| 1 | Biosorption mechanisms involved in immobilization of soil Pb by Bacillus subtilis DBM in a multi-metal-contaminated soil显示文摘Mechanisms of soil Pb immobilization by Bacillus subtilis DBM, a bacterial strain isolated from a heavy-metal-contaminated soil, were investigated. Adsorption and desorption experiments with living bacterial cells as well as dead cells revealed that both extracellular adsorption and intracellular accumulation were involved in the Pb2+removal from the liquid phase. Of the sequestered Pb(II), 8.5% was held by physical entrapment within the cell wall, 43.3% was held by ion-exchange, 9.7% was complexed with cell surface functional groups or precipitated on the cell surface, and 38.5% was intracellularly accumulated.Complexation of Pb2+with carboxyl, hydroxyl, carbonyl, amido, and phosphate groups was demonstrated by Fourier transform infrared spectroscopic analysis. Precipitates of Pb5(PO4)3OH, Pb5(PO4)3Cl and Pb10(PO4)6(OH)2that formed on the cell surface during the biosorption process were identified by X-ray diffraction analysis. Transmission electron microscopy–energy dispersive spectroscopic analysis confirmed the presence of the Pb(II)precipitates and that Pb(II) could be sequestered both extracellularly and intracellularly.Incubation with B. subtilis DBM significantly decreased the amount of the weak-acid-soluble Pb fraction in a heavy-metal-contaminated soil, resulting in a reduction in Pb bioavailability, but increased the amount of its organic-matter-bound fraction by 71%. | 2 | Major advances in studies of the physical geography and living environment of China during the past 70 years and future prospects显示文摘The natural environment provides material essentials for human survival and development. The characteristics,processes, regional differentiation and forcing mechanisms of the elements of the natural environment(e.g. geomorphology,climate, hydrology, soil, etc.) are the main objects of research in physical geography. China has a complex natural environment and huge regional differentiation and therefore it provides outstanding reserach opportunities in physical geography. This review summarizes the most important developments and the main contributions of research in the physical geography and human living environment in China during the past 70 years. The major topics addressed are the uplift of the Tibetan Plateau and the evolution of its cryosphere, the development of fluvial systems, the acidification of the vast arid region of the Asian interior, variations in the monsoon and westerly climate systems on multiple timescales, the development of lakes and wetlands, the watershed system model, soil erosion, past human-environment interactions, biogeography, and physical geographic zonality. After briefly introducing international research developments, we review the history of research in physical geography in China, focusing on the major achievements and major academic debates, and finally we summarize the status of current research and the future prospects. | 5 | An improved Landsat Image Mosaic of Antarctica显示文摘A revised Landsat Image Mosaic of Antarctica (LIMA) is presented, using the 1073 multi-band scenes of the original Landsat-7 ETM+ LIMA image collection available at the United States Geological Survey (USGS: http://gffzza64998f73d704ee0hv06vb9koob9565kn.ffgz.tsg.suse.edu.cn/). Three improvements have been applied during the data processing: (1) DN saturation is adjusted by adopting a linear regression, which has a lower root mean square error than the ratio regression used by LIMA; (2) solar elevation angle is calculated using pixel-level latitude/longitude and the acquisition time and date of the central pixel of the scene, improving slightly upon the bilinear interpolation of the solar elevation angles of scene corners applied in LIMA; and (3) two additional image bands, Band 5 and Band 7, are sharpened using the panchromatic band (Band 8) and a Gram-Schmidt Spectral Sharpening algorithm to more easily distinguish snow, cloud and exposed rocks. The final planetary reflectance product is stored in 16-bit bands to preserve the full radiometric content of the scenes. | 6 | Coordinated transcription of ANRIL and P16 genes is silenced by P16 DNA methylation显示文摘Objective: To investigate the relationship between the transcription of ANRIL, P15, P14 and P16 at the same locus and the regulation mechanism of ANRIL.Methods: Publicly available database of Cancer Cell Line Encyclopedia(CCLE) was used in bioinformatic analyses. Methylation of Cp G islands was detected by denaturing high performance liquid chromatography(DHPLC). Gene transcript levels were determined using quantitative real-time polymerase chain reaction(q RTPCR) assays. An engineered P16-specific transcription factor and DNA methyltransferase were used to induce P16-specific DNA demethylation and methylation.Results: The expression level of ANRIL was positively and significantly correlated with that of P16 but not with that of P15 in the CCLE database. This was confirmed in human cell lines and patient colon tissue samples. In addition, ANRIL was significantly upregulated in colon cancer tissues. Transcription of ANRIL and P16 was observed only in cell lines in which the P16 alleles were unmethylated and not in cell lines with fully methylated P16 alleles.Notably, P16-specific methylation significantly decreased transcription of P16 and ANRIL in BGC823 and GES1 cells. The synthesis parameters, such as the reaction temperature, the first aging temperature and the second aging temperature, were investigated. The textural properties of γ-Al2O3 were characterized by means of N2 adsorption-desorption isotherms, X-ray powder diffractometry(XRD), scanning electron microscopy(SEM), Fourier transform infrared(FTIR) spectroscopy and thermogravimetry(TG). The experimental results indicated that AACH and amorphous AlOOH were the precursors of alumina, which were formed via precipitation from solutions after reaction of aluminum sulphate with ammonium hydrogen carbonate. The precursor nanocrystallites grew and re-assembled during the secondary reforming process, which resulted in an increased pore size and pore volume and a decreased bulk density.
