| 1 | Flavonoid profiling of a traditional Chinese medicine formula of Huangqin Tang using high performance liquid chromatography显示文摘The quality control processes for herbal medicines have been problematic. Flavonoids are the major active components of Huangqin Tang(HQT, a traditional Chinese medicine formula). In this study, we used a combinative method approach consisting of chromatographic fingerprinting(high performance liquid chromatography; HPLC), quantitative methods and a pharmacodynamic evaluation model to analyze the flavonoids of HQT obtained from different sources. Ten batches of HQT were analyzed by the HPLC fingerprinting method and 26 common peaks were detected, of which 23 peaks corresponded with the chemical profile of HQT. In addition, 11 major compounds were identified by LC–MS analysis(liquid chromatography–tandem mass spectrometer; LC–MS^n) and quantified by the HPLC quantitative method approach. The studied10 batches of HQT were found to be homogeneous in their composition with a similarity between 0.990 and1.000. The distribution of the 11 identified compounds was found to be very similar among the batches. Only slight pharmacodynamic differences were detected between the different batches, confirming the homogeneity of HQT. | 2 | Effects of dietary Bacillus amyloliquefaciens supplementation on growth performance,intestinal morphology, inflammatory response, and microbiota of intra-uterine growth retarded weanling piglets显示文摘Background: The focus of recent research has been directed toward the probiotic potential of Bacillus amyloliquefaciens(BA) on the gut health of animals. However, little is known about BA's effects on piglets with intra-uterine growth retardation(IUGR). Therefore, this study investigated the effects of BA supplementation on the growth performance,intestinal morphology, inflammatory response, and microbiota of IUGR piglets.Methods: Eighteen litters of newborn piglets were selected at birth, with one normal birth weight(NBW) and two IUGR piglets in each litter(i.e., 18 NBW and 36 IUGR piglets in total). At weaning, the NBW piglet and one of the IUGR piglets were assigned to groups fed a control diet(i.e., the NBW-CON and IUGR-CON groups). The other IUGR piglet was assigned to a group fed the control diet supplemented with 2.0 g BA per kg of diet(i.e., IUGR-BA group). The piglets were thus distributed across three groups for a four-week period.Results: IUGR reduced the growth performance of the IUGR-CON piglets compared with the NBW-CON piglets. It was also associated with decreased vil us sizes, increased apoptosis rates, reduced goblet cel numbers, and an imbalance between pro-and anti-inflammatory cytokines in the smal intestine. Supplementation with BA improved the average daily weight gain and the feed efficiency of the IUGR-BA group compared with the IUGR-CON group(P < 0.05). The IUGR-BA group exhibited increases in the ratio of jejunal vil us height to crypt depth, in ileal vil us height, and in ileal goblet cel density. They also exhibited decreases in the numbers of jejunal and ileal apoptotic cel s and ileal proliferative cel s(P < 0.05). Supplementation with BA increased interleukin 10 content, but it decreased tumor necrosis factor alpha level in the smal intestines of the IUGR-BA piglets(P < 0.05). | 5 | Genetic and biological characteristics of the globally circulating H5N8 avian influenza viruses and the protective efficacy offered by the poultry vaccine currently used in China显示文摘The H5N8 avian influenza viruses have been widely circulating in wild birds and are responsible for the loss of over 33 million domestic poultry in Europe, Russia, Middle East, and Asia since January 2020. To monitor the invasion and spread of the H5N8 virus in China, we performed active surveillance by analyzing 317 wild bird samples and swab samples collected from 41,172 poultry all over the country. We isolated 22 H5N8 viruses from wild birds and 14 H5N8 viruses from waterfowls. Genetic analysis indicated that the 36 viruses formed two different genotypes: one genotype viruses were widely detected from different wild birds and domestic waterfowls;the other genotype was isolated from a whopper swan. We further revealed the origin and spatiotemporal spread of these two distinct H5N8 virus genotypes in 2020 and 2021. Animal studies indicated that the H5N8 isolates are highly pathogenic to chickens, mildly pathogenic in ducks, but have distinct pathotypes in mice. Moreover, we found that vaccinated poultry in China could be completely protected against H5N8 virus challenge. | 6 | Phase-field simulation for the evolution of solid/liquid interface front in directional solidification process显示文摘In this study, the phase field method was used to study the multi-controlling factors of dendrite growth in directional solidification. The effects of temperature gradient, propelling velocity, thermal disturbance and growth orientation angle on the growth morphology of the dendritic growth in the solid/liquid interface were discussed. It is found that the redistribution of solute leads to multilevel cavity and multilevel fusion to form