2Lycorine inhibits breast cancer growth and metastasis via inducing apoptosis and blocking Src/FAK-involved pathway显示文摘Breast cancer is the most commonly diagnosed cancer type worldwide among women and more than 90% of patients die from tumor metastasis. Lycorine, a natural alkaloid, has been widely reported possessing potential efficacy against cancer proliferation and metastasis. In our study, the anti-tumor potency on breast cancer was evaluated in vitro and in vivo for the first time. Our results indicated that lycorine inhibited breast cancer cells growth, migration and invasion as well as induced their apoptosis.In in vivo study, lycorine not only suppressed breast tumor growth in xenograft models and inhibited breast tumor metastasis in MDA-MB-231 tail vein model. More importantly, we found lycorine had less toxicity than first-line chemotherapy drug paclitaxel at the same effective dose in vivo. Furthermore, on mechanism, lycorine inhibited tumor cell migration and invasion via blocking the Src/FAK(focal adhesion kinase)-involved pathway. 4Characterization of a Putative New Semi-Dominant Reduced Height Gene, Rht_NM9, in Wheat(Triticum aestivum L.)显示文摘Plant height is an important agronomic trait in cereal crops, and can affect both plant architecture and grain yield. New dwarfing genes are required for improving the genetic diversity of wheat. In this study, a novel dwarf mutant, NM9, was created by treating seeds of the wheat variety NAU9918 with ethyl methanesulfonate(EMS). NM9 showed obvious phenotypic changes, which were distinct from those caused by other dwarfing genes, especially the reduced plant height, increased effective tiller number, and elongated spike and grain length. The reduced plant height in NM9 was attributable to a semi-dominant dwarfing gene Rht_NM9, which was flanked by two closely linked SNP markers, SNP34 and SNP41, covering an 8.86-Mb region on the chromosome arm 2AS. The results of gibberellic acid(GA) sensitivity evaluation, comparative genomics analysis and allelism test indicated that Rht_NM9 was neither allelic to Rht7 and Rht21 nor homoeoallelic to Rht8, so Rht_NM9 was proposed to be a new dwarfing locus on the homoeologous group 2 chromosomes of wheat. 5Feedrate scheduling method for constant peak cutting force in five-axis flank milling process显示文摘It is extremely important to select appropriate feedrates for the stable machining of parts with ruled surface in modern aviation industrial applications.However,the current studies take too much time to achieve this goal.Therefore,this paper presents an efficient feedrate optimization method for constant peak cutting force in five-axis flank milling process.The solution method of the instantaneous undeformed chip thickness(IUCT)is proposed using least squares theory with the cutter entry angle and feedrate as variables.Based on this method,an explicit analytical expression of the peak cutting force for each cutting point is established.Furthermore,a feedrate scheduling method is developed to quickly solve the appropriate feedrate under constant peak cutting force.To verify the proposed IUCT model,the fitting IUCT is compared with the accuracy data at different feedrates.Additionally,some experiments of five-axis flank milling are conducted to demonstrate the effectiveness of the peak force model and the feedrate scheduling method.And the surface roughness before and after feedrate scheduling is detected.The results show that the proposed feedrate scheduling method can quickly adjust the feedrate and ensure constant peak force during machining.At the same time,the surface quality is kept at a high level.Liping WANG Xing YUAN Hao SI Feiyu DUAN 2020Chinese Journal of Aeronautics2020,33,7:5
6Effect of addition of Al-Si eutectic alloy on microstructure and mechanical properties of Mg-12wt%Li alloy显示文摘Mg-12 Li, Mg-12 Li-3(Al-Si), Mg-12 Li-7(Al-Si) and Mg-12 Li-9(Al-Si) alloys(all in wt%) were fabricated by high frequency vacuum induction melting in a water cooled copper crucible. After subsequently hotrolling and annealing, their microstructure and mechanical properties were investigated. Experimental results show that mechanical properties of Mg-12 Li alloy were significantly improved by the addition of Al-Si eutectic alloy. Mg-12 Li-7(Al-Si) alloy shows the highest strength of 196 MPa of the investigated alloys, which is about 1.8 times of the strength of Mg-12 Li alloy, and maintains high elongation of 27%.The improved mechanical property with addition of Al and Si in the eutectic proportion into Mg-12 Li alloy was attributed to the solution strengthening effect of A1 and precipitation hardening effect from AlLi and Mg_2 Si precipitates.Zilong Zhao Xuegang Xing Jinyu Ma Liping Bian Wei Liang Yide Wang 2018Journal of Materials Science & Technology2018,34,9:5
