3Identification and Characterization of a LEA-like Gene, CaMF5,Specifically Expressed in the Anthers of Male-fertile Capsicum annuum显示文摘To improve the understanding of molecular mechanisms of anther and/or pollen development in Chili pepper, in the present study, fulllength cDNA and DNA sequences of the pollen development-related gene CaMF5 were obtained from the anthers of a Capsicum annuum nuclear male-fertile line. Sequence analysis indicated that the full length of CaMF5 was 747 bp, containing a maximum opening reading frame of 447 bp.Amino acid sequence alignment and phylogenetic analysis revealed that CaMF5 shared approximately 37%–77% homology with a series of uncharacterized or hypothetical proteins and late embryogenesis abundant(LEA) proteins from other plants. However, no LEA structural domain was detected in CaMF5, which indicated that it might be a new type of LEA gene. CaMF5 was only expressed in flower buds at stages 7 and 8 and in open flowers of the male-fertile line, whereas it exhibited no expression in any examined organs of the male-sterile line. In addition, CaMF5 showed the highest transcript abundance in the anthers of the male-fertile line, with no expression being detected in any other examined organs, such as the sepals, petals, pistils, roots, stems, or leaves. 6Spatial-temporal characteristics of drought detected from meteorological data with high resolution in Shaanxi Province, China显示文摘The spatial pattern of meteorological factors cannot be accurately simulated by using observations from meteorological stations(OMS) that are distributed sparsely in complex terrain. It is expected that the spatial-temporal characteristics of drought in regions with complex terrain can be better represented by meteorological data with the high spatial-temporal resolution and accuracy. In this study, Standard Precipitation Evapotranspiration Index(SPEI) calculated with meteorological factors extracted from ITPCAS(China Meteorological Forcing Dataset produced by the Institute of Tibetan Plateau Research, Chinese Academy of Sciences) was applied to identify the spatial-temporal characteristics of drought in Shaanxi Province of China, during the period of 1979–2016. Drought areas detected by SPEI calculated with data from ITPCAS(SPEI-ITPCAS) on the seasonal scale were validated by historical drought records from the Chinese Meteorological Disaster Canon-Shaanxi, and compared with drought areas detected by SPEI calculated with data from OMS(SPEI-OMS). Drought intensity, trend and temporal ranges for mutations of SPEI-ITPCAS were analyzed by using the cumulative drought intensity(CDI) index and the Mann-Kendall test. The results indicated that drought areas detected from SPEI-ITPCAS were closer to the historical drought records than those detected from SPEI-OMS. Severe and exceptional drought events with SPEI-ITPCAS lower than –1.0 occurred most frequently in summer, followed by spring. There was a general drying trend in spring and summer in Shaanxi Province and a significant wetting trend in autumn and winter in northern Shaanxi Province. On seasonal and annual scales, the regional and temporal ranges for mutations of SPEI-ITPCAS were different and most mutations occurred before the year 1990 in most regions of Shaanxi Province. 8Claroideoglomus etunicatum improved the growth and saline-alkaline tolerance of Potentilla anserina by altering physiological and biochemical properties显示文摘To investigate the effects of arbuscular mycorrhizal(AM)fungi on the growth and saline–alkaline tolerance of Potentilla anserina L.,the seedlings were inoculated with Claroideoglomus etunicatum(W.N.Becker&Gerd.)C.Walker&A.Schüßler in pot cultivation.After 90 days of culture,saline–alkaline stress was induced with NaCl and NaHCO_(3)solution according to the main salt components in saline–alkaline soils.Based on the physiological response of P.anserina to the stress in the preliminary experiment,the solution concentrations of 0 mmol/L,75 mmol/L,150 mmol/L,225 mmol/L and 300 mmol/L were treated with stress for 10 days,respectively.The mycorrhizal colonization rate,mycorrhizal dependence,chlorophyll content,malondialdehyde content,antioxidant enzyme activities,osmoregulation substances content and water status were measured.The results showed that with the increase of NaCl and NaHCO_(3)stress concentration,mycorrhizal colonization rate,colonization intensity,arbuscular abundance and vesicle abundance decreased,and reached the lowest value at 300 mmol/L.Strong mycorrhizal dependence was observed after the symbiosis with AM fungus,and the dependence was higher under NaHCO_(3)treatment.Under NaCl and NaHCO_(3)stress,inoculation with AM fungus could increase chlorophyll content,decrease malondialdehyde content,increase activities of superoxide dismutase,peroxidase and catalase,increase contents of proline,soluble sugar and soluble protein,increase tissue relative water content and decrease water saturation deficit.It was concluded that salt–alkali stress inhibited the colonization of AM fungus,but the mycorrhiza still played a positive role in maintaining the normal growth of plants under salt–alkali stress.YUNHUI ZHOU YAJIE LIU YUDAN WANG CHUNXUE YANG 2022BIOCELL2022,46,8:2