multistage solute segregation, and the increase of temperature gradient and propelling velocity can accelerate the dendrite growth of directional solidification, and also make the second dendrites more developed, which reduces the primary distance and the solute segregation. When the temperature gradient is large, the solid-liquid interface will move forward in a flat interface mode,and the thermal disturbance does not affect the steady state behavior of the directionally solidified dendrite tip. It only promotes the generation and growth of the second dendrites and forms the asymmetric dendrite. Meanwhile, it is found that the inclined dendrite is at a disadvantage in the competitive growth compared to the normal dendrite, and generally it will disappear. First, the wavelet transform is adopted to decompose the image of refuge chamber, of which low frequency component remains unchanged. Then, three high-frequency components are treated by bilateral filtering, and the image is reconstructed. The result shows that the combination of bilateral filtering and wavelet transform for image denoising can better retain the details which are included in the image, while providing better visual effect. This is superior to using either bilateral filtering or wavelet transform alone. | 11 | The FGOALS climate system model as a modeling tool for supporting climate sciences:An overview显示文摘Climate system models are useful tools for understanding the interactions among the components of the climate system and predicting/projecting future climate change. The development of climate models has been a central focus of the State Key Laboratory of Numerical Modeling for Atmospheric Sciences and Geophysical Fluid Dynamics, Institute of Atmospheric Physics, Chinese Academy of Sciences(LASG/IAP) since the establishment of the laboratory in 1985. In China, many pioneering component models and fully coupled models of the climate system have been developed by LASG/IAP. The fully coupled climate system developed in the recent decade is named FGOALS(Flexible Global Ocean-Atmosphere-Land System Model). In this paper, an application-oriented review of the LASG/IAP FGOALS model is presented. The improved model performances are demonstrated in the context of cloud-radiation processes, Asian monsoon, ENSO phenomena, Atlantic Meridional Overturning Circulation(AMOC) and sea ice. The FGOALS model has contributed to both CMIP5(Coupled Model Intercomparison Project-phase 5) and IPCC(Intergovernmental Panel on Climate Change) AR5(the Fifth Assessment Report). The release of FGOALS data has supported the publication of nearly 500 papers around the world. The results of FGOALS are cited ~106 times in the IPCC WG1(Working Group 1) AR5. In addition to the traditional long-term simulations and projections, near-term decadal climate prediction is a new set of CMIP experiment, progress of LAGS/IAP in the development of nearterm decadal prediction system is reviewed. The FGOALS model has supported many Chinese national-level research projects and contributed to the national climate change assessment report. | 12 | Benefit of everolimus as a monotherapy for a refractory breast cancer patient bearing multiple genetic mutations in the PI3K/AKT/mTOR signaling pathway显示文摘A postmenopausal patient with a diagnosis of estrogen receptor(ER)(+), progesterone receptor(PR)(+), and human epidermal growth factor receptor-2(HER2)(-) breast cancer was reported. The patient refused surgery and was resistant to conventional chemotherapy regimens. Computed tomography and the circulating tumor cell test indicated that the patient's tumor burden increased rapidly even after several chemotherapy sessions. Multiple genetic aberrances in the phosphatidylinositol3-kinases(PI3 K) signaling pathway were detected via next-generation sequencing(NGS)-based liquid biopsy, including a p. G1007 R missense mutation in exon 21 of PIK3 CA(33.61%), a p.L70 fs frameshift mutation in exon 3 of phosphatase and tension homolog deleted on chromosome ten(PTEN)(49.14%), and a p. D1542 Y missense mutation in exon 32 of mammalian target of rapamycin(m TOR)(1.66%). Therefore, only the m TOR inhibitor everolimus was administered to the patient. Partial remission(PR) was observed after 2 months, and sustained stable disease(SD) was observed after a year and a half. Subsequent sequencing showed that the mutation ratio of PIK3 CA decreased to 4.17%, and that the PTEN and m TOR mutations disappeared, which revealed the significant curative effect of everolimus. We report the first case of successful monotherapy treatment using everolimus in a patient with advanced breast cancer bearing mutations in genes involved in the PI3 K/ARK/m TOR signaling pathway. The experiments were conducted to study the dynamic distribution characteristics of suspended particles under different hydrodynamic conditions. The results show the self-similarity and scale invariance of particle size distribution. The study further proposed the concept of fractal dimension of particle size distribution and found out that fractal dimension changed in a similar way as residual turbidity did