7Genetic analysis of yield and fiber quality traits in upland cotton(Gossypium hirsutum L.) cultivated in different ecological regions of China显示文摘Background:Cotton is an important fiber crop worldwide.The yield potential of current genotypes of cotton can be exploited through hybridization.However,to develop superior hybrids with high yield and fiber quality traits,information of genetic control of traits is prerequisite.Therefore,genetic analysis plays pivotal role in plant breeding.Results:In present study,North Carolina II mating design was used to cross 5 female parents with 6 male parents to produce 30 intraspecific F1cotton hybrids.All plant materials were tested in three different ecological regions of China during the year of 2016-2017.Additive-dominance-environment(ADE)genetic model was used to estimate the genetic effects and genotypic and phenotypic correlation of yield and fiber quality traits.Results showed that yield traits except lint percentage were mainly controlled by genetic and environment interaction effects,whereas lint percentage and fiber quality traits were determined by main genetic effects.Moreover,dominant and additiveen vironine nt in teraction effects had more influence on yield traits,whereas additive and domi nance-e nviron ment interaction effects were found to be predominant for fiber traits.Broad-sense and its interaction heritability were significant for all yield and most of fiber quality traits.Narrow-sense and its interaction heritability were non-significant for boll number and seed cotton yield.Correlation analysis indicated that seed cotton yield had significant positive correlation with other yield attributes and non-significant with fiber quality traits.All fiber quality traits had signiflcant positive correlation with each other except micronaire.Conclusions:Results of current study provide important information about genetic control of yield and fiber quality traits.Further,this study identified that parental lines,e.g.,SJ48-1,ZB-1,851-2,and DT-8 can be utilized to improve yield and fiber quality traits in cotton.SHAHZAD Kashif LI Xue QI Tingxiang GUO Liping TANG Huini ZHANG Xuexian WANG Hailin ZHANG Meng ZHANG Bingbing QIAO Xiuqin XING Chaozhu WU Jianyong 2019Journal of Cotton Research2019,2,2:5
8The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. 9Exploration of presence/absence variation and corresponding polymorphic markers in soybean genome显示文摘This study was designed to reveal the genome‐wide distribution of presence/absence variation(PAV) and to establish a database of polymorphic PAV markers in soybean. The 33 soybean whole‐genome sequences were compared to each other with that of Williams 82 as a reference genome. A total of 33,127 PAVs were detected and 28,912 PAV markers with their primer sequences were designed as the database NJAUSoyPAV_1.0. The PAVs scattered on whole genome while only 518(1.8%) overlapped with simple sequence repeats(SSRs) in BARCSOYSSR_1.0database. In a random sample of 800 PAVs, 713(89.13%) showed polymorphism among the 12 differential genotypes. Using 126 PAVs and 108 SSRs to test a Chinese soybean germplasm collection composed of 828 Glycine soja Sieb. et Zucc. and Glycine max(L.) Merr. accessions, the per locus allele number and its variation appeared less in PAVs than in SSRs. The distinctness among alleles/bands of PCR(polymerase chain reaction) products showed better in PAVs than in SSRs, potential in accurate marker‐assisted allele selection. 10Current understanding of osteoarthritis pathogenesis and relevant new approaches显示文摘Osteoarthritis(OA)is the most common degenerative joint disease that causes painful swelling and permanent damage to the joints in the body.The molecular mechanisms of OA are currently unknown.OA is a heterogeneous disease that affects the entire joint,and multiple tissues are altered during OA development.To better understand the pathological mechanisms of OA,new approaches,methods,and techniques need to be used to understand OA pathogenesis.In this review,we first focus on the epigenetic regulation of OA,with a particular focus on DNA methylation,histone modification,and microRNA regulation,followed by a summary of several key mediators in OA-associated pain.We then introduce several innovative techniques that have been and will continue to be used in the fields of OA and OA-associated pain,such as CRISPR,scRNA sequencing,and lineage tracing.Next,we discuss the timely updates concerning cell death regulation in OA pathology,including pyroptosis,ferroptosis,and autophagy,as well as their individual roles in OA and potential molecular targets in treating OA.Finally,our review highlights new directions on the role of the synovial lymphatic system in OA.An improved understanding of OA pathogenesis will aid in the development of more specific and effective therapeutic interventions for OA.Liping Tong Huan Yu Xingyun Huang Jie Shen Guozhi Xiao Lin Chen Huaiyu Wang Lianping Xing Di Chen 2022Bone Research2022,10,4:4