15Neuronal injury and brain-derived neurotrophic factor expression in a rat model of amygdala kindling seizures Differences in brain regions and kindling courses显示文摘BACKGROUND:Studies have demonstrated that brain-derived neurotrophic factor(BDNF) has a dual effect on epilepsy.However,the relationship between epilepsy-induced brain injury and BDNF remains poorly understood.OBJECTIVE:According to ultrastructural and molecular parameters,to detect the degree of neuronal injury and BDNF expression changes at different brain regions and different kindling times to determine the effects of BDNF on epilepsy-induced brain injury.DESIGN,TIME AND SETTING:A randomized,controlled,animal experiment based on neuropathology and molecular biology was performed at the Department of Physiology and Department of Pathology,Basic Medical College of Jilin University in 2003.MATERIALS:UltraSensitive SP kit for immunohistochemistry(Fuzhou Maxim Biotechnology,China),BDNF antibody(concentrated type,Wuhan Boster Biological Technology,China),JEM-1000SX transmission electron microscopy(JEOL,Japan),and BH-2 light microscope(Olympus,Japan) were used in the present study.METHODS:Wistar rats were randomly assigned to control(n = 6),sham-surgery(n = 6),and model(n = 60) groups.The control group rats were not treated;an electrode was embedded into the amygdala in rats from the sham-surgery and model groups;an amygdala kindling epilepsy model was established in the model group.MAIN OUTCOME MEASURES:Pathological changes in the temporal lobe and hippocampus were observed by light and electron microscopy at 1,3,7,14,and 21 days following kindling,and BDNF expression in the various brain regions was determined by immunohistochemistry.RESULTS:In the model group,temporal lobe cortical and hippocampal neurons were swollen and the nuclei were laterally deviated.There were also some apoptotic neurons 3 days after kindling.The nucleoli disappeared and the nuclei appeared broken or lysed,as well as slight microglia hyperplasia,at 7 days.Electron microscopic observation displayed chromatin aggregation in the nuclei and slight mitochondrion swelling 3 days after kindling.Injury changes were aggravated at 7 days,characterized by broken cytoplasmic membrane and pyknosis.With the development of seizure,the number of BDNF-positive neurons in the hippocampus and temporal lobe increased and peaked at 7 days.Moreover,hippocampal and cortical temporal lobe injury continued.Following termination of electrical stimulation after 7 days of kindling,BDNF expression decreased,but continued to be expressed,up to 21 days of kindling.In addition,the number of temporal and hippocampal BDNF-positive neurons was greater than the control group.CONCLUSION:Brain injury and BDNF expression peaked at 7 days after kindling,and hippocampal changes were significant.Weihong Lin Hongmei Meng Yudan Lv Zan Wang Na Li Li Cui Baimin Zhang 2010Neural Regeneration Research2010,5,8:1

17Effect of cationic monomer structure on the aggregation behavior of amphoteric acrylic polymer around isoelectric point显示文摘Amphoteric polymer can be used as retanning agent in leather manufacture. It is particularly useful in chrome-free tanning systems since it can regulate the charge properties of chrome-free leather and enhance the fixation of anionic post-tanning chemicals in leather. However, the aggregation and precipitation of amphoteric polymer retanning agents around the isoelectric point (pI) hinder their wide application. Herein, we synthesized five amphoteric acrylic polymers (AAPs) by free radical copolymerization with acrylic acid and five different cationic acrylic monomers. The effect of cationic monomer structure on the aggregation behavior of AAPs was investigated. The aggregation of AAPs in aqueous solution showed pH and concentration dependence. Light scattering analysis showed that Poly (AA-co-MAPTAC) and Poly (AA-co-DMAPMA) were in the shape of coiled linear flexible chains with small particle size (Rg 7.6 nm and 14.8 nm, respectively) near the pI. However, Poly (AA-co-DAC), Poly (AA-co-DMC) and Poly (AA-co- DMAEMA) were in the shape of hollow spheres and exhibited serious aggregation. Quantum chemical calculations suggested that the amide groups in the cationic monomers MAPTAC and DMAPMA enhanced the nucleophilicity of AAPs. Thus the corresponding AAPs could carry a large number of cationic charges to slow their aggregation when the pH just climbed over the pI. 18Determination of Total Flavonoids in Acanthopanax senticosus(Rupr. et Maxim.) Harms by Enzymolysis and Its Technological Optimization显示文摘In order to optimize the ultrasonic extraction technique for the total flavonoid of leaf yellows plus, the contents of 21 leaf yellows plus total flavonoid from four regions in Heilongjiang Province were comparatively analyzed. The ultrasonic extraction technology was optimized by Box-Behnken response surface method, and the total flavonoid content of 21 kinds of Acanthopanax senticosus(Rupr. et Maxim.) Harms from different producing areas were analyzed by cluster analysis. The optimal process conditions were determined as ultrasonic time 30 min, solid-liquid ratio 1 : 12 and ultrasonic power 250 W, and the average yield of the total flavonoid was 1.453 mg·g^ (-1). 20Numerical and Experimental Study of a Tornado Mixer显示文摘A new design of a selective catalytic reduction(SCR)mixer called tornado was developed for a heavy-duty diesel engine to solve the urea deposition problem.A combination of CFD simulation and experimental studies was used to comprehensively evaluate the performance of the tornado mixer.According to the numerical simulations,this mixer can improve the front surface flow uniformity of the SCR carrier by 6.67%and the NH3 distribution uniformity by 3.19%compared to a traditional mixer.Similarly,steady state SCR conversion efficiency test results have shown that the tornado mixer can increase the average SCR conversion efficiency by 1.73%compared to a traditional mixer.Therefore,the tornado mixer outperforms traditional mixers in terms of mixing uniformity,resistance to deposition and impact on NOX emissions.In addition,a dimensionless parameter,the“limiting deviation rate”,is proposed in the present study to improve the mixing uniformity assessment method for SCR mixers(with the explicit intent to evaluate the mixing uniformity more accurately).Yibao Wang Dongsheng Wang Yudan Xue Dailong Shi Xiaoli Zhang Yang Chai Bang An 2021Fluid Dynamics & Materials Processing2021,17,6:0
