1Organellar genome assembly methods and comparative analysis of horticultural plants显示文摘Although organellar genomes(including chloroplast and mitochondrial genomes)are smaller than nuclear genomes in size and gene number,organellar genomes are very important for the investigation of plant evolution and molecular ecology mechanisms.Few studies have focused on the organellar genomes of horticultural plants.Approximately 1193 chloroplast genomes and 199 mitochondrial genomes of land plants are available in the National Center for Biotechnology Information(NCBI),of which only 39 are from horticultural plants.In this paper,we report an innovative and efficient method for high-quality horticultural organellar genome assembly from next-generation sequencing(NGS)data.Sequencing reads were first assembled by Newbler,Amos,and Minimus software with default parameters.The remaining gaps were then filled through BLASTN search and PCR.The complete DNA sequence was corrected based on Illumina sequencing data using BWA(Burrows–Wheeler Alignment tool)software.The advantage of this approach is that there is no need to isolate organellar DNA from total DNA during sample preparation.Using this procedure,the complete mitochondrial and chloroplast genomes of an ornamental plant,Salix suchowensis,and a fruit tree,Ziziphus jujuba,were identified.This study shows that horticultural plants have similar mitochondrial and chloroplast sequence organization to other seed plants.Most horticultural plants demonstrate a slight bias toward A+T rich features in the mitochondrial genome.In addition,a phylogenetic analysis of 39 horticultural plants based on 15 protein-coding genes showed that some mitochondrial genes are horizontally transferred from chloroplast DNA.Our study will provide an important reference for organellar genome assembly in other horticultural plants.Furthermore,phylogenetic analysis of the organellar genomes of horticultural plants could accurately clarify the unanticipated relationships among these plants.Xuelin Wang Feng Cheng Dekai Rohlsen Changwei Bi Chunyan Wang Yiqing Xu Suyun Wei Qiaolin Ye Tongming Yin Ning Ye 2018Horticulture Research2018,5,1:4

3Inhibiting Hv1 channel in peripheral sensory neurons attenuates chronic inflammatory pain and opioid side effects显示文摘Both opioids and nonsteroidal anti-inflammatory drugs(NSAIDS)produce deleterious side effects and fail to provide sustained relief in patients with chronic inflammatory pain.Peripheral neuroinflammation(PN)is critical for initiation and development of inflammatory pain.A better understanding of molecular mechanisms underlying PN would facilitate the discovery of new analgesic targets and the development of new therapeutics.Emerging evidence suggests that peripheral sensory neurons are not only responders to painful stimuli,but are also actively engaged in inflammation and immunity,whereas the intrinsic regulatory mechanism is poorly understood.Here we report the expression of proton-selective ion channel Hv1 in peripheral sensory neurons in rodents and humans,which was previously shown as selectively expressed in microglia in mammalian central nervous system.Neuronal Hv1 was up-regulated by PN or depolarizing stimulation,which in turn aggravates inflammation and nociception.Inhibiting neuronal Hv1 genetically or by a newly discovered selective inhibitor YHV98-4 reduced intracellular alkalization and ROS production in inflammatory pain,mitigated the imbalance in downstream SHP-1-pAKT signaling,and also diminished pro-inflammatory chemokine release to alleviate nociception and morphine-induced hyperalgesia and tolerance.Thus,our data reveal neuronal Hv1 as a novel target in analgesia strategy and managing opioids-related side effects.Qiansen Zhang Yimin Ren Yiqing Mo Peipei Guo Ping Liao Yuncheng Luo Jie Mu Zhuo Chen Yang Zhang Ya Li Linghui Yang Daqing Liao Jie Fu Juwen Shen Wei Huang Xuewen Xu Yanyan Guo Lianghe Mei Yunxia Zuo Jin Liu Huaiyu Yang Ruotian Jiang 2022Cell Research2022,32,5:3

6All-optical logic devices based on black arsenic-phosphorus with strong nonlinear optical response and high stability显示文摘The Kerr nonlinearity in two-dimensional(2D)nanomaterials is emerging as an appealing and intriguing research area due to their prominent light processing,modulation,and manipulation abilities.In this contribution,2D black arsenic-phosphorus(B-AsP)nanosheets(NSs)were applied in nonlinear photonic devices based on spatial self-phase modula-tion(SSPM)method.By applying the Kerr nonlinearity in 2D B-AsP,an all-optical phase-modulated system is proposed to realize the functions of“on”and“off”in all-optical switching.By using the same all-optical phase-modulated system,another optical logic gate is proposed,and the logical“or”function is obtained based on the 2D B-AsP NSs dispersions.Moreover,by using the SSPM method,a 2D B-AsP/SnS_(2) hybrid structure is fabricated,and the result