11Optical dating of the Jingshuiwan Paleolithic site of Three Gorges, China显示文摘The Jingshuiwan Paleolithic site occu- pies the second terrace on the right bank of the Yangtze River. The lithic assemblage is characterized by choppers and scrapers made on pebbles and large flakes——the typical lithic industry of South China. Deposits of the site are mainly composed of fluvial sediments. Quartz grains extracted from these fluvial sediments from which the artifacts were un- covered were dated using the optically stimulated luminescence/single-aliquot regenerative-dose tech- nique. The dating results show that ancient human activities at this site took place in the early Upper Pleistocene (ca. 70 ka). The successful age analysis of the Jingshuiwan site is considered as a major break- through in chronological analysis of Paleolithic open site in the Three Gorges region and even in South China. 12First experimental constraints on WIMP couplings in the effective field theory framework from CDEX显示文摘We present weakly interacting massive particles(WIMPs) search results performed using two approaches of effective field theory from the China Dark Matter Experiment(CDEX), based on the data from both CDEX-1B and CDEX-10 stages. In the nonrelativistic effective field theory approach, both time-integrated and annual modulation analyses were used to set new limits for the coupling of WIMP-nucleon effective operators at 90% confidence level(C.L.) and improve over the current bounds in the low mχregion. In the chiral effective field theory approach, data from CDEX-10 were used to set an upper limit on WIMP-pion coupling at 90% C.L. 13Pm67, a new powdery mildew resistance gene transferred from Dasypyrum villosum chromosome 1V to common wheat(Triticum aestivum L.)显示文摘Powdery mildew, caused by the biotrophic fungus Blumeria graminis f. sp. tritici(Bgt), is a global disease that poses a serious threat to wheat production. To explore additional resistance gene, a wheatDasypyrum villosum 1 V#5(1 D) disomic substitution line NAU1813(2 n = 42) with high level of seedling resistance to powdery mildew was used to generate the recombination between chromosomes 1 V#5 and1 D. Four introgression lines, including t1 VS#5 ditelosomic addition line NAU1815, t1 VL#5 ditelosomic addition line NAU1816, homozygous T1 DL·1 VS#5 translocation line NAU1817, and homozygous T1 DS·1 VL#5 translocation line NAU1818 were developed from the selfing progenies of 1 V#5 and 1 D double monosomic line that derived from F1 hybrids of NAU1813/NAU0686. All of them were characterized by fluorescence in situ hybridization, genomic in situ hybridization, 1 V-specific markers analysis, and powdery mildew tests at different developmental stages. A new powdery mildew resistance gene named Pm67 was physically located in the terminal bin(FL 0.70–1.00) of 1 VS#5. Lines with Pm67 exhibited seedling stage immunity and tissue-differentiated reactions at adult plant stage. The sheaths, stems, and spikes of the Pm67 line were still immune, but the leaves showed a low degree of susceptibility.Microscopic observation showed that most penetration attempts were stopped in association with papillae on the sheath, and colonies cannot form conidia on the susceptible leaf of Pm67 line at adult plant stage, suggesting that the defence layers of the Pm67 line is tissue-differentiated. 14Climatic significance of n-alkanes and their compound-specific dD values from lake surface sediments on the Southwestern Tibetan Plateau显示文摘The content and hydrogen isotope ratios of n-alkanes extracted from recent lake surface sediments sampled from nine lakes in three different climate zones on the Southwestern Tibetan Plateau were analyzed.Values were compared with n-alkanes in plants from lake drainages,and dD values of meteoric water,lake water,and mean annual precipitation.The results showed that n-C23was predominantly derived from aquatic plants,and n-C27–n-C33from terrestrial higher plants.The average carbon chain length of n-C27–n-C33(ACL27–33)was positively correlated with the mean annual precipitation.dD values of the long-chain n-alkanes n-C29and n-C31of terrestrial origin(varying between-214%and-169%,and-226%and-185%,respectively)were inversely correlated with mean annual precipitation;but in accordance with the average annual variations in dD(OIPC),dD values of n-C31were strongly related to the dD values of growing season meteoric water(R2=0.74).The large difference between dD values of n-C23of aquatic origin and n-C31(an average of about 27%)demonstrates the enrichment of the lake water dD compared to precipitation,caused by strong evaporation in the semiarid–arid areas of the southwestern Tibetan Plateau.Average value of en C25 31=p(-95%)is evidently higher than the value observed in European wet regions(-128%);besides,en C31(about-116%)is constant along the study transect(SD=9),which indicates that n-C31is a useful proxy for the environment.Xing Hu Liping Zhu Yong Wang Junbo Wang Ping Peng Qingfeng Ma Jianfang Hu Xiao Lin 2014Chinese Science Bulletin2014,59,24:3