and could excellently indicate the variation of coagulation effect. | 16 | Efficacy of acupuncture therapy for improving anorexia in tumor patients:a Meta-analysis显示文摘OBJECTIVE:To use evidence-based medicine to systematically evaluate the effectiveness and safety of acupuncture therapy for improving anorexia in tumor patients.METHODS:We queried the China National Knowledge Infrastructure Database(CNKI),China Science and Technology Journal Database(VIP),Wanfang Data,Pub Med,Cochrane Library,and Embase databases to identify reports of randomized controlled trials(RCTs)that applied acupuncture therapy to improve anorexia in tumor patients,and used Rev Man 5.3 software to conduct a Meta-analysis of the effective rate,appetite score,Karnofsky Performance Status(KPS)score,Functional Assessment of Anorexia/Cachexia Therapy(FAACT)appetite scale,and body weight in each study.Subgroup analysis was conducted based on whether radiotherapy or chemotherapy were also administered.RESULTS:A total of 10 RCTs were included with a total of 648 patients,including 343 patients in the treatment group and 305 patients in the control group.The Meta-analysis results showed that the clinical efficacy,appetite score,KPS score,and FAACT score of the treatment group(which received acupuncture to improve appetite)were better than those of the control group,and the difference was statistically significant(P<0.05);however,there was no statistically significant difference in body weight between the treatment group and the control group(P>0.05).The results of the subgroup analysis showed that the effective rate and appetite score for patients with long-term and chronic loss of appetite who underwent acupuncture were better than those of the control group,and the difference was statistically significant(P<0.05).CONCLUSIONS:Acupuncture therapy has good efficacy and safety in the treatment of anorexia in tumor patients,and it also has good efficacy and safety for long-term and chronic loss of appetite.The reliability and stability of the above results need to be confirmed by high-quality RCTs with larger sample sizes. | ZHAO Weipeng LI Jing ZHANG Yushuang LI He HUANG Jinchang BAI Jing LI Jianbo | 2021 | Journal of Traditional Chinese Medicine2021,41,4: | 4 |
| 17 | Adaptive cavity-enhanced dual-comb spectroscopy显示文摘Resolution and bandwidth are critical for cavity-enhanced dual-comb spectroscopy(CE-DCS). Here, we pioneer an adaptive approach in CE-DCS to improve the broadband as well as the resolution. Postcorrections to dualcomb interferograms adaptively compensate the relative phase jitters of the optical frequency combs and result in both a mode-resolved spectral resolution and a signal-to-noise ratio of 440:1 in 1 s. Meanwhile, an adaptive combcavity locking scheme exploits more than 90% of the comb modes, covering 340 cm-1(10 THz) at 6450 cm-1. For a single dual-comb interferogram, more than 40,000 comb teeth spaced by 250 MHz are measured in less than7.5 ms, contributing to a noise equivalent absorption per spectral element of 2 × 10-10 cm-1·Hz-1∕2. | 18 | Simulation of sea surface temperature changes in the Middle Pliocene warm period and comparison with reconstructions显示文摘The Middle Pliocene (ca 3.12–2.97 Ma) is a recent warm period in the Earth’s history. In many respects, the warmth of the Middle Pliocene is similar to the probable warm situation of the late 21st century predicted by climate models. Understanding the Middle Pliocene climate is important in predicting the future climate with global warming. Here, we used the latest reconstructions for the Middle Pliocene—Pliocene Research Interpretation and Synoptic Mapping (PRISM) version 3—to simulate the Middle Pliocene climate with a fully coupled model Fast Ocean Atmosphere Model. From comparison of the results of simulations with reconstructions, we considered two important scientific topics of Middle Pliocene climate modeling: extreme warming in the subpolar North Atlantic and a permanent El Ni?o in the tropical Pacific. Our simulations illustrate that the global annual mean sea surface temperature (SST) in the Middle Pliocene was about 2.3°C higher than that in the pre-industrial era. The warming was stronger at midand high latitudes than at low latitudes. The simulated SST changes agree with SST reconstructions in PRISM3 data, especially for the North Atlantic, North Pacific and west coast of South America. However, there were still discrepancies between the simulation of the SST and reconstructions for the subpolar North Atlantic and tropical Pacific. In the case of the Atlantic, the weakened meridional overturning circulation in the simulation did not support the reconstruction of the extremely warm condition in the subpolar North Atlantic. In the case of the tropical Pacific, the whole ocean warmed, especially the eastern tropical Pacific, which did not support the permanent El Ni?o suggested by the reconstruction. From evaluation of the modeling and reconstruction, we suggest that the above discrepancies were due to uncertainties in reconstructions, difficulties in paleoclimate modeling and deficiencies of climate models.
