1Resveratrol provides neuroprotection by regulating the JAK2/STAT3/PI3K/AKT/mTOR pathway after stroke in rats显示文摘Ischemic stroke is a common disease with high mortality and morbidity worldwide.One of the important pathophysiological effects of ischemic stroke is apoptosis.A neuroprotective effect is defined as the inhibition of neuronal apoptosis to rescue or delay the infarction in the surviving ischemic penumbra.Resveratrol is a natural polyphenol that reportedly prevents cerebral ischemia injury by regulating the expression of PI3K/AKT/mTOR.Therefore,this study aimed to elucidate the neuroprotective effect of resveratrol on cerebral ischemia/reperfusion injury and to investigate the signaling pathways and mechanisms through which resveratrol regulates apoptosis in the ischemic penumbra.Rats were subjected to middle cerebral artery occlusion for 2 h followed by 24 h reperfusion.Cerebral infarct volume was measured using 2%TTC staining.TUNEL staining was conducted to evaluate neuronal apoptosis.Western blotting and immunohistochemistry were used to detect the proteins involved in the JAK2/STAT3/PI3K/AKT/mTOR pathway.The results suggested that resveratrol significantly improved neurological function,reduced cerebral infarct volume,decreased neuronal damage,and markedly attenuated neuronal apoptosis;these effects were attenuated by the inhibition of PI3K/AKT with LY294002 and JAK2/STAT3 with AG490.We also found that resveratrol significantly upregulated the expression of p-JAK2,p-STAT3,p-AKT,p-mTOR,and BCL-2 and downregulated expression of cleaved caspase-3 and BAX,which was partially reversed by LY294002 and AG490.These results suggested that resveratrol provides a neuroprotective effect against cerebral ischemia/reperfusion injury,which is partially mediated by the activation of JAK2/STAT3 and PI3K/AKT/mTOR.Resveratrol may indirectly upregulate the PI3K/AKT/mTOR pathway by activating JAK2/STAT3.Yongying Hou Ke Wang Weijun Wan Yue Cheng Xia Pu Xiufeng Ye 2018Genes & Diseases2018,5,3:60
2NLRP3-dependent pyroptosis is required for HIV-1 gp120- induced neuropathology显示文摘The human immunodeficiency virus-1(HIV-1)envelope protein gp120 is the major contributor to the pathogenesis of HIVassociated neurocognitive disorder(HAND).Neuroinflammation plays a pivotal role in gp120-induced neuropathology,but how gp120 triggers neuroinflammatory processes and subsequent neuronal death remains unknown.Here,we provide evidence that NLRP3 is required for gp120-induced neuroinflammation and neuropathy.Our results showed that gp120-induced NLRP3-dependent pyroptosis and IL-1βproduction in microglia.Inhibition of microglial NLRP3 inflammasome activation alleviated gp120-mediated neuroinflammatory factor release and neuronal injury.Importantly,we showed that chronic administration of MCC950,a novel selective NLRP3 inhibitor,to gp120 transgenic mice not only attenuated neuroinflammation and neuronal death but also promoted neuronal regeneration and restored the impaired neurocognitive function.In conclusion,our data revealed that the NLRP3 inflammasome is important for gp120-induced neuroinflammation and neuropathology and suggest that NLRP3 is a potential novel target for the treatment of HAND.Xiaolong He Weijun Yang Zhijie Zeng Yi Wei Jie Gao Bao Zhang Li Li Liqun Liu Yu Wan Qing Zeng Zelong Gong Liting Liu Hanyun Zhang Yubin Li Shaojie Yang Tongtong Hu Lixian Wu Eliezer Masliah Shenghe Huang Hong Cao 2020Cellular & Molecular Immunology2020,17,3:11
3Fluid–solid coupling analysis of rock pillar stability for concealed karst cave ahead of a roadway based on catastrophic theory显示文摘In order to study the mechanism of water inrush from a concealed, confined karst cave, we established a fluid–solid coupling model of water inrush from a concealed karst cave ahead of a roadway and a strength reduction method in a rock pillar for preventing water inrush based on catastrophic theory. Fluid–solid coupling effects and safety margins in a rock pillar were studied. Analysis shows that rock pillar instability, exerted by disturbance stress and seepage stress, is the process of rock pillar catastrophic destabilization induced by nonlinear extension of plastic zones in the rock pillar. Seepage flow emerges in the rock pillar for preventing water inrush, accompanied by mechanical instability of the rock pillar. Taking the accident of a confined karst cave water-inrush of Qiyi Mine as an example, by studying the safety factor of the rock pillar and the relationship between karst cave water pressure and thickness of the rock pillar,it is proposed that rock pillar thickness with a safety factor equal to 1.5 is regarded as the calculated safety thickness of the rock pillar, which should be equal to the sum of the blasthole depth, blasting disturbance depth and the calculated safety thickness of the rock pillar. The cause of the karst water inrush at Qiyi Mine is that the rock pillar was so small that it did not possess a safety margin. 