1A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 5Infection with novel coronavirus(SARS-CoV-2)causes pneumonia in Rhesus macaques显示文摘The 2019 novel coronavirus(SARS-CoV-2)outbreak is a major challenge for public health.SARS-CoV-2 infection in human has a broad clinical spectrum ranging from mild to severe cases,with a mortality rate of^6.4%worldwide(based on World Health Organization daily situation report).However,the dynamics of viral infection,replication and shedding are poorly understood.Here,we show that Rhesus macaques are susceptible to the infection by SARS-CoV-2.After intratracheal inoculation,the first peak of viral RNA was observed in oropharyngeal swabs one day post infection(1 d.p.i.),mainly from the input of the inoculation,while the second peak occurred at 5 d.p.i.,which reflected on-site replication in the respiratory tract.Histopathological observation shows that SARS-CoV-2 infection can cause interstitial pneumonia in animals,characterized by hyperemia and edema,and infiltration of monocytes and lymphocytes in alveoli.We also identified SARS-CoV-2 RNA in respiratory tract tissues,including trachea,bronchus and lung;and viruses were also re-isolated from oropharyngeal swabs,bronchus and lung,respectively.Furthermore,we demonstrated that neutralizing antibodies generated from the primary infection could protect the Rhesus macaques from a second-round challenge by SARS-CoV-2.The non-human primate model that we established here provides a valuable platform to study SARS-CoV-2 pathogenesis and to evaluate candidate vaccines and therapeutics.Chao Shan Yan-Feng Yao Xing-Lou Yang Yi-Wu Zhou Ge Gao Yun Peng Lian Yang Xue Hu Jin Xiong Ren-Di Jiang Hua-Jun Zhang Xiao-Xiao Gao Cheng Peng Juan Min Ying Chen Hao-Rui Si Jia Wu Peng Zhou Yan-Yi Wang Hong-Ping Wei Wei Pang Zheng-Fei Hu Long-Bao Lv Yong-Tang Zheng Zheng-Li Shi Zhi-Ming Yuan 2020Cell Research2020,30,8:19
6Preventive and therapeutic effect of simvastatin on secondary inflammatory damage of rats with cerebral hemorrhage显示文摘Objective:To investigate the preventive and therapeutic effect and mechanism of simvastatin on secondary inflammatory damage of rats with cerebral hemorrhage.Methods:Sixty SD rat aged 9-12 weeks were chosen and divided into the control group,model group and simvastatintreated group randomly with 20 rats in each group.Rats in the model group and simvastatintreated group were infused with autologous fresh uncoagulated blood to the right brain tissue of the basal ganglia to build the cerebral hemorrhage model,while rats in the control group were treated with the same amount of normal saline.Then,rats in the simvastatin-treated group were given a gavage of 3 mg/kg of simvastatin once a day after modeling.Rats in the three groups were given nerve dysfunction score(NDS) and wet-dry weighting method was used to detect the brain water content(BWC) of brain tissues around the lesion of the rats.Then Nissl staining was conducted and the undamaged neurons were counted.Immunohistochemical SP method was applied to count the number of NF-d the immuno fluorκB,TLR4 and IL-1escence method wasβ positive cells in brain tissues around the lesions,an employed to determine the expression levels of NF-κB,TLR4 and IL-1me points were aβ proteins.Results:The NDS results of the simvastatin-treated group at all till significantly higher than those of the model group(P < 0.05);the BWC values of the simvastatin-treated group at all time points were all significantly lower than those of the model group at the same periods(P < 0.05);the number of the undamaged neurons around the lesions of the simvastatin-treated group at all time points were all significantly higher than those of the model group(P < 0.05);seven days after treatment,the number of the NF-κB,TLR4 and IL-1β positive cells in brain tissues around the lesions of the simvastatin-treated group were all significantly lower than those of the model group(P < 0.05),and its expression levels of NF-ower than those of the model group(κB,TLR4 and IL-1P < 0.05).Conclusioβ protein were also significantly lns:Simvastatin can inhibit the expressions of NF-κB,TLR4 and IL-1β proteins in rats with cerebral hemorrhage,and protect neurons and reduce secondary inflammatory damages by down-regulating the above protein-mediated inflammatory responses.Hong-Xia Zhou Ling-Huan Gao Ling-Li Meng Yu-Xin Zhang Zi-Feng Wei Dao-Wen Si 2017Asian Pacific Journal of Tropical Medicine2017,10,2:17