illustrates that the hybrid structure possesses the ability of the unidirectional nonlinear excitation,which helps in obtaining the function of spatial asymmetric light propagation.This function is considered an important prerequisite for the realization of diode functionalization,which is believed to be a factor in important basis for the design of isolators as well.The initial investig-ations indicate that 2D B-AsP is applicable for designing optical logical devices,which can be considered as an import-ant development in all-optical information processing.Leiming Wu Taojian Fan Songrui Wei Yijun Xu Ye Zhang Dingtao Ma Yiqing Shu Yuanjiang Xiang Jun Liu Jianqing Li Krassimir Panajotov Yuwen Qin Han Zhang 2022Opto-Electronic Advances2022,5,1:3
7Gedunin Degrades Aggregates of Mutant Huntingtin Protein and Intranuclear Inclusions via the Proteasomal Pathway in Neurons and Fibroblasts from Patients with Huntington’s Disease显示文摘Huntington's disease(HD) is a deadly neurodegenerative disease with abnormal expansion of CAG repeats in the huntingtin gene. Mutant Huntingtin protein(m HTT) forms abnormal aggregates and intranuclear inclusions in specific neurons, resulting in cell death. Here,we tested the ability of a natural heat-shock protein 90 inhibitor, Gedunin, to degrade transfected m HTT in Neuro-2 a cells and endogenous m HTT aggregates and intranuclear inclusions in both fibroblasts from HD patients and neurons derived from induced pluripotent stem cells from patients. Our data showed that Gedunin treatment degraded transfected m HTT in Neuro-2 a cells, endogenous m HTT aggregates and intranuclear inclusions in fibroblasts from HD patients, and in neurons derived from induced pluripotent stem cells from patients in a dose-and time-dependent manner, and its activity depended on the proteasomal pathway rather than the autophagy route. 9CsPbBr_(3) quantum dots photodetectors boosting carrier transport via molecular engineering strategy显示文摘Perovskite quantum dots (PQDs) require ligands on their surfaces to passivate defects and prevent aggregation. However, the ligands construct the interface relationship between the PQDs, which may seriously hinder the carrier transport. Hence, we propose a molecular engineering strategy of using 3,4-ethylenedioxythiophene (EDOT) to perfectly solve this problem, benefiting from its high conjugation and passivation ability to CsPbBr_(3) PQDs. Furthermore, EDOT on the surface of PQDs can be in-situ polymerized under the photocurrent of the photodetector, thus interconnecting the PQDs which enhanced the performance of the photodetectors up to 178% of its initial performance. We have thoroughly investigated the electropolymerization process of EDOT and its passivation effect on PQDs. The simple lateral photodetector based on EDOT PQDs exhibits a high responsivity of 11.96 A/W, which is 104 times higher than that of oleic acid caped PQDs. Due to the protection of poly(3,4-ethylenedioxythiophene) (PEDOT), the photodetector prepared from EDOT PQDs exhibited very high stability, retaining 94% of its performance after six months in air. 11Whole-brain patterns of the presynaptic inputs and axonal projections of BDNF neurons in the paraventricular nucleus显示文摘Brain-derived neurotrophic factor (BDNF) plays a crucial role in human obesity. Yet, the neural circuitry supporting the BDNF-mediated control of energy homeostasis remains largely undefined. To map key regions that might provide inputs to or receive inputs from the paraventricular nucleus (PVN) BDNF neurons, a key type of cells in regulating feeding and thermogenesis, we used rabies virus-based transsynaptic labeling and adeno-associated virus based anterograde tracing techniques to reveal their whole-brain distributions. We found that dozens of brain regions provide dense inputs to or receive dense inputs from PVN BDNF neurons, including several known weight control regions and several novel regions that might be functionally important for the BDNF-mediated regulation of energy homeostasis.Interestingly, several regions show very dense reciprocal connections with PVN BDNF neurons, including the lateral septum, the preoptic area, the ventromedial hypothalamic nucleus, the paraventricular thalamic nucleus, the zona incerta, the lateral parabrachial nucleus, the subiculum, the raphe magnus nucleus, and the raphe pallidus nucleus. These strong anatomical connections might be indicative of important functional connections. 