1A Chromosome-Scale Genome Assembly of Paper Mulberry(Bmussonetia papyrifera)Provides New Insights into Its Forage and Papermaking Usage显示文摘Paper mulberry(Broussonetia papyrifera)is a well-known woody tree historically used for Cai Lun papermaking,one of the four great inventions of ancient China.More recently,Paper mulberry has also been used as forage to address the shortage of feedstuff because of its digestible crude fiber and high protein contents.In this study,we obtained a chromosome-scale genome assembly for Paper mulberry using integrated approaches,including Illumina and PacBio sequencing platform as well as Hi-C,optical,and genetic maps.The assembled Paper mulberry genome consists of 386.83 Mb,which is close to the estimated size,and 99.25%(383.93 Mb)of the assembly was assigned to 13 pseudochromosomes.Comparative genomic analysis revealed the expansion and contraction in the flavonoid and lignin biosynthetic gene families,respectively,accounting for the enhanced flavonoid and decreased lignin biosynthesis in Paper mulberry.Moreover,the increased ratio of syringyl-lignin to guaiacyl-lignin in Paper mulberry underscores its suitability for use in medicine,forage,papermaking,and barkcloth making.We also identified the rootassociated microbiota of Paper mulberry and found that Pseudomonas and Rhizobia were enriched in its roots and may provide the source of nitrogen for its stems and leaves via symbiotic nitrogen fixation.Collectively,these results suggest that Paper mulberry might have undergone adaptive evolution and recruited nitrogen-fixing microbes to promote growth by enhancing flavonoid production and altering lignin monomer composition.Our study provides significant insights into genetic basis of the usefulness of Paper mulberry in papermaking and barkcloth making,and as forage.These insights will facilitate further domestication and selection as well as industrial utilization of Paper mulberry worldwide.Xianjun Peng Hui Liu Peilin Chen Feng Tang Yanmin Hu Fenfen Wang Zhi Pi Meiling Zhao Naizhi Chen Hui Chen Xiaokang Zhang Xueqing Yan Min Liu Xiaojun Fu Guofeng Zhao Pu Yao Lili Wang He Dai Xuming Li Wei Xiong Wencai Xu Hongkun Zheng Haiyan Yu Shihua Shen 2019Molecular Plant2019,12,5:18

4Mesenchymal stem cells and immune disorders: from basic science to clinical transition显示文摘As a promising candidate seed cell type in regenerative medicine, mesenchymal stem cells (MSCs) have attracted considerable attention. The unique capacity of MSCs to exert a regulatory effect on immunity in an autologous/allergenic manner makes them an attractive therapeutic cell type for immune disorders. In this review, we discussed the current knowledge of and advances in MSCs, including its basic biological properties, i.e., multilineage differentiation, secretome, and immunomodulation. Specifically, on the basis of our previous work, we proposed three new concepts of MSCs, i.e.,“subtotipotent stem cell” hypothesis, MSC system, and “Yin and Yang” balance of MSC regulation, which may bring new insights into our understanding of MSCs. Furthermore, we analyzed data from the Clinical Trials database (http://gffzzd3f38a9638994581hpwp9fqwnpvcx6q5v.ffgz.tsg.suse.edu.cn) on registered clinical trials using MSCs to treat a variety of immune diseases, such as graft-versus-host disease, systemic lupus erythematosus, and multiple sclerosis. 6Detection and identification of Vibrio parahaemolyticus by multiplex PCR and DNA-DNA hybridization on a microarray显示文摘In this paper,we developed a rapid and accurate method for the detection of Vibrio parahaemalyticus strains,using multiplex PCR and DNA—DNA hybridization.Multiplex PCR was used to simultaneously amplify three diagnostic genes(tlh,tdh and fla) that serve as molecular markers of V.parahaemolyticus.Biotinylated PCR products were hybridized to primers immobilized on a microarray,and detected by chemiluminesce with avidin—conjugated alkaline phosphatase.With this method,forty-five samples were tested.Eight known virulent strains (tlh^+/tdh^+/fla^+) and four known avirulent strains(tlh^+/tdh^-/fla^+) of the