).Here,we assess“Kap-centric”NPC models,which postulate that Kaps participate in establishing the selective barrier.We combine biomimetic nanopores,formed by tethering Nsp1 to the inner wall of a solid-state nanopore,with coarse-grained modeling to show that yeast Kap95 exhibits two populations in Nsp1-coated pores:one population that is transported across the pore in milliseconds,and a second population that is stably assembled within the FG mesh of the pore.Ionic current measurements show a conductance decrease for increasing Kap concentrations and noise data indicate an increase in rigidity of the FG-mesh.Modeling reveals an accumulation of Kap95 near the pore wall,yielding a conductance decrease.We find that Kaps only mildly affect the conformation of the Nsp1 mesh and that,even at high concentrations,Kaps only bind at most 8%of the FG-motifs in the nanopore,indicating that Kap95 occupancy is limited by steric constraints rather than by depletion of available FG-motifs.Our data provide an alternative explanation of the origin of bimodal NPC binding of Kaps,where a stable population of Kaps binds avidly to the NPC periphery,while fast transport proceeds via a central FG-rich channel through lower affinity interactions between Kaps and the cohesive domains of Nsp1. | Alessio Fragasso Hendrik W.de Vries John Andersson Eli Ovan der Sluis Erik van der Giessen Patrick R.Onck Cees Dekker | 2022 | Nano Research2022,15,11: | 0 |
