| 1 | Preliminary investigation of seismic damage to two steel space structures during the 2013 Lushan earthquake显示文摘Severe damage to steel space structures is rarely reported when compared to other structural systems damaged during past major earthquakes around the world.Two gymnasiums of steel space structures in downtown Lushan County that were damaged during the 2013 M7.0 Lushan earthquake in China were investigated and the observations are summarized in this paper.Typical damage to these two steel space structures ranges from moderate to severe.Moderate damage includes global buckling and dislocation of bolted connections of truss members,and inelastic elongation of anchor bolts and sliding of pedestal plates of supports.Severe damage includes member fracture caused by local buckling,and fracture failure of anchor bolts and welds.The distribution of structural damage to these two structures is described in detail and future research opportunities are suggested. | Dai Junwu Qu Zhe Zhang Chenxiao Weng Xuran | 2013 | Earthquake Engineering and Engineering Vibration2013,12,3: | 11 |

| 3 | Effect of herb-partitioned moxibustion on dopamine levels and dopamine receptor 1 expression in the colon and central nervous system in rats with Crohn’s disease显示文摘OBJECTIVE:To observe the effect of herb-partitioned moxibustion at the Tianshu (ST 25) and Qihai (CV 6) acupoints in rats with Crohn's disease,and explore the underlying mechanism from dopamine (DA) and dopamine receptor 1 (D1 R) in the colon,spinal dorsal horn and hypothalamus.METHODS:The rats were randomly divided into the normal,model (CD),herb-partitioned moxibustion (Mox) and mesalazine (Mesa) groups.Damage in the colons was scored and observed by hematoxylin and eosin staining.DA and D1R protein expression in the colonic mucosa were detected by immunohistochemistry.The concentrations of DA and D1R in the spinal dorsal horn and hypothalamus were measured by enzyme-linked immunosorbent assay,and D1R mRNA expression was evaluated by quantitative real-time polymerase chain reaction.RESULTS:In the colon,compared with the normal group,DA,D1 R protein expressions and D1 R mRNA expression were significantly higher in the model group,while decreased in the Mox group and the Mesa group.In the spinal dorsal horn and hypothalamus,compared with the normal group,the concentrations of DA and D1 R,and the D1R mRNA expressions were significantly higher in the model group,and decreased in the Mox group and the Mesa group.CONCLUSION:Herb-partitioned moxibustion at the Tianshu (ST 25) and Qihai (CV 6) acupoints relieved ulceration in CD rats,the underlying mechanism maybe relative with the regulation of DA and D1R in the colon,spinal dorsal horn and hypothalamus by moxibustion. | Lu Yuan Zheng Handan Lü Tingting Ma Zhe Wu Huangan Xu Manwen Weng Zhijun Feng Hui Zhang Fang Ding Guanghong Wu Luyi Liu Huirong | 2019 | Journal of Traditional Chinese Medicine2019,39,3: | 3 |
| 4 | Application of imputation methods to genomic selection in Chinese Holstein cattle显示文摘Missing genotypes are a common feature of high density SNP datasets obtained using SNP chip technology and this is likely to decrease the accuracy of genomic selection. This problem can be circumvented by imputing the missing genotypes with estimated genotypes. When implementing imputation, the criteria used for SNP data quality control and whether to perform imputation before or after data quality control need to consider. In this paper, we compared six strategies of imputation and quality control using different imputation methods, different quality control criteria and by changing the order of imputation and quality control, against a real dataset of milk production traits in Chinese Holstein cattle. The results demonstrated that, no matter what imputation method and quality control criteria were used, strategies with imputation before quality control performed better than strategies with imputation after quality control in terms of accuracy of genomic selection. The different imputation methods and quality control criteria did not significantly influence the accuracy of genomic selection. | 5 | Repurposing