V.parahaemolyticus were successfully detected,and no non-specific hybridization and cross-hybridization reaction were found from fifteen closely-related strains(tlh^-/tdh^-/fla^+) of the Vibrio spp.In addition,all the other eighteen strains of non-Vibrio bacteria(tlh^-/tdh^-/fla^-) gave negative results.The DNA microarray successfully distinguished V. parahaemolyticus from other Vibrio spp.The results demonstrated that this was an efficient and robust method for identifying virulent strains of V.parahaemolyticus.Rongzhi Wang Jiadong Huang Wei Zhang Guangmei Lin Junwei Lian Libin Jiang Hongcong Lin Songfa Wang Shihua Wang 2011Journal of Genetics and Genomics2011,38,3:11
7Petrogenesis, Tectonic Evolution and Geothermal Implications of Mesozoic Granites in the Huangshadong Geothermal Field, South China显示文摘Mesozoic multi-stage tectono-magmatic events produced widely distributed granitoids in the South China Block. Huangshadong(HSD) is located in south-eastern South China Block, where closely spaced hot springs accompany outcrops of Mesozoic granites. New data on whole-rock geochemistry, zircon U-Pb geochronology, and zircon Lu-Hf isotopes are presented, to study the petrogenesis and tectonic evolution of the granites, and to explore the relationship between granites and geothermal anomalies. Zircon U-Pb isotopes display three periods of granites in the HSD area: Indosinian(ca. 253 Ma, G4) muscovite-bearing monzonitic granite, early Yanshanian(ca. 175–155 Ma, G5 and G3) monzonitic granite and granodiorite, and late Yanshanian(ca. 140 Ma, G1 and G2) biotite monzonitic granite. In petrogenetic type, granites of the three periods are I-type granite. Among them, G1, G2, G3, and G4 are characterized by high fractionation, with high values of SiO2, alkalis, Ga/Al, and Rb/Sr, and depletion in Sr, Ba, Zr, Nb, Ti, REEs, with low(La/Yb)N, Nb/Ta, and Zr/Hf ratios and negative Eu anomalies. In terms of tectonic setting, 253 Ma G4 may be the product of partial melting of the ancient lower crust under post-orogenic extensional tectonics, as the closure of the Paleo-Tethys Ocean resulted in an intracontinental orogeny. At 175 Ma, the subduction of the Pacific Plate became the dominant tectonic system, and low-angle subduction of the Paleo-Pacific Plate facilitated partial melting of the subducted oceanic crust and basement to generate the hornblende-bearing I-type granodiorite. As the dip angle of the subducting plate increased, the continental arc tectonic setting was transformed to back-arc extension, inducing intense partial melting of the lower crust at ca. 158 Ma and resulting in the most frequent granitic magmatic activity in the South China hinterland. When slab foundering occurred at ca. 140 Ma, underplating of mantle-derived magmas caused melting of the continental crust, generating extensive highly fractionated granites in HSD. Combining the granitic evolution of HSD and adjacent areas and radioactive heat production rates, it is suggested that highly fractionated granites are connected to the enrichments in U and Th with magma evolution. The high radioactive heat derived from the Yanshanian granites is an important part of the crustal heat, which contributes significantly to the terrestrial heat flow. 9Construction of multiform scFv antibodies using linker peptide显示文摘Multiform single chain variable fragments (scFvs) including different length linker scFvs and bispecific scFv were constructed. The linker lengths of 0, 3, 5, 8, 12, and 15 amino acids between VH and VL of antideoxynivalenol (anti-DON) scFv were used to analyze the affinities of scFvs. The affinity constants of these scFvs increased when the linker was lower than 12 amino acids. The affinity constant would not change when the linker was longer than 12 amino acids. Fusion gene of anti-DON scFv and antizearalenone (anti-ZEN) scFv was also constructed through connection by a short peptide linker DNA to express a bispecific scFv. The affinity constants assay showed that the two scFvs of fusion bispecific scFv remained their own affinity compared to their parental scFvs. Competitive direct enzyme linked immunosorbent assay was used to detect DON and ZEN in contaminated wheat (Triticum aestivum L.) samples, and the results indicated that this bispecific scFv was applicable in DON and ZEN detection. 