6Alterations of endotoxin distribution across different biofluids and relevant inflammatory responses by supplementing L-theanine in dairy cows during heat stress显示文摘The present trial was performed to reveal the regulatory effects of L-theanine on the levels of lipopolysaccharide(LPS)endotoxin within different biofluids,as well as relevant inflammatory responses of dairy cattle under heat stress conditions.Thirty lactating Chinese Holstein dairy cattle(189±47 d in milk,and 2±1 parities)were allocated in a completely randomized design to each of 3 dietary treatments:the control(CON,0 g/d per cow L-theanine),the low L-theanine dosage treatment(LL,16 g/d per cow L-theanine),and the high L-theanine dosage treatment(HL,32 g/d per cow L-theanine).This trial consisted of 38 d(7 d for adaption and 31 d for data and sample collection),and sample collection for rumen liquid,blood plasma or serum,and milk were conducted on the d 27 and 38,respectively.Dairy cattle were constantly exposed to environmental heat stress during this experiment according to the recorded temperature-humidity index(THI).In the LL treatment,LPS concentration in rumen liquid was higher(P<0.05),whilst LPS densities in plasma and milk were lower(P<0.05)than those of the CON.Supplementing L-theanine at 2 dosages both significantly lowered(P<0.05)the level of interleukin(IL)-1βin the serum.Results of the present study suggested that L-theanine could be a promising additive in reducing the detrimental effects of heat stress on dairy cows,and L-theanine supplementation at 16 g/d per cow is preferred because it reduced the LPS translocation into the peripheral blood and LPS accumulation in the milk,as well as mitigated LPS-induced inflammatory reactions in dairy cows during heat stress.Further studies are necessitated to investigate the underlying mechanisms of L-theanine in LPS alteration and inflammation alleviation.Zuo Wang Lingmei Zhang Zhicai Li Yuannian Yu Lingyuan Yang Peihua Zhang Weijun Shen Fachun Wan Jianhua He Wenjun Xiao Dasheng Yang Zhiliang Tan Shaoxun Tang 2021Animal Nutrition2021,,4:1
7On the Consistency of Large Earthquake Moment and Strain Rate Inferred from GPS Data in North China显示文摘The new GPS data can map crustal strain rates over large areas with a useful degree of precision. Stable strain measurement results open the door for improved estimates of earthquake occurrence. The Kostrov’s formula (1974) translates the smoothed strain rates in North China into geodetic moment rates. In North China, the ratio of seismic moment released to moment accumulated from GPS measurement is 60.6% in NS direction, 68.9% in EW direction, and 104.1% in NE shear direction. The near unit ratio points to the reliability of GPS measurements there. 8Co-seismic Slip Distribution of the 2001 Kokoxili Earthquake Inverted by the GPS Data显示文摘The authors analyze co_seismic displacement field derived from the Global Position System (GPS) observations collected before and after the 2001 Kokoxili earthquake, western China. Using the co_seismic displacement data, and constrained with surface rupture data, they invert co_seismic slip distribution along the seismic fault. Their result shows that the earthquake ruptured the upper crust down to a depth of 13.1~22km (at 70% certainty), with its optimal estimate at 16.5km. A 2~3m left_lateral strike slip is resolved between the Sun Lake segment and the west end of the main rupture zone, although surface rupture has not been observed there. The surface rupture of this earthquake is ended at the Sun Lake to the west, but left_lateral slip of 1.5~2.0m seems to exist beyond the east end of surface rupture observed from field geology.