15First results on ^(76)Ge neutrinoless double beta decay from CDEX-1 experiment显示文摘We report the first results on ^(76)Ge neutrinoless double beta decay from stage one of the China dark-matter experiment(CDEX).A p-type point-contact high-purity germanium detector with a mass of 994 g has been installed to detect neutrinoless double beta decay events, as well as to directly detect dark matter particles. An exposure of 304 kg d has been analyzed over a wide spectral band from 500 keV to 3 MeV. The average event rate obtained was about 0.012 counts per keV per kg per day over the 2.039 MeV energy range. The half-life of ^(76)Ge neutrinoless double beta decay derived based on this result is T^(0ν)1/2>6.4×10^(22) yr(90% C.L.). 16Holocene Temperature Records from the East China Sea Mud Area Southwest of the Cheju Island Reconstructed by theU_(37)~K~' and TEX_(86) Paleothermometers显示文摘As an important marginal sea under the influences of both the Changjiang River and the Kuroshio,the East China Sea (ECS) environment is sensitive to both continental and oceanic forcing.Paleoenvironmental records are essential for understanding the long-term environmental evolution of the ECS and adjacent areas.However,paleo-temperature records from the ECS shelf are currently very limited.In this study,the'K37U and TEX86paleothermometers were used to reconstruct surface and subsurface temperature changes of the mud area southwest of the Cheju Island (Site F10B) in the ECS during the Holocene.The results indicate that temperature changes of F10B during the early Holocene (11.6 6.2 kyr) are associated with global climate change.During the period of 6.2 2.5 kyr,the similar variability trends of smoothing average of T (the difference between surface and subsurface temperature) of Site F10B and the strength of the Kuroshio suggest that the Kuroshio influence on the site started around 6.2 kyr when the Kuroshio entered the Yellow Sea and continued to 2.5 kyr.During the late Holocene (2.5 1.45 kyr) ,apparent decreases of'K37U sea surface temperature (SST) and T imply that the direct influence of the Kuroshio was reduced while cold eddy induced by the Kuroshio gradually controlled hydrological conditions of this region around 2.5 kyr.XING Lei JIANG Yiqing YUAN Zineng ZHANG Hailong LI Li ZHOU Liping ZHAO Meixun 2013Journal of Ocean University of China2013,12,4:3
17Genome-wide analysis of Rf-PPR-like(RFL)genes and a new InDel marker development for Rf1 gene in cytoplasmic male sterile CMS-D2 Upland cotton显示文摘Background: Cytoplasmic male sterility in flowering plants is a convenient way to use heterosis via hybrid breeding and may be restored by nuclear restorer-of-fertility(Rf) genes. In most cases, Rf genes encoded pentatricopeptide repeat(PPR) proteins and several Rf genes are present in clusters of similar Rf-PPR-like(RFL) genes. However, the Rf genes in cotton were not fully characterized until now.Results: In total, 35 RFL genes were identified in G. hirsutum, 16 in G. arboreum, and 24 in G. raimondii. Additionally,four RFL-rich regions were identified; the RFL-rich region in Gh_D05 is the probable location of Rf-PPR genes in cotton and will be studied further in the future. Furthermore, an insertion sequence was identified in the promoter sequence of Gh_D05 G3392 gene in the restorer line, as compared with the CMS-D2 line and maintainer lines. An InDel-R marker was then developed and could be used to distinguish the restorer line carrying Rfl from other genotypes without the Rf1 allele.Conclusion: In this study, genome-wide identification and analysis of RFL genes have identified the candidate Rf-PPR genes for CMS in Gossypium. 