10A comparative study of CuO/TiO_2-SnO_2,CuO/TiO_2 and CuO/SnO_2 catalysts for low-temperature CO oxidation显示文摘Nanometer SnO2 particles were synthesized by sol-gel dialytic processes and used as a support to prepare CuO supported catalysts via a deposition-precipitation method. The samples were characterized by means of TG-DTA,XRD,H 2-TPR and XPS. The catalytic activity of the CuO/TiO2-SnO2 catalysts was markedly depended on the loading of CuO,and the optimum CuO loading was 8 wt.%(T 100 = 80 C) . The CuO/TiO2-SnO2 catalysts exhibited much higher catalytic activity than the CuO/TiO 2 and CuO/SnO 2 catalysts. 11Differentiation of illicit drugs with THz time-domain spectroscopy显示文摘The terahertz time-domain spectroscopy(THz-TDS) was used for sensing and identifying illicit drugs.The absorption spectra of seven illicit drug samples(morphine and its hydrochloride,cocaine hydrochloride,codeine phosphate,papaverine hydrochloride,pethidine hydrochloride,and thebaine) were studied by THz-TDS at 0.3-2.0 THz at room temperature.The geometric structure and vibration frequencies of morphine were calculated by density functional theory.The four absorption features were dominated by intra-/inter-molecular collective or lattice vibration modes.Each illicit drug has a distinct signature in its THz spectra.The results indicate that the THz-TDS can be used to identify and discriminate illicit drugs by their characteristic fingerprints.LIU Guifeng MA Shihua JI Te ZHAO Hongwei WANG Wenfeng 2010Nuclear Science and Techniques2010,21,4:7

15A Diagnostic Analysis on the Effect of the Residual Layer in Convective Boundary Layer Development near Mongolia Using 20th Century Reanalysis Data显示文摘Although the residual layer has already been noted in the classical diurnal cycle of the atmospheric boundary layer,its effect on the development of the convective boundary layer has not been well studied. In this study, based on 3-hourly20 th century reanalysis data, the residual layer is considered as a common layer capping the convective boundary layer. It is identified daily by investigating the development of the convective boundary layer. The region of interest is bounded by(30°–60° N, 80°–120° E), where a residual layer deeper than 2000 m has been reported using radiosondes. The lapse rate and wind shear within the residual layer are compared with the surface sensible heat flux by investigating their climatological means, interannual variations and daily variations. The lapse rate of the residual layer and the convective boundary layer depth correspond well in their seasonal variations and climatological mean patterns. On the interannual scale, the correlation coefficient between their regional averaged(40°–50°N, 90°–110° E) variations is higher than that between the surface sensible heat flux and convective boundary layer depth. On the daily scale, the correlation between the lapse rate and the convective boundary layer depth in most months is still statistically significant during 1970–2012. 16Mesenchymal stem cell-derived small extracellular vesicles mitigate oxidative stress-induced senescence in endothelial cells via regulation of miR-146a/Src显示文摘Senescent endothelial cells (ECs) could impair the integrity of the blood vessel endothelium, leading to vascular aging and a series of diseases, such as atherosclerosis, diabetes. Preventing or mitigating EC senescence might serve as a promising therapeutic paradigm for these diseases. Recent studies showed that small extracellular vesicles (sEV) have the potential to transfer bioactive molecules into recipient cells and induce phenotypic changes. Since mesenchymal stem cells (MSCs) have long been postulated as an important source cell in regenerative medicine, herein we investigated the role and mechanism of MSC-derived sEV (MSC-sEV) on EC senescence. In vitro results showed that MSC-sEV reduced senescent