12Influence of cementite spheroidization on relieving the micro-galvanic effect of ferrite-pearlite steel in acidic chloride environment显示文摘The corrosion behavior of the as-received steel and the spheroidized steel in acidic chloride environment was investigated. The results indicate the corrosion mode and corrosion rate of two steels are diverse due to their difference in microstructure. For as-received steel with ferrite-pearlite microstructure, severe localized corrosion happens on the pearlite regions, and plenty of cathodic cementite remains in the pits, further strengthening the micro-galvanic effect and accelerating the corrosion rate. 13Relationship between gravity change and Yangbi M_(S)6.4 earthquake显示文摘Based on the relative and absolute gravity measurements in the southern South-North Seismic Belt since 2015,we analyzed the dynamic change of the regional gravity field and its relationship with the Yangbi M_(S)6.4 earthquake that occurred on May 21,2021.The results show that:(1)The regional gravity field changes are closely related to the Weixi-Qiaohou fault,which reflects the surface gravity field changes caused by the fault activity from 2015 to 2021;(2)The gravity field change related to the preparation of Yangbi earthquake has experienced the evolution process of'steady state-regional gravity anomaly-local gravity anomaly-four-quadrant distribution-large area positive anomaly-earthquake occurring in the reverse change process';(3)The cumulative and differential change images of the gravity field show that there were significant gravity changes in the two years preceding the Yangbi earthquake,and the earthquake occurred in the high-gradient belt of gravity variation,the center of the four-quadrant,and close to the zero contour turn;(4)The dynamic evolution image of the gravity field can well reflect the precursory phenomena during the preparation for the Yangbi MS6.4 earthquake.Based on the anomaly change of mobile gravity,a certain degree of medium-term prediction was made before the Yangbi M_(S)6.4 earthquake,especially the determination of strong earthquake location.Xiong Yang Yiqing Zhu Yunfeng Zhao Shouchun Wei 2023Geodesy and Geodynamics2023,14,4:1

17The synergy between cementite spheroidization and Cu alloying on the corrosion resistance of ferrite-pearlite steel in acidic chloride solution显示文摘In this work,the corrosion behavior of medium-carbon steels(45,45 Cu and 45 Cuq steels)in acidic chloride environment was investigated.The results indicated that the micro-galvanic effect between the anodic ferrite matrix phase and the cathodic cementite secondary phase notably affected the corrosion resistance of the three steels.For 45 steel,serious pitting corrosion happened in and around the pearlite regions,and a large number of lamellar cementite was fixed in the corrosion pits.Meanwhile,the continuously increasing superficial area of cathodic cementite enhanced the micro-galvanic corrosion,resulting in a rapidly increase in corrosion rate with time.While for 45 Cu and 45 Cuq steels,macroscopic uniform corrosion occurred,and the cementite accumulation was markedly reduced as compared with 45 steel,thus the micro-galvanic effect was weakened and the corrosion rate was decreased accordingly.Among these,45 Cuq steel showed the most stable and excellent corrosion resistance during long-term corrosion,indicating the occurrence of a synergistic effect between cementite spheroidization and Cu alloying,thereby significantly improving the corrosion resistance of 45 steel.Hu Liu Jie Wei Junhua Dong Yangtao Zhou Yiqing Chen Yumin Wu Subedi Dhruba Babu Aniefiok Joseph Umoh Wei Ke 2021Journal of Materials Science & Technology2021,,25:1

20Transcriptomic characteristics and impaired immune function of patients who retest positive for SARS-CoV-2 RNA显示文摘Coronavirus disease 2019(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection,has become a global public health crisis.Some patients who have recovered from COVID-19 subsequently test positive again for SARS-CoV-2 RNA after discharge from hospital.How such retest-positive(RTP)patients become infected again is not known.In this study,30 RTP patients,20 convalescent patients,and 20 healthy controls were enrolled for the analysis of immunological characteristics of their peripheral blood mononuclear cells.We found that absolute numbers of CD4^(+)T cells,CD8^(+)T cells,and natural killer cells were not substantially decreased in RTP patients,but the expression of activation markers on these cells was significantly reduced.The percentage of granzyme B-producing T cells was also lower in RTP patients than in convalescent patients.Through transcriptome sequencing,we demonstrated that high expression of inhibitor of differentiation 1(ID1)and low expression of interferon-induced transmembrane protein 10(IFITM10)were associated with insufficient activation of immune cells and the occurrence of RTP.These findings provide insight into the impaired immune function associated with COVID-19 and the pathogenesis of RTP,which may contribute to a better understanding of the mechanisms underlying RTP.Dongyao Wang Dong Wang Min Huang Xiaohu Zheng Yiqing Shen Binqing Fu Hong Zhao Xianxiang Chen Peng Peng Qi Zhu Yonggang Zhou Jinghe Zhang Zhigang Tian Wuxiang Guan Guiqiang Wang Haiming Wei 2021Journal of Molecular Cell Biology2021,13,10:0