econazole as a pharmacological autophagy inhibitor to treat pancreatic ductal adenocarcinoma显示文摘Pancreatic ductal adenocarcinoma(PDAC)is characterized by the highest mortality among carcinomas.The pathogenesis of PDAC requires elevated autophagy,inhibition of which using hydroxychloroquine has shown promise.However,current realization is impeded by its suboptimal use and unpredictable toxicity.Attempts to identify novel autophagy-modulating agents from already approved drugs offer a rapid and accessible approach.Here,using a patient-derived organoid model,we performed a comparative analysis of therapeutic responses among various antimalarial/fungal/parasitic/viral agents,through which econazole(ECON),an antifungal compound,emerged as the top candidate.Further testing in cell-line and xenograft models of PDAC validated this activity,which occurred as a direct consequence of dysfunctional autophagy.More specifically,ECON boosted autophagy initiation but blocked lysosome biogenesis.RNA sequencing analysis revealed that this autophagic induction was largely attributed to the altered expression of activation transcription factor 3(ATF3).Increased nuclear import of ATF3 and its transcriptional repression of inhibitor of differentiation-1(ID-1)led to inactivation of the AKT/mammalian target of rapamycin(m TOR)pathway,thus giving rise to autophagosome accumulation in PDAC cells.The magnitude of the increase in autophagosomes was sufficient to elicit ER stress-mediated apoptosis.Furthermore,ECON,as an autophagy inhibitor,exhibited synergistic effects with trametinib on PDAC.This study provides direct preclinical and experimental evidence for the therapeutic efficacy of ECON in PDAC treatment and reveals a mechanism whereby ECON inhibits PDAC growth. | Ningna Weng Siyuan Qin Jiayang Liu Xing Huang Jingwen Jiang Li Zhou Zhe Zhang Na Xie Kui Wang Ping Jin Maochao Luo Liyuan Peng Edouard C.Nice Ajay Goel Suxia Han Canhua Huang Qing Zhu | 2022 | Acta Pharmaceutica Sinica B2022,12,7: | 2 |

| 8 | Somatic mutation contributing to clonal haematopoiesis is a risk factor of recurrent stroke in first-ever acute ischaemic stroke:a prospective cohort study显示文摘Background Somatic mutation contributes to clonal haematopoiesis of indeterminate potential(CHIP)is related to age and associated with a higher risk of stroke and atherosclerotic cardiovascular disease.Here,we investigated the prognostic significance of CHIP in a large first-ever acute ischaemic stroke(AIS)cohort and explored the underlying mechanisms.Methods We studied a prospective cohort of 6016 patients who had a first-ever AIS in China.Whole-genome sequencing was performed to identify CHIP.High-sensitivity C reactive protein(hs-CRP)levels above 3 mg/L at baseline were defined as hyperinflammation.Recurrent stroke during the 3-month follow-up was the primary outcome.Results Among the 6016 patients who had a first-ever AIS,with a median age was 62 years(IQR,54.0‒70.0),3.70%were identified as CHIP carriers.The most common mutations occurred in the DNMT3A(30.0%)and TET2(11.4%)genes.During a follow-up of 3 months,the presence of CHIP was associated with recurrent stroke(HR 1.62,95%CI 1.04 to 2.51,p=0.03),recurrent ischaemic stroke(HR 1.64,95%CI 1.04 to 2.58,p=0.03)and combined vascular events(HR 1.58,95%CI 1.02 to 2.44,p=0.04)after adjusting for hsCRP levels at baseline in patients who had a first-ever AIS.Subgroup analysis demonstrated that CHIP was only associated with recurrent stroke when patients under hyperinflammation(OR 3.10,95%CI 1.92 to 5.00,p<0.001)but not in those without hyperinflammation(OR 0.18,95%CI 0.03 to 1.04,p=0.06,Pinteraction=0.002).Conclusion Our results suggest that somatic mutations contributing to CHIP increase the risk of short-term recurrent stroke in patients who had a first-ever AIS.Hyperinflammation may be important in the relationship between CHIP and recurrent stroke. | Xin Qiu Yalun Dai Si Cheng Hong-Qiu Gu Yong Jiang Xia Meng Yilong Wang Xingquan Zhao Yingyu Jiang Zhe Xu Xinying Huang Meng Wang Tian Jie Lyu Yubo Wang Jiaxu Weng Lingyun Cui Yi Shangguan Hao Li Yongjun Wang Zixiao Li | 2023 | Stroke & Vascular Neurology2023,8,2: | 1 |