biomarkers, decreased senescence-associated secretory phenotype (SASP), rescued angiogenesis, migration and other dysfunctions in senescent EC induced by oxidative stress. In the In vivo natural aging and type-2 diabetes mouse wound-healing models (both of which have senescent ECs), MSC-sEV promoted wound closure and new blood vessel formation. Mechanically, miRNA microarray showed that miR-146a was highly expressed in MSC-sEV and also upregulated in EC after MSC-sEV treatment. miR-146a inhibitors abolished the stimulatory effects of MSC-sEV on senescence. Moreover, we found miR-146a could suppress Src phosphorylation and downstream targets VE-cadherin and Caveolin-1. 17COP9 signalosome subunit PfCsnE regulates secondary metabolism and conidial formation in Pestalotiopsis fici显示文摘The COP9 signalosome(CSN) is a highly conserved multiprotein complex in all eukaryotes and involved in regulation of organism development. In filamentous fungi, several lines of evidence indicate that fungal development and secondary metabolism(SM) are mediated by the fifth subunit of CSN, called CsnE. Here we uncover a connection with CsnE and conidial formation as well as SM regulation in the plant endophytic fungus Pestalotiopsis fici. A homology search of the P. fici genome with CsnE, involved in sexual development and SM in Aspergillus nidulans, identified PfCsnE. Deletion of PfcsnE resulted in a mutant that stopped conidial production, but the conidia are recovered in a PfcsnE complemented strain. This indicates that PfCsnE is required for the formation of conidia. Secondary metabolite analysis demonstrated that the ΔPfcsnE strain produced more chloroisosulochrin, less ficiolide A production in comparison to wild type(WT). Transcriptome analysis of WT andΔPfcsnE strains indicated that PfcsnE impacts the expression levels of 8.37% of 14,797 annotated genes. Specifically, nine biosynthetic gene clusters(BGCs) were up-regulated and three BGCs were down-regulated by PfCsnE. 18Effects of GS3 and GL3.1 for Grain Size Editing by CRISPR/Cas9 in Rice显示文摘Grain size is one of key agronomic traits associated with grain yield and grain quality. Both major quantitative trait loci GS3 and GL3.1 play a predominant role in negative regulation of grain size. In this study, a CRISPR/Cas9-mediated multiplex genome editing system was used to simultaneously edit GS3 and GL3.1 in a typical japonica rice Nipponbare. In T1 generation, we found that gs3 formed slender grain with lower chalkiness percentage, while gs3gl3.1 produced larger grain with higher chalkiness percentage. In terms of other agronomic traits, flag leaf size, grain number and grain yield of both gs3 and gs3gl3.1 mutants were affected. It is noteworthy that gs3 and gs3gl3.1 mutants both led to dramatical reduction of grain number, thereby decreased grain yield. 20Short-term climatic impacts of afforestation in the East Asian monsoon region显示文摘The influence of changes in vegetation cover on short-term climate over the East Asian monsoon region is simulated using the Community Climate System Model Version 3.5.The results show the annual mean surface air temperature significantly decreases by 0.93°C in response to afforestation over the East Asian monsoon region.Also,surface air temperature decreases by 1.46 and 0.40°C in summer and winter,respectively.The cooling is caused by enhanced evapotranspiration(ET) produced by increased forest cover.Evapotranspiration is greater in summer than in winter,so summer cooling is greater than winter cooling.The annual mean precipitation increases in response to afforestation,with a maximum of 7% in April.Water vapor increases significantly because of greater latent heat flux release.Meanwhile,afforestation leads to higher surface roughness,which decreases surface wind speed and induces an ascending air motion.These factors can produce more clouds and precipitation.Moreover,the surface albedo and the reflective solar radiation are reduced in response to afforestation.MA Di LIU ZhengYu Lü ShiHua MICHAEL Notaro RONG XinYao CHENG GuangShan WANG FuYao 2013Chinese Science Bulletin2013,58,18:3