7Highly Stretchable,Elastic,and Sensitive MXene-Based Hydrogel for Flexible Strain and Pressure Sensors显示文摘Electronic skin is driving the next generation of cutting-edge wearable electronic products due to its good wearability and high accuracy of information acquisition.However,it remains a challenge to fulfill the requirements on detecting full-range human activities with existing flexible strain sensors.Herein,highly stretchable,sensitive,and multifunctional flexible strain sensors based on MXene-(Ti_(3)C_(2)T_(x)-)composited poly(vinyl alcohol)/polyvinyl pyrrolidone double-network hydrogels were prepared.The uniformly distributed hydrophilic MXene nanosheets formed a three-dimensional conductive network throughout the hydrogel,endowing the flexible sensor with high sensitivity.The strong interaction between the double-network hydrogel matrix and MXene greatly improved the mechanical properties of the hydrogels.The resulting nanocomposited hydrogels featured great tensile performance(2400%),toughness,and resilience.Particularly,the as-prepared flexible pressure sensor revealed ultrahigh sensitivity(10.75 kPa^(-1))with a wide response range(0-61.5 kPa),fast response(33.5 ms),and low limit of detection(0.87 Pa).Moreover,the hydrogel-based flexible sensors,with high sensitivity and durability,could be employed to monitor fullrange human motions and assembled into some aligned devices for subtle pressure detection,providing enormous potential in facial expression and phonation recognition,handwriting verification,healthy diagnosis,and wearable electronics.Yao Lu Xinyu Qu Wen Zhao Yanfang Ren Weili Si Wenjun Wang Qian Wang Wei Huang Xiaochen Dong 2020Research2020,,1:14
8Genesis of sandstone-type uranium deposits along the northern margin of the Ordos Basin,China显示文摘Sandstone-type U mineral resources are among the important sources for nuclear energy.The U deposits in the Ordos Basin in China form part of the northern segment of the sandstone-hosted Central Asian Uranium MegaProvince.Two types of mineralizations are recognized in this basin:'phreatic permeable type'and'interlayer permeable type',both exhibiting features equivalent to roll-front subtypes.The'interlayer permeable type'is widely accepted as the dominant mineralization type for sandstone-type uranium deposits within large-scale basins,also designated as the'interlayer oxidation zone type',based on the horizontal color zoning model representing changing redox conditions.Here we synthesize data from several drill holes within the Ordos Basin,which suggest that major Mesozoic tectonic movements controlled the evolution of the sedimentary system in the basin.These tectonic movements contributed to the formation of three angular unconformities and four parallel unconformities as inferred from the stratigraphic relationships.In addition,other features such as vertical color zoning,paleo-channel controlled tabular or lentoid ore bodies(without roll-type)and a group interlayer horizontal zoning of altered minerals are also documented.Sequence stratigraphic analysis indicates that the Ordos Basin generally witnessed four cycles of water level variations during Mesozoic.During the variations,three high water level and three low water level events were recorded.Biological characteristics imply that the Ordos Basin went through multiple arid to humid climatic evolutions during Mesozoic.Combining the newly documented features with some novel concepts on the hydrodynamic mechanism for supergene ore-forming fluids,we propose a metallogenic model which invokes the importance of tectonic movements and water level fluctuations to explain the genesis of uranium deposits along the northern margin of the Ordos Basin.Ruoshi Jin Xueming Teng Xiaoguang Li Qinghong Si Wei Wang 2020Geoscience Frontiers2020,11,1:13