18Comparative performance of hybrid generations reveals the potential application of F_(2)hybrids in upland cotton显示文摘Background:The utilization of heterosis has greatly improved the productivity of cotton worldwide.However,a major constraint for the large-scale promotion of F_(1) hybrid cotton is artificial emasculation and pollination.This study proposed the potential utilization of F_(2) hybrids to improve upland cotton production through a comparative evaluation of hybrid generations.Results:Eight upland cotton varieties were analyzed and crosses were made according to NCII incomplete diallel cross-breeding design in two cotton belts of China.Variance analysis revealed significant differences in agronomic,yield,and fiber quality in both generations and environments.The broad-sense heritability of agronomic and yield traits was relatively higher than quality traits.Furthermore,the narrow-sense heritability of some traits was higher in F_(2) than in the F_(1) generation in both cotton belts.Overall,parental lines Zhong901,ZB,L28,and Z98 were observed with maximum combining ability while combinations with strong special combining ability were ZB×DT,L28×Z98,and ZB×851.The yield traits heterosis was predominant in both generations.However,the level of heterosis was altered with trait,hybrid combination,generation,and environment.Interestingly,L28×Z98 performed outstandingly in Anyang.Its lint yield(LY)was 24.2%higher in F_(1) and 11.6%in F_(2) than that of the control Ruiza 816.The performance of SJ48×Z98 was excellent in Aral which showed 36.5%higher LY in F_(2)and 10.9%in F_(2)than control CCRI 49.Further results revealed most hybrid combinations had shown a low level of heterosis for agronomic and fiber quality traits in both generations.Comparatively,ZB×DT and L28×Z98 showed hybrid vigor for multiple traits in both generations and cotton belts.It is feasible to screen strong heterosis hybrid combinations with fine fiber in early generations.In the two environments,the correlation of some traits showed the same trend,and the correlation degree of Anyang site was higher than that of Aral site,and the correlation of some traits showed the opposite trend.According to the performance of strong heterosis hybrid combinations in different environments,the plant type,yield and fiber traits associated with them can be improved according to the correlation.Conclusions:Through comparative analysis of variance,combining ability,and heterosis in F_(2)and F_(2)hybrids in different cotton belts,this study proposed the potential utilization of F_(2)hybrids to improve upland cotton productivity in China.CHEN Liangliang TANG Huini ZHANG Xuexian QI Tingxiang GUO Liping SHAHZAD Kashif WANG Hailin QIAO Xiuqin ZANG Rong ZHANG Meng WU Jianyong XING Chaozhu 2022Journal of Cotton Research2022,5,2:2
19Mechanism of variations in environmental magnetic proxies of lake sediments from Nam Co, Tibet during the Holocene显示文摘High-resolution environmental records from the Tibetan Plateau are essential to understand past global climatic and environmental changes. Magnetic minerals in lake sediments are important proxies to reconstruct environmental and climatic changes. Nam Co (lake) is a typical great lake in the transitional region of southwest monsoon in the Tibetan Plateau. Previous studies have extensively focused on geochemistry, microfossils, sedimentology and biochemistry analysis of Nam Co, which provides sound interpretation of paleoclimatic and paleoenvironmental changes. However, up to now, no systematic environmental magnetic studies have been carried out. Therefore, high-resolution and systematic magnetic studies combined with geochemical parameters were carried on lake sediments of core NC 08/01 from Nam Co for the Holocene period (11.3 cal ka BP) in order to explore how magnetic properties of the sediments respond to climatic changes. Based on variations of magnetic proxies, the sequence can be separated into 3 units. Unit 1 (236-199 cm, 11.3-7.8 cal ka BP) contains dominantly coarse-grained magnetite with homogeneous grain size. A positive correlation between magnetite and Ti strongly suggests that these coarse-grained detrital magnetites reflect detrital input signals due to insignificant effects of postdepositional dissolution processes on these coarse-grained magnetite particles. For Unit 2 (198-102 cm, 7.8-2.1 cal ka BP), magnetic grain size is finer and the corresponding concentration of magnetite is also reduced. This is mainly due to significant dissolution of these fine-grained detrital magnetite particles, which were transported under reduced water flow conditions during this period. For Unit 3 (101-0 cm, 2.1-0 cal ka BP), the bulk magnetic properties are dominated by a mixture of single domain biogenic magnetite and detrital magnetite. The concentration of magnetic minerals is not correlated with the Ti content. In conclusion, the preservation of magnetic minerals in the lake sediment and thus the corresponding magnetic properties are related to the initial grain size. Combination of magnetic properties (including variation of grain size and concentration) and other proxies of detrital inputs (e.g. Ti) can be used to infer the variation of redox conditions in Nam Co.