9Patterns,magnitude,and controlling factors of hydraulic redistribution of soil water by Tamarix ramosissima roots显示文摘Tamarix spp.(Saltcedar)is a facultative phreatophyte that can tolerate drought when groundwater is not accessed.In addition to deep water uptake,hydraulic redistribution(HR)is another factor contributing to the drought tolerance of Tamarix spp.In this study,data on soil volumetric moisture content(θ),lateral root sap flow,and relevant climate variables were used to investigate the patterns,magnitude,and controlling factors of HR of soil water by roots of Tamarix ramosissima Ledeb.in an extremely arid land in Northwest China.Results showed evident diurnal fluctuations in θ at the depths of 30 and 50 cm,indicating 'hydraulic lift'(HL).θ increased remarkably at 10 and 140 cm but decreased at 30 and 50 cm and slightly changed at 80 cm after rainfall,suggesting a possible 'hydraulic descent'(HD).However,no direct evidence was observed in the negative flow of lateral roots,supporting HR(including HL and HD)of T.ramosissima.The HR pathway unlikely occurred via lateral roots;instead,HR possibly occurred through adventitious roots with a diameter of 2-5 mm and a length of 60-100 cm.HR at depths of 20-60 cm ranged from 0.01-1.77 mm/d with an average of 0.43 mm/d,which accounted for an average of 22% of the estimated seasonal total water depletion at 0-160 cm during the growing season.The climate factors,particularly vapor pressure deficit and soil water potential gradient,accounted for at least 33% and 45% of HR variations with depths and years,respectively.In summary,T.ramosissima can be added to the wide list of existing species involved in HR.High levels of HR may represent a considerable fraction of daily soil water depletion and substantially improve plant water status.HR could vary tremendously in terms of years and depths,and this variation could be attributed to climate factors and soil water potential gradient.TengFei YU Qi FENG JianHua SI HaiYang XI Wei LI 2013Journal of Arid Land2013,5,3:12

11Multiscale carbon foam confining single iron atoms for efficient electrocatalytic CO2 reduction to CO显示文摘Electrocatalytic CO2 reduction to CO is a sustainable process for energy conversion.However,this process is still hindered by the diffusi limited mass transfer,low electrical conductivity and catalytic activity.Therefore,new strategies for catalyst design should be adopted to solve these problems and improve the electrocatalytic performa nee for CO production.Herein,we report a multiscale carb on foam confining〔single iron atoms prepared with the assistant of S1O2 template.The pore-enriched environment at the macro-scale facilitates the diffusion of reacta nts and products.The graphe ne nano sheets at the nano-scale promote the charge tran sfer duri ng the reaction.The single iron atoms con fined in carb on matrix at the atomic-scale provide the active sites for electrocatalytic CO2 reductio n to CO.The optimized catalyst achieves a CO Faradaic efficiency of 94.9%at a moderate potential of-0.5 V vs.RHE.Furthermore,the performance can be maintained over 60 hours due to the stable single iron atoms coordi nated with four n itroge n atoms in the carb on matrix.This work provides a promising strategy to improve both the activity and stability of single atom catalysts for electrocatalytic CO2 reduction to CO.Zheng Zhang Chao Ma Yunchuan Tu Rui Si Jie Wei Shuhong Zhang Zhen Wang Jian-Feng Li Ye Wang Dehui Deng 2019Nano Research2019,12,9:11
12Recombinant adenovirus vector Ad-hIL-10 protects grafts from cold ischemia-reperfusion injury following orthotopic liver transplantation in rats显示文摘BACKGROUND:Interleukin 10(IL-10),a Th2 type cytokine,modulates inflammatory responses by inhibiting the production of proinflammatory cytokines.This study was designed to investigate the protective effects of adenovirus- mediated human IL-10(Ad-hIL-10)gene transfer on protecting grafts from cold ischemia-reperfusion injury following orthotopic liver transplantation in rats. METHODS:Adenoviruses encoding hIL-10 orβ-galactosidase (Ad-lacZ)were injected via the superior mesenteric vein into prospective donor animals.The donor liver was harvested 48 hours after transduction,and stored for 12 hours at 4℃ in lactated Ringer's solution prior to transplantation.The rats were divided into saline,Ad-lacZ,and Ad-hIL-10 groups.Liver function test,histopathological examination,reverse transcriptase-polymerase chain reaction(RT-PCR),and Western blotting were performed at 24 hours after transplantation in the three groups. RESULTS:Liver function(ALT and AST)was significantly improved,and the Suzuki score was significantly decreased in the Ad-hIL-10 group.The levels of hepatic TNF-α,MIP-2,ICAM-1 mRNA,and NF-κB protein in the Ad-hIL-10 group were significantly decreased.The expression of hIL-10 mRNA was detected by RT-PCR in Ad-hIL-10-treated grafts but not in controls treated with saline or Ad-lacZ. 13Identification and Fine Mapping of a Gene Related to Pale Green Leaf Phenotype near the Centromere Region in Rice(Oryza sativa)显示文摘A thermo-insensitive pale green leaf mutant (pgl2) was isolated from T-DNA inserted transgenic lines of rice (Oryza sativa L. subsp. japonica cv. Nipponbare). Genetic analysis indicated that the phenotype was caused by a recessive mutation in a single nuclear-encoded gene. To map the PGL2 gene, an F2 population was constructed by crossing the mutant with Longtefu (Oryza sativa L. subsp. indica). The PGL2 locus was roughly linked to SSR marker RM331 on chromosome 8. To finely map the gene, 14 new InDel markers were developed around the marker, and PGL2 was further mapped to a 2.37 Mb centromeric region. Analysis on chlorophyll contents of leaves showed that there was no obvious difference between the mutant and the wild type in total chlorophyll (Chl) content, while the ratio of Chl a / Chl b in the mutant was only about 1, which was distinctly lower than that in the wild type, suggesting that the PGL2 gene was related to the conversion between Chl a and Chl b. 14First observation of tropospheric nitrogen dioxide from the Environmental Trace Gases Monitoring Instrument onboard the GaoFen-5 satellite显示文摘The Environmental Trace Gases Monitoring Instrument(EMI)is the first Chinese satellite-borne UV–Vis spectrometer aiming to measure the distribution of atmospheric trace gases on a global scale.The EMI instrument onboard the GaoFen-5 satellite was launched on 9 May 2018.In this paper,we present the tropospheric nitrogen dioxide(NO2)vertical column density(VCD)retrieval algorithm dedicated to EMI measurement.We report the first successful retrieval of tropospheric NO_(2) VCD from the EMI instrument.Our retrieval improved the original EMI NO_(2) prototype algorithm by modifying the settings of the spectral fit and air mass factor calculations to account for the on-orbit instrumental performance changes.The retrieved EMI NO_(2) VCDs generally show good spatiotemporal agreement with the satellite-borne Ozone Monitoring Instrument and TROPOspheric Monitoring Instrument(correlation coefficient R of ~0.9,bias<50%).A comparison with ground-based MAX-DOAS(Multi-Axis Differential Optical Absorption Spectroscopy)observations also shows good correlation with an R of 0.82.The results indicate that the EMI NO_(2) retrieval algorithm derives reliable and precise results,and this algorithm can feasibly produce stable operational products that can contribute to global air pollution monitoring.Chengxin Zhang Cheng Liu Ka Lok Chan Qihou Hu Haoran Liu Bo Li Chengzhi Xing Wei Tan Haijin Zhou Fuqi Si Jianguo Liu 2020Light(Science & Applications)2020,9,1:11
15Generation of a Hutchinson-Gilford progeria syndrome monkey model by base editing显示文摘Many human genetic diseases,including Hutchinson-Gilford progeria syndrome(HGPS),are caused by single point mutations.HGPS is a rare disorder that causes premature aging and is usually caused by a de novo point mutation in the LMNA gene.Base editors(BEs)composed of a cytidine deaminase fused to CRISPR/Cas9 nickase are highly efficient at inducing C to T base conversions in a programmable manner and can be used to generate animal disease models with single amino-acid substitutions.Here,we generated the first HGPS monkey model by delivering a BE mRNA and guide RNA(gRNA)targeting the LMNA gene via microinjection into monkey zygotes.Five out of six newborn monkeys carried the mutation specifically at the target site.HGPS monkeys expressed the toxic form of lamin A,progerin,and recapitulated the typical HGPS phenotypes including growth retardation,bone alterations,and vascular abnormalities.Thus,this monkey model genetically and clinically mimics HGPS in humans,demonstrating that the BE system can efficiently and accurately generate patient-specific disease models in non-human primates.Fang Wang Weiqi Zhang Qiaoyan Yang Yu Kang Yanling Fan Jingkuan Wei Zunpeng Liu Shaoxing Dai Hao Li Zifan Li Lizhu Xu Chu Chu Jing Qu Chenyang Si Weizhi Ji Guang-Hui Liu Chengzu Long Yuyu Niu 2020Protein & Cell2020,11,11:11

17Response of Populus euphratica Oliv.sap flow to environmental variables for a desert riparian forest in the Heihe River Basin,Northwest China显示文摘Being an important desert riparian forest in the lower reaches of the Heihe River Basin,Populus euphratica Oliv.forest functions as a natural barrier in maintaining and preserving the stability of local oases.Accordingly,accurately estimating the water use of P.euphratica is important for vegetation protection and water resource allocation.To date,little data are available for evaluating the hysteretic effects between sap flow and environmental variables,and for estimating the water use of desert riparian forest.In this study,we measured the sap flow velocity(V_s)of P.euphratica using the heat ratio method during the growing season of 2012.Based on the response of V_s to solar radiation(R_s)and vapor pressure deficit(VPD),we estimated the hourly Vsand daily V_s using the multivariable linear regression and a modified Jarvis-Stewart(JS)model,respectively.Hysteretic response of V_s to environmental variables was then evaluated using a sap flow model.We found the thresholds of V_s responses to R_s and VPD at both hourly and daily scales during the growing season,and successfully estimated the seasonal variations of hourly V_s and daily V_s using the JS model.At an hourly scale,the maximum V_s occurred earlier than the maximum VPD by approximately 0.5 h but later than the maximum R_s by approximate 1.0 h.At a daily scale,the maximum V_s lagged behind the maximum VPD by approximately 2.5 h while occurred earlier than the maximum R_s by approximately 2 h.However,hysteretic response of V_swas weakened when R_s and VPD were measured together using the JS model at both hourly and daily scales.Consequently,short-term and intensive field campaigns,where V_s and environmental variables can be measured,may be used to estimate short-run sap flow and stand transpiration using only two environmental variables.LI Wei SI Jianhua YU Tengfei LI Xiaoyan 2016Journal of Arid Land2016,8,4:9
18Effect of nanopore size on poly(dT)_(30) translocation through silicon nitride membrane显示文摘The nanopore size effect on translocation of poly(dT)30through Si3N4 membrane is investigated.In this paper,we report that the speed of the poly(dT)30 transport through Si3N4 nanopores can be slowed down by half through increasing the nanopore diameter from 4.8 nm to 10.8 nm.The results are consistent with our simulation results.Besides,the current blockage induced by DNA passing through the nanopore is less obvious as pore diameter is larger,which is in good agreement with the theoretical prediction.The conclusion about DNA transport through nanopores is beneficial for the design of DNA sequencing devices.SI Wei SHA JingJie LIU Lei QIU YingHua CHEN YunFei 2013Science China(Technological Sciences)2013,56,10:9
